    Notes: A method was proposed to prepare a peptide-type low-phenylalanine, high-tyrosine food for curing phenylketonuria. A fish protein concentrate (FPC) and a soybean protein isolate (SPI) were used as starting materials. Each was limitedly hydrolyzed with a very small amount of pepsin The peptic hydrolysate was further hydrolyzed with Pronase under an unconventional pH-condition to liberate aromatic amino acids. These were removed by Sephadex G-15 with the aid of its adsorp tion activity. To the resulting aromatic amino acid-free fraction were added ethyl esters of L-tyrosine and L-tryptophan and the mixture was incubated with papain under such conditions as its reverse process called “plastein synthesis reaction” proceeded efficiently. The reaction product was treated by ultrafiltration to obtain a plastein as a fraction having the lowest molecular weight of 500. The yields were 69.3% from FPC and 60.9% from SPI. Phenylalanine, tyrosine and tryptophan contents were 0.05%, 7.82% and 2.98% in the FPC plastein and 0.23%, 7.69% and 2.80% in the SPI plastein. Each plastem did not contain any free amino acids and was almost completely flat in taste and odor.

    Notes: SUMMARY— A proteolyzate obtained by treating an isolated soybean protein preparation with Molsin, a crude preparation of aspergillopeptidase A (APase A), was less bitter and contained larger amounts of free amino acids, especially hydrophobic amino acids. A proteolyzate obtained by treating this protein preparation with crystallized APase A was much more bitter and contained smailer amounts of free amino acids, mainly consisting of hydrophilic amino acids. The latter was richer in peptides than the former, bearing hydrophobic amino acid residues near the C-termini. Difference in N-terminal amino acid composition apparently has not been found between the 2 proteolyzates. These results indicate that Molsin per se contains a certain carboxypeptidase which decomposes the C-terminal structures and, consequently, lessens the bitterness (debittering effect). This carboxypeptidase was found to be identical with aspergillus acid carboxypeptidase (AACPase). Abase A, as well as MO/sin, was effective in removing odor ants, i.e., n-hexanal, n-hexanol and n-heptanol, from the isolated soybean protein preparation Ideodorization effect). AACPase seemed to have no deodorization effect. A method was suggested to prepare a deodorized and debittered proteolyzate by a combination use of APase A and AACPase.

    Notes: SUMMARY– The formation of N-substituted pyrrole-2-aldehydes in the browning reaction between D-xylose and selected amino acids was investigated.In order to facilitate the separation of the reaction products, the carbonyl compounds formed in the browning solution were converted into 2,4-dinitrophenylhydrazones (2,4-DNPs) and then esterified with diazomethane. Isolation of the esterified 2,4-DNP was accomplished by column chromatography with alumina. Chemical structures of the isolated 2,4-DNPs were confirmed by elementary analyses, ultraviolet and visible absorption spectra, and nuclear magnetic resonance spectra.The following N-substituted pyrrole-2-aldehydes were isolated as methyl ester of 2,4-DNP, respectively: (2-formylpyrrol-I-ybacetic acid from the reaction mixture of D-xylose and glycine, 3-(2-formylpyrrol-1-y1)propionic acid from β-alanine, and 2-(2-formylpyrrol-1-y1)-4-methyl-valeric acid from L-leucine.The extent of pyrrolealdehyde formation from D-xylose and selected alkylamine or amino acid was in the following order which corresponded to the extent of melanoidin formation: n-butylamine 〉 methylamine, β-alanine 〉 glycine 〉 DL-alanine. N-substituted pyrrole-2-aldehyde was rather stable and was not considered to be an important intermediate of melanoidins. However, some correlationships between the formation of pyrrolealdehyde and that of melanoidins were demonstrated and discussed.

    Notes: Effects of gamma irradiation on the chemical properties of actin and actomyosin during aging of meats have been studied. Meats were irradiated with 4 × 106 rad of cobalt-60 gamma rays at 40° F in three steps, i.e., immediately after slaughter, at maximum rigor, and at “rigor off.” Actin and two kinds of actomyosin (AMS and AML, previously reported by authors) were isolated from irradiated and control meats. The contents of sulfhydryl groups and amino acids, viscosity, the activity of ATPase and ATP sensitivity of actin and actomyosin were determined. From the results, actin is thought to be only slightly sensitive to irradiation as compared with actomyosin, and the latter, especially AML (a long-time extracted actomyosin) is considered to be very sensitive. In this case, it is inferred that the depolymerization must have taken place in actomyosin molecule by irradiation on meats. The effects of the time of irradiation on the chemical properties of actomyosin in meats seem to be more remarkable at rigor off than at the other two steps.

    Notes: Chemical and organoleptic tests showed that partial digestion of soybean curd and defatted soybean flour by aspergillopeptidase A preparation removed flavor compounds and related fatty materials from these soybean products. Amounts of ether-soluble compounds, total carbonyl compounds and volatile reducing substances released during incubation of soybean curd with the enzyme were higher than those released during incubation without the enzyme. Amounts of volatile flavor compounds, including n-hexanal and n-hexanol, released during the incubation of defatted soybean flour with the enzyme were also higher than those released during incubation without the enzyme. Related compounds, i.e., lipids, pigments, saponins, etc., also were released during the enzymation. The enzymic digestion product from either soybean curd or defatted soybean flour had less odor, taste and color. Stability of the enzymic digestion product from the defatted soybean flour against autoxidation during the preservation, measured by TBA-reaction and organoleptic test, was superior to that of the product treated similarly without the enzyme.

    Notes: Isolation and characterization of 7 sorts of diffusable bitter peptides in peptic hydrolyzate of soybean protein were presented. One liter of dilute HCI solution containing 25 g of soybean cold-insoluble protein and 250 mg of pepsin was incubated at 37°C for 24 hr. The resulting peptic hydrolyzate was dialyzed against water and the diffusable fraction was, after being concentrated at 40°C, investigated by gel permeation chromatography using Sephadex G-10, ionexchange chromatography of Dowex 50 (w)-X2 (pyridinium) and thin-layer chromatography with silica gel G. Bitterness of the diffusable fraction was attributable to free amino acids having bitter flavor, such as isoleucine, leucine, phenylalanine and valine; also to peptides such as H. Gly-Leu. OH, H. Leu-Phe. OH, H. Ser-Lys-Gly-Leu . OH, H-Leu-Lys . OH, H. Phe-(lle, Leu2)- Gin-Gly-Val . OH, H. Arg-Leu-Leu . OH and H. Arg-Leu . OH. It was characteristic that almost all diffusable bitter peptides investigated here had leucine at the termini. Through carboxypeptidase A treatment on these peptides, the bitterness decreased considerably. Furthermore, applying carboxypeptidase A or Nagane, a bacterial neutral proteinase, to the diffusable fraction of the peptic hydrolyzate was also effective in removing the bitterness somewhat.

    Notes: To elucidate the changes of “myosin B” during storage of rabbit muscle, investigation was made of the dissociation of “myosin B” into myosin A and actin, their content of “myosin B,” and the interaction of “myosin B” with ATP. The approximate contents of myosin A and actin in “myosin B” were variable, showing a maximum content of actin in “myosin B” extracted from the muscle at 2 days after slaughter. This can also be inferred from changes of viscosity and the salting-out curve of “myosin B,” as recently shown. Moreover, the interaction between actin and myosin A in “myosin B” became less strong with the progress of aging.

    Notes: Abstract We herein report on a 64-year-old Japanese female patient who presented with a splenic artery aneurysm (SAA) associated with systemic lupus erythematosus (SLE). The saccular aneurysm, which measured 3 cm in diameter, was located in the proximal third of the splenic artery from the pancreas with a portosystemic shunt. A double ligation of the splenic artery (the distal and proximal sides of the aneurysm) was performed without a splenectomy. The postoperative course showed acute pancreatitis without either splenic infarction or portal thrombus. To our knowledge, the closed association of SLE with an aneurysmal dilatation of the splenic artery has not been previously reported. Both the pathogenesis and the management of SAA associated with SLE are discussed following the presentation of this case. This is the first reported case of SAA associated with SLE.

    Description / Table of Contents: Résumé Nous avons analysé, aux points de vue métastases ganglionnaires et survie, 530 cas de cancers gastriques traités par chirurgie curative. Pendant cette période, les résections à visée curative ont représenté 75.9% de toutes les résections pour cancer gastrique (530/698) et la survie à 5 ans a été de 50.6% (268/530). Au total, 15,739 ganglions régionaux enlevés en cours d'opération (en moyenne 30 par pièce opératoire) ont été examinés: 16.0% renfermaient des métastases. Sur 339 malades avec cancer avancé ayant effracté les limites de l'organe, 74.9% avaient des métastases ganglionnaires; les survies à 5 ans ont été de 63.5% dans les cas sans métastase ganglionnaire et de 29.9% dans les cas avec métastase. Sur l'ensemble des survivants à 5 ans, 58.5% avaient des ganglions envahis. La résection presque totale d'au moins les premier et deuxième relais ganglionnaires drainant le cancer gastrique est un élément essentiel du traitement à visée curatrice. Pour les lésions des tiers supérieur et moyen de l'estomac qui envahissent la séreuse de la paroi gastrique postérieure, il faut réaliser une gastrectomie totale avec splénectomie et hémipancréatectomie gauche pour enlever en bloc les ganglions régionaux.
    Notes: Abstract A total of 530 cases of gastric cancer treated by curative surgery was analyzed with regard to lymph node metastases and survival rate. During this period, curative resections made up 75.9% of all resections (530/698) for gastric cancer, and the 5-year survival rate was 50.6% (268/530). A total of 15,739 regional lymph nodes (an average of 30 per specimen) removed at surgery were examined histologically for metastases, and 16.0% were found to be positive. Of 339 patients with advanced, transmural cancers, 74.9% had lymph node métastases; those with negative nodes had a 5-year survival rate of 63.5%, while those with positive nodes had a 5-year survival rate of 29.9%. The 5-year survivors with positive nodes made up 58.5% of all 5-year survivors. The almost complete removal of at least the primary and secondary lymph node groups draining a gastric neoplasm is an essential part of the curative surgical treatment of gastric cancer. For lesions in the upper and middle portions of the stomach that invade the serosa of the posterior wall, total gastrectomy with a caudal hemipancreatectomy and splenectomy should be aggressively performed, so as to accomplish complete en bloc removal of the lymph nodes in these regions.

    Description / Table of Contents: Résumé Les effets del'administration pré-opératoire de l'association bléomycine (BLM) et radiothérapie ont été étudiés par les auteurs. Sur 206 malades atteints de cancer de l'oesophage dont la tumeur a été réséquée et étudiée histologiquement, 109 ont été traités en pré-opératoire par cette association. Les résultats ont été comparés avec ceux des malades qui ont été traités seulement par la radiothérapie ou par la bléomycine ou qui n'ont pas subi de traitement pré-opératoire. Par comparaison l'association BLM-radiothérapie a donné de meilleurs résultats que la radiothérapie isolée. L'efficacité sur les cellules néoplasiques et sur les adénopathies associées s'est traduite par un meilleur pronostic: 32,2 pour cent de survie à 5 ans dans le premier groupe et seulement 16,3 pour cent dans le second groupe (avec une moyenne globale de 26,2 pout cent) La dose nécessaire de bléomycine s'élève à 75mg (7.5mg par jour pendant 10 jours) et celle des rayons à 3 000 rads (200 rads pendant 15 jours). Le traitement pré-opératoire s'étend sur 3 semaines cependant que l'opération est entreprise de une à trois semaines après son arrêt. La BLM peut être remplacé par d'autres agents thérapeutiques actifs sur les cancers de type malphighien.
    Abstract: Abstracto Se acometió la evaluación de la terapia preoperatoria combinada con Bleomicina (BLM) y radioterapia para carcinoma esofágico. De 206 pacientes con carcinomas esofágicos resecados y examinados histológicamente, 109 fueron tratados con BLM e irradiación (grupo BR) y comparados con otros grupos: el grupo R, fué tratado con irradiación sólamente, el grupo B con BLM sólamente y el grupo N no recibió tratamiento preoperatorio. La terapia combinada de BLM e irradiación en comparación, particularmente con la radioterapia simple, fué considerada como de mejor efectividad histológica sobre las células carcinomatosas, especialmente en los ganglios linfáticos remotos, y de mejores resultados pronósticos representados por las tasas de supervivencia a 5 años: 32.2% para el grupo tratado con la primera forma de terapia y 16.3% para el grupo tratado con la ultima (promedid, 26.2%). La dosis ideal total de BLM puede ser 75 mg (intramuscular, 7.5 mg por 10 días) y la irradiación 3.000 rads (200 rads × 15 días), en ambos casos en 3 semanas, y la operación puede ser realizada una a dos semanas después de completado este tratamiento. La BLM puede ser reemplazada por otros agentes quimioterapéuticos que se crea son efectivos en carcinoma escamocelular.
    Notes: Abstract Evaluation of the preoperative combined bleomycin (BLM) and radiation therapy for esophageal carcinoma was undertaken. Of 206 patients with esophageal carcinomas resected and histologically examined, 109 were treated with BLM and radiation (BR group) and compared with the other groups consisting of group R which was treated with radiation alone, group B with BLM alone, and group N with no preoperative treatment. The combined BLM and radiation therapy in comparison particularly with simple radiation therapy was considered to have better histological effectiveness on carcinoma cells, particularly in remote lymph nodes, and better prognostic results represented by the 5-year survival rates: 32.2% for the group treated with the former therapy and 16.3% for the group with the latter (average, 26.2%). The ideal total dosage of BLM may be 75 mg (intramuscular, 7.5 mg × 10 days) and that of radiation 3,000 rads (200 rads × 15 days), both in 3 weeks, and the operation may be performed 1 or 2 weeks after completion of this treatment. BLM may be replaced by other chemotherapeutic agents that are believed effective in squamous cell carcinomaf.
