    Notes: Zusammenfassung Einmalige Injektion von Reserpin bewirkt beim Meerschweinchen dosis-abhängige Abnahme der histochemischen Reaktivität der enterochromaffinen Zellen und Verminderung des Gesamt-5HT-Gehaltes im Dünndarm. Durch Vorbehandlung mit Iproniazid, nicht aber mit Isoniazid, wird die Reserpin-bedingte Abnahme von histochemischer Reaktivität der enterochromaffinen Zellen und Gesamt-5HT-Gehalt des Dünndarms vermindert. Dies spricht dafür, daß Iproniazid die Freisetzung von 5HT nach Reserpin hemmt und daß bei der 5HT-Freisetzung durch Reserpin Monoaminoxydase beteiligt ist.

    Notes: Zusammenfassung Die fortgesetzte Verabreichung von nicht spezifisch knochenmarksaktiven Arzneimitteln an Ratten führt bei Verwendung großer Dosen zu histopathologisch faßbaren Veränderungen des Knochenmarkes. Diese bestehen in einer Atrophie und Umgestaltung des Fettgewebes mit reaktiver Gefäßerweiterung und Ödembildung. Schwere toxische Schäden äußern sich außerdem in Knochenmarksblutungen und Kernpyknose der Megakaryocyten. Diese Reaktionen des Knochenmarksstromas werden — genügend hohe Dosierung vorausgesetzt — mit verschiedensten, gut verträglichen Arzneimitteln beobachtet, sind somit unspezifisch und lassen nicht auf eine knochenmarksschädigende Wirkung bei therapeutischer Anwendung am Menschen schließen. Dagegen sind die beschriebenen Markveränderungen Ausdruck für die „allgemeine Verträglichkeit“ von Arzneimitteln. Ausdehnung und Schweregrad der Markveränderungen verhalten sich zur Überlebenszeit der Versuchstiere reziprok, wie am Beispiel von Isonicotinsäurehydrazid und einiger seiner Derivate gezeigt wird.

    Notes: Abstract The possibility that the exposure of the embryo to certain chemical substances can lead to behavioral disturbances is known from human epidemiological studies, e.g., in chronic poisoning with mercury and ethanol. Therefore, efforts are made to develop toxicological techniques with which new behavioral teratogens can be recognized. The review describes the most important experimental methods which are presently explored, and which are based on a rich body of knowledge accumulated by experimental psychologists. Most of the tests were developed with small animals, mostly with rats. They range from a rather straightforward determination of various reflexes to complex behavioral situations involving mechanical devices, operant conditioning techniques and procedures evaluating social behavior. In applying these methods in routine toxicology, it is important to remember, that many behavioral effects determined in newborn and adult animals are subtle. Moreover, they are influenced by a large variety of environmental factors affecting the health and the behavior of the mothers and of the offspring in the early and later phases of development. Therefore, the experiments must be conducted under highly standardized conditions and must be controlled rigorously. It is concluded that the best experimental strategy for the evaluation of potential behavioral teratogens is not yet established. Therefore, it would be premature to decide on a fixed protocol to be included in routine animal safety experiments for drugs and other chemical substances.

    Notes: Zusammenfassung Die kardiotoxische Wirkung von Adriamycin wurde im Elektrokardiogramm von Ratten nach wiederholten i.p. Injektionen nachgewiesen. Bei gleichzeitiger Behandlung mit Acetyldaunomycin waren die elektrokardiographischen Veränderungen signifikant geringer. Die letale Wirkung grosser Adriamycindosen wurde durch Acetyldaunomycin ebenfalls abgeschwächt. Dagegen hatte Acetyldaunomycin keinen Einfluss auf die durch Adriamycin erzeugte Leukopenie.

    Notes: Abstract Daunomycin, adriamycin and 5 semisynthetic anthracycline antibiotics inhibited oxygen consumption or ATP production of rat heart mitochondria in vitro. The no-effect levels varied depending on the substrate used and ranged from 1 nmole to 800 nmole per mg mitochondrial protein. Mitochondrial functions were also studied in hearts of rats treated with repeated i.p. injections of the 7 antibiotics. Decrease in oxygen consumption without change in ATP production was observed with adriamycin and NSC-149584. Daunomycin, NSC-164011, NSC-143496 NSC-143114 affected primarily ATP production. The most potent compounds were daunomycin and adriamycin which damaged mitochondrial funciton at cumultive doses of approximately 10mg/kg. ECGs were monitored in groups of equally treated rats. Cardiotoxicity manifested itself by progressive widening of the QRS complex often followed by the development of a S-wave trough. The most toxic compounds also induced intraventricular block, bradycardia and heart failure. The development of the ECG changes showed a good correlation with the impairment of mitochondiral function.

    Notes: Abstract Chronic oral administration of imipramine to rats caused characteristic changes of the electrocardiogram (ECG), i.e. prolongation of the PR interval, widening of the QRS complex, and increase in T-wave voltage. The cardiotoxic anthracycline antibiotic adriamycin induced dose-dependent widening of the QRS complex. This effect on intraventricular conduction was not enhanced in rats receiving both drugs. The high adriamycin dose (5×4 mg/kg) abolished imipramine-induced prolongation of the PR interval and T-wave elevation. This was not seen with the low adriamycin dose (20×1 mg/kg). Imipramine prolonged survival time of rats treated with toxic doses of adriamycin, but enhanced growth retardation in animals receiving the low adriamycin dose. Chronic treatment with increasing doses of strophanthin induced significant flattening of the T wave in rats with and without imipramine therapy, but did not influence the changes of the ECG or body weight gain caused by imipramine. It is concluded that the combined use of imipramine and adriamycin or strophanthin did not lead to a serious enhancement of the toxicity of the tricyclic antidepressant.
