    Notizen: Objectives In a cohort of term infants with cerebral depression at delivery, to investigate the association of perinatal signs of birth asphyxia, particularly abnormal fetal heart rate patterns in labour, acidaemia, and serious neonatal encephalopathy, with neurodevelopmental outcome at age five years.Design Five year follow up study of a birth cohort.Setting Regional maternity hospital.Subjects One hundred and eighty-four singleton infants with a 1 min Apgar score ≤ 3, born at term between January 1984 and September 1985.Main outcome measures Neonatal death, cerebral palsy, and scores on a battery of neurodevelopmental tests at age five.Results Seven infants had a cluster of perinatal signs suggestive of birth asphyxia; all included serious neonatal encephalopathy. Three of these infants died neonatally, three had spastic quadriparesis with profound developmental delay, and one was unimpaired at the age of five. Among the remaining infants, no association was found between severely abnormal fetal heart rate patterns in labour and scores on neurodevelopmental tests, or between acid–base status at delivery and test scores.Conclusions Birth asphyxia, identified by a cluster of abnormal perinatal signs, including serious neonatal encephalopathy, has a poor prognosis. If serious encephalopathy is not present, cerebral depression at birth preceded by abnormal fetal heart rate patterns in labour, or with acid–base derangement, is not predictive of later impairment.

    Notizen: The stresses in the aluminum oxide formed during high-temperature oxidation of a bond-coated superalloy are shown to be measurable through zirconia thermal barrier coatings. The basis for the measurements is the piezospectroscopic shift in the R-line fluorescence (photoluminescence) from Cr3+ impurities incorporated into the growing aluminum oxide scale. Measurements through the thermal barrier coating are feasible because (partially stabilized) zirconia coatings have some transparency at both the excitation and at fluorescence frequencies. © 1996 American Institute of Physics.

    Notizen: Tests of the electron beam injector for the Boeing/Los Alamos Average Power Laser Experiment have demonstrated first time operation of a photocathode radio frequency gun accelerator at 25% duty factor, exceeding previous photocathode operating parameters by three orders of magnitude. The macropulse format was 30 Hz and 8.3 ms with a micropulse frequency of 27 MHz. Average beam currents of up to 32 mA have been accelerated to 5 MeV for an average beam power of 160 kW. The macropulse peak current was 128 mA. The 32 mA average beam current exceeds previous cathode performance by a factor of 1000. Emittance measurements demonstrate excellent electron beam quality.

    Notizen: Abstract Recent research from 1993 on performance evaluations in work settings is reviewed and integrated with the prior reset and historical bases. Contemporary research reflects several themes: General models of job performance are being developed, the job performance domain is being expanded, research continues to explore the psychometric characteristics of performance ratings, research is developing on potential bias in ratings, rater training is examined, and research continues in terms of efforts to attach utility values to rated performance. We conclude that research is progressing in traditional content areas as well in the exploration of new ground. Researchers are recognizing that job performance is more than just the execution of specific tasks and that it involves a wider array of important organizational activities. There is also an increased optimism regarding the use of supervisory ratings and recognition that such "subjective" appraisal instruments do not automatically translate into rater error or bias.

    Notizen: Kisspeptin is the peptide product of the KiSS-1 gene and the endogenous agonist for the GPR54 receptor. Recent evidence suggests the kisspeptin/GPR54 system is a key regulator of the reproductive system. We examined the effect of intracerebroventricular (i.c.v.) and peripheral administration of the active kisspeptin fragment, kisspeptin-10, on circulating gonadotrophins and total testosterone levels in adult male rats. The effect of kisspeptin-10 in vitro on the release of hypothalamic peptides from hypothalamic explants and gonadotrophins from anterior pituitary fragments was also determined. The i.c.v. administration of kisspeptin-10 dose-dependently increased plasma luteinizing hormone (LH) and increased plasma follicle stimulating hormone (FSH) and total testosterone at 60 min postinjection. In a separate study investigating the time course of this response, i.c.v. administered kisspeptin-10 (3 nmol) significantly increased plasma LH at 10, 20 and 60 min, FSH at 60 min and total testosterone at 20 and 60 min postinjection. Kisspeptin-10 stimulated the release of luteinizing hormone-releasing hormone (LHRH) from in vitro hypothalamic explants. Peripheral administration of kisspeptin-10 increased plasma LH, FSH and total testosterone. However, doses of 100–1000 nM kisspeptin-10 did not influence LH or FSH release from pituitary fragments in vitro. Kisspeptin therefore potently stimulates the hypothalamic-pituitary-gonadal axis. These effects are likely to be mediated via the hypothalamic LHRH system.

    Notizen: Orexin immunoreactive fibres are abundant in the hypothalamus suggesting a neuroendocrine regulatory role. Intracerebroventricular (ICV) administration of orexin A suppressed plasma prolactin in male rats by 71% at 20 min post-injection and 83% at 90 min post-injection (P 〈 0.005 vs saline at both time points). To investigate whether this effect was through the tuberoinfundibular dopaminergic (TIDA) system, a supra-maximal dose of domperidone, a dopamine receptor antagonist, was injected intraperitoneally (i.p.) prior to ICV injection of orexin A. ICV orexin A significantly suppressed domperidone (9 mg/kg)-stimulated plasma prolactin levels, by up to 40% (i.p. domperidone + ICV orexin A 3 nmol 34.5 ± 7.4 ng/ml and i.p. domperidone + ICV orexin A 20 nmol 43.5 ± 4.3 ng/ml, both P 〈 0.005 vs i.p. domperidone + ICV saline 57.9 ± 2.7 ng/ml). Orexin A, 100 nM, significantly stimulated release of neurotensin, vasoactive intestinal polypeptide, somatostatin, corticotropin releasing factor and luteinizing hormone releasing hormone, but had no effect on release of dopamine, thyrotropin releasing hormone (TRH), vasopressin or melanin-concentrating hormone from hypothalamic explants in vitro. Orexin A did not alter basal or TRH stimulated prolactin release in dispersed pituitary cells harvested from male rats. The data suggest that ICV administration of orexin A suppresses plasma prolactin in part through a pathway independent of the dopaminergic system.

    Notizen: The kisspeptins are the peptide products of the KiSS-1 gene and the endogenous agonists for the GPR54 receptor. Although KiSS-1 was initially discovered as a metastasis suppressor gene, recent evidence suggests the kisspeptin/GPR54 system is a key regulator of the reproductive system. Disrupted GPR54 signalling causes hypogonadotrophic hypogonadism in rodents and man. Central or peripheral administration of kisspeptin potently stimulates the hypothalamic-pituitary-gonadal axis, increasing circulating gonadotrophin concentrations in a number of animal models. These effects appear likely to be mediated via the hypothalamic gonadotrophin-releasing hormone system, although kisspeptins may have direct effects on the anterior pituitary gland. Hypothalamic KiSS-1 expression is regulated by circulating sex steroids. The precise physiological role of the kisspeptin system in the regulation of reproductive function remains to be elucidated.

    Notizen: A number of neuropeptides implicated in the hypothalamic regulation of appetite are synthesized in the arcuate nucleus (Arc). Neuropeptide Y (NPY) and agouti-related protein (Agrp) are orexigenic. The pro-opiomelanocortin (POMC) product alpha-melanocyte-stimulating hormone (α-MSH) is anorectic. Intracerebroventricular administration of cocaine- and amphetamine-regulated transcript (CART) decreases food intake. However, recent results show that CART is orexigenic when injected into discrete hypothalamic nuclei. There is almost complete coexpression of NPY and Agrp mRNA in Arc neurones, and the majority of CART-containing neurones in the Arc also contain POMC mRNA. We investigated possible interactions between these neuropeptides in vitro using a rat hypothalamic explant system. Administration of 1, 10 and 100 nm of NPY to hypothalamic explants significantly increased release of Agrp(83-132)-immunoreactivity (IR). NPY (10 and 100 nm) significantly increased the release of CART(55-102)-IR and α-MSH-IR from hypothalamic explants. Agrp(83-132) (10 nm) administered to hypothalamic explants significantly increased the release of NPY-IR. Agrp(83-132) (10 and 100 nm) significantly decreased the release of CART(55-102)-IR from hypothalamic explants. Administration of 1, 10 and 100 nm CART(55-102) to hypothalamic explants resulted in a significant increase in NPY-IR release. Administration of 10 nm CART(55-102) to hypothalamic explants significantly increased the release of Agrp(83-132)-IR. NDP-MSH (10 nm) administered to hypothalamic explants significantly increased the release of NPY-IR. NDP-MSH (10 and 100 nm) significantly increased the release of Agrp(83-132)-IR from hypothalamic explants. These data suggest that orexigenic neuropeptides in the arcuate nucleus stimulate the release of each other, perhaps reinforcing orexigenic behaviour via a positive-feedback loop. Our results are also in keeping with the possibility that the melanocortin-3 receptor in the arcuate nucleus may influence the release of arcuate neuropeptides.

    Notizen: Many studies have shown the negative effects of cocaine on neuropsychological and cognitive performance in drug-dependent individuals, but little is known about the underlying neuroanatomy of these dysfunctions. The present study addressed attention switching between items held in working memory (WM) with a task in which subjects were required to store and update two items held in verbal or visuospatial WM. Attention-switching frequency varied between trials, thereby allowing us to isolate the switching component of task performance. Behavioural data revealed that cocaine addicts performed worse than healthy controls in all tasks. On the visuospatial task addicts performed at chance levels revealing particular impairment in visuospatial WM. On the verbal task, in which controls and users could be matched for performance, we identified attenuated responses in prefrontal and cingulate cortices and in striatal regions, while other areas such as dorsolateral prefrontal cortex did not differ between healthy controls and users. The results reveal that addiction may be accompanied by specific rather than ubiquitous hypoactivation in prefrontal and subcortical areas and suggest a compromised ability in users to control their attention to their thoughts as might be particularly relevant when required to switch away from drug-related thoughts, and thus the dysfunction in attention switching may contribute to the maintenance of addiction.
