    Notizen: The diatomic iron–noble gas complexes FeAr+, FeKr+, FeXe+, and FeRn+ have been theoretically investigated by means of quantum-chemical calculations including an extensive treatment of electron correlation. Potential energy curves and spectroscopic constants for the lowest 4Δ and 6Δ states are derived from an open-shell coupled-cluster approach and the relative energies of all seven low-lying electronic states are evaluated by the multireference configuration interaction method. While in FeAr+ the lowest quartet and sextet states are found to be energetically almost degenerate, the heavier Fe+–noble gas molecules are predicted to exhibit 4Φ ground states. From a qualitative point of view bonding in these species is shown to be electrostatic in origin with intrinsically higher interaction energies for the quartet as compared to the sextet states. For calibration purposes, also an accurate calculation of the 4F(4s03d7)–6D(4s13d6) energy difference in the atomic Fe+ cation is provided. © 1995 American Institute of Physics.

    Notizen: We present a new design of a scanning force microscope (SFM) for operation at low temperatures in an ultrahigh vacuum (UHV) system. The SFM features an all-fiber interferometer detection mechanism and can be used for contact as well as for noncontact measurements. Cooling is performed in a UHV compatible liquid helium bath cryostat. The design allows in situ cantilever and sample exchange at room temperature; the subsequent transport of the microscope into the cryostat is done by a specially designed transfer mechanism. Atomic resolution images acquired at various temperatures down to 10 K in contact as well as in noncontact mode are shown to demonstrate the performance of the microscope. © 1998 American Institute of Physics.

    Notizen: The efficient generation of specific brain cells in vitro may serve as a source of cells for brain repair in several devastating neurological diseases. Production of dopaminergic neurons from precursor cells for transplantation in Parkinson's disease has become a major research goal. We found that murine mesencephalic neurospheres were viable and proliferated, preserved telomerase activity, pluripotency and dopaminergic commitment for many weeks when cultured in 3% O2, whereas exposing these cells to 21% oxygen prohibited long-term expansion. Microarray data suggest that a variety of genes related to the cell cycle, cell maturation and apoptosis are differentially regulated in midbrain-derived precursors cultured in 3 versus 21% oxygen after 1–2 months. Taken together, we hypothesize that sustained high oxygen has deleterious effects on the self-renewal capacity of mesencephalic neural precursors, possibly accelerating maturation and senescence resulting in overall cell loss. Gene regulation governed by low oxygen tension may be relevant to the normal development and survival of midbrain neurons.

    Notizen: Abstract There is strong evidence for a complex network-like interaction between cytokines, growth factors and other mediators being responsible for cell growth and differentiation as well as for the outcome of an inflammatory reaction. Therefore, the regulation of the production of the ubiquitous proinflammatory cytokine interleukin 6 (IL-6) by transforming growth factor β (TGFP) was investigated. Human peripheral blood mononuclear cells (PBMC), human normal keralinocytes (HNK), and an epidermoid carcinoma cell line (KB) were treated with TGFβ or TGFβ2 and subsequently IL-6 secretion was evaluated. Addition of TGFβ1 as well as TGFβ2 to PBMC, HNK and KB cells resulted in a significantly increased release of IL-6 activity. The inducing effect of TGFβ was dose dependent and maximal when supernatants were harvested 48 h after stimulation. In addition, upon Western blot analysis using a monoclonal IL-6 antibody significantly increased amounts of IL-6 protein were detected in KB cell supernatants following stimulation with TGFβ 1. These results were further confirmed at the transcriptional level using a cDNA probe specific for IL-6 and Northern blot analysis. Accordingly, an increased IL-6 inRNA expression in PBMC or KB cells was detected following TGFβl treatment. These findings indicate that TGFβ in contrast to its antiinflammatory capacities also may stimulate IL-6 production in PBMC and keratinocytes. This further supports the possibly important immunoregulatory role of growth factors such as TGFβ.

    Notizen: Abstract Irradiation with ultraviolet (UV) B light results in the formation of apoptotic keratinocyles called sunburn cells (SC). Although generation of SC appears to be one of the most characteristic features of UV-induced skin damage and has been a well-known phenomenon for a long time, the mechanisms involved are not quite clear. Recently, it was demonstrated that tumor necrosis factor α (TNFα) appears to be involved in the formation of SC since neutralization of TNFα both in vitro and in vivo reduced UVB-induced apoptosis of keratinocytes. Pentoxifylline is a methylxanthine derivative suppressing the release of TNFα. Therefore, we studied whether PTX is able to prevent the formation of SC. Addition of PTX to UVB-exposed HaCaT cells reduced DNA-fragmentation as examined by nick translation evaluated by floweytometry. To prove whether PTX also reduces UVB-induced apoptosis in vivo. BALB/c mice were exposed to UVB on their abdomens, skin biopsies performed 24 h later and SC counted. A single dose of 2000 J/m2 caused a significant induction of SC which were remarkably reduced when PTX was injected intraperitoneally 3 h before and 12 h after UVB exposure. In summary, the data demonstrate that PTX can reduce the formation of SC both in vitro and in vivo and thus further support that TNFα is involved in UVB-induced apoptosis of keratinocytes.

    Notizen: Abstract: It is almost three decades ago that it was discovered that ultraviolet radiation (UV) can compromise the immune system. UV suppresses immune responses in several ways. It inhibits the function of antigen-presenting cells, induces T cells with suppressor activity and induces the release of immunosuppressive cytokines. The latter phenomenon is mainly responsible for systemic immunosuppression. Although UV can also target cytoplasmic and cell membrane components, UV-induced DNA damage has been recognized as the most important molecular structure in mediating UV-induced immunosuppression. Recently, it was observed that interleukin-12 (IL-12), which antagonizes UV-induced immunosuppression, can accelerate the removal of UV-induced DNA lesions, probably via inducing DNA repair. Hence, it is tempting to speculate that the activity of IL-12 to reduce UV-induced immunosuppression may be due at least partially to this new biological activity of IL-12.
