Notizen: For the purpose of investigating the interdiffusion properties between thin Y–Ba–Cu–O films of compositions near YBa2Cu3O7−y and MgO substrates, time evolution of both Rutherford backscattering spectra and scanning electron microscope images during annealing were observed. Actual diffusivities of Y, Ba, and Cu elements in the MgO substrates have been found to be extremely smaller than the values previously reported. The origins of the discrepancies between the present and the previously reported results are discussed in connection with the morphology variation of the thin Y–Ba–Cu–O films on the MgO substrates during heat treatments.

Notizen: Interdiffusion between sputter-deposited thin Y-Ba-Cu-O films and Al2O3 substrates has been studied by applying 3.2 MeV 4He+ Rutherford backscattering spectrometry. The effects of morphology variation of the films during annealing have been successfully excluded using the data of thickness distribution measured by applying atomic force microscopy. The formalism used for the analysis is presented. The obtained values of the diffusivities are more than 20 times smaller than those reported by other workers.

Notizen: To characterize the genes for enzymes involved in the biosynthesis of polyamines (PAs), their expression profiles were investigated and the levels of PAs in Arabidopsis thaliana quantified. In the Arabidopsis genome, eight genes involved in PAs biosynthesis were identified and the expression profiles of these genes were analysed, not only under abiotic stress to determine whether they were stress-inducible, constitutive, or stress-repressible, but also in various organs to show their tissue specificity. AtADC2 and AtSPMS mRNAs, encoding arginine decarboxylase and spermine synthase, clearly increased in response to NaCl and dehydration and abscisic acid treatments. Stress-inducible accumulation of AtADC2 mRNA correlated with putrescine (Put) accumulation under NaCl and dehydration treatments. In a cold condition, AtSAMDC2 mRNA increased significantly. AtADC2 and AtSAMDC2 mRNA were expressed in sexual organs such as flowers, buds and immature siliques. PAs also accumulated in sexual organs. These results suggest that the transcripts of eight genes involved in PA biosynthesis show different profiles of expression not only in response to environmental stress but also during plant development.

Notizen: Background The mtist cell is one of the important cells In the pathogenesis of allergic disorders. However, isolating human mast cells is a laborious procedure. Recently, cultured human mast cells raised from umbilical cord blood cells have become available. It is necessary to examine whether these cells are useful in investigating the role of mast cells in human diseases. Objective The phenotype of mast cells depends on their anatomical sites. To examine which phetiotype of mast cells these cultured mast cells most closely resemble, their ability to release was investigated. Methods The mast cells were raised from human umbilical cord blood cells in the presence of stem cell factor and interIeukin-6. To determine the mast cell subtypes, the mast cells were immunocytochemically stained for tryptase and chymase. The cultured mast cells were then stimulated with various secretagogues, and histamine release was measured by a fluorometric technique using high-performance liquid chromatography. Results The immunocytochemical staining for mast cell proteases revealed that virtually all cells contained tryptase, the definitive marker of mast cells, and that about a quarter of the cells contained chymase. Anti-TgE effectively stimulated these mast cells to release histamine in a dose-dependent, lime-dependent manner. The release was completed in about 30 min. One of the non-specific stimuli, calcium ionophore A23I87. also induced histamine release in a dose-dependent, time-dependent manner. In contrast, compound 48/80 and substance P failed to induce histamine release from these cells. Conclusion Cultured human mast cells resemble lung mast cells in their ability to release histamine. They will help in studying the functional properties of human mast cells and may contribute to clarifying the pathophysiology of human allergic diseases.

Notizen: summary  Occlusal conditions at the retruded contact position (RCP) have been thought to be among the aetiological factors for temporomandibular disorders. However, the role of these occlusal factors still remain unclear. The purpose of a series of studies was to investigate the locations of RCP contacts and to discuss their implication by the authors. In the study reported here, the influence of the operator's skill on bite registration was surveyed. Eighty-six dental students were selected as subjects. One specialist in prosthetic dentistry moved the subject's mandible up and down while guiding it gently backward until it rotated on its retruded axis. Once the initial tooth contacts were established, the operator checked them by using a thin articulating paper (direct method). On the other hand, six younger operators following precise instruction from the specialist about the technique of jaw guidance examined each of the subjects for location of RCP contacts using the indirect method. Silicone-based impression material was used for bite registration while the subject kept the jaw at this contact position (indirect method). Wilcoxon signed rank test was applied to evaluate differences between the expert operator and six younger operators regarding locations of RCP contacts. Results from the expert operator (direct method) showed that tooth contact at the first premolar was most frequently observed and the number of subjects who possessed unilateral tooth contacts was more than that with bilateral tooth contacts. On the other hand, in the case of the indirect method performed by younger operators, tooth contact patterns significantly differed from those of the direct method.

Notizen: Three types of retainers (wrought wire clasp, Aker's cast clasp, and conical crown telescopic retainer) designed for distal-extension removable partial dentures (RPDs) were assessed in two Kennedy class I patients’ mouths. The assessment, included the ratio of denture base shearing load and mobility of the terminal abutments when loaded on a free-end RPD occlusal surface. The mean values of denture base shearing ratios of wrought wire clasp, Aker's clasp and conical crown telescope were 60, 42 and 20%, respectively. The abutment mobility of the three types of retainers were all within the ‘mobile ability area’ except the wrought wire clasp for patient A's right side. The greatest tooth mobility was observed with the wrought wire clasps, followed by Aker's clasp and the conical crown telescopic retainer. From the analysis the following was concluded: (i) different retainers do influence the occlusal load distribution; (ii) the occlusal load distributed to the free-end saddle is closely related to the connecting rigidity of the retainer; (iii) mucosal support has an indispensable role in sharing the occlusal load with various retainers, even the rigid telescopic retainer.

However, the role of these occlusal factors remains still unclear. The purpose of this series of studies was to investigate the locations of RCP contacts and to discuss their meaning. In this part of the study, two different methods for bite registration at the RCP have been compared. Twenty dentists were selected as subjects for this study. Statistical difference was not shown between the two registration methods regarding locations of RCP contacts.
