    Notizen: The hypothalamic suprachiasmatic nucleus, the primary circadian pacemaker in mammals, and the retinohypothalamic tract, the retinal afferent fibres to the suprachiasmatic nucleus, both mature during early postnatal life. The establishment of circadian rhythms is thought to depend on input from the retina, but the mechanism remains unknown. Here we examined developmental changes in the expression of the Ca2+-binding proteins calbindin-D28k and calretinin in the mouse hypothalamus. Robust calbindin-D28k immunoreactivity was observed in the dorsomedial suprachiasmatic nucleus and the supraoptic nucleus in neonatal mice (postnatal day 3). The calbindin-D28k immunoreactivity decreased significantly in the suprachiasmatic nucleus but not in the supraoptic nucleus during postnatal days 9–15, when retinohypothalamic tract projections to the suprachiasmatic nucleus are completed. Calretinin immunoreactivity was low in the neonatal suprachiasmatic nucleus and increased with development in the ventrolateral suprachiasmatic nucleus, in parallel with the developmental reduction of calbindin-D28k immunoreactivity observed in the dorsomedial suprachiasmatic nucleus. Developmentally stable calretinin immunoreactivity was also observed in retinohypothalamic tract fibres. Organotypic slice cultures of the suprachiasmatic nucleus were prepared from postnatal day 3 mice to examine the effect of the absence of retinohypothalamic tract inputs on developmental changes in calbindin-D28k and calretinin expression. After 12 days in vitro, the cultured suprachiasmatic nucleus slices exhibited dense calbindin-D28k immunoreactivity similar to neonatal mice, and calretinin immunoreactivity in the ventrolateral suprachiasmatic nucleus similar to young adult mice. These results demonstrate a developmental reduction in calbindin-D28k expression that paralleled retinohypothalamic tract formation and a developmental increase in calretinin expression that is independent of retinohypothalamic tract connections to suprachiasmatic nucleus neurons.

    Notizen: We examined synaptic plasticity at intrasuprachiasmatic nucleus (SCN) γ-aminobutyric acid (GABA)ergic synapses by measuring the paired-pulse ratio between pairs of evoked inhibitory postsynaptic currents (IPSCs). Interstimulus intervals were chosen to represent the range of spontaneous action potential firing frequencies found in SCN neurons. A majority of synapses studied during the day exhibited paired-pulse depression (PPD), whereas a majority of synapses studied during the night showed no PPD. Two types of PPD were found. Type 1 PPD expresses the greatest inhibition at shorter interstimulus intervals, is predominant in the early morning and is likely to be a result of vesicle depletion. Type 2 showed the greatest inhibition at interstimulus intervals between 175 and 225 ms, is found throughout the day yet rarely at night and is likely to be a result of a Ca2+-dependent mechanism that is independent of pertussis toxin-sensitive G-proteins. Thus, multiple mechanisms of synaptic plasticity modulate intra-SCN communication throughout the diurnal cycle.

    Notizen: This study was designed to test whether the α-amino-3-hydroxy-5-methylisoxazole-4-propionate (AMPA) receptor-facilitating drug, aniracetam, could potentiate photic responses of the biological clock in the suprachiasmatic nucleus (SCN) of rodents. Using the whole-cell patch technique, we first demonstrated that AMPA currents elicited by either local AMPA application or optic chiasm stimulation were augmented by aniracetam in the neurons of the SCN. The AMPA application-elicited increase of intracellular Ca2+ concentration in SCN slices was also enhanced by aniracetam treatment. The systemic injection of aniracetam dose-dependently (10–100 mg/kg) potentiated the phase delay in behavioral rhythm induced by brief light exposure of low intensity (3 lux) but not high intensity (10 or 60 lux) during early subjective night. Under the blockade of NMDA receptors by (+) MK801, aniracetam failed to potentiate a light (3 lux)-induced phase delay in behavioral rhythm. Aniracetam increased the photic induction of c-Fos protein in the SCN that was elicited by low intensity light exposure (3 lux). These results suggest that AMPA receptor-mediated responses facilitated by aniracetam can explain enhanced photic responses of the biological clock in the SCN of rodents.

    Notizen: Abstract The coastal prairie ecoregion is located along the northwestern coastal plain of the Gulf of Mexico in North America. Because of agricultural and urban development, less than 1% of the original 3.4 million ha of this ecosystem type remains in native condition, making it one of the most endangered ecosystems in North America. The objective of this study was to characterize the vegetation and environmental relationships in a relatively pristine example of lowland coastal prairie in order to provide information for use in conservation and restoration. The study area was a small, isolated prairie located near the southern boundary of the coastal prairie region. Samples were taken along three parallel transects that spanned the prairie. Parameters measured included species composition, elevation, soil characteristics, indications of recent disturbance, above-ground biomass, and light penetration through the plant canopy. Fifty-four species were found in the 107 0.25-m2 plots and a total of 96 species were found at the site. Only two non-native species occurred in sample plots, both of which were uncommon. Cluster analysis was used to identify six vegetation groups, which were primarily dominated by members of the Poaceae or Asteraceae. A conspicuous, natural edaphic feature of the prairie was the presence of `mima' mounds, which are raised areas approximately 0.5 to 1 m high and 5 to 10 m across. Indicator species analysis revealed a significant number of species that were largely restricted to mounds and these were predominately upland and colonizing species. Ordination was performed using nonmetric, multidimensional scaling. The dominant environmental influence on species composition was found to be elevation and a host of correlated factors including those associated with soil organic content. A secondary group of factors, consisting primarily of soil cations, was found to explain additional variance among plots. Overall, this prairie was found to contain plant associations that are now rare in the surrounding landscape. Within the prairie, plant groups were largely separated by a suite of environmental conditions associated with topography. These results suggest that conservation and restoration efforts will need to carefully consider local topographic influences in order to be successful.
