    Notes: This paper reviews the principle of the phase-Doppler particle sizer by means of examples of the effects of varying the various experimental parameters. The examples are computed by means of a comprehensive numerical model of a complete phase-Doppler system including many practical aspects such as aperture size, polarization filters and detector properties. First the phase-Doppler principle is explained as a logical development of an interferometric measurement of the phase of the light scattered from a particle. Then the standard phase-Doppler analyzer (PDA) is decribed followed by some newer or less known variations of the principle intended to measure, e.g., in the backscatter direction, to size very small particles or to determine the refractive index of a particle. Finally, the trajectory problem in PDA measurements is briefly discussed.

    Notes: Light scattering by particles is often used to determine velocities or concentrations of particles in gaseous or liquid streams. Within the Lorenz-Mie theory, light scattering is well understood both for a single compact spherical particle and a single multilayered particle in a non-absorbing surrounding medium. However, in some cases of practical importance the Lorenz-Mie theory in its present form may fail to describe the scattering because the host medium is absorbing (e.g. water droplets in oil). In this case, a new treatment of the scattering theory is required. In previous work, solutions were obtained in the far-field of the scattering sphere. In this paper, a rigorous solution is derived from the calculation of the total absorption rate of the particle in the host medium, which is valid for all distances from the surface of the encapsulated particle. It is shown that it is necessary to consider finite sizes R of the integrating sphere when dealing with absorbing host media. Cross-sections are defined which are characteristic quantities not only for the particle, depending on the size of a conceptual sphere around the scatterer and the imaginary part of the refractive index of the host medium. The results obtained are discussed for the case of non-absorbing host media and in the far-field approximation. Some numerical examples are given which are also related to experimental results.

    Notes: After having recalled basic theoretical results concerning the extension of generalized Lorenz-Mie theory to the case of multilayered spheres, results connected with phase-Doppler anemometry are considered, showing the influence of Gaussian beam intensity profiles on the light scattering properties of these particles. Particular emphasis is placed on the case of water-coated carbon core particles, for which the possibility of obtaining simultaneous size measurements of the core and outer diameters is discussed. The sensitivity of the technique to particles with a refractive index profile is also considered, showing that this technique is more sensitive to changes in the average refractive index of the particles than to refractive index profiles, such as produced by high pressure and temperature stresses, at least for the studied geometry.

    Notes: Microcontamination of product surfaces by deposited particles is an important problem in clean technologies. A most sensitive product to contamination by particles is a wafer during chip production. Therefore, methods for monitoring particle deposition on wafer surfaces have been developed in the last decade. A wafer with an unstructured and reflecting surface is inserted into the process equipment. After some time, depending on the process, this wafer is removed from the process equipment and is analysed with respect to the number of deposited particles using a wafer scanner. However, in situ particle detection in a process chamber is not possible with this technique. This would be possible if, instead of a monitor wafer, a transparent glass plate is mounted, e.g. in the housing of the process equipment. Then the illuminating and scattered light detection equipment can be mounted outside the process equipment. Since both the illuminating laser beam and the scattered light have to be transmitted through the glass plate, losses will occur, which will reduce the lower limit of detection with respect to particle size. In this article we estimate the detection possibilities theoretically and experimentally.A simple model based on Mie and vector scattering theory has been developed to describe the light-scattering behavior of a single spherical particle on a glass plate with random surface irregularities. The scattered light of individual particles of four particle sizes (1.03, 1.6, 2.92 and 4.23 μm) on the same glass surface and from the uncontaminated area of the glass plate was measured for unpolarized and normally incident light. The values of the scattered light from this model were compared with the experimental results. The comparison shows a reasonable agreement of the angular distribution of the scattered light. The developed model is used to predict the lower limit of detection for particles on a transparent surface. The theoretical estimations show that it should be possible to detect particles of a diameter down to 0.2 μm with the described measurement technique.

    Notes: The ability to routinely make simultaneous measurements of the size and velocity of individual spherical particles is identified as a crucial component of advances in the science and technology of multiphase systems and processes. Since it is the dual beam interferometer identified here as the phase-Doppler particle-sizing velocimeter (PD) that has provided this capability, and since 1995 marks twenty years from a seminal paper by Durst and Zaré, a historical review is warranted and is presented. The roots and context of the development of PD are traced from the days of laser Doppler velocimetry (LDV) in the early 1970's through work culminating at several laboratories in the mid 1980's. A number of important contributions and the engineers and scientists responsible for those contributions are recognized.

    Notes: The Scattering of focused laser beams by arbitrarily shaped dielectric bodies is investigated theoretically. The beam description is based on Davis third-order beam approximation for the field components. The scattering problem can be solved on a spherical basis by the extended boundary condition method or by the so-called modified version of the extended boundary condition method. For spheroidal particles with small eccentricities, a perturbation technique for the internal subproblem of the extended boundary condition method is described. This procedure is applied to analyse the influence of the particle shape and orientation on the response of a phase-Doppler system.

    Notes: A new technique for the calculation of the optical properties of agglomerated spherical primary particles is presented. By means of the so-called virtual refractive index, the multiple scattering within the agglomerate is decribed by a modified Mie solution, thus avoiding restrictions with respect to the primary particle size. An additional benefit of the method is the small numerical expense required, permitting practical use for complex agglomerate structures such as real combustion aerosols. Utilizing this method, attention is focused on the calculation of the extinction cross-section of real combustion aerosols in order to discuss the results of multiple-wavelength extinction measurements within combustion systems. It is found that approximately the primary particle diameter is determined. Furthermore, the error that occurs can be avoided by means of the method presented without a detailed knowledge of the agglomerate size. Thus, the present work eliminates some current uncertainties inherent in optical combustion aerosol characterization.

    Notes: The principle of the dual burst technique (DBT) based on phase-Doppler anemometry (PDA) is proposed for simultaneous particle refractive index, size and velocity measurements. This technique used the trajectory effects in PDA systems to separate the two contributions of the different scattering processes. In the case of forward scattering and refracting particles, it is shown that from the phase of the reflected contribution, the particle diameter can be deduced, whereas from the refracted contribution the particle refractive index and velocity can be obtained. Furthermore, the intensity ratio of these two scattering processes can be used for absorption measurements. Simulations based on generalized Lorenz-Mie theory and experimental tests using monodispersed droplets of different refractive indices and absorption coefficients have validated this technique.

    Notes: Rainbow refractometry allows the determination of the refractive indices of droplets if a homogeneous refractive index can be assumed throughout the whole droplet. The influence of gradients on the angular position of the rainbow was examined for refractive index distributions with constant gradients and for distributions as they occur during cooling, heating and burning of droplets. Geometrical optics were used to determine rainbow positions. For a given value of refractive index at the surface, the rainbow position is in many cases a measure for the gradient itself. It was found that information obtained for constant gradients can often be used in cases with more complex distributions, which are found, for instance, for droplets undergoing heat-transfer processes. Significant errors may be obtained only for a very short initial phase of the process. Experimental results obtained with monosized droplet streams, which confirm theoretical predictions of the influence of gradients on the rainbow position, are presented.

This paper investigates sources of measurement error in each of these quantities and provides means to eliminates or minimize these errors. A new PDA configuration, termed dual-mode PDA, is introduced as an implementation of these suggestions. Example measurements illustrate the advantages of the system over conventional arrangements.

For solving the RTE with characteristics depending on the propagation direction of the radiation beam, an algorithm is developed based on the method of layer doubling. Calculations are presented of angle correlations for intensities of radiation in a layer with Fresnel reflection on the boundaries, and reflection and transmission coefficients.

    Notes: Catalyst surface characterizations have been carried out to investigate the role of dispersion on catalyst activity and to probe the occurrence of oscillations in coking levels with cycle number generally observed during multiple deactivation and regeneration schemes. The titrations were done, cycle by cycle, at 430°C after oxidation and at the same temperature (430°C) after reduction at 500°C. Results show the usually observed trend - that the dispersions after oxidation are higher than those after reduction. The average decline in dispersion from oxidation to reduction was calculated to be 39.25%. It was observed that the cycles with high toxic coke removal were characterised by high deactivation times. The deactivation times were still high even for cycles subsequent to those with low dispersion. At high dispersions the catalyst had short deactivation times, that is the small crystallites deactivate faster than large ones. The nature of reducebale coke and the efficiency of its removal is a much more determinant factor of catalyst activity than the level of metal disperision. Thus prolonged toxic coke reduction at the high temperature of 500°C, though resulting in an apparent lowering of dispersion, does not affect the quality of the catalyst. The dispersion before reduction could be retained on oxidation. Hence reduction at 500°C did not introduce sintering.

    Notes: Reasons are given why the axial dispersion in a gas flowing through a packed bed may be influenced by the elasticity - or compressibility - of the fluid. To support this hypothesis, experiments have been done in a packed column at pressures from 0.13 to 2.0 MPa. The elasticity E of a gas is proportional to the pressure P and the compressibility to 1/P. The axial dispersion coefficients as determined were found to be a function of the pressure in the packed bed in the turbulent flow region of 3 〈 Rep 〈 150 if the Bodenstein number is plotted as a function of the particle Reynolds number. This is shown to be an artifact. The pressure influence is eliminated, if Bom, ax is plotted versus the ratio of the kinetic forces over the elastic forces ϱu2/E. Regrettably, Bom, ax seems to be independent of ϱu2/E. For the moment we only can conclude that Bom, ax in the turbulent region is a unique function of the velocity of the gas which flows through the packed bed. Although the fact that a constant Bo value is obtained when plotted against ϱu2/E, the experimental results are so intriguing we wanted to make them public already now. The experimental work proceeds.

    Notes: A unified gas/ solid reaction model has been formulated for multi reactions with multi gas and solid species. The transient formulation has been followed by finite-difference method and subsequent solution via an iterative procedure.     Notes: Calcination and carbonation behaviour of dolomite has been studied in a pressurised thermo balance at pressures in the range of 1.1 to 2.3 MPa and for temperatures ranging from 730 to 840°C. The atmosphere consisted of nitrogen containing up to 20 vol.-% carbon dioxide. The calcination of dolomite has been studied as a function of temperature, particle size, and Pco2. An investigation concerning possible mass transfer restrictions for the experimental system and a model investigation of the calcination rate in CO2 atmosphere has been made. Further, the effect of calcination on the BET surface and the BET surface including the surface structure due to successive temperature cycles has been studied. The temperature for the initialisation of calcination of dolomite in CO2 atmosphere was independent of both Pco2 and the particle size. No conclusive indications on mass transfer restrictions have been detected for the experimental system. A significant difference in calcination rate as a function of particle size existed for decomposition in CO2 atmosphere. A model for homogeneously progressing chemical reaction parallel with a shrinking core chemical reaction together with a mass transfer control mechanism was found to describe the calcination rate in CO2 atmosphere most accurately.

The evaluation algorithms of fluctuating transmission signals are well known in the case of single particle measurement. By evaluating fluctuating transmission signals the particle characterization is also possible at high particle concentrations (0.01 to 30 vol.-%). In comparison with conventional photometers a significant gain of information can be received. Fluctuating signals are caused by the statistical probability of a limited number of particles being present in a defined measuring volume. The evaluation of such signals allows the determination of particle concentration, particle size of monodisperse particles (extinction diameter) and information about structure of agglomerates independent from each other. Mathematical fundamentals and practical ways to measure these parameters are shown. Experimental results are examplarily presented for monodisperse, polydisperse and agglomerated suspensions.

    Notes: For highly viscous products, multiple shaft agitators represent an interesting alternative to expensive special mixers. A triple shaft agitator with a volume of 250 liters is used to determine the powder consumption of the impellers and its relationship with the process parameters. The result shows that the power consumption of a single impeller running in multiple shaft mode is significantly smaller than of one running in single shaft mode. But the power required for the whole multiple shaft machine is much higher than that for a single shaft machine. A model is presented for detailed description of the interactions between the individual agitators in multiple shaft mode.

    Notes: A Comparative study of different Lewis acids (metal chlorides of valences, I, II, III, IV) has been made. These catalysts are used in the synthesis of a high molecular weight ester in a stirred tank reactor using octanoic acid and n-octyl alcohol as reactants. It was found that the best efficiency of the formatted ester (n-octyl octanoate) was obtained with ferric chloride. This could be explained by the high electronegativity and the existence of free d orbitals in the Lewis acid.

    Notes: The results of recent researches on twin-fluid atomization are briefly reviewed, with particular emphasis on the effects of air and liquid properties on mean drop size. It is stressed that no single equation for the mean drop sizes produced in twin-fluid atomization can satisfactorily correlate all the experimental data from any given atomizer. This is because for all types of atomizers the mechanism of liquid breakup changes from one basic mode to another with changes in atomizer operating conditions and/or liquid properties. These different modes of atomization exhibit different dependencies on atomizer dimensions and various physical properties such as liquid viscosity and air density. Equations for estimating the mean drop sizes produced in the various modes of twin-fluid atomization are presented and discussed.

    Notes: The avalanching behaviour of two coal types was determined, one of good and the other of poor handleability characteristics. This revealed significant differences in the nature of flow of the coals. Analysis of the strange attractors of the weight series of avalanches enabled quanitification of this difference, and the establishment of practical, relative criteria for the measurement of coal handleability. These criteria will now enable a structured study of the variables that affect handleability, for example proportion of fines and moisture content, and industrial consumers of coal to specify minimum acceptable limits for its transport characteristics.

    Notes: A concern about all reinforcing fibers is the extent to which they are respirable. The degree of respirability of a particle is frequently estimated in terms of its aerodynamic equivalent diameter being less than a certain value (typically 3.5 to 7 μ). However, the aerodynamic equivalent diameter, like the Stokes diameter, is only defined exactly for spheres, which is far from the shape of acicular fibers. Thus, it is useful to be able to calculate, based on actual fiber diameter and aspect ratio, the effective Stokes and aerodynamic equivalent diameters of various fibers. The present paper derives simple expression for relating the two effective diameters to actual fiber dimensions.Calculated results are compared with experimental data for glass fiber and three fractions of phosphate fiber [1], which is a short, reinforcing inorganic fiber. Agreement is seen to be good. Phosphate fiber was fractionated using a sedimentation technique described in the paper. The resulting fractions were characterized by semi-manual measurements of micrographic dimensions and compared with results obtained from particle size instruments based on sedimentation principles. Based on these results, a large weight fraction of phosphate fiber is predicted not to be respirable.

    Notes: When properly measured and interpreted angle of repose (AOR) can be a useful method of characterising a powder. Unfortunately, a multitude of definitions and measuring equipment is used. A review of the inherent problems is presented in the first part of this paper.The second part of the paper describes the improvements made to a measuring device developed originally for measuring the angle of repose of soda ash, so that a wider range of powders can be tested. In the improved design, the tendency of cohesive and semi-cohesive powders to block the device's funnel has been eliminated.A total of 73 powders consisting of four different materials, and covering Groups A, A/C and C of Geldart's classification, have been tested using the new equipment. The results show that AOR increases systematically with decrease in mean particle size. Combination of AOR measured in the new device with aerated bulk density provides a quick, sensitive and effective method for characterising a wide range of powders.

    Notes: This paper gives a theoretical basis for calculating Fraunhofer diffraction patterns of arbitrary polyhedron particles. It is shown that this solution can be used for calculating a scatter matrix adapted to the particle shape in a straightforward manner. Some simulations were made to show the difference between the size distribution by volume obtained with a scatter matrix for spheres and with that for the appropriate shape. Finally, some experimentally measured signatures from platelets and rods are evaluated in order to show that the spherical equivalent diameter could be accurately retrieved.

    Notes: Many different methods for size measurement are known and since they differ in their physical principles, different results are also obtained. In the recent past, there were rapid developments in new measurement techniques and one can now quickly and routinely determine particle sizes in the very fine range. Smaller particle sizes, on the other hand, tend to increase the probability of agglomeration. The measurement of the particle sizes for magnetic materials is not well understood and this paper reports results regarding the size determination of very fine magnetic materials. Because of their dipole moment, these particles tend to agglomerate even more, which in turn causes certain difficulties during the measurements. Wet and dry laser measurement systems were compared and also magnetic materials with different permeabilities in order to establish the influence of individual factors on the measurement process and on the accuracy of the results obtained.

    Notes: Within the past 20 years, particle size analysis with laser diffraction (LD) has been subject to rapid development, extending the size range stepwise from 1-200 μm to about 0.1-3500 μm. The limits of LD are discussed in terms of light sources, the influence of the beam diameter, special Fourier optics and a new detector design.It is shown that the size range is not only restricted by the wavelength of the laser and the transmission limits of the medium. Its extension is mainly related to improvements in the measurement of the angular intensity distribution. Influences from stability and flow dominate on the coarse side of the measuring range. On the fine side, the spatial extension of aerosols and the resulting demand for extended working distances can be covered only in a parallel laser beam. Extended Fourier optics in combination with an adapatable beam expansion technique and a detector with virtual borders between semicircular elements overcome the existing limits and extend the size range to a lower limit of about 0.05 μm and an upper limit above 10 mm.The sensititivity limit of LD is approaching that of single particle counting techniques. For medical spray and inhaler applications, a 0.1% optical concentration can be converted to particle size distributions even for time-resolved analyses with sample intervals of a few milliseconds.The reproducibility of the sensor, with a standard deviation typically much less than 0.5%, is no longer the limiting factor. The reproducibility of the results is mainly dominated by the reproducibility of sampling, sample splitting, dispersion and the contamination of the optical path. The latter can be improved by the control of flow, especially for in-line and inhaler applications.

    Notes: Dynamic light scattering has become a standard technique of investigating colloidal suspensions and polymer solutions. The experimental field autocorrelation function ĝ1 (t) can often be well modelled by a Laplace transform relating ĝ1 (t) to a distribution of decay times A(τ). In simple systems A(τ) can usually be directly related to a distribution of molecular weights, particle sizes, diffusion coefficients or other physically relevant quantities, With constrained regularization methods, the parameter-free estimation of A(τ) has become straightforward. In complex systems, the resulting A(τ) may contain several components the identification of which is not always obvious. The problem often originates in a superposition of diffusive and angle-independent components that have different variations of their respective decay times with the scattering vector. A method is presented based on a simultaneous fit of several autocorrelation functions measured at several different scattering angles, which, using simple and reasonable assumptions, yields a robust analysis of the spectra of decay times. The application of the method is illustrated on simulated autocorrelation functions and also on real experimental data obtained on a variety of different polymer systems.

    Notes: A light-scattering instrument that is normally used for particle size analysis was applied to obtain information about particle shape. Micro-cuboids of dimensions a × b × c in the range between 10 and 1000 μm, which can be accurately manufactured employing special technology, were used as labelled samples for the determination of 3-D features. Particle projections at different orientations due to turbulent motion were measured sequentially. The laser diffraction of cuboids rotating in a turbulent flow is described theoretically using a simplified model, i.e. rotation of a rectangle. The change in the projection areas of rotating cuboids is connected with their rotational velocity. The power spectrum density of rotational velocity of cuboids and spheres was determined and the influence of particle shape on the power spectrum density established. The frequency distribution of projection sizes was found to be suitable for particle shape estimation.

    Notes: It is well established that particle size and shape substantially influence the bulk properties of powdered materials. Although these characteristics are closely interrelated, the tendency has been to analyse the particle size independently from particle shape. The aim of this work was the assessment of particle shape through particle sizing data. For this purpose, three different particle shape materials - glass beads (spherical), crushed glass (moderately irregular) and mica (lamellar) - were tested, employing four particle sizers. The shape factor selected was the Wadell's sphericity, which was evaluated utilizing distinct equations. The comparison of the results reveals problems arising from the use of approximations rather than more fundamental equations.

    Notes: A phenomenological model with 12 parameters for rotary drum granulation with the help of material balance equations taking into account the growth and death functions was developed. Experiments were conducted and the data were used to obtain model parameters with reference to the operating conditions and for validation of the model. It was concluded that the proposed model is adequate for the representation of dynamics and hence this model can be recommended for dynamic simulation leading to improved control of granulation circuits.

    Notes: A novel technique to determine the size of particles suspended in a stirred vessel is investigated. The method uses microphotography to obtain a still image of the particles in situ. The equivalent circular diameter of the particles is obtained from the photographs using digital image analysis. The particles used for the test were certified particle size standards of a very small tolerance in diameter deviation. The size comparison was determined as a percent error between the measured particle diameter (equivalent circular diameter) and the diameter established by the particle manufacturer. To determine the limiting ranges of particle size and concentrations, spherical particles in a size range of approximately 1 to 10.0 μm (microns) in diameter were employed in the test. Concentrations varied from 0.00005% to 0.1% (mass basis). For each particle size, an aqueous solution of the particles was prepared at the desired concentration and placed in a stirred vessel. Photographs of the solutions were made at 25.5x, 40x, 60x and 80x magnification. For all sizes of particles, the entire range of concentration was examined.The results indicate a minimum size resolution of approximately 3μm, in a corresponding concentration range of 0.0001% to 0.05%. Similar limits on concentration were found for the larger diameter particles, although a true upper limit for the largest particle sizes was not established. The functional concentration range appears to shift towards higher mass concentrations with increasing particle size. For 2.92 μm diameter particles, the error in size measurement was found to be below 10% for a concentration range of 0.0005% to 0.05%. Additionally, a relationship between turbidity and the error was seen at low particle concentration levels where decreasing levels of turbidity generated increasing levels of error.

    Notes: A new phase Doppler anemometry (PDA) signal processing method based on a Hilbert transform algorithm is introduced and analysed. By generating a 90° phase-shifted burst signal in the time domain, the envelope of the Doppler burst can be determined. In addition, this envelope is approximated by a Gaussian exponential function. The difference of the maxima of these Gaussian approximations for two related PDA bursts gives an estimate of the time difference between these time shifted signals. With the introduction of this estimation method, the restriction to the [0,360°] interval resulting from conventional signal analysis may be avoided in many cases. To investigate the dependence on SNR, burst position, burst frequency and sampling rate, results of computer simulations are presented. The feasibility of the method is demonstrated briefly by experimental results. Phase differences of more than 2000° arising from the measurement of monodisperse droplets by a conventional PDA setup could be determined.

    Notes: Using a monodisperse poly(methyl methacrylate) dispersion it was shown that light reflection at the sample cuvette walls may greatly influence the results of both static (SLS) and dynamic (DLS) light scattering experiments. Considering SLS, this reflection phenomenon mostly causes an overestimation of the scattered intensity at high scattering angles, which may give rise to the emergence of an additional, artificial peak in the lower region of the particle size distribution. On the other hand, the influence of reflection on DLS experiments was shown to be particularly important in the upper region of the particle size distribution. The experimentally observed phenomena were explained from basic principles of both particle sizing methods. Finally, it was shown that the disturbing effect of reflection could be avoided by modifying either the hardware or the software of the SLS and DLS techniques.

    Notes: An instrument is proposed for the visualisation and measurement of mixtures of small and large particles. The original concept was to use spectral extinction for sub-micron particles and scattering patterns for larger sizes. However, the former method is sensitive to the refractive index, which may be unknown or vary with wavelength in an unknown way. The experiments demonstrated this to be a problem. The forward scatter, on the other hand, is not so sensitive to this. It is shown here that the combination of scattered intensity as a function of both angle and Wavelength provides scatter diagrams for which there is good agreement between theory and experiment. The influence of quite small volume fractions of particles as small as 0.3μm mixed with 90μ particles is easily seen. The scatter diagrams suggest the use of image analysis for size measurement and quality control.

    Notes: A planar phase Doppler system is used to measure submicron droplets generated by an electrospray. Measured drop dia-meters are correlated with the liquid properties and the condition for transition of the spray from the single-jet mode to the multi-jet mode is introduced. In another set of measurements using a standard phase Doppler system, combined size and velocity data are employed to deduce the drag force on the drops. In a situation where the drag force is balanced primarily by the electric force, the phase Doppler measurements allow to estimate the power-law relationship between the charge on a drop and its diameter and hence, many provide insights into the underlying atomization mechanisms.

    Notes: Only a few optical techniques are available to measure simultaneously the velocity and size of coarse particles dispersed in a fluid. Since particles are randomly shaped with a rough surface and inhomogeneous with respect to the refractive index, phase-based sizing techniques are not reliable. The applied optical sizing technique is based on light intensity measurement of a scattering particle crossing a top-hat laser beam. This optical set-up was first described by Gréhan et al. In the range of geometrical optics, the sampled light at the receiving system is proportional to the square of the equivalent particle diameter within the diameter range 20-700 μmVelocity information is obtained by using a smaller concentric laser Doppler probe volume. Coincidence of the signals is guaranteed by the optical set-up, since the smaller probe volume diameter defines the triggering event. To demonstrate the performance of the system, some diameter distributions measured with the top-hat beam system are compared with results for a commercial laser diffraction system for different materials. The system is applied to obtain size-correlated velocity information of a particle-laden two-phase flow at a single-sided backward-facing step.

    Notes: Imaging techniques in particle sizing have always been important. Nevertheless, owing to the progress in CCD cameras, these techniques have found new potential which strongly depends on the capability to describe the image formation. Based on a combination of the generalized Lorenz-Mie theory and the diffraction theory, the image characteristics are described, taking into account the beam size, particle location, direction of observation and collecting optics characteristics. In the case of forward detection, a finite coherence is introduced and the results are compared with those obtained by a convolution approach, with the point spread function assumed to be Gaussian. Then some experimental results are displayed which underline the improvement achieved in particle size measurement.

    Notes: A Diesel spray injected into a high-pressure test chamber was investigated with two different holographic techniques. The usual off-axis recording geometry was used to investigate the very early beginning of the injection. Double pulsing of the recording laser facilitated velocity measurements. By using two different reference angles for the two recording the two images can be separated upon reconstruction of the holograms. Thus superior image quality could be achieved. Processes leading to jet formation could be identified: the spray tip is periodically axially decelerated and radially accelerated. Consequently, liquid moves at the tip off axis. The spray consists of fast central region surrounded by a slower outer jet region. The outer jet region exhibits periodic droplet concentration fluctuations which are interpreted as a result of the periodic processes at the spray tip. Some high concentration regions in the outer jet region were investigated using a dark-field holographic technique. This technique permits single droplet velocity measurements and local droplet concentration determination at extremely high droplet concentrations. The most important result of these investigations is that no correlation between the local droplet concentration and the droplet velocity could be found.

    Notes: In Saccharomyces cerevisiae, the structural genes PDC1, PDC5 and PDC6 each encode an active pyruvate decarboxylase. Replacement mutations in these genes were introduced in a homothallic wild-type strain, using the dominant marker genes APT1 and Tn5ble. A pyruvate-decarboxylase-negative (Pdc-) mutant lacking all three PDC genes exhibited a three-fold lower growth rate in complex medium with glucose than the isogenic wild-type strain. Growth in batch cultures on complex and defined media with ethanol was not impaired in Pdc- strains. Furthermore, in ethanol-limited chemostat cultures, the biomass yield of Pdc- and wild-type S. cerevisiae were identical. However, Pdc- S. cerevisiae was unable to grow in batch cultures on a defined mineral medium with glucose as the sole carbon source. When aerobic, ethanol-limited chemostat cultures (D = 0·10 h-1) were switched to a feed containing glucose as the sole carbon source, growth ceased after approximately 4 h and, consequently, the cultures washed out. The mutant was, however, able to grow in chemostat cultures on mixtures of glucose and small amounts of ethanol or acetate (5% on a carbon basis). No growth was observed when such cultures were used to inoculate batch cultures on glucose. Furthermore, when the mixed-substrate cultures were switched to a feed containing glucose as the sole carbon source, wash-out occurred. It is concluded that the mitochondrial pyruvate dehydrogenase complex cannot function as the sole source of acetyl-CoA during growth of S. cerevisiae on glucose, neither in batch cultures nor in glucose-limited chemostat cultures.

    Notes: We have investigated the effects on Saccharomyces cerevisiae of a novel antitumour agent (FCE24517 or Tallimustine) which causes selective alkylations to adenines in the minor groove of DNA. Tallimustine, added to wild-type cells for short periods, reduced the growth rate and increased the percentage of budded cells and delayed the cell cycle in the late S+G2+M phases. In the rad9Δ null mutant cells, Tallimustine treatment did not affect growth rate and the percentage of budded cells but greatly reduced cell viability compared to isogenic cells. Consistent with a role of RAD9 in inducing a transient delay in G2 phase which preserves cell viability, the potent cytotoxic effect of the drug on rad9Δ cells was alleviated by treatment with nocodazole. Tallimustine was also found to delay the resumption from G1 arrest of wild-type but not of rad9Δ cells. These data indicate that the effects of Tallimustine on cell cycle progression in yeast are mediated by the RAD9 gene product. From our data it appears that yeast could be a valuable model system to study the mode of action of this alkylating drug and of minor groove alkylators in general.

This map has been constructed by restriction analysis of a clone from a YCp50 genomic library and by use of pre-existing and new sequence data from this region.     Notes: A sequence of 31431 bp located on the left arm of chromosome (chr.) XIV from Saccharomyces cerevisiae was analysed. A total of 18 open reading frames (ORFs) could be identified. Twelve ORFs are new, two of which are most likely ribosomal protein genes, leaving ten ORFs of unknown function. Nine of the 18 ORFs show either at least 20% overall amino acid identity or significant regional homology to other S. cerevisiae ORFs. Additionally, six of these nine ORFs have homologues of similar size and the same transcriptional orientation within a stretch of 50 kb on chromosome IX. The degree of homology ranges from 90% overall identity to 23% in 375 amino acids. The homologues on chromosome IX are grouped in two blocks that are separated by relatively long ORFs. This is the first example of a multi-gene duplication in S. cerevisiae not linked to a centromere or subtelomere region. The sequence has been deposited in the EMBL data library under Accession Number X86470.

    Notes: Genomic comparison of two sibling yeast species, Saccharomyces bayanus and Saccharomyces cerevisiae, was performed by Southern blot analysis with various S. cerevisiae gene probes following electrophoretic karyotyping. Fifteen genes on chromosome IV of S. cerevisiae were examined and classified into two groups. Gene probes of CEN4 and TRP1, as well as six other genes located on the left arm of the chromosome hybridized to a 1100-kb chromosome of S. bayanus that is smaller than chromosome IV of S. cerevisiae. On the other hand, probes of seven genes located on the right arm of chromosome IV hybridized to a 1350-kb chromosome that is homeologous to chromosome IV, judging from its size. Two genes located on the left arm of chromosome II hybridized to the 1350-kb chromosome, while four genes on the right arm hybridized to the 1100-kb chromosome. These pieces of evidence indicate that chromosomes II and IV of S. cerevisiae are rearranged into 1350-kb and 1100-kb chromosomes in S. bayanus. Furthermore, it is suggested that chromosome XV is rearranged into two chromosomes (800 and 850 kb in size) in S. bayanus. The translocation points of chromosomes II and IV were delimited using S. cerevisiae prime clone membranes. The results indicated that the translocation points are located close to the FUR4 locus on chromosome II and close to the RAD57 locus on chromosome IV.

    Notes: The vacuolar aspartyl protease proteinase A (PrA) of Saccharomyces cerevisiae is encoded as a preproenzyme by the PEP4 gene and transported to the vacuole via the secretory route. Upon arrival of the proenzyme proPrA to the vacuole, active mature 42 kDa PrA is generated by specific proteolysis involving the vacuolar endoprotease proteinase B (PrB). Vacuolar activation of proPrA can also take place in mutants lacking PrB activity (prb1). Here an active 43 kDa species termed pseudoPrA is formed, probably by an autocatalytic process. When the PEP4 gene is overexpressed in wild-type cells, mature PrA can be found in the growth medium. We have found that prb1 strains overexpressing PEP4 can form pseudoPrA extracellularly. N-terminal amino acid sequence determination of extracellular, as well as vacuolar pseudoPrA showed that it contains nine amino acids of the propeptide, indicating a cleavage between Phe67 and Ser68 of the preproenzyme. This cleavage site is in accordance with the known substrate preference for PrA, supporting the notion that pseudoPrA is formed by autoactivation. When a multicopy PEP4 transformant of a prb1 mutant was grown in the presence of the aspartyl protease inhibitor pepstatin A, a significant level of proPrA was found in the growth medium. Our analyses show that overexpression of PEP4 leads to the secretion of proPrA to the growth medium where the zymogen is converted to pseudoPrA or mature PrA in a manner similar to the vacuolar processing reactions. Amino acid sequencing of secreted proPrA confirmed the predicted cleavage by signal peptidase between Ala22 and Lys23 of the preproenzyme.

All conventional actins that have been identified so far are 374-376 amino acids in size and exhibit at least 70% amino acid sequence identity when compared with one another. In the yeast Saccharomyces cerevisiae, one conventional actin gene ACT1 and three so-called actin-related genes, ACT2, ACT3 and ACT5, have been identified. We report here the discovery of a new actin-related gene in this organism, which we have named ACT4. The deduced protein, Act4, of 449 amino acids, exhibits only 33·4%, 26·7%, 23·4% and 29·2% identity to Act1, Act2, Act3 and Act5, respectively. In contrast, it is 68·4% identical to the product of the Schizosaccharomyces pombe Act2 gene and has a similar level of identity to other Sch. pombe Act2 homologues. This places Act4 in the Arp3 family of actin-related proteins. ACT4 gene disruption and tetrad analysis demonstrate that this gene is essential for the vegetative growth of yeast cells. The act4 mutants exhibit heterogenous morphological phenotypes. We hypothesize that Act4 may have multiple roles in the cell cycle.     Notes: We compared the ability of signal sequences from various Bacillus or yeast secreted proteins to direct Bacillus subtilis levansucrase into the secretion pathway of the yeast Saccharomyces cerevisiae. The efficiency of these sequences correlated with the overall hydrophobicity of their h-domain and was independent of their origin. Furthermore, the net charge of the proximal protein sequence downstream from the signal sequence contributed to the competence of the heterologous proteins to be secreted by yeast. Modification of this net charge allowed the protein to be translocated under the control of the yeast invertase signal sequence. Moreover, glycosylation of levansucrase did not modify significantly the fructosyl polymerase activity.

    Notes: A temperature-sensitive mutation (act1-1) in the essential actin gene of Saccharomyces cerevisiae can be suppressed by mutations in the SAC3 gene. A DNA fragment containing the SAC3 gene was sequenced. SAC3 codes for a 150 kDa hydrophillic protein which does not show any significant similarities with other proteins in the databases. Sac3 therefore is a novel yeast protein. A nuclear localization of Sac3 is suggested by the presence of a putative nuclear localization signal in the Sac3 sequence. A SAC3 disruption mutation was constructed. SAC3 disruption mutants were viable but grew more slowly and were larger than wild-type cells. In contrast to the sac3-1 mutation, the SAC3 disruption was not able to suppress the temperature sensitivity and the osmosensitivity of the act1-1 mutant. This demonstrates that act1-1 suppression by sac3-1 is not the result of a simple loss of SAC3 function. Furthermore, we examined the act1-1 and the sac3 mutants for defects in polarized cell growth by FITC-Concanavalin A (Con A)-labelling. The sac3 mutants showed a normal ConA-labelling pattern. In the act1-1 mutant, however, upon shift to non-permissive temperature, newly synthesized cell wall material, instead of being directed towards the bud, was deposited at discrete spots in the mother cell.

    Notes: The authors of the first yeast chromosome sequence defined a minimum threshold requirement of 100 codons, above which an open reading frame (ORF) is retained as a putative coding sequence. However, at least 58 yeast genes shorter than 100 codons have an assigned protein function. Therefore, the yeast genome may contain other tiny but functionally important genes that are discarded from analyses by this simple filtering rule.We have established discriminant functions from the in-phase hexamer frequencies of functional genes and of simulated ORFs derived from a stationary Markov chain model. Fifty-two out of the 58 genes were recognized as coding ORFs by our discriminating method. The test was also applied to all the small ORFs (36 to 100 codons) found in the intergenic regions of published chromosomes. It retained 140 new potential tiny coding sequences, among which we identified seven new genes by similarity searches. Our method, used conjointly with similarity searches, can also highlight sequencing errors resulting from the disruption of the coding frame of longer ORFs. This method, by its ability to detect potential coding ORFs, can be a very useful tool for functional analysis.

    Notes: Starvation for nitrogen in the absence of a fermentable carbon source causes diploid Saccharomyces cerevisiae cells to leave vegetative growth, enter meiosis, and sporulate; the former nutritional condition also induces expression of the YVH1 gene that encodes a protein phosphatase. This correlation prompted us to determine whether the Yvh1p phosphatase was a participant in the network that controls the onset of meiosis and sporulation. We found that expression of the IME2 gene, encoding a protein kinase homologue required for meiosis- and sporulation-specific gene expression, is decreased in a yvh1 disrupted strain. We also observed a decrease, albeit a smaller one, in the expression of IME1 which encodes an activator protein required for IME2 expression. Under identical experimental conditions, expression of the MCK1 and IME4 genes (which promote sporulation but do not require Ime1p for expression) was not affected. These results demonstrate the specificity of the yvh1 disruption phenotype. They suggest that decreased steady-state levels of IME1 and IME2 mRNA were not merely the result of non-specific adverse affects on nucleic acid metabolism caused by the yvh1 disruption. Sporulation of a homozygous yvh1 disruption mutant was delayed and less efficient overall compared to an isogenic wild-type strain, a result which correlates with decreased IME1 and IME2 gene expression. We also observed that expression of the PTP2 tyrosine phosphatase gene (a negative regulator of the osmosensing MAP kinase cascade), but not the PTP1 gene (also encoding a tyrosine phosphatase) was induced by nitrogen-starvation. Although disruption of PTP2 alone did not demonstrably affect sporulation or IME2 gene expression, sporulation was decreased more in a yvh1, ptp2 double mutant than in a yvh1 single mutant; it was nearly abolished in the double mutant. These data suggest that the YVH1 and PTP2 encoded phosphatases likely participate in the control network regulating meiosis and sporulation. Expression of YVH1 and PTP2 was not affected by nitrogen source quality (asparagine compared to proline) suggesting that nitrogen starvation-induced YVH1 and PTP2 expression and sensitivity to nitrogen catabolite repression are on two different branches of the nitrogen regulatory network.

    Notes: The yeast SEC14 gene product is required for the transport of proteins from the Golgi complex. We have cloned the homologous Candida albicans SEC14 gene (CaSEC14) by functional complementation of a Saccharomyces cerevisiae thermosensitive mutant, sec14. Some putative TATA boxes have been identified in CaSEC14 and, contrary to S. cerevisiae SEC14, no introns were found in the Candida homologue. Sequence analysis revealed that CaSec14p is a 301 amino acid protein, 67% identical to S. cerevisiae and Kluyveromyces lactis Sec14p, and 61% identical to the 300 amino-terminal residues of Yarrowia lipolytica Sec14p. Hydrophatic profile analysis of CaSec14p suggests a soluble protein without transmembrane domains, as has been described for the S. cerevisiae counterpart. While it was easy to disrupt one allele of SEC14 in C. albicans, repeated attempts to disrupt the second allele were unsuccessful, thus suggesting that the gene could be essential for vegetative growth in C. albicans.     Notes: The synthesis of mevalonate, a molecule required for both sterol and isoprene biosynthesis in eukaryotes, is catalysed by 3-hydroxy-3-methylglutaryl coenzyme A (HMG-CoA) reductase. Using a gene dosage approach, we have isolated the gene encoding HMG-CoA reductase, hmg1+, from the fission yeast Schizosaccharomyces pombe (Accession Number L76979). Specifically, hmg1+ was isolated on the basis of its ability to confer resistance to lovastatin, a competitive inhibitor of HMG-CoA reductase. Gene disruption analysis showed that hmg1+ was an essential gene. This result provided evidence that, unlike Saccharomyces cerevisiae, S. pombe contained only a single functional HMG-CoA reductase gene. The presence of a single HMG-CoA reductase gene was confirmed by genomic hybridization analysis. As observed for the S. cerevisiae HMG1p, the hmg1+ protein induced membrane proliferations known as karmellae. A previously undescribed ‘feed-forward’ regulation was observed in which elevated levels of HMG-CoA synthase, the enzyme catalysing the synthesis of the HMG-CoA reductase substrate, induced elevated levels of hmg1+ protein in the cell and conferred partial resistance to lovastatin.The amino acid sequences of yeast and human HMG-CoA reductase were highly divergent in the membrane domains, but were extensively conserved in the catalytic domains. We tested whether the gene duplication that produced the two functional genes in S. cerevisiae occurred before or after S. pombe and S. cerevisiae diverged by comparing the log likelihoods of trees specified by these hypotheses. We found that the tree specifying post-divergence duplication had significantly higher likelihood. Moreover, phylogenetic analyses of available HMG-CoA reductase sequences also suggested that the lineages of S. pombe and S. cerevisiae diverged approximately 420 million years ago but that the duplication event that produced two HMG-CoA reductase genes in the budding yeast occurred only approximately 56 million years ago. To date, S. pombe is the only unicellular eukaryote that has been found to contain a single HMG-CoA reductase gene. Consequently, S. pombe may provide important opportunities to study aspects of the regulation of sterol biosynthesis that have been difficult to address in other organisms and serve as a test organism to identify novel therapies for modulating cholesterol synthesis.

    Notes: Saccharomyces cerevisiae contains two double-stranded RNA (dsRNA) viruses (L-A and L-BC) and two different single-stranded (ssRNA) replicons (20S RNA and 23S RNA). Replicase (dsRNA synthesis on a ssRNA template) and transcriptase (ssRNA synthesis on a dsRNA template) activities have been described for L-A and L-BC viruses, but not for 20S or 23S RNA. We report the characterization of a new in vitro RNA replicase activity in S. cerevisiae. This activity is detected after partial purification of a particulate fraction in CsCl gradients where it migrates at the density of free protein. The activity does not require the presence of L-A or L-BC viruses or 23S RNA, and its presence or absence is correlated with the presence or absence of the 20S RNA replicon. Strains lacking both this RNA polymerase activity and 20S RNA acquire this activity when they acquire 20S RNA by cytoduction (cytoplasmic mixing). This polymerase activity converts added ssRNA to dsRNA by synthesis of the complementary strand, but has no specificity for the 3′ end or internal template sequence. Although it replicates all tested RNA templates, it has a template size requirement, being unable to replicate templates larger than 1kb. The replicase makes dsRNA from a ssRNA template, but many single-stranded products due to a terminal transferase activity are also formed. These results suggest that, in contrast to the L-A and L-BC RNA polymerases, dissociation of 20S RNA polymerase from its RNA (or perhaps some cellular factor) makes the enzyme change its specificity.

    Notes: HM, an HMG1-like mitochondrial DNA-binding protein, is required for maintenance of the yeast mitochondrial genome when cells are grown in glucose. To better understand the role of HM in mitochondria, we have isolated several multicopy suppressors of the temperature-sensitive defect associated with an abf2 null mutation (lacking HM protein). One of these suppressors, SHM1, has been characterized at the molecular level and is described herein. SHM1 encodes a protein (SHM1p) that shares sequence similarity to a family of mitochondrial carrier proteins. On glycerol medium, where mitochondrial function is required for growth, shm1 deletion mutants are able to grow, whereas shm1 abf2 double mutants are severely inhibited. These results suggest that SHM1p plays an accessory role to HM in the mitochondrion. The GenBank Accession Number for the SHM1 sequence is U08352.

    Notes: The endoplasmic reticulum (ER) of eukaryotic cells contains a quality control system, that is required for the proteolytic removal of aberrantly folded proteins that accumulate in this organelle. We used genetic and biochemical methods to analyse the involvement of N-glycosylation in the degradation of a mutant derivative of carboxypeptidase yscY in the ER of the yeast Saccharomyces cerevisiae. Our results demonstrate that N-glycosylation of this protein is required for its degradation since an unglycosylated species is retained stably in the ER. Cells that were devoid of the ER-processing α1,2-mannosidase showed reduced degradation of the glycosylated substrate protein. Disruption of CNE1, a gene encoding a putative yeast homologue for calnexin, did not exhibit any effects on the degradation of this substrate protein in vivo. Also, the α1,2-mannosidase-dependent reduction in the degradation rate did not show any correlation with the function of the CNE1 gene product. Our results suggest that the ER of yeast contains a glycosylation-dependent quality control system, as has been shown for higher eukaryotic cells.

    Notes: Recent studies suggest that intracellular membrane traffic relies upon families of related proteins which confer specificity to individual transport reactions but which operate in tandem with a ubiquitous fusogenic complex containing the N-ethylmaleimide-sensitive fusion protein (NSF). The extent to which components of this process are functionally conserved is apparent from the finding that yeast Sec18 protein (Sec18p) can substitute for mammalian NSF in intra-Golgi transport reactions. Here we report that yeast cytosol can support mammalian endosomal vesicle fusion, demonstrating conservation of cytosolic components required for this reaction. Furthermore, under conditions in which the fusion reaction is NSF-dependent we show that yeast Sec18p can functionally substitute for NSF, showing that the yeast protein is capable of catalysing at least two distinct mammalian membrane fusion events. In addition we exploit the complex pattern of sensitivity of the mammalian reaction to N-ethylmaleimide (NEM), coupled with the use of yeast cytosol, to dissect a number of factors required for fusion. We reveal at least three novel NEM-sensitive activities. One of these can be restored by yeast cytosol suggesting that it is functionally conserved.

    Notes: The DEX gene encoding an extracellular dextranase was isolated from the genomic DNA library of Penicillium minioluteum by hybridization using the dextranase cDNA as a probe. Comparison of the gene and cDNA sequences revealed that the DEX gene does not contain introns. Amino acid sequences comparison of P. minioluteum dextranase with other reported dextranases reveals a significant homology (29% identity) with a dextranase from Arthrobacter sp. CB-8. The DEX gene fragment encoding a mature protein of 574 amino acids was expressed in the methylotrophic yeast Pichia pastoris by using the SUC2 gene signal sequence from Saccharomyces cerevisiae under control of the alcohol oxidase-1 (AOX1) promoter. Over 3·2g/l of enzymatically active dextranase was secreted into the medium after induction by methanol. The yeast product was indistinguishable from the native enzyme in specific activity and the N-terminus of both proteins were identical.

    Notes: A binding site for the transcription factor Abf1p was identified as an important promoter element of the gene that encodes Rpo26, a subunit common to all three yeast nuclear RNA polymerases (RNAP). Mutations in the Abf1p binding site were identified among a pool of rpo26 mutant alleles that confer synthetic lethality in combination with a temperature-sensitive mutation (rpo21-4) in the gene that encodes the largest subunit of RNAPII (Rpo21p). In the presence of the wild-type allele of RPO21 these rpo26 promoter mutations confer a cold-sensitive growth defect. Electrophoretic mobility-shift assays using purified Abf1p demonstrated that Abf1p binds to the RPO26 promoter and that the promoter mutations abolish this binding in vitro. Quantitation of the amount of RPO26 mRNA showed that mutations in the Abf1p binding site reduce the expression of RPO26 by approximately 60%. Mutations that affect Abf1p binding also result in a shift of the RPO26 transcriptional start sites to positions further upstream than normal. These results suggest that binding of the Abf1p transcription factor to the RPO26 promoter is important not only in establishing the level of transcription for this gene, but also in positioning the initiation sites of transcription.

    Notes: We have sequenced a DNA fragment of 39 411 bp which includes part of the left telomere of chromosome VII of Saccharomyces cerevisiae. We have identified 19 open reading frames (ORFs); six correspond to known yeast genes (ADH4, FZF1, HKB, RTG2, HFM1 and PDE1), nine have similarity with other genes and four exhibit no significant similarity with any known gene. The average size of these ORFs seems to be related to their location, the eight ORFs nearest the telomere being shorter than the 11 others. These two groups of genes are separated by a region of 4·5 kb devoid of significant ORFs. One ORF, NRF120, is a new member of the seripauperine family, represented once in all sequenced yeast chromosomes, in a subtelomeric location. This sequence has been entered in the EMBL data library under accession number X94357.

    Notes: We present here the sequence analysis of a DNA fragment (cosmid pUOA1258) located on the right arm of chromosome XV. The 22 956 bp sequence reveals 14 open reading frames (ORFs) longer than 300 bp and the 201 bp RPS33 gene. Among the 14 large ORFs, two overlapping frames are likely to be non-expressed and one corresponds to the known GLN4 gene encoding glutaminyl-tRNA synthetase. Two ORFs, O3571 and O3620, encode putative transcriptional regulators with a Zn(2)-Cys(6) DNA binding domain characteristic of members of the GAL4 family. Among the nine remaining ORFs, five (O3568, O3575, O3590, O3615 and O3625) present significant similarity to proteins of unknown function and four (O3580, O3595, O3630 and O3635) lack homology to sequences present in the databases screened. This sequence has been deposited in the GenBank database under Accession Number U55021.

    Notes: We have determined the complete nucleotide sequence of a 23 kb segment from the left arm of chromosome IV, which is carried by the cosmid 1L10. This sequence contains the 3′ coding region of the STE7 and RET1 (COP1) genes, and 13 complete open reading frames longer than 300 bp, of which ten correspond to putative new genes and three (CLB3, MSH5 and RPC53) have been sequenced previously. The sequence from cosmid 1L10 was obtained entirely by a combined subcloning and walking primer strategy.

    Notes: We have determined the nucleotide sequence of a fragment of chromosome XV of Saccharomyces cerevisiae cloned into cosmid pEOA048. The analysis of the 26 857 bp sequence reveals the presence of 19 open reading frames (ORFs), and of one RNA-coding gene (SNR17A). Six ORFs correspond to previously known genes (MKK1/SSP32, YGE1/GRPE/MGE1, KIN4/KIN31/KIN3, RPL37B, DFR1 and HES1, respectively), all others were discovered in this work.Only five of the new ORFs have significant homologs in public databases, the remaining eight correspond to orphans (two of them are questionable). O5248 is a probable folylpolyglutamate synthetase, having two structural homologs already sequenced in the yeast genome. O5273 shows homology with a yeast protein required for vanadate resistance. O5268 shows homology with putative oxidoreductases of different organisms. O5257 shows homology with the SAS2 protein and another hypothetical protein from yeast. The last one, O5245, shows homology with a putative protein of Caenorhabditis elegans of unknown function. The present sequence corresponds to coordinates 772 331 to 799 187 of the entire chromosome XV sequence which can be retrieved by anonymous ftp (ftp. mips. embnet. org).

    Notes: In this study we used genetically manipulated strains in order to identify polypeptide spots of the protein map of Saccharomyces cerevisiae. Thirty-two novel polypeptide spots were identified using this strategy. They corresponded to the product of 23 different genes. We also explored the possibilities of using peptide-mass fingerprinting for the identification of proteins separated on our gels. According to this strategy, proteins contained in spots are digested with trypsin and the masses of generated peptides are determined by matrix-assisted laser desorption-ionization mass spectrometry (MALDI-MS). The peptide masses are then used to search a yeast protein database for proteins that match the experimental data. Application of this strategy to previously identified polypeptide spots gave evidence of the feasibility of this approach. We also report predictions on the identities of nine unknown spots using MALDI-MS.

    Notes: A cDNA clone specifying a cell wall protein was isolated from a Yarrowia lipolytica cDNA library. The cDNA library was constructed in the expression vector λgt11, with the RNA isolated from actively growing mycelial cells. The deduced amino acid sequence shows that the encoded protein contains an N-terminal hydrophobic signal peptide. We have designated this protein YWP1 for Yarrowia lipolytica cell Wall Protein. Northern hybridization identified YWP1 transcript only when Y. lipolytica was growing in the mycelial form. The encoded protein seems to be covalently bound to the glucan cell wall since it is not released from the cell walls by sodium dodecyl sulphate extraction, but it is solubilized following partial degradation of β-glucan by Zymolyase digestion. The protein is localized in the outer surface on the tip of the growing mycelial cells and is found partially cryptic in sub-apical locations, suggesting that it participates directly in the mycelial wall architecture.

    Notes: In this paper is described the DNA sequence of cosmid 14-5 from chromosome XIV of Saccharomyces cerevisiae. The sequence is 38 855 bases long and contains 21 open reading frames (ORFs) plus three internal ORFs. Six ORFs correspond to known yeast genes (SLA2, ZWF1, BLH1, KEX2, SIN4 and URE2); two other ORFs had already been sequenced because they are adjacent to known genes; the remaining 12 ORFs are novel genes. Of these, one ORF (N1142) is particularly interesting since it shows a significant similarity to mammalian globin. Another ORF (N1254) displays two zinc finger motifs as well as a DNAJ motif. The cosmid sequence has been submitted to the EMBL data library under Accession Number Z69381.

    Notes: We report the sequence of a 20 300 bp DNA fragment from the left arm of Saccharomyces cerevisiae chromosome IV. This segment contains 13 complete open reading frames (ORFs) and part of another ORF, altogether covering 84·2% of the entire sequence, five of which correspond to the previously characterized KIN28, MSS2, PHO2, POL3/CDC2 and DUN1 genes. One putative protein, D2358p, shares considerable homology with an O-sialoglycoprotein endopeptidase from Pasteurella haemolytica serotype A1. The putative product of D2325 contains the characteristic consensus motif of triacylglycerol lipases. D2320p and D2352p have a putative ‘leucine-zipper’ structure and a RNA-binding region Rnp-1 signature, respectively.     Notes: We report the sequence of a 30 469 bp long DNA fragment on the left arm of chromosome XV of Saccharomyces cerevisiae. The fragment contains 15 open reading frames (ORFs) of at least 300 bp. Five previously sequenced yeast genes, PHO80, TIR2, SLG1, the gene encoding the subtilisin-like protease III precursor and the gene coding for ATP-dependent permease, are found among these ORFs. By DNA sequence comparison, two ORFs identified previously reported expressed sequence tags from yeast. Of the proteins encoded by the remaining eight ORFs, six show similarities to proteins from different organisms and two lack detectable similarity with any amino acid sequence described in public data banks.     Notes: The complete DNA sequence of cosmid clone 31A5 containing a 35 452 bp segment from the left arm of chromosome IV from Saccharomyces cerevisiae, was determined from an ordered set of subclones in combination with primer walking on the cosmid. The sequence contains 23 open reading frames (ORFs) of more than 100 amino acid residues and the tRNA-Val2a gene. Five ORFs corresponded to the known yeast genes SNQ2, SES1, GCV1, RPL2B and RPS18A. The DNA sequence for RPS18A is interrupted by an intron. One ORF corresponded to a part of the yeast gene HEX2 at the end of the cosmid insert. Four ORFs encoded putative proteins which showed strong homologies to other previously known proteins, three of yeast origin and one of non-yeast origin. Two ORFs were classified as having borderline homologies: one had similarity to two protein families and another to two protein products of unknown function from other species. The remaining 11 ORFs bore no significant similarity to any published protein. The complete DNA sequence has been submitted to the EMBL data library, Accession Number X95966.

    Notes: We report a 29,445 bp sequence from the right arm of yeast chromosome XV. It contains the genes MYO2, SNC2, PDR10, SCD5 (also called FTB1), MIP1, VMA4, MRS2, ALA1, KRE5, TEA1, and a homologue of YAL034c. Several discrepancies with previously published sequences were found. PDR10 encodes a protein highly similar to the pleiotropic drug resistance protein Pdr5p. This sequence contig forms part of a region of extended similarity to part of the left arm of chromosome I, which is a relic of an ancient duplicated chromosomal region.

    Notes: In a shotgun approach we sequenced the cosmid pEOA284 containing a fragment derived from the right arm of chromosome XV of Saccharomyces cerevisiae. An analysis of the sequence revealed that it contained open reading frames (ORFs) corresponding to the known genes SLY41, SPS4, COT1, FAA1, PMT3, PRO2 and MYO2. Of the 18 unknown ORFs, five are contained totally within, and two, O6105 and O6163, partially overlap other ORFs. ORF O6116 and O6139 have putative introns. Regions of similarity with chromosomes I and XIII have been uncovered. Interestingly, most of the paired ORFs encode proteins of the same gene family. The relatedness of these ORFs suggests gene duplication. The sequence has been entered into the public data libraries under Accession Number X90565.

    Notes: A 13 421 bp fragment located near the left telomere of chromosome XV (cosmid pEOA461) has been sequenced. Seven non-overlapping open reading frames (ORFs) encoding polypeptides longer than 100 residues have been found (AOB859, AOC184, AOE375, AOX142i, AOE423, AOA476 and AOE433). An additional ORF (AOE131) is found within AOA476. Three of them (AOC184, AOA476 and AOE433) show no remarkable identity with proteins deposited in the data banks. ORF AOB859 is quite similar to a hypothetical yeast protein of similar size located in chromosome VI, particularly within the C-terminal half. AOE375 encodes a new member of the glycogen synthase kinase-3 subfamily of Ser/Thr protein kinases. AOX142i is the gene encoding the previously described ribosomal protein L25. AOE423 codes for a protein virtually identical to the MDH2 malate dehydrogenase isozyme. However, our DNA sequence shows a single one-base insertion upstream of the reported initiating codon. This would produce a larger ORF by extending 46 residues the N-terminus of the protein. The existence of this insertion has been confirmed in three different yeast strains, including FY1679. The complete nucleotide sequence of the 13·4 kbp fragment has been deposited at the DNA databases (Accession Number U41293).

    Notes: We have determined the sequence of a 10 624 bp DNA segment located in the left arm of chromosome XV of Saccharomyces cerevisiae. The sequence contains eight open reading frames (ORFs) longer than 100 amino acids. Two of them do not present significant homology with sequences found in the databases. The product of ORF o0553 is identical to the protein encoded by the gene SMF1. Internal to it there is another ORF, o0555 that is apparently expressed. The proteins encoded by ORFs o0559 and o0565 are identical to ribosomal proteins S19.e and L18 respectively. ORF o0550 encodes a protein with an RNA binding signature including RNP motifs and stretches rich in asparagine, glutamine and arginine. The nucleotide sequence determined has been deposited in the EMBL data library under Accession Number X95258.

    Notes: We report the sequence of an 8·8 kb segment of DNA from the left arm of chromosome VII of Saccharomyces cerevisiae. The sequence reveals seven open reading frames (ORFs) G1651, G1654, G1660, G1663, G1666, G1667 and G1669 greater than 100 amino acids in length and the tRNALys1 gene. ORF G1651 shows 100% identity with the ROK1 protein which is a putative RNA helicase of the ‘DEAD box’ protein family. ORF G1654 exhibits a motif highly conserved in ATP/GTP binding proteins generally referred to as ‘P-loop’. From FastA analysis, G1660 and G1666 were found to be previously sequenced genes, respectively SUA5 and PMR1. The three other ORFs identified are partially (G1663) or completely (G1667 and G1669) overlapping with the PMR1 sequence on the complementary strand. This feature, together with their low codon adaptation indexes and the absence of significant homology with known proteins suggest that they do not correspond to real genes. The nucleotide sequence of the 8·8 kb fragment is available through the EMBL data library under the Accession Number X85757.
