    Notizen: Two rabbit arylamine N-acetyltransferases (NAT1 and NAT2, EC 2.3.1.5) have been cloned and characterized recently in this laboratory. They catalyze the acetylation of primary arylamine and hydrazine drugs and other substrates in the liver, including sulfamethazine, ρ-aminosalicylic acid, and ρ-aminobenzoic acid. In the pineal gland, serotonin is metabolized to N-acetylserotonin by an unknown N-acetyltransferase. Similarity of the liver enzymes and the pineal gland arylalkylamine N-acetyltransferase (AA-NAT) has been suggested, because pineal gland homogenates were shown to metabolize arylamine substrates as ρ-phenetidine, aniline, or phenylethylamine, and liver homogenates or partially purified liver enzyme preparations catalyzed the N-acetylation of serotonin. The present study was undertaken to elucidate the possible role of NAT1 or NAT2 in serotonin acetylation in the pineal gland. We transiently expressed rNAT1 and rNAT2 genes in COS cells, studied the kinetics of the enzymes produced with various substrates, and compared these data with activities of rabbit pineal glands and livers. These enzymatic studies were complemented with western blot analysis with antibodies against NAT1 and NAT2. Cross-hybridization of rNAT1 or rNAT2 to the gene for the pineal gland AA-NAT was tested by Southern blot studies of genomic rabbit DNA. Our results indicate that although NAT1 is expressed in the pineal gland, it is not involved in the physiologically important step of N-acetylation of serotonin.

    Notizen: Oestradiol-17ß, in its positive feedback mode, as well as stimulating the release of luteinizing hormone-releasing hormone. (LHRH), also stimulates LHRH mRNA synthesis in medial preoptic neurons. Our aim was to determine whether the action of oestradiol might be mediated in part by inhibiting arcuate proopiomelanocortin (POMC) neurons which terminate on LHRH neurons. Female rats were ovariectomized at about 1000 h of dioestrus, given a subcutaneous injection of oil or oestradiol benzoate and killed at 1600 h the same day (presumptive dioestrus), presumptive pro-oestrus and oestrus and at 1000 h of presumptive pro-oestrus. In situ hybridization of brain sections was carried out with a [35S]UTP-labelled riboprobe containing 538 bases of a cDNA identical to exon 3 of the POMC gene, which includes the ß-endorphin coding domain. The amount of POMC mRNA in cells of the anterior arcuate nucleus, as assessed by grain counts, was significantly decreased in oestradiol benzoate- compared with oil-treated animals at 1600 h of pro-oestrus, but at none of the other times studied. This finding, together with other data, suggests that the positive feedback stimulation of LHRH mRNA synthesis by oestradiol-17ß may be due, in part, to the disinhibition of LHRH neurons consequent on inhibition of POMC mRNA biosynthesis in arcuate neurons.

    Notizen: Soft tissue chondromas are rare benign tumours unrelated to bone that arise primarily in the distal extremities. Lesions in the soft tissues of the oral cavity are extremely rare although several condromatous lesions have been reported in the tongue. A case is presented of a chondroma arising in the buccal mucosa. It was composed of a lobulated mass of myxoid tissue showing central areas of chondroid differentiation. Immunocytochemistry confirmed the lesion to be mesenchymal and not epithelial in origin. In the differential diagnosis it is important to exclude chondromatous change in a pleomorphic adenoma and also the possibility of a metastasis from an osseous chondrosarcoma.

    Notizen: A system for collecting and measuring X-ray diffraction data from protein crystals has been developed for a multiwire area detector. Computer programs run concurrently on two microcomputers, which collect and reduce detector data to integrated intensities. The self-contained system consists of an X-ray area detector, a rotation/oscillation camera, and two microcomputers connected by a high-speed Ethernet network. One microcomputer is dedicated to operation of the detector, control of the camera, and storage of the raw data. The second microcomputer automatically integrates the data as they are collected and allows the user to monitor the quality of data as they are processed. The integration programs are written in Fortran 77 and have been designed to be portable. Additional programs for crystal alignment, detector and camera control, and graphics are written in the C programming language. A description of the system, some characteristics of the detector, and the results of data collection are presented.

    Notizen: Comprehensive knowledge of the complicated physical behavior of the induction furnace with cold crucible (IFCC) is required to utilize the advantages of this melting aggregate in melting and casting chemically high-reactive materials, like titanium-aluminides (TiAl). Practical experiences show that the overheating temperature of the melt is decisive for the quality of the cast products. Therefore, a systematic analysis of the electromagnetic and in particular, the hydrodynamic and thermal behavior of the IFCC is carried out. The examinations of the influence of the construction elements as well as the process parameters on the temperature field and finally the overheating temperature in the IFCC are performed using specifically developed numerical models. The evaluation of the numerical results is done by experimental investigations, where aluminum serves as a model melt for the experimental determination of the thermal and hydrodynamic field of the melt. The analysis of the influence of construction-elements on the overheating temperature is focused on the design of the crucible wall and the crucible bottom, on the height-diameter ratio of the crucible and on the axial inductor position. The inductor current, the operation frequency and the crucible filling level are found to be very important for reaching a high overheating temperature.
