    Notes: Abstract The effects of acrylamide intoxication on the numbers of granule and Purkinje cells and the volume of Purkinje cell perikarya have been evaluated with stereological methods. The analysis was carried out in the cerebella of rats that had received a dose of 33.3 mg/kg acrylamide, twice a week, for 7.5 weeks. The total numbers of cerebellar granule and Purkinje cells were estimated using the optical fractionator and the mean volume of the Purkinje cell perikarya was estimated with the vertical rotator technique. The volumes of the molecular layer, the granular cell layer and the white matter were estimated using the Cavalieri principle. The mean weight of the cerebellum of the intoxicated rats was 7   % lower than that of the controls rats (2P = 0.001). The numbers of the Purkinje cells and granule cells were the same in both groups, but the mean volume of the perikarya of the Purkinje cells in the intoxicated rats was 10.5   % less than that of the control group (2P = 0.004). The volume of the granular cell layer was reduced by 15   % (2P = 0.006) but there were no differences in the volumes of the molecular layer and the white matter in the intoxicated and control animals.

    Notes: Modern diagnostic instrumentation produces a vast amount of data that often requires substantial analysis efforts. New methods are needed to improve the efficiency of the analysis process. Artificial neural networks have been applied to a variety of signal processing and image recognition problems. The feed-forward, back-propagation technique is well suited for the analysis of scientific laboratory data, which is viewed as a pattern-matching problem. We summarize the concepts and algorithms as implemented on a personal computer, and illustrate the method using a nonlocal thermodynamic equilibrium theoretical atomic physics model for k-shell x-ray spectroscopy of a high density, high temperature aluminum plasma. Extensions to other types of spectroscopy data analysis are discussed.

    Notes: Prospectively, serum levels of IgE, specific IgE antibodies (AB) to whole cow milk protein (CMP), bovine se-albumin, bovine immunoglobulin, bovine lactoferrin, bovine lactalbumin and beta-lactoglobulin (BLG), IgG and IgG subclass antibodies to ovalbumin (OA) and BLG, and IgG4 RAST to CMP (bovine whey) were measured in 39 infants with cow milk protein allergy (CMPA) at birth (cord blood), at time of diagnosis and before and after milk challenge at the age of 12 months. Immunological measurements were also undertaken in 33 control infants without CMPA at birth, at 6 months and at 18 months. At no time, were differences found between the levels of IgG and IgG subclass AB to OA and BLG in control versus infants with CMPA. In the 39 infants with CMPA no correlation was found between the levels of IgE, IgG and IgG subclass AB in cord blood and subsequent levels of these values, irrespective of the type of CMPA (IgE-mediated (CMA) or non-IgE-mediated (CMI)), and irrespective of whether remission had occurred. In cord blood 25/33 (76%) of the infants with CMPA had specific IgE-AB to one or more of the bovine milk proteins indicating a prenatal intrauterine sensitization to cow milk protein. At 6 months the frequency of specific IgE-AB to bovine milk proteins was significantly (p〈0.05) higher in infants with CMA versus CMI, and at 12 months total serum-IgE and the increase of these specific IGE-AB and RAST to CMP were significantly higher (p〈0.05) in infants with persistent CMA. From 6 to 12 months withholding milk resulted in a significant fall in specific IgE-AB to CMP, and IgG, IgG, and IgG4 anti-BLG followed by an increase after milk challenge. Decreasing levels of IgG anti-OA from birth to 6 months reflect passive maternal transfer of IgG through the placenta, and increasing levels of IgG anti-BLG, already from birth to 6 months, may represent an early exposure to CMP in all infants. Significantly higher levels (p〈0.05) of IgG anti-OA AB, IgG, and IgG4 anti-BLG AB were found in infants with persistent CMA, indicating a close relation between the synthesis of IgE and IgG and between IgE and IgG subclasses (IgG, and IgG4) in symptomatic cow milk-allergic individuals. Determination of IgG AB and IgG subclass AB (IgG, and IgG4) to BLG and bovine whey in cord blood appears unable to discern infants at high risk of development of CMPA. However, infants with persistent CMPA have an increased antibody response of specific IgE and IgG subclasses (IgG, and IgG4) to CMP exposure.

    Notes: Long-period equivalent scalar magnetotelluric impedance functions from 22 magnetic observatories distributed over the Earth's surface are inverted to test the hypothesis that there exists significant lateral heterogeneity in the electrical conductivity structure of the mid-mantle. Limited spaces of extremal models are examined, and strict bounds are placed on the range of permissible models. Joint D+ inversion of combined response functions from observatory pairs proves the requirement for global heterogeneity, while exploration of H+ layered-earth model space suggests systematic regional heterogeneity, indicating that enhanced upper mantle conductivity is found within the Pacific basin and Baikal Rift zone and is absent elsewhere. The results of these extremal inverse solutions are compared with the Backus–Gilbert-like Schmucker inversion and a heuristic inversion.

A total of 517 Vibrio anguillarum strains isolated from discased fish together with 14 V. anguillarum serogroup O2 and V. ordalii type strains were serotyped using the European scrotyping system. Marked species differences were recorded. In isolates from salmonids serovar O1 (70.2%) and O2 (20.2%) were dominantwhilst a minor proportion belonged to other serogroups or were non-typeable. Figures for turbot were similar to those from salmonids. In 32 isolates from sea bass, sea bream and mullet, most strains belonged to serogroup O1. while one was O2a. one O7, and the rest non-typeable In cod serovar O2 was dominant while only a minor proportion belonged to other serogroups or were non-typeable. The ecl isolates belonged equally to serovars O2 and O3. All O2 strain were subtyped with absorbed O2a and O2b antisera. O2a was dominant in all fish species. but in cod. the relative number of O2b isolates was considerably higher than in other fish species. The applicability of the European serotyping system is discussed and compared with other serotyping systems.

    Notes: Abstract. A collection of 130 strains of the bacterial fish pathogen Aeromonas salmonicida subsp. salmonicida isolated from diseased salmonids in Denmark, Norway, North America and Scotland has been characterized with regard to protein patterns, antibiotic resistance and exoprotease activity. Whole cell and outer membrane protein profiling could distinguish three different profiles in A. salmonicida. Eight outer membrane proteins were demonstrated (49, 40, 38, 37, 33, 31, 30 and 29 kDa). One protein profile was deficient in a 38 kDa outer membrane protein and instead contained an outer membrane protein of 37 kDa which was not detectable among the other protein profiles. Strains with the 37 kDa outer membrane protein showed multiple low-level antibiotic resistance towards cephalothin, penicillin, chloramp-henicol, tetracycline and quinolones. In addition, these strains were exoprotease deficient. Strains with the 37 kDa protein were unable to degrade cattle and trout serum proteins and displayed a delayed degradation of casein. Haemolysis on cattle blood agar plates was similarly delayed. In vivo examination of extracellular products from a normal protein profile strain and one with the 37 kDa outer membrane protein demonstrated major differences in pathological effects in rainbow trout. The strain possessing the 37 kDa outer membrane protein produced almost no pathological effects while the normal protein profile strain produced typical furuncles.

    Notes: This study presents a technique for measuring shapes of three dimensional surface cracks continuously during fatigue crack growth. The technique uses a laser interferometric system to measure crack compliance and a photomicroscopic system to measure surface crack length. Using a compliance expression for surface cracks, valid for the range of crack aspect ratios (a/c) of 0.2 to 2.0, aspect ratio calculations employing compliance and surface crack length measurements are demonstrated for cracks growing from EDM notches of different geometries (shallow or deep). The experimentally determined aspect ratio variations during cyclic crack growth are shown to be in good agreement with the expected variations in aspect ratio, predicted using the stress intensity factor equations for surface cracks. The effects of deviations in the compliance measurement location from the center of the surface crack due to assymetric crack growth are also accounted for through corrections of compliance measurements for crack-asymmetry. The fatigue crack growth rates of surface cracks, after incorporating the variations in aspect ratio in the calculations, agreed with the large-crack growth data for all crack geometries.
