    Notizen: Abstract Tangential flow filtration of a biotransformation medium of 6-α-methylhydrocortisone to 6-α-methylprednisolone (medrol) was carried out to study the recovery of the product, from heattreated cell suspensions of Arthrobacter simplex. A higher product recovery rate was achieved when concentration of the medium was performed prior to the diafiltration. The concentration of the product in the permeate was also higher when concentration is the first mode of operation. Combined anionic and cationic microsized polymeric particles were used as filtration aids. These resins flocculated cells of A. simplex and allowed a 5-fold enhancement in the recovery rate of the product, through a 0.22 μm Durapore membrane, by increasing the diafiltration flux from 20 dm3/hm2 to 125 dm3/hm2. The medrol rejection coefficient remained constant when the polymeric particles were used, being highly dependent on the cell concentration.

    Notizen: Abstract The aetiopathology of extrahepatic portal vein obstruction is unknown. In retrospective studies, umbilical vein cannulation and sepsis have been alleged to cause portal thrombosis. This prospective study was undertaken to detect whether thrombosis and consequent obstruction of the splenoportal venous system develops after umbilical vein catheterization for exchange transfusion in newborns using Doppler ultrasound. Forty children (M = 24; F = 16) who had undergone exchange transfusion for hyperbilirubinaemia were studied at school age. Maximal duration of the venous umbilical cannulation was 120 min and sepsis did not occur. Clinical, biological and sonographic examinations were normal, except in 3 children. In 2 the left branch of portal vein could not be identified (normal variant). Conclusion Our results show that, in these children, umbilical vein catheterization did not lead to development of portal vein thrombosis. However, when other risk factors such as umbilical infection, traumatic catheterization are associated, children should be screened for obstruction of the portal vein.

    Notizen: Vortex flow filtration experiments with bovine serum albumin (BSA) solutions and Arthrobacter simplex cell suspensions in a rotary membrane device are modeled in the pressure-dependent flux and mass-transfer-controlled regions. An expression for the concentration-dependent diffusion coefficients accounting for hydrodynamic and long-range potential interactions between rigid spherical macromolecules is used for BSA. A shear-induced diffusion coefficient, strongly concentration-dependent on the cell concentration, is assumed to describe the diffusion behavior of the cell suspensions. The adjusted model parameters for albumin solutions in the pressure-dependent region of flux predict the permeation flux behavior for A. Simplex cell suspensions. When the mass-transfer correlation is calculated using cell diffusion coefficients, based on a volumetric cell fraction that is around half of the average value of the calculated membrane surface fractions, the fluxes are estimated within less than 10.5% relative error. In the mass-transfer-controlled region of flux, the volume fractions of cells at the membrane surface are between 0.16 and 0.35, depending on bulk concentration and hydrodynamic conditions.
