    Notizen: With Staphylococcus xylosus FAX-1, metmyoglobin in MRS broth (pH 5.8) was found to undergo conversion to hexacoordinate nitric oxide (NO) complex of Fe(II) myoglobin. When the pH of the MRS culture containing myoglobin changed from 5.8 to 4.0, it affected the conversion from hexacoordinate to pentacoordinate NO complex of Fe(II) myoglobin. This conversion process was reversible. Salami without nitrite or nitrate addition was prepared by inoculating S. xylosus FAX-1, and pentacoordinate NO complex of Fe(II) myoglobin (nitrosylmyoglobin formed in cured meat) was formed in the salami.

    Notizen: The red pigment of Parma ham was compared with the myoglobin derivatives present in meat and meat products. Spectral patterns of 75% acetone extracts and electron spin resonance spectra from Parma ham differed from those of the myoglobin derivatives. Staphylococci isolated from Parma ham generated red myoglobin derivative from metmyoglobin. Model fermented sausage prepared by inoculation with the isolates developed a more desirable red color without nitrite or nitrate treatment. The red pigment of Parma ham and the model sausage appeared to be the same myoglobin derivative. The reddening of Parma ham is probably caused by the action of bacteria.

    Notizen: Chitosanase, an enzyme which hydrolyzes chitosan, isolated from Bacillus circulans MH-K1, was crystallized by a vapor-diffusion procedure at 293 K using ammonium sulfate as a precipitant. Rod-shaped colorless crystals, which grew to 0.05 × 0.15 × 1.2 mm within a week, belong to the orthorhombic system and the space group P2221 or P21212 with unit-cell dimensions of a = 43.3, b = 57.7, and c = 128.0 Å. The asymmetric unit is thought to contain one chitosanase molecule (29 024 Da). The crystals diffract X-rays to at least 2.3 Å resolution and are suitable for high-resolution X-ray structure analysis.

    Notizen: Summary Since D-serine occurs at high concentrations in mammalian forebrains, the brain D-serine content was analyzed in hyperkinetic and ataxic mutant mice as well as normal control mice in a search for a physiological role. The concentrations of free D-serine (nmol/g wet weight) were 392 ± 114 (mean ± S.D.), 43 ± 17 and 18 ± 8.4 in the cerebrum, brain stem and cerebellum of the BUS mouse, respectively; and 336 ± 93,58 ± 11 and 18 ± 8.5 in the cerebrum, brain stem and cerebellum of the Rolling mouse, respectively. These values were not significantly different from those for each control animal. The present results suggest that brain D-serine may not be a cause of the abnormal movements of the mutant mice. On the contrary, among many amino acids examined, tyrosine level was found to be lower in the brain stem of BUS mouse compared to the normal control animal by amino acid analysis.

    Notizen: γ-Hexachlorocyclohexane (γ-HCH; also called BHC or lindane) is one of the highly chlorinated pesticides which can cause serious environmental problems. Sphingomonas paucimobilis UT26 degrades γ-HCH under aerobic conditions. The unique degradation pathway of γ-HCH in UT26 is revealed. In the upstream pathway, γ-HCH is transformed to 2,5-dichlorohydroquinone (2,5-DCHQ) by two different dehalogenases (LinA and LinB) and one dehydrogenase (LinC) which are expressed constitutively. In the downstream pathway, 2,5-DCHQ is reductively dehalogenated, and then ring-cleaved by enzymes (LinD and LinE, respectively) whose expressions are regulated. We have cloned and sequenced five structural genes (linA, linB, linC, linD, and linE) directly involved in this degradation pathway. The linD and linE genes form an operon, and its expression is positively regulated by the LysR-type transcriptional regulator (LinR). The genes linA, linB, and linC are constitutively expressed, and are present separately from each other in the UT26 genome. Cell fractionation analysis, Western blotting, and immuno electron microscopy revealed that LinA and LinB are localized in the periplasmic space of UT26.

    Notizen: Abstract Oxidative stress is suggested as a significant causative factor forpathogenesis of neuronal degeneration on spinal cord of human ALS. Wemeasured some enzymic activities implicating neuronal degenerationprocess, such as cytochrome c oxidase (CO), superoxidedismutase (SOD), and transglutaminase (TG) in spinalcord of an animal model of ALS, motor neuron degeneration(Mnd) mouse, a mutant that exhibits progressivedegeneration of lower spinal neurons during developmental growth, andcompared them with age-matched control C57BL/6 mice. CO activity inMnd spinal cord decreased during early postnatal period, whileSOD activity reduced in later stage. In Mnd tissue, TG activityin lumbar cord was increasing during early stage, but tended to declinein later period gradually. These biochemical alterations became evidentprior to the appearance of clinical motor dysfunction which wereobserved in later stages of development in Mnd spinal cord.

    Notizen: Abstract A rare case of primary large cell carcinoma of the submandibular gland is reported. A 53-year-old man presented with a tumor in the left submandibular region extending to the supraclavicular fossa. After unsuccessful radiochemotherapy at another hospital, he was referred to our department for combined therapy of radiation, hyperthermia, and intraarterial anticancer drug infusion. Although the local tumor decreased in size after therapy, the patient died of respiratory insufficiency due to rapid progression of pulmonary metastases. Autopsy showed that tumor cells in both the submandibular gland and the lung were compatible with undifferentiated large cell carcinoma without tubular formation or laminar structure. The submandibular tumor was considered to be the primary site because cicatricial tissue surrounding the lesion suggested that had formed over a long period. The pulmonary lesions were considered to be metastases because necrosis and intravascular carcinoma cell embolism were noted. The above findings led to the final diagnosis of primary large cell carcinoma of the submandibular gland with pulmonary metastases. This condition has rarely been reported in the literature.

With the availability of a high-dose-rate (HDR) remote afterloading device, a Phase I/11 protocol was initiated at our institution to assess the toxicity and efficacy of HDR intracavitary brachytherapy, using molds, in the treatment of squamous cell carcinomas of the oral cavity. Methods. Eight patients with squamous cell carcinoma of the oral cavity were treated by the technique. The primary sites of the tumors were the buccal mucosa, oral floor, and gingiva. Two of the buccal mucosal cancers were located in the retromolar trigon. For each patient, a customized mould was fabricated, in which two to four afterloading catheters were placed for an 192Ir HDR source. Four to seven fractions of 3–4 Gy, 5 mm below the mould surface, were given following external radiation therapy of 40–60 Gy/2 Gy. The total dose of HDR brachytherapy ranged from 16 to 28 Gy. Results. Although a good initial complete response rate of 7/8 (88%) was achieved, there was local recurrence in four of these seven patients. Both of the retromolar trigon tumors showed marginal recurrence. No serious (e.g., ulcer or bone exposure) late radiation damage has been observed thus far in the follow up period of 15–57 months. Conclusions. High-dose-rate brachytherapy using the mold technique seems a safe and useful method for selected early and superficial oral cavity cancer. However, it is not indicated for thick tumors and/or tumors located in the retromolar trigon.
