    Notes: Abstract Lectins conjugated to horseradish peroxidase in combination with selected exoglycosidase digestion procedures were used to localize fucoglycoconjugates in the bovine submandibular gland. In particular, sequential treatments were employed to determine the distribution of neutral and acidic fucose-containing oligosaccharides that were previously shown to be present by biochemical techniques. Information was obtained on the distribution of the acidic oligosaccharide A-1a, α-Fuc(1→2)-β-Gal-(1→4)-β-GlcNAc-(1→3)-[α-NeuAc-(2→6)]-GalNAc-ol, which was sequenced in situ and localized in acinar cells.

    Notes: Summary Previous findings have demonstrated the presence of muramic acid and the lack of sialic acid in gastropod glycoconjugates from different tissues. The present study investigated the composition of muramyl derivatives in Mollusca Gastropoda tissue from the foot, mantle and periesophageal ganglia, using HRP-labeled lectins (LTA, UEA I, GSA IB4, GSA II, DBA, SBA, RCA II, WGA, PNA, ConA) and glycosidase digestion (neuraminidase, lysozyme, α-l-fucosidase, β-N-acetylglucosaminidase, α-N-acetylgalactosaminidase). Muramyl derivatives from the tissue examined showed some differences related to the composition of the terminal disaccharides. Indeed, foot and mantle mucocytes exhibited muramic acid in a terminal position, linked to (subterminal) N-acetylgalactosamine, whereas in neuron cells muramic acid was present in an internal position and linked to N-acetylglucosamine. Diversities also occurred between foot and mantle mucocytes with respect to the receptor sugar for penultimate N-acetylgalactosamine.

    Notes: Summary We employed sialidase procedures followed by lectin stainings combined with oxidizing and deacetylating agents to visualize the distribution and sequentiate sialoglycoconjugates in the bovine submandibular gland. In particular we evidenced in acinar and ductal cells the dishomogeneous presence of sialic acids acetylated in the polyhydroxy side chain (C7, C8, C9), whereas O-acetyl substituents at position C1 and/or C4 were not found. Sialoglycoderivatives were also differentiated by the occurrence of penultimate sugars; indeed the dimers sialic acid-(α2→3,6)-β-galactose and sialic acid-(α2→6)-α-N-acetylgalactosamine were identified. Using such technique we supported further the possibility to develop methods for the identification of the positions of Oacetyl groups and the reconstruction of terminal disaccharides within surface and cytoplasm glycoconjugates.

    Notes: Summary The authors describe a case report of a patient suffering from thrombosis and recurrent abortions. Routine coagulation screening was abnormal and investigation showed the presence of a circulating anticoagulant. High titres of anticardiolipin antibodies were demonstrated. Antinuclear antibodies were slightly positive and antinative DNA antibodies were present. Lupus anticoagulant activity and anticardiolipin antibodies seem to be strong markers of a thrombotic tendency.

    Notes: Summary Cell surface and cytoplasmic glycoconjugates were characterized in embryonic human explant cells (a transformed heteroploid line) cultured in iso-osmotic medium (0.137m NaCl) and in hyperosmotic medium (0.274m NaCl) for 10 days in order to study the changes induced in these compounds by hyperosmoticity. Cytochemical and ultracytochemical staining selective for glycoconjugates was carried out. The following results were obtained: (1) glycoproteins, glycosaminoglycans and glycolipids are present on the cell surface and in the cytoplasm of the explant cells; (2) lectin histochemistry combined with glycosidase digestion demonstrated the presence of the disaccharides fucose-N-acetylglucosamine and sialic acid-β-galactose as terminal sequences; (3) histophotometric evaluation of lectin labelling showed a noticeable decrease in histochemical reactivity of adapted cells; (4) plasma membrane cell coat decreased in adapted cells, which was emphasized by ultracytochemical reactions and a rearrangement of glycolipids in the cytoplasm.
