    Notes: Cryostat-cut sections of unamputated and amputated-regenerating limbs of the adult newt were examined following the Nile blue test for acidic and non-acidic lipids, the acid hematein and plasmal tests for phospholipids, and a Schultz test for cholesterol.Triglycerides (Nile blue test) are prominent in dermis and macrophages: triglyceride droplets are scattered in epidermis, wound epithelium, and regeneration blastema. Fatty acids (Nile blue test) are present in all tissues of the normal and regenerating limb: nerve myelin contains relatively little free fatty acid, while macrophages appear to contain the least amount of this lipidic substance.Plasmalogens (plasmal test) are prominent in the myelin of nerves, and macrophages: a weak cytoplasmic reaction obtains in the epidermis, subcutaneous glands, striated muscle, tunics of blood vessels, wound epithelium, blastema cells, chondrocytes, perichondrium and periosteum.Mitochondria responding for cephalin, lecithin, and sphingomyelin (acid hematein test) are ubiquitously distributed among the cells and tissues of the normal and regenerating limb. These phosphatides are prominent in nerve myelin, macrophages, and in dermal droplets: a variable response obtains from the myofibrils of striated muscle.Cholesterol (Schultz test) was demonstrated only in nerve myelin and in macrophages associated with injured nerves.Particular attention was paid to the lipid responses of the regeneration blastema, and the conclusion was reached that not all of the lipid previously demonstrated with sudan dyes was characterized by the current series of lipid tests.A modified Nile blue sulfate test that promises greater specificity in distinguishing between acidic and non-acidic lipids is introduced.

    Notes: We have examined the fine structure of injured, repairing and regenerating tissues of the amputated forelimbs of the adult newt: observations were made on wound epithelium, nerves, striated muscle, the regeneration blastema, connective tissues and cartilage.The wound epithelium shows little modulation from the adjacent epidermis: a subjacent glycocalyx and hemidesmosomes are initially absent. Some cells reveal phagocytic activity. As regeneration progresses, a glycocalyx, hemidesmosomes, and an adepidermal reticulum of fibers develop.Nerve fiber degeneration and regeneration lie side by side in the same nerve bundle. Myelin degeneration is evident within 24 hours; neural connective tissues loosen and fibers appear to spread apart.Striated muscle fibers display a nonuniform response to trauma, due to unequal degree of injury inflicted by amputation. The moderately and greatly injured fibers lose their distal fibril organization. There is a numerical increase in mitochondria, and occasionally a pyknotic nucleus is present. The myofiber glycocalyx appears to resist destruction, even when cell degeneration is extensive. Satellite cells have been observed intimately in contact with injured myofibers. Myogenesis is accompanied by an increase in collagen fibrillogenesis associated with adjacent fibroblasts. Many of these latter cells appear to contain intracellular fibrils: the significance of these observations are discussed, and a basis for intracellular fibrillogenesis and tropocollagen accretion is proposed.The fine structure of the regeneration blastema cell agrees with the description offered by others for the adult newt limb regeneration.Early prochondral condensation of blastema cells is described briefly.

    Notes: The development of the cranial and branchial skeleton of the surfperch Amphistichus argenteus, a member of the family Embiotocidae, is described, and phylogenetic and functional aspects of the skull development of this species are discussed. The earliest bones to appear are those dermal elements of the branchial skeleton involved with feeding, and the bones, both dermal and endochondral, located in the basicranial region of the neurocranium. These are followed by dermal bones associated with the lateral line system and finally by the remainder of the bones of the branchial skeleton and the cartilaginous bones of the otic capsules. The last bone to develop is the ethmoid.

    Notes: Spermatozoa from the caput epididymis are known to be much less capable of fertilization when compared to sperm from more distal segments of the epididymis. The purpose of this study was to determine if two micromanipulative techniques, zona drilling (ZD) and a modification of partial zona dissection (PZD), could be used to enhance fertilization with caput epididymal sperm. A mouse in vitro fertilization model was used. Inseminating oocytes with 500-1,000 sperm/oocyte from the cauda epididymis as a control resulted in fertilization of 98 of 300 (32.6%) oocytes. Of those fertilized, 47 developed to the blastocyst stage (47.9%). Caput sperm fertilized 13 of 116 (11.2%) nonmanipulated oocytes. Only 1 of 13 developed into a blastocyst, while with oocyte ZD, caput sperm fertilized 24 of 144 (16.7%) oocytes, 50% of those fertilized developing to blastocyst (P=0.0129). When modified PZD was performed on oocytes, only one of 23 was fertilized, with no blastocyst development. These results indicate that acid Tyrode ZD enhances both fertilization and early embryonal development when caput epididymal sperm are used for insemination. These mouse studies suggest that ZD or other micromanipulation techniques may prove clinically useful in men with proximal epididymal obstruction where only caput sperm are available.

    Notes: Electron microscopic studies on magnetosomes in magnetotactic bacteria have revealed much information on their composition, structure, and even the formation of their mineral phase. The mineral phases of the magnetosomes are of two general types: iron oxides and iron sulfides. Iron oxide-type magnetosomes contain particles of the ferrimagnetic mineral magnetite (Fe3O4) while the iron sulfide-type contain ferrimagnetic greigite (Fe3S4), greigite and non-magnetic pyrite (FeS2), or possibly ferrimagnetic pyrrhotite (Fe7S8). Regardless of their composition, the crystalline particles in magnetosomes have a narrow size range: approximately 35 to 120 nm. Magnetite crystals in this size range are single-magnetic-domains and confer a permanent magnetic dipole moment to the cell. The single-domain size range for greigite is not known but is probably similar to that for magnetite.The morphology of the particles in the bacterial magnetosomes appears to be species-specific. Morphologies of magnetite crystals in different species of magnetotactic bacteria include cubooctahedra, parallelepipedal (truncated hexahedral or octahedral prisms), and tooth- or bullet-shaped (anisotropic). Morphologies of greigite particles include cubo-octahedra and rectangular prismatic. The greigite-pyrite particles are generally pleomorphic with no consistent crystalline morphology. A membrane has been shown to surround the particles in some organisms and may be involved in the formation of the crystalline phase while also providing physical constraints on the size and the shape of the crystal. These results clearly indicate that the biomineralization process involved in the bacterial magnetosome, a good example of a self-assembled structure on a nanometer scale, is highly controlled by the organism. © 1994 Wiley-Liss, Inc.

    Notes: Many previous reports have demonstrated the development of aberrant neural connections in response to neonatal brain lesions. This investigation was undertaken to study possible alterations, particularly axonal sprouting, in rodent rubrospinal projections after neonatal destruction of the corticospinal tract through frontal cortical ablation. The neonatal ablations were made by aspiration in 1 to 2-day-old rats under hypothermic anesthesia. At three to six months after neonatal surgery, the rubrospinal tracts were ablated bilaterally in these same animals as well as in controls, by stereotaxically transecting the ventral tegmental decussation. Animals were killed two to six days after adult surgery, and rubrospinal projections were demonstrated using the Fink-Heimer degeneration stain. No differences in the pattern of rubrospinal projections were observed between animals with neonatal cortical lesions and controls. In all animals rubrospinal projections were located primarily in Rexed's lamina VI with a slight distribution into lamina V and the dorsal portion of lamina VII. Various hypotheses explaining the lack of rubrospinal sprouting after neonatal cortical lesions are presented, along with possible experiments to test these hypotheses.

    Notes: Retrograde transport of horseradish peroxidase (HRP) was used to define the origin of afferents to the inferior olivary complex (IOC) in rats. Using both ventral and dorsal surgical approaches to the brainstem, HRP was injected into the IOC through a micropipette affixed to the tip of a 1-μl Hamilton syringe. After a 2-day postoperative survival, animals were sacrificed by transcardiac perfusion with a 1% paraformaldehyde-1.25% gluteraldehyde solution, and brains were processed according to the DeOlmos protocol (1977), using o-dianisidine as the chromogen.Labeled cells were found at many levels of the nervous system extending from lumbar spinal cord to cerebral cortex. This wide-ranging input from numerous regions clearly underscores the complexity of the IOC and its apparent involvement in several functions.Within the spinal cord, labeled neurons were identified from cervical to lumbar but not at sacral levels. These neurons were found contralaterally in the neck region of the dorsal horn and in the medial portions of the intermediate gray.In the caudal brainstem, reactive cells in the dorsal column nuclei, the spinal trigeminal nucleus, and the subnucleus y of the vestibular complex were observed primarily contralateral to the injection sites. Labeling within the gigantocellular, magnocellular, ventral, and lateral reticular nuclei and the nucleus prepositus hypoglossi was primarily ipsilateral. Reactive neurons in the medial and inferior vestibular nuclei were predominantly ipsilateral or contralateral to HRP injections into the caudal or rostral IOC, respectively.The dentate and interposed nuclei of the cerebellum contained small, lightly labeled neurons primarily contralateral to the injection site, while the fastigial nuclei contained a few relatively large, heavily labeled cells bilateral to caudal olivary injections.Ipsilaterally labeled mesencephalic regions included the periaqueductal gray, interstitial nucleus of Cajal, rostromedial red nucleus, ventral tegmental area, medial terminal nucleus of the accessory optic tract, nucleus of the optic tract, and the lateral deep mesencephalic nucleus. The caudal part of the pretectum and small cells of the stratum profundum of the superior colliculus were labeled predominantly contralateral to the injection.In the caudal diencephalon labeled neurons were most numerous within the nucleus of Darkschewitsch and the subparafascicular nucleus, primarily ipsilateral to olivary injections. Scattered reactive neurons were also found within the ipsilateral zone incerta. With the exception of the zona incerta, all labeled mesencephalic and diencephalic nuclei had some bilateral representation of labeled cells.No labeled neurons were identified within the basal ganglia, while numerous reactive cells were found bilaterally within layer V of the frontal and parietal cerebral cortex.

    Notes: The development of intestinal mucosal capillaries was studied in Wistar rats from the fourteenth day of gestation through the second postnatal day with the transmission electron microscope. The microvessels develop first as continuous capillaries without a basal lamina and become fenestrated during later stages of gestation. In the thickened areas of the cytoplasm of endothelial cells at early stages, there is a paucity of pinocytotic vesicles; but some vesicles are observed which are larger than those in the adult. Later in gestation these larger vesicles can still be seen, but the frequency of their occurrence decreases with the maturation of the vessels. The intercellular junctions vary in length and shape, and the space between the outer leaflets of the apposed cell membranes is usually 20 nm. Surrounding the abluminal surface of the endothelium, the incipient basal lamina appears attenuated and incomplete until at least the second day of neonatal life.

    Notes: The function of the mural components involved in vascular exchange (pinocytotic vesicles, fenestrations, intercellular junctions, and the basal lamina) was assessed by intravascular injection of tracers (carbon, carbon plus histamine, ferritin, hemoglobin, and myoglobin) on separate gestational days in fetal and neonatal rat intestinal capillaries. The continuous capillaries present through day 17 of gestation were impermeable, even at junctions, to all the tracers within the maximum circulation time studied (3 minutes). Most of the luminal pinocytotic vesicles were labeled within 3 minutes with myoglobin (3.3-nm diameter) or hemoglobin (5.5-nm diameter), while only occasional vesicles contained ferritin (11-nm diameter) and none contained carbon (25-50nm diameter). Even though luminal pinocytotic vesicles contained the smaller tracers, no vesicular transport and discharge were ever seen. The fenestrated vessels present from day 18 of gestation allowed a rapid extravasation of ferritin and hemoglobin via diaphragmed fenestrations. At no time period did extravasation of carbon take place, nor did the carbon plus histamine solution induce vascular leakage. Once a tracer gained access to the extravascular space, the developing basal lamina did not seem to retard its movement.

    Notes: The structure and function of colonic mucosal lymphoid organs remain largely unexplored, especially in the rectum hidden within the pelvic vault. Two-month-old female BALB/c mice were anesthetized, and the entire colon was removed from cecum to anus. Distal colonic patches were then prepared for electron microscopy or were quick-frozen and sectioned for immunoperoxidase localization of B cells and T cell subsets. Aggregated lymphoid follicles were distributed irregularly along the entire colon with an average of 1.4 patches per centimeter of colon length. There were large collections of follicles opposite the ileocecal valve (cecal patches), variable numbers of patches throughout the colon, and at least one patch within 10 mm of the anus (rectal patch). Follicles were adjacent to branching crypts lined by epithelium infiltrated by lymphoid cells and containing few goblet cells. In electron micrographs, M cells were identified by their short, irregular microvilli; intraepithelial lymphoid cells; reduced lysosomal dense bodies; and an expanded tubulovesicular network. Small germinal centers were seen. Cytoarchitectural components of colonic lymphoid follicles and Peyer's patch follicles were remarkably similar, despite differences in surrounding mucosa and luminal microbial exposure. The presence of organized lymphoid tissue with M cells and germinal centers suggests that transepithelial particle transport and antigen recognition can take place in the rectum. Whether such tissue has the capacity for uptake of luminal microorganisms is of particular interest, not only because colonic follicles may be sites for local initiation of immune responses but also because they may be important entry points for systemic infection.
