    Notizen: Abstract: Positron emission tomography (PET) with labeled neuroleptics has made possible the study of neurotransmitter-receptor systems in vivo. In this study we investigate the kinetics of the 3, 4-dihydroxyphenylethylamine (dopamine) receptor-ligand binding using PET data from a series of experiments in the baboon with the 18F-labeled drugs spiperone, haloperidol, and benperidol. Models used to describe these systems are based on first-order kinetics which applies at high specific activity (low receptor occupancy). The parameters governing the uptake and loss of drug from the brain were found by fitting PET data from regions with little or no receptor concentration (cerebellum) and from experiments in which specific binding was blocked by pretreatment with the drug (+)-butaclamol. Receptor constants were determined by fitting data from receptor-containing structures. Correcting the arterial plasma activities (the model driving function) for the presence of drug metabolites was found to be important in the modeling of these systems.

    Notizen: Abstract: The application of positron emission tomography (PET) to the study of biochemical transformations in the living human and animal body requires the development of highly selective radiotracers whose concentrations in tissue provide a record of a discrete metabolic process. l-N-[11C-methyl]Dtprtnyl ([11C]l-deprenyl), a suicide inactivator of monoamine oxidase (MAO) type B, has been developed as a radiotracer for mapping MAO B in the living human and animal brain. In this investigation, [11C]l-deprenyl (1) and [11C]l-deprenyl-α,α-2H2 (2) have been compared in three different baboons by PET measurement of carbon-11 uptake and retention in the brain and the measurement of the amount of unchanged tracer in the arterial plasma over a 90-min time interval. For one baboon, N-[11C-methyl-2H3]h-de-prenyl (3) was also studied. Kinetic parameters calculated using a three-compartment model revealed a deuterium isotope effect of 3.8 ± 1.1. Comparison of the two tracers (1 and 2) in mouse brain demonstrated that deuterium substitution significantly reduced the amount of radioactivity bound to protein. HPLC and GLC analysis of the soluble radioactivity in mouse brain after injection of [11C]l-deprenyl showed the presence of [11C]methamphetamine as a major product along with unidentified labeled products. Sodium dodecyl sulfate-polyacrylamide electrophoresis with carbon-14-labeled l-deprenyl showed that a protein of molecular weight 58,000 was labeled. These results establish that MAO-catalyzed cleavage of the α carbon-hydrogen bond on the propargyl group is the rate limiting (or a major rate contributing) step in the retention of carbon-11 in brain and that the in vivo detection of labeled products in brain after the injection of [11C]L-deprenyl provides a record of MAO activity.

    Notizen: The magnitude of local stress applied to a polymer surface in the buffing process, which is a critical method to uniformly align liquid crystals for displays, has been evaluated from the polarizing optical microscopic investigation of nematic liquid crystals in contact with polyimide surfaces which were suitably buffed in order to visualize the individual tracks of the buffing rayon fibers. The estimated magnitude of local stress is most likely to be far greater than the yield stress of the polymer film, thereby causing permanent molecular orientation along the buffing direction. The surfaces of poly(amic acid) and cured polyimide films, subjected to different buffing and cure profiles, have been examined by atomic force microscopy. The ability of these buffed polymer surfaces to align nematic and chiral smectic C (ferroelectric) liquid crystals were also investigated. These studies show that the presence of microgrooves on buffed polymer surfaces is not necessary for alignment of liquid crystals. The contribution of polymer molecules from the buffing fibers, deposited and oriented on the polyimide surfaces during buffing, can also be ruled out. Rather, it is concluded that the liquid-crystal alignment is mainly caused by the anisotropic interactions between liquid-crystal molecules and the polymer segments oriented through plastic deformation, with little contribution from frictional heating. For the alignment of smectics, both the degree of order and mechanical properties of polyimide films are found to be important factors. © 1996 American Institute of Physics.

    Notizen: Interferometric thermometry is a promising noncontact technique for measuring the temperature of transparent substrates (with polished front and back surfaces) from thermally induced changes in sample thickness and refractive index. However, for substrates of uniform thickness, the technique is not sensitive to the direction of temperature change, thus restricting its use to situations in which the temperature variation is monotonic. In this paper, we present some new schemes for interferometric thermometry based on the wavelength modulation capabilities of the distributed feedback laser diode. These schemes allow both the magnitude and direction of temperature change to be determined. One scheme utilized to measure temperature changes in a silicon wafer during thermal cycling is described in detail. In addition, the calibration factors required to convert the thermally induced reflectance oscillations ("fringes'') of known-thickness samples into temperature change are provided for Si and GaAs at wavelengths near 1.5 μm.

    Notizen: Scanning tunneling microscopy (STM) was employed to measure the three-dimensional surface topography of radial servo grooves etched in a glass substrate. The effect of groove depth (nominal values of 20, 40, and 70 nm) on tracking servo signals at a track pitch of 1.6 μm is examined. Using an optical disk tester, three groove related signals were measured for each groove depth: the on-track signal ratio, cross-track ratio, and the push-pull ratio. In a key feature of this study, scalar diffraction theory was used to calculate the groove related signals from the STM profiles and the beam profile of the optical disk tester. The calculated groove related signals were found to be in nearly quantitative agreement with those measured with the disk tester.

Room-temperature angularly tuned type I phase matching for doubling of 1.064 μm radiation was achieved with a deff =27 pm/V. Favorable crystal growth habits were observed.

    Notizen: Recently, there have been significant advances in demonstrating the possible application of scanning probe techniques for high density data storage. While many obstacles have yet to be overcome, one of the more promising applications of such devices is for very high density read-only disk drives. Such an application requires the means to pattern and replicate nanometer scale features over macroscopic dimensions. We have developed a technique for both mastering and replicating data patterns for use in such a device at a density 100 times that of current read-only disks. The process consists of using electron beam lithography to write data features as small as 50 nm and a photopolymerization process to faithfully replicate the written marks. © 1996 American Institute of Physics.

    Notizen: We have demonstrated a new planarized all-refractory technology for low Tc superconductivity (PARTS). With the exception of the Nb-AlOx-Nb trilayer preparation, the processing is done almost exclusively within an advanced Si technology fabrication facility. This approach has allowed us to leverage highly off of existing state-of-the-art lithography, metal etching, materials deposition, and planarization capabilities. Using chemical-mechanical polish as the planarization technique we have fabricated Josephson junctions ranging in size from 0.5–100 μm2. Junction quality is excellent with the figure of merit Vm typically exceeding 70 mV. PARTS has yielded fully functional integrated Josephson devices including magnetometers, gradiometers, and soliton oscillators.
