    Notes: We have measured the static dipole polarizability of Nay−xLix clusters (with y=2, 3, 4, and 8) by molecular beam deflection technique. For a given size, the polarizability of pure lithium clusters is smaller than the polarizability of pure sodium clusters. For mixed clusters, a smooth decrease in the polarizability is observed as the proportion of lithium atoms increases. For the NaLi molecule, both experimental permanent dipole and average polarizability have been obtained. Experimental results are compared to results of density functional theory and configuration interaction single and double (CISD) ab initio calculations. © 1999 American Institute of Physics.

    Notes: The metallicity of lithium rich LinHm〈sup ARRANGE="STAGGER"〉 + cluster ions [1≤m≤6, n≤22, and (n–m)〉3] is studied by measuring unimolecular dissociation rates. These clusters are found to decompose by evaporation of a Li atom or a Li2 molecule. The evaporative rates of mixed clusters display features characteristic of metallic clusters. This confirms and extends to a larger size range the previous results obtained by photoionization and absorption cross-section measurements. The simulation of the evaporative rates, with a statistical Rice–Ramsperger–Kassel (RRK) model which has been adapted to mixed clusters, confirms that there is a clear separation between a metallic Lin–m〈sup ARRANGE="STAGGER"〉 + part and an insulating (LiH)m part. © 1997 American Institute of Physics.

    Notes: We present the first direct measurement of the electric polarizability of isolated C60 molecules by molecular beam deflection technique. We have obtained a value of 76.5±8.0 Å3 which is consistent with most of the recent calculations and slightly lower than the value of the polarizability of C60 measured in fullerite crystals. © 1999 American Institute of Physics.

    Notes: The second order nonlinear response from 4 nm diam gold clusters embedded in an alumina matrix deposited on a pure silica substrate has been recorded as a function of the wavelength of the fundamental incident beam. The spectrum exhibits a narrow resonance band peaked at 520 nm as a result of the coupling of the second harmonic field with the surface plasmon of the particles. The nonlinear second harmonic response of gold clusters is found to be blueshifted compared to the bulk, due to the finite size effects on the cluster optical properties, as already observed with the linear response. Furthermore, this nonlinear response appears to be well described with a simple free electron model where the valence electrons only participate through the screening of the ionic cores, owing to the weakness of the interband transition contribution. © 1998 American Institute of Physics.

    Notes: Abstract: Amphiphysin I and II are nerve terminal-enriched proteins that display src homology 3 domain-mediated interactions with dynamin and synaptojanin. It has been demonstrated that the amphiphysins also bind to clathrin, and we have proposed that this interaction may help to target synaptojanin and dynamin to sites of synaptic vesicle endocytosis. To understand better this potential functional role, we have begun to characterize clathrin-amphiphysin interactions. Using PCR from adult human cortex cDNA, we have cloned a number of amphiphysin II splice variants. In in vitro binding assays, the amphiphysin II splice variants display differential clathrin binding and define a 44-amino acid region mediating the interaction. Amphiphysin II truncation and deletion mutants identify two distinct clathrin-binding domains within this region: one with the sequence LLDLDFDP, the second with the sequence PWDLW. Both domains are conserved in amphiphysin I, and saturation binding analysis demonstrates that both sites bind clathrin with approximately equal affinity. The elucidation of clathrin as a splice-specific binding partner for amphiphysin II begins to address the potential functional role(s) for the multiple amphiphysin II splice variants and further supports an important function for clathrin-amphiphysin interactions in protein targeting during endocytosis.

    Notes: The metastable decomposition of hydrogenated lithium cluster ions (LiH)nLi+m(m=0, 1 and 3; n≤15) is studied by using a reflectron mass spectrometer. These clusters are found to decompose by evaporation of a LiH or a Li2H2 molecule. The binding energy of these clusters are determined, using a statistical model which has been adapted to mixed clusters. Comparison with other mixed clusters suggests that (LiH)nLi+ clusters form compact cubic structure similar to pieces of a crystal lattice. For (LiH)nLi+3 clusters, the dissociation channels are more surprising, and the localization of the two excess electrons is discussed, as well as the possible existence of an energy barrier for the dissociation. © 1996 American Institute of Physics.

    Notes: Diazaborine and isoniazid are, at first sight, unrelated anti-bacterial agents that inhibit the enoyl-ACP reductase (ENR) of Escherichia coli and Mycobacterium tuberculosis respectively. The crystal structures of these enzymes including that of the diazaborine-inhibited E. coli ENR have been obtained at high resolution. Site-directed mutagenesis was used to study the importance of amino acid residues in diazaborine susceptibility and enzyme function. The results show that drug binding and inhibition require the presence of a glycine residue at position 93 of E. coli ENR or at the structurally equivalent position in the plant homologue, which is naturally resistant to the drug. The data confirm the hypothesis that any amino acid side-chain other than hydrogen at this position within the three-dimensional structure of these enzymes will affect diazaborine resistance by encroaching into the drug binding site. Substitutions of Gly-93 by amino acids with small side-chains, such as serine, alanine, cysteine and valine, hardly affected the catalytic parameters and rendered the bacterial host resistant to the drug. Larger amino acid side-chains, such as that of arginine, histidine, lysine and glutamine, completely inactivated the activity of the enzyme.

    Notes: Abstract The Escherichia coli fabH gene encoding 3-ketoacyl-acyl carrier protein synthase III (KAS III) was isolated and the effect of overproduction of bacterial KAS III was compared in both E. coli and Brassica napus. The change in fatty acid profile of E. coli was essentially the same as that reported by Tsay et al. (J Biol Chem 267 (1992) 6807–6814), namely higher C14:0 and lower C18:1 levels. In our study, however, an arrest of cell growth was also observed. This and other evidence suggests that in E. coli the accumulation of C14:0 may not be a direct effect of the KAS III overexpression, but a general metabolic consequence of the arrest of cell division. Bacterial KAS III was expressed in a seed- and developmentally specific manner in B. napus in either cytoplasm or plastid. Significant increases in KAS III activities were observed in both these transformation groups, up to 3.7 times the endogenous KAS III activity in mature seeds. Only the expression of the plastid-targeted KAS III gene, however, affected the fatty acid profile of the storage lipids, such that decreased amounts of C18:1 and increased amounts of C18:2 and C18:3 were observed as compared to control plants. Such changes in fatty acid composition reflect changes in the regulation and control of fatty acid biosynthesis. We propose that fatty acid biosynthesis is not controlled by one rate-limiting enzyme, such as acetyl-CoA carboxylase, but rather is shared by a number of component enzymes of the fatty acid biosynthetic machinery.

Complementation of a mutation affecting bacterial membrane lipid biosynthesis by a plant ARF protein, could indicate the existence of as yet unidentified bacterial equivalents of this ubiquitous eucaryotic GTP-binding protein.
