Notes: Patients with germ cell tumors, hepatomas and other malignant tumors were studied throughout their courses of surgical, radiological and medical treatments with serial determinations of AFP, CEA, HCG and basic fetoprotein (BFP), which was newly found by M. Ishii. In general, these carcinoembryonic or trophoblastic markers were observed to be well correlated with the fate of the tumor in the body and to be useful in evaluating the effect of treatment or in predicting the prognosis of the patient. Except for a postoperative transient increase in serum BFP and CEA, the levels fell after surgery and rose again with recurrence of the tumor. Although we had some patients with teratocarcinoma, hepatoma or gastric carcinoma, whose serum AFP showed a transient parallelism with their clinical courses, an obvious dissociation in these markers occurred later in their course. Similar dissociations were also observed with these carcinofetal or trophoblastic markers in patients with chemotherapy or radiotherapy for their teratocarcinomas. We conclude that each carcinoembryonic protein is a useful but limited marker for malignancy but should be used clinically in combination with other tumor markers.

Notes: A new human fetal antigen, common to several types of carcinomas was found in fetal serum and fetal gut extract. Because this antigen was shown to have an isoelectric point of 9.3, it has been designated basic fetoprotein (BFP). BFP was purified from ascites of a patient with hepatoma by the techniques of QAE-Sephadex A-50 chromatography. affinity chromatography. isoelectrofocusing and gel filtration. The purified BFP had a γ-mobility by electrophoresis and a molecular weight of 124,000 by SDS polyacrylamide gel electrophoresis. Monospecific anti-BFP was prepared by immunizing a rabbit with the purified BFP.A radioimmunoassay was developed based on a coprecipitation-inhibition technique using 125I-BFP and monospecific anti-BFP. BFP in human serum was determined by the radioimmunoassay. BFP was detected in sera from all normal subjects. Serum BFP levels were lower than 100 ng/ml in 100 out of 101 normal subjects, lower than 200 ng/ml in 180 out of 204 patients with non-malignant diseases and higher than 200 ng/ml in 94 out of 251 patients with malignant neoplasms. The determination of BFP in serum was demonstrated to be useful for the diagnosis of various carcinomas.

Notes: Novel γ-glutamyl transpeptidase (γ-GTP) was purified from ascites or sera obtained from hepatoma patients by means of following procedures including ammonium sulfate fractionation, separation by 2 cycles of preparative polyacrylamide disc electrophoresis and elimination by a technique of immuno-absorption using affinity chromatography. The purified γ-GTP was shown to be homogeneous as judged by disc electrophoresis. Some physicochemical properties of the partially purified enzyme obtained at the stage of 2 cycles of preparative disc electrophoresis such as heat stability, pH optimum. Km value for substrate and effect of some cations are reported.

Notes: In view of carcinoembryonic properties with respect to γ-GTP, we investigated whether or not hepatoma specific γ-GTP isoenzyme was found in the sera of patients with hepatoma. Fractionations of serum γ-GTP were carried out in sera from 229 patients with various hepatobiliary diseases including 68 with hepatoma by polyacrylamide gradient gel electrophoresis. The band II which was seen in the region near to ceruloplasmin could be detected in 25 out of the 68 patients with hepatoma (37%), but not in the patients with other diseases. In addition to the II band, an extra band (II) was found in 15 of these 25 patients. The band II and II′ could not be found in any groups of patients studied except hepatoma patients. It was presumed that band II and II′ could be specific for hepatoma.

Notes: A new antigen, basic fetoprotein (BFP) found by M. Ishii in human fetus and in some cancer tissue of adult, was quantitated in patients' sera by radioimmunoassay. In 167 of 377 patients (44.3%) with malignant diseases, serum BFP exceeded 100 ng/ml and 87 cases (23.0%) were over 200 ng/ml. In contrast, 49 of 230 (21.3%) with benign diseases exceeded 100 ng/ml and 11 cases (4.8%) were over 200 ng/ml. No normal control exceeded 100 ng/ml.A higher incidence and higher serum-concentration of BFP were observed in patients with carcinoma of the liver, bile duct, esophagus and kidney, but it was also found in various other malignancies including leukemias and sarcomas. Generally, after a successful operation, BFP fell to normal levels, and with tumor recurrence, BFP levels rose higher particularly with massive metastasis. In the benign group, BFP was higher in patients with acute and chronic active hepatitis.Although BFP has less organ specificity than other fetal substances, and it appears in some benign diseases too, BFP is a new member of the carcinofetal protein family, which may be a useful marker in cancer pathophysiology.

Notes: The contributions of the internal modes of urea molecules, CH4N2O, to the vibration tensors of their atoms are calculated at 0, 100 and 300 K. The calculations are based on IR and Raman date from the literature, and the force constants determined are of the Urey-Bradley type. The temperature dependence of the vibration tensors of the internal modes (diagonal components) is represented graphically in the 0-300 K region (interpolation over nine calculated points); in this region it is weak. Since, for urea, there are vibration tensors available which were determined by neutron diffraction, the contributions of the internal modes could be estimated. On average over the C, N, O atoms it is 1.8% at 300 K, 4.4% at 98 K and 5.8% at 60 K; on average over the H atoms it is 22.6% at 300 K, 39.9% at 98 K and 46.3% at 60 K.

Notes: A lattice-parameter measurement technique using two lattice planes and monochromatic X-ray beams has been investigated. Accuracy, measurement correction and extensions of the technique are discussed. With this technique, the lattice parameters of Gd3Ga5O12, single crystals, grown under different conditions, have been measured to an accuracy of ±0.0003 Å. From the results obtained for Gd3Ga5O12, single crystals, it has been found that the technique has several advantages in comparison with conventional methods.

Notes: Summary The possible changes in the subarachnoid space after subarachnoid haemorrhage were studied in animals by using a scanning electron microscope (SEM). About 1 ml/kg of autogenous blood was injected intracisternally in 36 adult mongrel dogs to investigate changes in the subarachnoid space, over periods ranging from immediately after the injection to as long as 6 months. We have come to the conclusion that the injected blood disappears in about one to two weeks; the fibrosis or thickening of the arachnoid membrane appears in one to three weeks, and then returns to normal in a month in instances of rapid recovery, but there are some cases in which fibrosis persists for a long period and becomes chronic. The fact that an increase of fibrous tissue was found in the parietal region, where the injected blood had hardly reached, appears to indicate that the fibrosis is not always limited to the site of the haemorrhage but can occur in remote regions. We also discuss the usefulness of the SEM in the observation of the subarachnoid space, and the finding that vascular specimen preparations can be made by perfusing the brain with 2–10% phosphate-buffered formaldehyde solution.

Notes: Summary Scanning electron microscopic observations of the subarachnoid space were made in dogs focussing upon the fibre components in both the normal subarachnoid space and in areas of post-haemorrhagic fibrosis. It was concluded that the fibrous tissue originates from the arachnoid membrane itself, while organized haematoma is considered to form a component of the fibrosis. Perfusion of the subarachnoid space of dogs with a solution of 0.1% Toluidine Blue was also done. This showed that cerebrospinal fluid (CSF) is carried from the subarachnoid space directly to the dural sinuses through a fine string-like structure, which is conceivably one of the collateral CSF absorptive pathways.
