    Notes: Summary The binding of disopyramide (DSP) to plasma (or serum) proteins was determined using an ultrafiltration technique in three patients undergoing haemodialysis. An increase in the free fraction (FF) of DSP during dialysis occurred together with elevation of the free fatty acid (FFA) level in plasma. The effect of FFA on protein binding in vitro was examined using DSP- and FFA-spiked solutions containing human α1-acid glycoprotein and serum albumin. The FF of DSP rose in proportion to increasing FFA levels, supporting the in vivo observations. The findings suggest that the free concentration of DSP should be routinely monitored, especially in haemodialysis patients.

    Notes: Summary The effect of three new fluoroquinolones on theophylline kinetics and the urinary excretion of metabolites was studied in 5 healthy subjects (3 male, 2 female). All subjects received serial, single i.v. infusions of theophylline (aminophylline, 250 mg) over 60 min after 200 mg doses of a quinolone (enoxacin, ofloxacin, norfloxacin) every 8 h for 3 consecutive days, the quinolone being administered up to the day following theophylline administration. Pretreatment with ofloxacin and norfloxacin did not influence theophylline disposition, but theophylline clearance fell from 0.054 to 0.027 l·h−1·kg−1 in the presence of enoxacin, without a change in the apparent volume of distribution. Enoxacin, too, was the sole compound to increase the urinary excretion of theophylline (33.2 vs 43.9 mg, before vs after treatment), and significantly to decrease the excretion of 3-methylxanthine (3-MX), 1-methyluric acid (1-MU) and 1,3-dimethyluric acid (1,3-DMU) in 24-h urine samples (from 19.8 to 7.16 mg, from 28.3 to 10.3 mg and from 68.8 to 49.5 mg, respectively). The effect of the quinolones on hepatic drug metabolizing enzyme activity was investigated in each subject using the ratios of 6-hydroxycortisol to total 17-hydroxycorticosteroids and to free cortisol in 24-h urines as an index of the hepatic P-450-dependent enzyme system. No significant difference in ratio was observed between control and other treatments. It is concluded that the theophylline-enoxacin interaction was largely due to inhibition of a metabolic system other than the common hepatic P-450 system.

    Notes: Point defects have been observed directly in non-stoichiometric niobium oxides, Nb12O29, by high- resolution electron microscopy at a resolution of 3-4 Å. A proposed model for the point defect is a complex composed of two interstitial metal atoms and two interstitial oxygen atoms. Observations have been made on the annealing out of these defects under electron irradiation. Evidence on the density of these defects offers a semi-quantitative explanation of how the non-stoichiometric oxygen excess in these oxides is accommodated in the matrix structure.

    Notes: The crystal structure of bismuth tungstate, Bi2W2O9, has been examined by 1 MV high-resolution electron microscopy. The symmetry is orthorhombic with lattice parameters a = 5.43, b = 5.41 and c = 23.7 Å and space group Pna21. The point group is determined directly from the images in which each cation site is resolved. The structure is similar to that of conventional layered Bi compounds with respect to the Bi2O2 sheets normal to the c axis; however, the layer between the sheets is of a deformed ReO3 type. The strong chemical bonds between the Bi ions and the apex O atoms cause the rotation of the WO6 octahedron about the c axis. W ions are displaced antiparallel in the WO3 layer.

    Notes: With the use of high-resolution electron microscopy, defects have been observed in crystals of Nb22O54: two types of point defects and extended Wadsley-type defects having a structure corresponding to Nb12O29. For one type of point defect a model is proposed which contains two displaced metal atoms and two interstitial oxygen atoms within a 'block'. For the other, one metal atom and three oxygen atoms are considered to be missing from a tetrahedral site. Contributions made by the three types of defects to the non-stoichiometry of the crystals are discussed.

    Notes: The crystal structure of a bismuth titanate, Bi7Ti4NbO21, was determined on the basis of super-high- resolution images, taken by a recently constructed 1000 kV electron microscope, in which each cation site was approximately defined. It was found that the crystal has a mixed-layer structure, i.e. Bi4Ti3O12 and Bi3TiNbO9-like layers are alternately intergrown. The symmetry is orthorhombic with space group I2cm. The lattice parameters are a = 5.45, b = 5.42 and c = 58.1 Å. That the periodicity along c is twice that expected from a simple intergrowth can be accounted for by the rotation of octahedra. The lattice defects found are discussed in terms of twinning of the crystal.

    Notes: Activity of fish muscle glutathione peroxidase, which presumably protects muscle from oxidative deterioration during storage and processing, was found in both Japanese jack mackerel and skipjack tuna. Activity of the peroxidase and level of reduced glutathione (enzyme substrate) decreased during 5 days storage at 4°C. Lipid hydroperoxides were substantially formed in the fish muscles during the storage.
