    Notizen: Macrocilia are thick compound ciliary organelles found on the lips of the ctenophore Beroë. Each macrocilium contains several hundred axonemes enclosed by a single common membrane around the shaft of the organelle. Macrocilia are activated to beat rapidly and continuously in the normal direction by stimulus-triggered Ca influx through voltage-dependent Ca channels (Tamm, 1988). Heat-dissociated macrociliary cells are spontaneously active without depolarizing stimuli, providing Ca is present (Tamm, 1988). Here we investigate the spatial distribution of macrociliary Ca channels by iontophoretic application of extracellular Ca to different sites along quiescent, “potentially activated” macrocilia of dissociated cells in Ca-free medium. We find that Ca sensitivity for eliciting motility is highest or resides exclusively on the basal portion of the macrociliary surface. This is the first demonstration of local differences in Ca morphologically with a reticulum of unfused ciliary membranes at the base of the macrocilium. This ciliary rete is in direct communication with the surrounding sea water. It is likely that the ciliary rete provides the necessary Ca influx to trigger beating by virtue of its greater Ca conductance (i.e., density of Ca channels) and/or greater total membrane area.

    Notizen: Studies of the microtubular cytoskeleton during sporogenesis in the hepatic Conocephalum conicum (Bryophyta) have revealed several unusual phenomena that contribute to understanding the cytokinetic apparatus in plant cell division. Although a typical phragmoplast forms in the interzonal microtubules of the first division spindle and expands to the cell periphery, no cell plate develops. There is no evidence of predetermined division sites and the orientation of both first and second meiotic spindles is imprecise. Simultaneous division of the cytoplasm follows second nuclear division. Equal apportionment of the cytoplasm appears to be a function of the establishment of cytoplasmic domains in the coenocyte, the boundaries of which are delimited by interaction of postmeiotic microtubule systems radiating from the four nuclei. Primary phragmoplasts are initiated in phragmoplasts that are initiated between nonsister nuclei. Depending upon the arrangement of nuclei in the nonpolar sporocyte, from one to three secondary phragmoplasts develop in the zones of contact between opposing sets of microtubules. Except for the site and time of initiation, the two types of phragmoplasts are identical. Eventually the phragmoplasts become confluent and cell plates form in all second division phragmoplasts. It is clear that typical functional phragmoplasts can form in sites determined by interaction of postmeiotic microtubule systems as well as in interzonal spindles as is common in plant cell division.

    Notizen: The 51-kD protein, a protein component of the mitotic apparatus in sea urchin eggs, is involved in the aster-forming activity previously shown in vitro [Toriyama et al., 1988]. Postembedding immunofluorescent labelings of eggs from fertilization through first cleavage showed that the 51-kD protein is localized in sperm asters, centrosomal regions, spindles, basal regions of astral microtubules, and regions surrounding daughter nuclei at telophase in situ. Immunofluorescence and immunoblot analyses detected the 51-kD protein uniformly in unfertilized eggs, but not in spermatozoa. When unfertilized eggs were treated with taxol, the 51-kD protein was shown to be associated with taxol-induced cytasters. Immunoblot analysis revealed that similar protein species are present in the mitotic apparatus of other species of sea urchin. It was suggested that the 51-kD protein may be involved in microtubule nucleation and microtubule matrix in sea urchin eggs in vivo.

    Notizen: A high-molecular-weight polypeptide, named B-band, was partially purified from sea urchin sperm flagella using selective extraction, hydroxylapatite chromatography, and sucrose density gradient centrifugation. The molecular weight of the B-band was 440,000 by continuous system of sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE). Sedimentation coefficient of the B-band protein was 10.5 S, and its Stokes radius was 10 nm. When examined by low-angle rotary shadowing electron microscopy, this molecule appeared to be composed of four globular heads and two curved linkers (“double headphone shape”), which was quite different from the shape of 21 S dynein, the outer arm dynein. Flagellar axonemes were also subjected to several chemical dissections. The B-band was not extracted with treatments that remove both arm structures but was solubilized with treatments that extract other components such as radial spokes and nexin links. The B-band protein in the axoneme was also more susceptible to trypsin digestion than the arm structures. These results suggest that the B-band protein is a “double headphone-shaped” component of the axonemal structures and makes up the elastic structure that might regulate the active sliding between adjacent doublet microtubules.

    Notizen: Fluorescence microscopy, uniquely, provides the ability to examine specific components within intact, even living, cells. Unfortunately, high-resolution conventional fluorescence microscopy is intrinsically a two-dimensional technique and performs poorly with specimens thicker than about 0.5 μm. Probing the spatial organization of components within cells has required the development of new methods optimized for three-dimensional data collection, processing, display, and interpretation. Our interest in understanding the relationship between chromosome structure and function has led us to develop the necessary methodology for exploring cell structures in three dimensions. It is now possible to determine directly the three-dimensional spatial organization of diploid chromosomes within intact nuclei throughout most of the mitotic the cell cycle.

    Notizen: Bacterial flagella have rotary motors at their base; embedded in the cytoplasmic membrane and powered by transmembrane ion gradients instead of ATP. Assays have been developed to measure the torque output of individual motors over a wide regime of load, to correlate the energizing proton flux with rotation speed and relate through genetic analysis motor structure to function. These assays promise substantial advances in understanding mechanochemical coupling in these motors. Here, I summarize the present status of our understanding of energy transduction in bacterial flagella and compare this with the case for muscle.

    Notizen: Video and digital image processing have been used to amplify the contrast of light microscopic images, making it possible to observe in real time the diffraction images of cell structures 10 times smaller than the Raleigh resolution limit of 0.2 μm. In this paper we discuss how quantitative analysis of diffraction images can be used to extract information about motion or structure at the nanometer level. This issue is considered in the context of a method for tracking the motion of kinesin-coated beads on microtubules with 1-2 nm precision (Gelles et al.: Nature 331:450-453, 1988).

    Notizen: The compartmentalization of eukaryotic cells by internal membranes and the subcellular localization of endogenous macromolecules by specific binding mechanisms are familiar concepts. In this report we present evidence that the cytoplasmic ground substance, which surrounds and contains the membranebound compartments, may also be compartmentalized by local differentiations of its submicroscopic structure that sort subcellular particles on the basis of size. The subcellular distribution of size-fractionated, fluorescent tracer particles was studied in living cells by ratio imaging and fluorescence recovery after photobleaching (FRAP). Large and small particles showed different distributions within the cytoplasmic volume, suggesting that the large particles were relatively excluded from some domains. While the structural basis for this phenomenon is not yet understood in detail, ratio imaging of large and small particles can be used as an empirical tool to identify cytoplasmic compartments for further study. The cytoplasmic diffusion coefficient (Dcyto) and % mobile fraction of the large particles showed considerable spatial variation over the projected area of the cell, while Dcyto and % mobile fraction of the small particles did not. A model is presented to account for this difference. Based on this model, a method is proposed by which FRAP can be used to detect sol-gel transitions in the cytoplasmic ground substance of living cells.

    Notizen: Recent advances in thin film optical coating technology significantly improve the filters available for fluorescence spectroscopy. Bandpass and long- and shortpass filters with very sharply defined edges can provide from 10-5 to 10-6 blocking within 10-15 nm of the transmission region and are ideal for use as excitation and emission filters. A variety of nonpolarizing dichroic beamsplitters for use in epi-illumination configurations or in multiple emission configurations provides optimum longpass, shortpass, band reflection, or bandpass spectral control. These dichroics, used with high-performance bandpass, longpass, or shortpass filters, form matched sets that optimize the signal-to-noise ratio and system efficiency for fluorescence spectroscopic systems in single or multiple dye applications. Specially designed dichroic beamsplitters are used to reduce excitation filter overheating. Other dichroic beamsplitters efficiently separate two planes of polarization in a narrow wavelength band. Rejection band filters can be used to measure the fluorescent dye Indo 1 with very low emission signals.

    Notizen: A computer-assisted Dynamic Morphology System (DMS) is described that allows the rapid quantitation of more than 30 parameters of motility and dynamic morphology for up to 40 amebae in parallel. This system also generates “difference pictures” for characterizing the dynamics of pseudopod formation. A 3-D DMS is described, and application of DMS to problems of motility and chemotaxis in normal and mutant cells of Dictyostelium discoideum is reviewed.

    Notizen: We used video-fluorescence microscopy to directly observe the sliding movement of single fluorescently labeled actin filaments along myosin fixed on a glass surface. Single actin filaments labeled with phalloidin-tetramethyl-rhodamine, which stabilizes the filament structure of actin, could be seen very clearly and continuously for at least 60 min in O2-free solution, and the sensitivity was high enough to see very short actin filaments less than 40 nm long that contained less than eight dye molecules. The actin filaments were observed to move along double-headed and, similarly, single-headed myosin filaments on which the density of the heads varied widely in the presence of ATP, showing that the cooperative interaction between the two heads of the myosin molecule is not essential to produce the sliding movement. The velocity of actin filament independent of filament length (〉1 μm) was almost unchanged until the density of myosin heads along the thick filament was decreased from six heads/14.3 nm to 1 head/34 nm. This result suggests that five to ten heads are sufficient to support the maximum sliding velocity of actin filaments (5 μm/s) under unloaded conditions. In order for five to ten myosin heads to achieve the observed maximum velocity, the sliding distance of actin filaments during one ATP cycle must be more than 60 nm.

    Notizen: Amoebae of the cellular slime mold Dictyostelium discoideum are an excellent model system for the study of amoeboid chemotaxis. These cells can be studied as a homogeneous population whose response to chemotactic stimulation is sufficiently synchronous to permit the correlation of the changes in cell shape and biochemical events during chemotaxis. Having demonstrated this synchrony of response, we show that actin polymerization occurs in two stages during stimulation with chemoattractants. The assembly of F-actin that peaks between 40 and 60 sec after the onset of stimulation is temporally correlated with the growth of new pseudopods. F-actin, which is assembled by 60 sec after stimulation begins, is localized in the new pseudopods that are extended at this time. Both stages of actin polymerization during chemotactic stimulation involve polymerization at the barbed ends of actin filaments based on the cytochalasin sensitivity of this response. We present a hypothesis in which actin polymerization is one of the major driving forces for pseudopod extension during chemotaxis. The predictions of this model, that localized regulation of actin nucleation activity and actin filament cross-linking must occur, are discussed in the context of current models for signal transduction and of recent information regarding the types of actin-binding proteins that are present in the cell cortex.

    Notizen: To move, leukocytes extend portions of their cortical cytoplasm as pseudopods. These pseudopods are filled with a three-dimensional actin filament skeleton, the reversible assembly of which in response to receptor stimulation is thought to play a major role in providing the mechanical force for these protrusive movements. The organization of this actin skeleton occurs at different levels within the cell, and a number of macrophage proteins have been isolated and shown to affect the architecture, assembly, stability, and length of actin filaments in vitro. The architecture of cytoplasmic actin is regulated by proteins that cross-link filaments in higher-order structures. Actin-binding protein plays a major role in defining network structure by cross-linking actin filaments into orthogonal networks. Gelsolin may have a central role in regulating network structure. It binds to the sides of actin filaments and severs them, and binds the “barbed” filament end, thereby blocking monomer addition at this end. Gelsolin is activated to bind actin filaments by μM calcium. Dissociation of gelsolin bound on filament ends occurs in the presence of the polyphosphoinositides, PIP and PIP2. Calcium and PIP2 have been shown to be intracellular messengers of cell stimulation.

    Notizen: Microtubules are the major cytoskeletal component of foraminiferan reticulopodia. Video-enhanced differential interference contrast light microscopy has demonstrated that the microtubules serve as the intracellular tracks along which rapid bidirectional organelle transport and cell surface motility occurs. Microtubules appear to move, both axially and laterally within the pseudopodial cytoplasm, and these microtubule translocations appear to drive the various reticulopodial movements. F-actin is localized to discrete filament plaques form at sites of pseudopod-substrate adhesion. Correlative immunofluorescence and electron microscopy reveals a structural interaction between microtubules and the actin-containing filament plaques. Our recent data on reticulopodial motility are discussed in an historical context, and a model for foram motility, based on motile microtubules, is presented.

    Notizen: In goldfish xanthophores, the formation of pigment aggregate requires: (1) that a pigment organelle (carotenoid droplet) protein p57 be in the unphosphorylated state; (2) that self-association of pigment organelles occur in a microtubule-independent manner; and (3) that pigment organelles via p57 associate with microtubules. In the fully aggregated state, the pigment organelles are completely stationary. Pigment dispersion is initiated by activation of a cAMP-dependent protein kinase, which phosphorylates p57 and allows pigment dispersion via an active process dependent on F-actin and a cytosolic factor. This factor is not an ATPase, and its function is unknown. However, its abundance in different tissues parallels secretory activity of the tissues, suggesting a similarity between secretion and pigment dispersion in xanthophores. The identity of the motor for pigment dispersion is unclear. Experimental results show that pigment organelles isolated from cells with dispersed pigment have associated actin and ATPase activity comparable to myosin ATPase. This ATPase is probably an organelle protein of relative molecular mass ∼72,000, and unlikely to be an ion pump. Isolated pigment organelles without associated actin have 5× lower ATPase activity. Whether this organelle ATPase is the motor for pigment dispersion is under investigation. The process of pigment aggregation is poorly understood, with conflicting results for and against the involvement of intermediate filaments.

    Notizen: Previous studies with the hagfish, a primitive vertebrate, have shown that the gland thread cells (GTCs) each contain a single thread (∼60 cm long in average-sized cells) in the form of a concisely coiled cytoskeletal entity destined for export by holocrine secretion. The thread in relatively immature GTCs consists almost entirely of intermediate filaments (IFs) bundled in parallel alignment with far fewer microtubules (MTs). The three thread polypeptides described earlier (α, basic; β acidic; γ, most acidic; each with a Mr of 63-64 kD) are now further evaluated with respect to in vitro assembly, cross-reactivity with IF polypeptides from higher vertebrates, and peptide sequence homology with known IF polypeptides. The overall results mainly suggest that the hagfish polypeptides are keratinlike substances but lamins or a new type of IF is not ruled out. However, cross-reactivity is weak with mammalian keratins; the 8-11-nm filaments formed from mixtures of α and γ in vitro are generally linear rather than the curvilinear structures usually formed by keratin and nonkeratin IFs; and mixtures of α and β tend to yield 9-12-nm granules or granular strings. Polypeptide analyses on GTCs segregated on the basis of maturational stage show a progressive increase in β/γ values which correlates with cell maturation, but the α/(β+γ) ratios remain near 1. Inasmuch as β and γ have many similar properties, the documented increase in the amount of the β component in aging GTCs might in part be the result of a failure in a posttranslational modification system and may contribute to the ultrastructural changes that accompany thread maturation in preparation for holocrine secretion and subsequent modulation of the viscoelastic properties of mucus.

    Notizen: We investigated the nature of the asymmetric positioning and attachment of Chaetopterus oocyte meiotic spindles to the animal pole cortex by micromanipulation. The manipulated spindle's behavior was analyzed in clarified oocyte fragments using video-enhanced polarized light microscopy. As the spindle was drawn towards the cell interior with a microneedle, the cell surface dimpled inwards adjacent to the outer spindle pole. As the spindle was pulled further inwards, the dimple suddenly receded indicating a rupture of a mechanical link between the cell cortex and outer spindle pole. The spindle paused briefly when released from the microneedle; then it spontaneously migrated back to the original attachment site and reassociated with the cell cortex. Positive birefringent astral fibers were seen running between the outer spindle pole and the cortex during the migration. The velocity of the spindle during its migration tended to increase as it came closer to the cortex. Velocities as high as 1.25 μm/sec. were measured. If removed too far from the attachment site cortex ( 〉 35 μm), the spindle remained stationary until pushed closer to the original attachment site. Spindles, inverted by micromanipulation, migrated and reattached to the cortical site by their former inner pole; thus either spindle pole can seek out and migrate to the original attachment site. However, spindle poles pushed against other cortical regions did not attach demonstrating that there is only one unique, localized attachment site for spindle attachment.

    Notizen: Mammalian respiratory ciliary outer arm dyneins isolated as the major ATPase peak migrating at 19S on sucrose density gradients were examined by transmission electron microscopy of negatively stained samples and scanning transmission electron microscopy of unstained samples. The predominant discrete particle structure observed was composed of two globular heads apparently connected by amorphous or indistinct material. The heads were either circular or slightly elliptical of mean 13 ± 1×10 ± 2 nm dimensions. The mass of this structure averaged 1.22 ± 0.34 million daltons with the individual globular heads averaging 310 ± 77 kilodaltons (kD). Negative staining revealed that one or both of the globular heads often contained a central accumulation of stain measuring 2.5 ± 1 nm across. A second type of structure, appearing with lesser frequency in the 19S fraction than in the unfractionated dynein preparation loaded onto the sucrose gradient, was a single globular head of 13 ± 1×10 ± 2 nm often with 2 ± 1 nm centrally accumulated stain and with or without an appendage. This one-headed particle thus resembled one-half of the two-headed particle. Mass measurements were lower, however, for isolated, single globular heads, averaging 220 ± 111 kD. A third type of particle observed was a ring-like structure with 4 ± 1 nm centrally accumulated stain and without appendages. The ring structure was slightly larger in diameter, 14 ± 1 nm, and had a greater peripheral accumulation of negative stain than either of the one- or two-headed particles, suggesting that it was not derived therefrom. Structures appearing as three globular heads were the least frequently observed. Mammalian respiratory ciliary outer arm 19S dynein is therefore a two-headed structure and conforms to descriptions of outer arm dyneins from mammalian spermatozoa flagella and from invertebrate cilia or flagella.

    Notizen: In most physiological situations, cell contact with a substratum is mediated by proteins of extracellular matrix. Therefore, an increasing number of cellsubstratum adhesion studies employ substrata covered with one or more proteins of extracellular matrix. To visualize the most adhesive cell structures, focal contacts and focal adhesions, the interference reflection microscopy has been widely used. It has been generally accepted that these strongly adhesive structures can be seen as black streaks in interference reflection microscopy. Calculations are presented herein, which although simplified, suggest that when cells are plated on protein-covered substrata, their focal contacts may not always appear black in interference reflection microscopy.

    Notizen: We report the primary structures of two mRNA species (GH1 and GH2), each predicted from the cloned cDNA and genomic gene sequences, that encode growth hormone in rainbow trout (Salmo gairdneri). Both GH1 and GH2 mRNA contain open reading frames comprising 630 nucleotides and encode 210 amino acid residues, of which 11 are variant. The translated regions of mRNA are flanked by a short 5′-untranslated sequence, which is highly conserved, and a relatively long 3′-untranslated sequence, which is highly divergent. The differences at the 3′-untranslated regions suggest that the GH1 and GH2 mRNA originate from different loci. RNA blot analysis of trout pituitary RNA using an oligonucleotide probe specific for the GH2 sequence indicates that the cloned gene is expressed. The GH1 and GH2 mRNA likely are transcribed from two distinct loci, which were duplicated during tetraploidization of the salmonid genome between 50 and 100 million years ago.

    Notizen: The mass of tubulin protein in developing embryos of the sea urchin Lytechinus pictus was measured using a radiodilution immunoassay based on densitometric analysis of immunoprecipitated tubulins resolved electrophoretically. The tubulins constitute an average of 360 ± 35 pg per egg, or 0.66% of the total protein, and there is no significant change in their concentration during embryogenesis. The masses of soluble and polymerized tubulins were measured for extracts prepared under conditions that stabilize microtubules. In eggs, a maximum of 14% of the tubulin is insoluble, and this increases throughout embryogenesis to 67% at pluteus stage (72 hr). The concentration of tubulin in eggs is at least 500 μg/ml, well above the critical concentration for tubulin assembly in vitro, yet microtubules have not been observed in eggs. The mass of newly synthesized tubulin, estimated from the mass of tubulin mRNA per embryo, accounts for a small fraction of the total tubulin by the end of gastrulation but for over half of the tubulin by the 72-hr pluteus stage. These observations are consistent with a model in which the declining level of unpolymerized tubulin controls the stability of tubulin mRNA, providing an autogenous regulation of the ontogenetic pattern of tubulin synthesis during sea urchin embryogenesis (Gong and Brandhorst, Development 102: 31-43).

    Notizen: Unfertilized eggs of several species of giant moths contain a substantial cortical cytoskeleton. This structure is assembled during oogenesis, and contains actin as a major fibrillar component. The presence of actin was confirmed by gel electrophoresis and binding to phalloidin, DNase 1, and a monoclonal antibody against cytoskeletal actin. Several lines of evidence suggest that the fat body is a source of the actin in the oocyte and that the transport and acquisition of actin by the ovary are similar to the mechanism of vitellogenin acquisition. A possible role for the cortical cytoskeleton in directing early embryogenesis is discussed.

    Notizen: The expression of the mRNA for mouse testicular lactate dehydrogenase (LDH-X) was examined by Northern analyses of meiotic and postmeiotic spermatogenic cell populations. Silver grains accumulated in cells inside the second layer from the periphery of the seminiferous tubule, confirming previous findings that LDH-X mRNA first appears in the spermatocyte and continues to accumulate until the late spermatid stage. Northern analyses showed that meiotic and postmeiotic cells contained 1.2 and 1.3 kb classes of hybridizing mRNA, respectively. RNase H digestion of oligo(dT)-hybridized RNA and poly(U)-Sepharose column chromatography with differential elution by formamide revealed that the difference in size of the two classes of mRNAs was due to the poly(A) tail length of the LDH-X mRNA. When the distribution of the LDH-X mRNA was examined across polysome gradients, both mRNAs were partially associated with polysomes. These results suggest that the changes in the polyadenylation of LDH-X mRNA were associated with the meiotic division during spermatogenesis in the mouse. They raise the possibility that the stable accumulation of the LDH-X mRNAs in the postmeiotic cells is enhanced by poly(A) tails of increased length.

    Notizen: To investigate the decrease of transcription and to determine the localization of RNA molecules during spermiogenesis in the fern Scolopendrium, we have used the enzyme-gold electron microscope method. During cell differentiation, the labeling decreases over the cytoplasm and increases over the nucleus. In the latter, the nucleolus disappears at the beginning of differentiation and the gold particles that were at first situated over the dispersed chromatin are then located over the condensed chromatin. In mature gametes, gold particles linked to RNA are abundant over the dense and homogeneous nucleus and sparse over the cytoplasm. By using in situ hybridization of RNA-poly(A) tails with radioactive poly(U) probes, we have shown that these RNA molecules are mRNA; they are located in the condensed chromatin of the mature sperm nucleus.

    Notizen: Specific changes in the two dimensional gel electrophoretic pattern of mouse oocyte phosphoproteins precede germinal vesicle breakdown (GVBD). We report that changes in the relative abundance of phosphoamino acids occurred prior to GVBD. We also report data that further strengthen the close association of the changes in phosphoprotein patterns with resumption of meiosis. The calmodulin antagonist W7, which transiently inhibits GVBD, inhibited partially at least two of the maturation-associated phosphoprotein changes, the dephosphorylation of a 60,000 Mr 24,000 and 28,000 occurred; all these changes occurred, however, in oocytes from juvenile mice that were competent to resume meiosis. The microinjection of the heat-stable inhibitor of cyclic adenosine monophosphate (cAMP)-dependent protein kinase (PKI), which induces GVBD in fully grown oocytes, did not induce GVBD in meiotically incompetent oocytes. Microinjected PKI did not induce the increased protein phosphorylations associated with maturation, but it did induce the dephosphorylation of the 60,000 Mr phosphoprotein. These results provide molecular markers for commitment to resume meiosis in GV-intact oocytes and indicate a potential basis for meiotic incompetence.

    Notizen: Endogenous and foreign DNA sequences can be detected in an extremely small number of cells via sequence amplification in vitro. The polymerase chain reaction (PCR) technique applied in multiple cycles allows the amplification of specific short regions of the genome to levels that can be detected by DNA blotting techniques. Cow and mouse blastocysts were analyzed by PCR for the presence of an endogenous single copy gene or an integrated foreign gene. The endogenous single-copy gene encoding the β chain of bovine luteinizing hormone was detectable in cow blastocysts and in purified bovine genomic DNA representing as few as 25 cells. To determine whether exogenous genes (transgenes) can be detected in preimplantation embryos, transgenic male mice hemizygous for the prokaryotic gene encoding neomycin resistance where bred to nontransgenic females, and the resulting blastocysts were analyzed. The neo gene was detected in approximately half of the embryos. The capability to identify specific gene sequences in a limited number of embryonic cells affords investigators the opportunify to study genetics in early development.

    Notizen: The pronephros in juvenile brown trout (Salmo trutta) consists of a large ovoid renal corpuscle and a pair of tubules. The corpuscle is retained for 11 months, after which the glomerulus regresses. The glomerular arteries come directly from the dorsal aorta. The interstitium is permeated with venous blood vessels that arise from the anterior cardinal veins and are closely apposed to the tubules. Two distinct segments of the pronephric tubular system are distinguished by the histological and ultrastructural features of their component cells: (1) a short, transitional neck in which cells change from capsular epithelium to columnar epithelium, typical of tubules; (2) the convoluted segment composed of cells similar to first proximal tubular cells of the opisthonephros with well-formed brush borders, apical vesicles that vary in size and number along this segment, and lysosomes. Pinocytosis and exocytosis are also evident in this segment. The tubular system increases in length and in its convolutions until about week 9, when the opisthonephros develops. Distally each tubule connects with a Wolffian duct, with cells marked by the absence of apical inclusions and the presence of a uniform brush border, numerous mitochondria, and elaborate infolding of the basalar membrane. Nephrostomes, which are often characteristic of pronephroi, are not present. Cells with long cilia are found throughout the tubular system but are most characteristic of the neck and Wolffian-duct segments.

    Notizen: In anuran amphibians, cranial bones typically first form at metamorphosis when they rapidly invest or replace the cartilaginous larval skull. We describe early development of the first three bones to form in the Oriental fire-bellied toad, Bombina orientalis - the parasphenoid, the frontoparietal, and the exoccipital - based on examination of serial sections. Each of these bones is fully differentiated by Gosner stage 31 (hindlimb in paddle stage) during premetamorphosis. This is at least six Gosner developmental stages before they are first visible in whole-mount preparations at the beginning of prometamorphosis. Thus, developmental events that precede and mediate the initial differentiation of these cranial osteogenic sites occur very early in metamorphosis - a period generally considered to lack significant morphological change. Subsequent development of these centers at later stages primarily reflects cell proliferation and calcified matrix deposition, possibly in response to increased circulating levels of thyroid hormone which are characteristic of later metamorphic stages. Interspecific differences in the timing of cranial ossification may reflect one or both of these phases of bone development. These results may qualify the use of whole-mount preparations for inferring the sequence and absolute timing of cranial ossification in amphibians.

    Notizen: The primary and secondary elements of the cephalic vascular system in some sea snakes are similar to those of the generalized ophidian pattern. The three species examined in this study revealed only minor variations in vascular morphology; these variations appear to be correlated with myological differences among the three species. For example, in Hydrophis melanocephalus it appears that the depressor mandibulae artery is displaced by the cranially expanded insertion of the semi-spinalis and spinalis muscles. A preliminary hypothesis is put forth that explains the apparent constancy of the cephalic vascular system of ophidians in terms of possible constraints due to cranial kinesis.

    Notizen: In order to evaluate the differences in the cleavage patterns of the glossiphoniid leeches Glossiphonia complanata and Theromyzon tessulatum, previously studied by Müller ('32) and Schmidt ('17, '41), the cleavage of Theromyzon tessulatum was reexamined. For the period of the first 29 hours of development embryos were observed, photographed, and serially sectioned for light microscopy at each developmental stage. The exact cell lineage until completion of teloblast formation is reported. Besides some other not previously reported features, we show that the mesoteloblast precursor cell in the glossiphoniid leeches, as probably in most Annelida, is not the cell 3D, but cell 4d formed by an additional division of cell 3D. The results further indicate that all glossiphoniid leeches likely share a common cleavage pattern, and that major differences between Glossiphonia complanata and Theromyzon tessulatum do not exist. A comparison between the cleavage patterns of some Oligochaeta and Hirudinea is made, and plesiomorphic characters in the cleavage of a clitellate ancestor species and their deviations in present day species are discussed.

    Notizen: A serotonin-like substance in the organ of Bellonci in the eyestalks of embryos, larvae, and adults of the prawn Palaemon serratus was visualized by the use of two specific antisera against serotonin (5-hydroxytryptamine; 5-HT) in combination with peroxidase-antiperoxidase (PAP). The organ of Bellonci, characterized by compact onion bodies distally and degenerating onion bodies proximally, was the only site of the serotonin-like substance in adults, as well as during development in embryos and larvae. Variations in the content of the 5 HT analogue in the adult were detected during the molting cycle. There was more immunoreactivity in specimens fixed at night than in those fixed in daytime. Likewise, colchicine and nialamide injections enhanced the immunoreactivity of the serotonin-like substance. Extirpations of the medulla externa X organ (MEX), a neurosecretory cell group of the optic ganglion medulla externa, produced the same effect.

    Notizen: Neurosecretory (Nsy) cells within the cerebral ganglion of Lumbricus terrestris were classified ultrastructurally. The Nsy cells within the subesophageal ganglion, nerve cord ganglion, and the peripheral nervous system were also examined. A comparative survey of Nsy cells of four other species of oligochaetes, Eisenia feotida, octolasion cyaneum, Dendrobeona subrubicunda, and Allolophora longa, was also carried out. Seven cell types (A1, A2, A3, A4, A5, C, and SEF), distinguished by special cytological and ultrastructural features, were found within the cerebral ganglion. Distribution of these cells inside and outside the cerebral ganglion was studied in detail by light and electron microscopy. The nerve terminals of each cell type were followed into the neuropile region. Exocytosis from cell bodies appears to be the main release mechanism for the Nsy granules, whereas small Nsy vesicles are released through synapses in the neuropile. Peripheral fibers of some cell types (A1, A2, and A3) extend through the capsule to the pericapsular epithelium. It is possible that Nsy cells secrete hormones from their cell bodies and peripheral processes and that their centrally directed axons release modulators/transmitters within the neuropile.

    Notizen: The development of the oral structures of six species of anuran tadpoles with four different types of mouth parts and the metamorphic atrophy of these structures in two species with different mouth parts are described. The oral labia of typical tadpoles, oral flaps of microhylids, and lateral oral folds of Rhinophrynus are assumed to be homologous. We also suggest that the barbels of the tadpoles of Rhinophrynus are homologs of the marginal papillae of species with an oral disc. Developmental patterns and sequences of the oral structures of all tadpoles examined follow a common pattern: stomodeal invagination, oral pad development, jaw sheeth delimitation, tooth row ridge development, jaw sheath keratinization, and labial tooth keratinization. Developmental patterns remain constant, while interspecific differences are apparent because of truncations of ontogeny at specific stages. Metamorphic atrophy of oral structures occurs roughly in the reverse order of development, although the procedure is rapid and more haphazard than development.

    Notizen: The vertebral centra of Hiodon, Elops, and Albula are direct perichordal ossifications (autocentra) which enclose the arcocentra as in Amia. An inner ring of ovoid cells forms in late ontogeny from the intervertebral space inside the autocentrum. The chordacentrum is reduced or completely absent in centra of adult Elops, whereas it forms an important portion of the centra in adult Hiodon. The posterior portion of the compound ural centrum 3+4+5 is partially (Hiodon) or fully formed by the chordacentrum (Elops, Albula). The haemal arches and hypurals are fused medially by cartilage or bone trabecles of the arcocentrum with the centra, even though they appear autogenous in lateral view in Elops and Albula. The composition of the caudal skeleton of fossil teleosts and the ontogeny of that of Hiodon, Elops, and Albula corroborate a one-to-one relationship of ural centra with these dorsal and ventral elements. The first epural (epural 1) of Elops relates to ural centrum 1, whereas the first epural (epural 2) of Hiodon and Albula relates to ural centrum 2. In Albula, the first ural centrum is formed by ural centrum 2 only. With 4 uroneurals Hiodon has the highest number within recent teleosts. Juvenile specimens of Hiodon have eight, the highest number of hypurals within recent teleosts; this is the primitive condition by comparison with other teleosts and pholidophorids. Reduction of elements in the caudal skeleton is an advanced feature as seen within elopomorphs from Elops to Albula. Such reductions and fusions occur in osteoglossomorphs also, but the lack of epurals and uroneurals separates most osteoglossomorphs (except Hiodon) from all other teleosts.

    Notizen: Paraphalanges of gekkonid lizards are cartilaginous structures associated with interphalangeal joints. Their form and structure have been investigated by dissection, cleared-and-stained specimens, routine histoloty, and radiography. A family-wide survey revealed that paraphalangeal elements occur in at least 57 species in 16 genera of the subfamily Gekkoninae. The distribution and structure of these elements suggests multiple origins among gekkonine geckos. In most instances, they are present in species with expanded subdigital climbing pads, divided scansors, and a markedly raised penultimate phalanx that is elevated from, or free of, the pad. Thus, they seem to be associated with placement of the scansors onto the locomotor substrate. In two genera, Uroplatus and Palmatogecko, paraphalanges at the more proximal interphalangeal joints are associated with muscles that run between them. In these cases, the paraphalanges appear to be involved in grasping abilities of the foot associated with digging and climbing modifications.

    Notizen: Primary spermatogonia have highly lobate nuclei and can be distinguished as pale and dark types on the basis of nuclear and cytoplasmic features. Nuclei of secondary spermatogonia are also lobate. Primary spermatocytes have spherical nuclei and display synaptinemal complexes in late zygotene-pachytene. Spermatocytes are connected by intercellular bridges, which persist through spermiogenesis. During spermiogenesis no acrosomal granule is formed. The acrosomal vesicle is large and forms in the apical part of the cell. A helical system of perinuclear microtubules accompanies the phase of nuclear elongation. Microtubules disappear in late spermatids and there forms a compact bundle of filaments which extends into the subacrosomal area. These filaments probably derive from the breakdown of the microtubules. A mitochondrial sleeve is formed around the proximal portion of the tail and much of it is cast off in the mature spermatid. The tail is composed of a spirally coiled contractile element and a stiff fibrous axial rod connected together by an undulating membrane. The axial rod and the axoneme-associated rodlet derive from a dense, juxtacentriolar fibrous mass. Sertoli cells surrounding the spermatogonial and spermatocyte cysts are slender and have oblong nuclei. In contrast, those associated with spermatids are columnar and have deeply indented nuclei. They possess many Golgi complexes, elongated mitochondria, cisternae of smooth endoplasmic reticulum, lysosome-like bodies, masses of glycogen particles, few lipid droplets, and an array of microtubules running longitudinally around the elongating spermatid nuclei. Desmosomes are formed between adjacent Sertoli cells.

    Notizen: Microscopic studies show that the capsule of the hip joint in the vervet monkey (Cercopithecus aethiops pygerythrus) is preponderantly collagenous. Among the collagen fiber bundles are varying quantities of elastic fibers that demonstrate a definite, differential regional distribution. The highest concentration of elastic tissue is found in the posterior, postero-inferior, and inferior aspects of the hip joint capsule, whereas the anterior and superior aspects of the capsule are preponderantly collagenous. It is postulated that this regional distribution of elastic tissue is related to the differential functional requirements of the posterior, postero-inferior, and inferior aspects of the capsule for flexibility and stretchability. These requirements appear to be a consequence of the habitual postures and locomotory positions assumed at the hip joint by these primarily quadrupedal primates. Collagen, on the other hand, being much more resistant to deformation and relatively noncompliant, is the predominant tissue in the anterior and superior aspects of the joint.

    Notizen: Fluorescence histochemsitry was used to study the adrenergic innervation of the large arteries and veins at six points along the body of the semiarboreal rat snake Elaphe obsoleta. Apart from the vessels adjacent to the heart, there was a marked contrast in the density of adrenergic innervation of anterior and posterior systemic arteries and veins. The anterior arteries and veins have little adrenergic innervation in contrast to the extremely dense innervation of the arteries and veins posterior to the heart. The innervation pattern is consistent with known physiological adjustments to gravity and suggests a mechanism for regulating dependent blood flow via sympathetic nerves. In comparison to the posterior systemic arteries, parallel segments of pulmonary artery taken from the same body position of Elaphe contained a much sparser innervation by adrenergic nerves. The sparser innervation can be correlated with less gravitational disturbance in the pulmonary artery, which is relatively short in this and in other arboreal snakes.

    Notizen: Electromyography and cinematography were used to determine the activity of epaxial muscles of colubrid snakes during terrestrial and aquatic lateral undulatory locomotion. In both types of lateral undulation, at a given longitudinal position, segments of three muscles (Mm. semispinalis-spinalis, longissimus dorsi, and iliocostalis) usually show synchronous activity. Muscle activity propagates posteriorly and generally is unilateral. With each muscle, large numbers of adjacent segments (30 to 100) show simultaneous activity. Terrestrial and aquatic undulation differ in two major respects. (1) During terrestrial undulation, muscle activity in a particular region begins when that portion of the body has reached maximal convex flexion and ends when it is maximally concave; this phase relation is uniform along the entire snake. During swimming, however, muscle activity passes posteriorly faster than the wave of vertebral flexion, causing the relation of muscle activity to flexion to change along the length of the snake. (2) In the terrestrial mode, the block of active muscle segments remains approximately constant in size as it passes down the snake, whereas during swimming the number of adjacent active muscle segments increases posteriorly. Despite the fact that Elaphe obsoleta has nearly twice as many body vertebrate as Nerodia fasciata (240 vs. 125), the only difference observed in the swimming of these two species is that a larger number of adjacent muscle segments is simultaneously active in comparable regions of Elaphe obsoleta than in Nerodia fasciata.

    Notizen: The horseradish peroxidase (HRP) histochemical technique was used to examine the peripheral distribution and afferent projections of the trigeminal nerve in the goldfish, Carassius auratus. Sensory fibers of the trigeminal nerve distribute over the head via four branches. The ophthalmic branch distributes fibers to the region above the eye and naris. The maxillary and mandibular branches innervate the regions of the upper and lower lip, respectively. A fourth branch of the trigeminal nerve was demonstrated to be present in the hyomandibular trunk.Upon entering the medulla the trigeminal afferent fibers divide into a rostromedially directed bundle and a caudally directed bundle. The rostromedially directed bundle terminates in the sensory trigeminal nucleus (STN) located within the rostral medulla. The majority of fibers turn caudally, forming the descending trigeminal tract. Fibers of the descending trigeminal tract terminate within three medullary nuclei: the nucleus of the descending trigeminal tract (NDTV), the spinal trigeminal nucleus (Spv), and the medial funicular nucleus (MFn). All projections, except for those to the MFn, are ipsilateral. Contralateral projections were observed at the level of the MFn following the labeling of the ophthalmic and maxillomandibular branches. All branches of the trigeminal nerve project to all four of the trigeminal medullary nuclei. Projections to the STN and MFn were found to be topographically organized such that the afferents of the ophthalmic branch project onto the ventral portion of these nuclei, while the afferents of the maxillo- and hyomandibular branches project to the dorsal portion of these nuclei.Cells of the mesencephalic trigeminal nucleus were retrogradely labeled following HRP application to the ophthalmic, maxillary, and mandibular branches of the trigeminal nerve.     Notizen: Natural maturation of the oocytes of Oxycomanthus japonicus can be predicte in advance, and the multiple ovaries permit unintrusive serial biopsies. Ovaries were fixed for transmission electron micrscopy at 15-min intervals before, during, and after oocyte maturation. The start of maturation of each oocyte is signaled by the breakdown of the germinal vesicle and the disruption of a macula adhaerens associating the oocyte with nongerminal cells of the ovary. This disruption is followed by an ovulation of the oocyte into the ovarian lumen. Ovulation takes about 1 hr, and a continuous vitelline coat is produced around the oocyte during this interval. Within the oocyte cytoplasm, patches of nuage and the annulate lamellae disappear at 30 and 45 min after the start of oocyte maturation, respectively. Micropapillae become transiently abundant at the oocyte surface both at the time of germinal vesicle breakdown and around the time when the first and second polar bodies are produced. Oocyte maturation takes about 2 hr from start to finish, and the emission of the second polar body marks the beginning of the stage of the ovum. Within the cytoplasm of the ovum, the haploid chromosomes develop into chromosome-containing vesicles, which later fuse into a single female pronucleus. Pronuclear ova are retained in the ovarian lumen for about 1 hr and are then spawned into the surrounding seawater, where fertilization takes place.

    Notizen: Morphometric analysis of the cochlea was performed in wild and laboratory murids: Mus musculus, Apodemus sylvaticus, Rattus rattus, R. norvegicus, NMRI mouse, and Wistar rat. Results are based on light microscopic examination of surface specimens and serial sections and on three-dimensional computer reconstruction. The cochleae have 1.75-2.2 coils. The length of the basilar membrane varies from 6.0 to 12.1 mm. Mean density of outer hair cells ranges between 363 and 411, inner hair cells 98 and 121, neurons 1,230 and 1,760 per 1 mm. Following parameters change from base to apex: basilar membrane width 66.0 (±8.2) to 175.0 (±24.7) μm, basilar membrane thickness 17.0 (±2.6) to (±0.1) μm, width of triad of outer hair cells 13.2 (±0.7) to 28.8 (±4.4) μm. The given numbers are mean “murid” values (with respective standard deviations). Maximum of dimensions of scalae is located at 10-15%, that of density of outer hair cells at 65%, density of inner hair cells at 2.8 mm, maximum of innervation density at 40-60% from the base. The following parameters are correlated with pinna size: length and maximum width of basilar membrane, dimensions of scalae, total number of receptors, and probably resolution capabilities. The following parameters are correlated with body size: maximum width of traid of outer hair cells, density and total number of neurons, ratio of neurons to receptors, apicobasal difference in basilar membrane stiffness and width of triad of triad of outer hair cells; inversely proportional is receptor density and ratio of outer to inner hair cells and probably low-frequency cut-off. Thickness, and minimum width of basilar membrane and triad of outer hair cells and probably high-frequency cutoff are species-specific and independent of pinna or body size. The parameters mentioned indicate that the examined murids are acoustically unspecialized mammals and their cochleae approximate the generalized plan for a mammalian cochlea. Differences between domesticated and wild murids are stated.

    Notizen: Jenynsia lineata retains its embryos within the ovarian cavity for a prolonged gestation. In the absence of egg envelopes, maternal - embryonic transfer occurs through ovarian fluid across apposed epithelia, relatively lining the ovarian lumen and the surface of the embryos. There are no hypertrophied extraembryonic structures that could provide expanded exchange surfaces for the passage of nutrients beyond the 8-mm stage, but structural specializations of the ovary then form, and these may sustain embryogenesis. Outgrowths of the inner lining of the ovary, villi ovariales, enter the pharyngeal cavity of the embryos via an opercular cleft remaining from early stages of development, after depletion of yolk reserves, until shortly before term. The ovary and its villi are lined by a monolayer of squamous cells showing evidence of vesicular transport of macromolecular substances both on the apical surface and at the basolateral pole. It serves for transcellular passage of maternally derived substances rather than as a source of secretory products. Most adjacent cells interdigitate, and the epithelium is continuous except for few gaps at the villous tips, which allow paracellular passage of particulate matter. These epithelial cells contain abundant filaments, electron-dense granules within the cytoplasm and the nucleus, sparse elements of the rough endoplasmic reticulum, a Golgi apparatus, and different sorts of vacuoles. The capillaries in the intraovarian lining are spaced most densely at the ovarian wall, less so toward the tips of the villi. The villi ovariales contain a network of connective tissue that forms endotheliumlike septa, which divide the interior into numerous different-sized loculi.

    Notizen: We have examined the embryonic development of the major neuromast lines of the lateral-line system in the urodele Ambystoma mexicanum both in vivo (using microsurgical techniques to transplant placodes) and in preserved embryos using scanning electron microscopy (SEM). We have compared this to SEM observations of embryos of the anuran Rana pipiens. We have determined the approximate locations of the lateral-line placodes in A. mexicanum and the approximate timing of both the migration of the lateral line primordia and the neuromast eruption in both species. We find that, at hatching, all primary neuromasts are present and fully formed in Ambystoma, while migration of the primordia is just beginning in Rana. The neuromast systems in both species are fully formed by the time feeding begins. If neuromast eruption is considered in relation to developmental events other than hatching, fewer differences are found between species, suggesting that hatching is precocious in Rana. We find no evidence of heterochrony to account for the morphological differences observed in these lateral-line systems. Orthogonal neuromasts on the head, a derived feature of urodeles, appears to be the result of lateral neuromast movement subsequent to the rostral migration of the primordia. This process was not observed in the anuran. In addition, we observe that ciliated epidermal cells disappear from the area immediately around each of the lines and suggest that neuromasts cause the regression of cilia in their immediate vicinity.

    Notizen: Spermatogenesis of the sea urchin Paracentrotus lividus has been studied at the ultrastructural level after conventional staining of thin sections and after en bloc silver staining. Cytoplasmic dense bodies are present in all steps of spermatogenesis except in late spermatids and spermatozoa. These bodies are closely associated with the development of centrioles and of the acrosomal vesicle during spermiogenesis. After it first appears, acrosomal vesicle is linked to the nuclear envelope by electron-dense material and subsequently acquires a dense core. Later the acrosomal vesicle moves to the apical pole of the cell while maintaining its connection to the nucleus. Although chromatin was highly condensed in the head of spermatozoa, one or more nuclear vacuoles within the nucleus were found to contain uncondensed chromatin fibers. Silver nitrate stains several nuclear and cytoplasmic structures. In the nucleus it stains the nucleolus, specific regions at the periphery of the chromatin, the synaptonemal complexes, the nuclear basal fossa, and the nuclear vacuoles of spermatozoa. In the cytoplasm, silver stains the cytoplasmic dense bodies, the material that connects the acrosomal vesicle to the spermatid nucleus, the spermatozoan subacrosomal and periacrosomal materials, the intercellular bridges of spermatids, and the centrioles. Silver staining is abolished by pretreatment with pronase E, suggesting that silver staining is due to protein.

The mean absolute area occupied by mitochondria and the mean number of mitochondria per cell increases in a linear fashion from pachytene through dictyate.     Notizen: The homology of branchial arch segments in salamanders has been a matter of controversy since the last century. Many investigators term the most medial paired elements of salamander branchial arches “ceratobranchials” and the next distal paired elements “epibranchials.” This suggests that the first two segmental elements of the salamander branchial arch are not homologous with elements occupying the same position in ray-finned fishes, Latimeria, “rhipidistians,” and lungfishes, in which these bones are called hypobranchials and ceratobranchials, respectively. Three lines of evidence suggest that it is more parsimonious to interpret urodele branchial arch segments as being homologous with those of other vertebrate clades-(1) comparative osteology, (2) comparative myology, and (3) the discovery of cartilaginous structures forming a third segmental unit that we interpret as atavistic epibranchials of the branchial arch in one population of the salamander Notophthalmus viridescens. These structures possess all the defining attributes of atavisms, and illustrate the special role that atavistic features play in resolving questions of homology recognition.

    Notizen: Proteoglycans were studied in developing rat fetal kidney using cytological and biochemical techniques. These compounds were detected with immunoperoxidase in the nephron basement membranes from the earliest stages of differentiation. In the glomerular basement membrane, immunostaining appeared as both diffuse and granular deposits, as long as this membrane consisted of loose material; however, as soon as a three-layered membrane had formed, staining was confined to the laminae rarae as regularly arranged granules. The same pattern of staining was observed during differentiation of the basement membrane of the proximal tubule. In Bowman's capsule, immunostaining appeared as granules, which were sparsely distributed in the developing glomerulus and then regularly lined the stacked laminae when differentiation was complete. In all basement membranes, anionic sites (disclosed by polyethyleneimine) were colocated with immunostained granular diposits. Total glycosaminoglycan content gradually increased from the beginning of metanephros development to birth. During this period, the relative proportions of glycosaminoglycans changed: heparan sulfate increased and hyaluronic acid deceased as differentiation proceeded. The possible relationship between morphological observations and biochemical changes in glycosaminoglycan content is discussed.

    Notizen: Mastication has been studied by cinematography and quantitative electromyography while flying foxes, Pteropus giganteus, were freely feeding on standardized pieces of apple, soaked raisin, and banana.The primarily orthal mandibular movements are caused by mainly bilaterally symmetrical firing of all the masticatory muscles. Asymmetric activity in the superficial and deep masseter and medial pterygoid causes slight protrusion early in opening. Slight lateral deviations at the end of opening and at the start of closing are caused by asymmetric and asynchronous activity in the pterygoids and digastrics, and by asynchronous firing of the deep temporalis and zygomaticomandibularis. Food consistency affects movement characteristics as well as characteristics of muscular activity.In this study electromyograms were digitized and the number of spikes and mean amplitude per interval (set by the filming rate) recorded. Although a significant correlation exists between these descriptors, the product thereof appears to be the best predictor of certain kinematic variables (cycle length and maximum excursion of the mandible). On the other hand, the changes in magnitude of muscular activity as a function of the position of a cycle in the reduction sequence and as a function of food consistency are more translated in a variation of the mean amplitude than in a variation of the number of spikes per interval. Observed variation differs among muscles studied. It is most apparent in the superficial and deep masseter and least in the temporalis and zygomaticomandibularis.Late cycles of apple and raisin mastication are long and exhibit large gapes but almost no anterior movement. The adductor activity frequently shows a synchronized, pulsatile pattern leading to an unfused tetanus.

    Notizen: Embryonic growth and trophotaenial development are examined in two species of goodeid fish, Ameca splendens and Goodea atripinnis. During gestation of A. splendens, embryonic dry mass may increase from 0.21 mg at the onset of development to 31.70 mg at term. In G. atripinnis, embryonic dry mass ranges from 0.25 mg at the onset of development to 3.15 mg at term. Increase in mass is primarily due to the uptake of maternally derived nutrients by trophotaeniae, externalized embryonic gut derivatives. Trophotaenial development in both species is divisible into five phases. During the first phase, the anus is formed. The second phase involves dilation of the anus, enlargement of the perianal lips, differentiation of the hindgut absorptive epithelium, and formation of the trophotaenial peduncle. The third phase is characterized by a further marked hypertrophy and lateral expansion of the perianal lips that results in the formation of short trophotaenial processes. During the fourth phase, there is continued outward expansion of the inner mucosal surface of the trophotaenial peduncle that results in its eversion and lobulation. Placental function is established by this phase. Axial elongation and dichotomous branching of trophotaenial processes occurs during the fifth phase. Development of rosette and ribbon trophotaeniae differ in the degree of axial elongation during the fifth and final phase.

    Notizen: The calanoid copepod genus Pleuromamma is easily distinguished from all other copepods by the presence of a rounded, dark-red cuticular structure ( = pigment knob) that protrudes from the left or right side of the second thoracic segment in both sexes. The present study of the pigment knob reveals a complex ultrastructure consisting of various cell types within three distinct areas that are bathed by hemolymph from the lateral sinus. The knob is covered by a greatly expanded cuticle through which a pore passes. The pore appears to connect with a centrally positioned pigment cell containing a large mass of darkly staining granules. This suggests that the knob may have a secretory function. Observations of live animals and dissected pigment knobs, however, indicates that the knob does not secrete a luminescent material nor does it luminesce internally.

    Notizen: Oocyte development occurs in a temporal and spatial pattern in the ovary of the pipefish, Syngnathus scovelli. Cytological observations combined with [3H]thymidine uptake and cell culture were used as criteria to identify six stages of oocyte development in this species. Stage I - Oogonia, found within the germinal ridge. Stage II - Oocytes in primary growth, subdivided into (1) chromatin-nucleolus oocytes in early prophase I of meiosis, also found within the germinal ridge, and (2) perinucleolar oocytes, which possess multiple nucleoli and are within definitive follicles (the Balbiani vitelline body is the main cytoplasmic component in these latter oocytes). Stage III - Oocytes distinguished by the appearance of cortical alveoli, the vitelline envelope, and lipid. Stage IV - Vitellogenic oocytes, which accumulate yolk proteins and possess primary, transitional, and mature yolk spheres. Stage V - Maturational oocytes, which are competent to undergo final meiotic maturation in vivo and in response to the steroid 17 α-hydroxy-20β-dihydroprogesterone in vitro. Stage VI - Ovulated mature eggs, which are present in the ovary lumen and are capable of being fertilized. These studies provide a staging series for oogenesis and oocyte development in the pipefish and relate these stages to the unusual ovary in this animal.

    Notizen: Larval salivary gland cells of seven Drosophila species from the melanogaster group were studied during the early third-instar period. Similar cytoplasmic organization was seen in both the distal and proximal parts of the gland. The cytoplasm contained a large number of free ribosomes, but only a few rough endoplasmic reticulum profiles; the nucleolus was very large. Golgi complexes consisted mainly of vesiculated cisternae. Small secretory granules (diameter, 0.23-0.32 μm) are produced during this period and in some species contain both granular and filamentous material. These granules appeared to be secreted by a peculiar “apocrine-type” secretion after enclosure of granules into microvillar “lacunae.” A digestive function is attributed to the secretory material. During the third instar, a close association between the salivary gland and the fat body also was observed. The physiological significance of this association seems to be related to the transfer of nutrients, enzymes, or membranous materials from fat body to salivary gland.

    Notizen: Nuclear and cytoplasmic volume changes as well as the elimination of residual spermatid cytoplasm were investigated in the red-ear turtle (Pseudemys scripta) and the rooster (Gallus domesticus). Nuclei of newly formed spermatids which were originally centrally located became eccentrically located within the cell in both species. Shortly thereafter the nuclear pole of the spermatid was found situated within deep crypts of a Sertoli cell. The cytoplasm of elongating spermatids was displaced along the nonacrosomal region of the nucleus and the proximal flagellum. In both species sheetlike Sertoli cell processes indented spermatid cytoplasm adjacent to the nucleus and appeared to segregate small packets of the cytoplasm. In the turtle, these packets of cytoplasm were separated from the spermatid. In both the turtle and rooster, a portion of the spermatid cytoplasm was displaced forward over the acrosomal region of the spermatid to resemble a hood. As spermatids were transported to the seminiferous tubular lumen, cytoplasmic lobes which projected forward of the spermatid head were formed by preferential flow of cytoplasm into one aspect of the cytoplasmic hood. In both species, at sperm release the cytoplasmic lobe was disengaged from the spermatid head to form a large residual body that was internalized and degraded within the Sertoli cell. Medium-sized cytoplasmic lobes were pinched from the head and neck region of the turtle and rooster spermatids, respectively. In the turtle, small-sized mitochondrial-rich cytoplasmic fragments budded from the caudal head and midpiece of the spermatids and were phagocytosed by the Sertoli cell. Thus, cytoplasmic elimination occurred through (1) segregation of cytoplasmic packets by Sertoli penetrating processes (turtle), (2) elimination of large and medium-sized residual bodies from the head (turtle and bird), and (3) budding of small mitochondrial-rich cytoplasmic fragments from the region of the midpiece (turtle). In the turtle a 79% reduction in total cell volume occurred during spermiogenesis which was the result of an 84% cytoplasmic reduction and a 78% nuclear reduction. During spermiogenesis, the rooster lost 97% of its total cell volume due to a 97% cytoplasmic volume change and a 96% nuclear volume change.

    Notizen: The process involved in the reduction of both nuclear and cytoplasmic volume was investigated in the bluegill (Lepomis macrochirus), a teleost fish. Young spermatids contained centrally positioned nuclei which, with time, moved toward the cell surface to become eccentrically positioned. Chromatin condensation was initiated from a region near the implantation fossa, whereas at the opposite pole of the nucleus an area sparse in heterochromatin (clear area) was noted. The nuclear membrane lying adjacent to the clear area dissolved and subsequently reformed, yielding a nucleus with a reduced volume. During this process, packets of cytoplasm surrounded by a double membrane were formed along the future midpiece. The packets of cytoplasm migrated toward the cell surface, protruded from the surface, and were extruded into the spermatocyst lumen. These structures, termed residual bodies, were subsequently endocytosed, accumulated into large phagocytic vocuoles, and eventually degraded by the nearby Sertoli cell. When the spermatocyst ruptured, spermatozoa containing sparse cytoplasm were released into the excurrent duct system. During spermiogenesis, both the nuclear and cytoplasmic volumes decreased substantially (80%, 92% respectively) leading to an overall 87% reduction in total cell volume.

    Notizen: Light and transmission electron microscopy of the spermatozoa and spermatogenesis of 16 species (in three genera, Patella, Helcion, Cellana) of patellid limpet have shown that head lengths of the sperm range from 3 to 13 μm, and each species has a sperm with a unique morphology, indicating that the spermatozoa can be used as a taxonomic character. Although spermatozoon structure is species specific, five types can be recognized, based on the size, shape, and structure of the nucleus and acrosome. The occurrence of five morphological types of sperm, one of which (Cellana capensis) is particularly different from other patellids, suggests that the taxonomy of the family Patellidae be re-examined. The morphological changes that occur during spermatogenesis are very similar in all species, although two patterns of chromatin condensation are found. Those species with sperm that have short squat nuclei (length:breadth 〈 3.5:1) have a granular pattern of condensation. Species with sperm that have more elongate nuclei (length:breadth 〉 5:1) have an initial granular phase followed by the formation of chromatin fibrils. These fibrils become organized along the long axis of the elongating nucleus. The absence of a manchette suggests that nuclear elongation is brought about from within the nucleus.

These characteristics are discussed in functional terms and correlated with the diving ability of the species.

    Notizen: Tadpoles of Hyla lanciformis live in midwater and have a normal, beaked, anteroventral, subterminal mouth. Overall, they are generalized pond-type anuran larvae that show no specific modification for a particular habitat. The chondrocranium and ossification sequences of this hylid frog were studied, utilizing cleared and Alcian blue-alizarin red-stained tadpoles. The chondrocranium is similar to that of other unspecialized tadpoles. No larval otic process was found and a large process projecting from the otic capsule is considered homologous with the larval crista parotica described for other anurans. The pattern of cranial and postcranial ossification is reported. The sequence of cranial ossification is explained in the framework of functional changes of the skull through metamorphosis. Comparison with the available information for other hylid frogs shows clear differences in the ossification sequences. The sequence of postcranial ossification is described for the first time for any species of hylid frog.

The tongues of aquatic species have small lingual papillae or lack them entirely, while terrestrial species have tongues with numerous glandular papillae. The pharynx and the esophagus also have papillae in some species. These either facilitate swallowing in which case they are long, pointed, keratinized, and occur commonly in marine turtles, or they are vascular and nonkeratinized, facilitate respiratory gas exchange and are found in the Trionychidae, Dermatemyidae, and Carettochelyidae. The morphology of the buccopharyngeal mucosa of turtles reflects their diet, feeding behavior, habitat, and relationships. Convergence in the morphology of the buccopharyngeal mucosa occurs among families, especially among the Emydidae and other familes of turtles. Intergeneric parallelism is also seen within the Emydidae.

    Notizen: In uloborid spiders, eye loss is accompanied by increased visual angles, optical material investment, and potential visual acuity of the retained eyes. Relative to carapace volume, the six-eyed Hyptiotes cavatus and two four-eyed Miagrammopes species have greater retinal hemisphere areas and lens volumes than do the eight-eyed uloborids Waitkera waitkerensis, Uloborus glomosus, and Octonoba sinensis. In Waitkera, in which the eyes have little visual overlap, and in Miagrammopes, in which eye loss simplifies the spiders' patterns of visual overlap, increased retinal cell density enhances potential visual acuity. However, this occurs at the expense of potential retinal cell sensitivity.

    Notizen: The anterior vasculature of the pony eye was examined by the corrosion cast method. The anterior segment of the pony eye has a vascular pattern which is similar but distinct from that of other mammalian species. Large iridal veins collateralized directly with the anterior vortex venous system. The intrascleral plexus was present but formed a fine, radially oriented, extensive network. This contrasted to the circumferential larger-diameter intrascleral plexus noted in dogs and the canal of Schlemm present in primates. The intrascleral plexus only drained posteriorly in the pony as compared to that in other species where anterior collateralizations have been described. The pattern of aqueous outflow was found to have relevance clinically in veterinary medicine and could provide insight into the frequent occurrence and treatment of equine recurrent uveitis as well as diseases of the posterior segment.

    Notizen: The development of the sensory epithelium of the saccular macula of Opsanus tau was studied with transmission electron microscopy. In the 10-12 somite embryo all cells of the newly formed otocyst are morphologically undefined, having an apically placed cilium with an underlying basal body and parabasal body. Junctional complexes are characterized primarily by tight junctions and a few desmosomes. In the 17-somite embryo the sensory cells begin to differentiate and are definable by the development of microvilli, which lack a cuticular plate. When the embryo has approximately 25-30 somites, ganglion cells differentiate and send their nerve processes toward the thin, disrupted basal lamina and the developing rhombencephalon. Desmosomes are more definable in the sensory regions at this age.As the myotomes begin forming (approximately 5-8 days before hatching), the nerves invade the sensory epithelium, and the developing sensory cells contain dense bodies surrounded by clear, membrane-bound vesicles. Clear synapticlike vesicles are also found throughout the infranuclear region of the sensory cells. However, afferent fibers lack a postsynaptic density. Three to 6 days prior to hatching a cuticular plate begins forming under the ciliary bundles and support and peripheral cells begin to morphologically differentiate. Two to 4 days before hatching the cuticular plate is well formed, desmosomes are numerous, afferent synapses and complete, and the sensory cells are in the upper two-thirds of the epithelium. Seven to 10 days after hatching, sensory cells have efferent synapses and ganglion cells and nerves show a myelin coat. These results suggest that sensory cells begin their development prior to VIIth nerve innervation, although the orientation and pattern development of these cells may be related to the formation of the cuticular plate, desmosomes, afferent innervation, and basal lamina formation.

    Notizen: Two groups of external excretory pores associated with glandular units (AU and LPU) were observed on the labrum, one pair laterally and three pairs posteriorly. Each external pore leads to an underlying conical, flask-shaped epidermal chamber. The wide base of this chamber is perforated by an internal pore that delivers secretions from the excretory duct of a glandular unit. The chambers serve to protect the internal pores from turbulence in the outside environment. Expulsion of secretions from the chambers is probably brought about by contraction of labral striated muscles, which synchronizes opening of the AU and LPU pores. A complex funnel-shaped structure forms the internal end of the excretory duct between each chamber and the corresponding pole of accumulation for the secretory product of a glandular unit. This structure, composed of an epidermal syncytium lined by a sleeve of several aligned auxiliary cells, probably ensures a tight connection between the epidermal chamber and the syncytium. The dorsalmost glandular units (LDU) have no pores in the vicinity of their poles of accumulation. Instead they secrete through cuticular ducts delimited by aligned auxiliary cells. External pores for these canals have not yet been located. The secretions of lateral pores may be mucopolysaccharides that play an essential role in agglutination of food particles soon after capture, while the secretions of posterior pores may contain glycoproteins that mix with food only after ingestion into the buccal cavity and probably start the process of digestion.

    Notizen: The process of metamorphosis in tiger salamanders, Ambystoma tigrinum, is used to investigate motor pattern conservatism in vertebrates. Specifically, we examined cranial muscle activity to determine if changes in the motor pattern are correlated with the morphological or environmental changes that occur at metamorphosis.Twenty-three variables were measured from electromyographic recordings from six cranial muscles in 13 tiger salamanders. These variables described the configuration of the motor pattern: the peak amplitude of activity, duration, relative onset, and time to peak amplitude were measured for each of the six muscles. Univariate and multivariate statistical analyses showed that there was no change in the mean motor pattern associated with the morphological transformation at metamorphosis: larval and metamorphosed individuals feeding in the water have very similar motor patterns. This was true despite significant morphological changes in the design of the feeding mechanism at metamorphosis and despite a significant decrease in aquatic feeding performance following metamorphosis.There was a change in the mean motor pattern to jaw muscles when metamorphosed individuals fed in water and on land: metamorphosed terrestrial feedings tend to have longer bursts of muscle activity then do aquatic feedings. The environmental changes in the motor pattern cannot be attributed to effects of differing fluid density or viscosity between water and air and are instead related to the shift to feeding by tongue projection on land.The decrease in aquatic feeding performance that occurs after metamorphosis is not correlated with changes in the motor pattern. Instead, the results suggest that changes in behavioral performance during ontogeny are associated with the transformation of hydrodynamic design of the feeding mechanism from uni- to bidirectional, and that motor patterns driving complex rapid behaviors may be conserved when behavior is altered by changes in peripheral morphology.

    Notizen: Salamanders in the family Plethodontidae show a unique behavior (nose-tapping) and have unique structures (nasolabial grooves) that may be used specifically to convey chemicals to the vomeronasal organ. The nasal structure of Plethodon cinereus was studied to determine if there is enhanced development of the vomeronasal organ compared with other salamander families that would correlate with use of these unique features. The vomeronasal organ in salamanders is found in a ventrolateral diverticulum of each main olfactory organ. P. cinereus has a more anteriorly placed vomeronasal organ within the diverticulum, and the posterior limit of each nasolabial groove is adjacent to the anterior limit of the vomeronasal organs. This suggests that the grooves deliver chemicals preferentially to the vomeronasal organs instead of to the main olfactory organs. In addition, the vomeronasal sensory epithelium is thickest anteriorly and is at its thinnest at about the level corresponding to the location of the vomeronasal organ in other salamander families. These adaptations suggest a specific mechanism of odorant delivery to the vomeronasal organ in plethodontid salamanders not found in other salamander families.

    Notizen: Light microscopy (LM) and scanning (SEM) and transmission electron microscopy (TEM) disclose seven types of hemocytes in adults of the German cockroach, Blattella germanica (Dictyoptera: Blattellidae) - namely, prohemocytes (PR), granulocytes (GR), plasmatocytes (PL), spherulocytes (SP), adipohemocytes (AD), coagulocytes (CO), and oenocytoids (OE). Their shape, size, surface appearance, and internal ultrastructure are characterized here. The shapes and relative sizes of hemocytes noted in vitro have been compared with those flowing through the wing veins (in vivo). The GRs constitute more than 90% of the hemocyte population and have roles in the defense system such as encapsulation and phagocytosis. They are characterized by numerous cytoplasmic granules and marginal microtubular bundles, which are commonly noted in the blood cells of both vertebrates and invertebrates, but have been rarely reported in insect hemocytes. The PRs are characterized by being spherical and small, having a relatively large nucleus and a small area of cytoplasm. The polymorphic PLs have some micropapillae on the cell surface and fewer granules than the GRs. The SPs are characterized by an oblate shape, raspberrylike appearance in SEM, and many spherules in the cytoplasm. The ADs are spherical and have large, refringent fat droplets. The COs possess a distinctive cartwheellike appearance when observed by LM but resemble GRs in TEM preparations. The OEs are opaque and often have an eccentric nucleus. Of the seven types of hemocytes, all, except the COs, can be recognized both in vivo and in vitro by LM; the COs can be recognized only in vitro. SEM does not allow the COs to be distinguished from the GRs, except in samples in which coagulation has occurred. All hemocyte types can be recognized by TEM. Thus, all three types of microscopy allow the various types of hemocytes to be recognized; unequivocal discrimination among the cells is best achieved by combining all the three kinds of microscopy.

    Notizen: The role of actin filaments in the development of cellular shape in the mesenteric mesothelium of the bullfrog was studied by using a simple, new technique for making en face preparations of mesothelial sheets. By using these mesothelial cell preparations, the distribution of actin was determined by means of fluorescence microscopy with 7-nitrobenz-2-oxa-1,3-diazole (NBD)-phallacidin and that of myosin by means of immunofluorescence microscopy. Although fluorescence produced by both NBD-phallacidin and antimyosin staining was found exclusively along the margins of the cells, its intensity was altered in correspondence with changes in cell shape. For instance, tadpole-type mesothelial cells with either and irregular or very slender cell shape showed very weak fluorescence. On the other hand, frog-type mesothelial cells with a polygonal shape showed intense fluorescence at their margins and had circumferential bundles of acting filaments at their apices. Furthermore, intercellular junctions between the mesothelial cells developed as the cell shape became polygonal during metamorphosis. The present study showed that development of circumferential bundles of acting filaments and intercellular junctions may serve to establish and maintain the definitive polygonal cellular pattern in the mesenteric mesothelium of the bullfrog.

    Notizen: Extant bovids inhabit a wide diversity of environments that range from forest to savanna and display locomotor patterns that are habitat specific. I report here on an investigation of the linkage between these locomotor patterns and habitat based on a study of the morphology of the bovid femur. Femoral head shape, shaft dimensions, and knee structure are examined and support a statistically significant separation of the different morphological complexes present in bovids from forest, broken cover, and savanna habitats. Morphological differences are primarily related to locomotor patterns as reflected in the degree of cursoriality displayed by bovids in different habitats. Cursorial bovids from savanna environments have laterally expanded femoral heads that act to limit the degree of abduction and axial rotation at the hip, and elliptically shaped distal femora that increase the moment arm of the extensor muscles that cross the knee. Forest bovids have spherically shaped femoral heads. This morphology permits a much higher degree of abduction and axial rotation at the hip and appears to provide greater maneuverability in a vegetationally complex habitat. Bovids living in broken cover environments that fall between the extremes of closed canopy forest and savanna display an intermediate set of femoral characters. This approach to the relationship between habitat and locomotion offers a potentially powerful means with which to examine the interplay between structural form and function in bovid evolution.

    Notizen: In Hydra viridis, cordons of male germ cells lie in gonadal compartments, which are enlarged spaces between the elongated and “spongy” epidermal cells. The germ cells are surrounded by these cells, except for small areas where the interstitial cells and spermatogonia are in direct contact with the mesoglea. Cells from both epidermis and gastrodermis project cytoplasm into the mesoglea, where they contact each other and form trans-mesogleal bridges. The latter exhibit gap junctions, which are particularly abundant at the spermary region. Here, the mesoglea is thinner then elsewhere in the body. Both epithelia are joined by septate junctions toward their apical ends, which are totally impermeable to horseradish peroxidase (HRP). HRP gained entry to the cells of both epithelia by pinocytosis. Incorporation into the cells was high at the basal disk, in the tentacles, and in the mesoglea in the lower part of the body stalk. The tracer was never found within the gonadal space of the testis during spermatogenesis. In mature spermaries during spermiation, tracer-filled intracellular vacuoles fused with the gonadal spaces as the thin cytoplasmic columns of the epidermal cells ruptured; HRP thus gained access to the germ cells.During spermatogenesis, germ cells of Hydra viridis are in a closed compartment. The barrier that controls the access of metabolites to the germ cells is formed by epidermal cells, thinned-out mesoglea, and numerous transmesogleal interepithelial bridges. The presumed role of the barrier is the control of the environment (1) where interstitial cells are differentiating into spermatogonia and meiosis occurs and (2) in which ripe spermatozoa are kept immotile until spermiation.

    Notizen: The isopod Sphaeroma hookeri and many other isopods and peracarids have a sensory spine with laterally inserting sensory hair, positioned in the apical region of the propodal palm of pereopod 1. This spine is innervated by five to eight sensory cells (each giving rise to one cilium) the dendrites of which can be divided into an inner and outer dendritic segment. The cilia are surrounded by an extracellular, electron-dense dendritic sheath. Thirteen enveloping cells are present. The outer dendritic segment (structure beyond the basal bodies) contains two receptor lymph cavities; the inner one lying within the dendritic sheath is homologous with the inner receptor lymph cavity of insects.Scolopales, or tubular bodies, are lacking; their function is probably accomplished by the dendritic sheath. Apically the sensory hair does not have a pore, and the spine is heavily sclerotized. The inner dendritic segment begins with a basal body from which rootlets of different length and thickness extend into the dendrite. In the latter is an accumulation of vesicles. The dendrites keep close contact with other dendrites and the enveloping cells by desmosomal membrane structures. The possible importance of the sensory spine for phylogenetic studies is discussed.

    Notizen: The nephron of the one-humped camel Camelus dromedarius was investigated by light and transmission electron microscopy. Besides the many features common to other mammalian kidneys, the nephron of the camel is unique in having an unusually thick basal lamina underlying the epithelial cells of the nephron, the thickest being found in part of the parietal layer of Bowman's capsule and the thin limb of the loop of Henle. In the latter, the membrane usually appears lamellated and contains numerous tiny vesicles. In other parts of the nephron, the basal lamina usually has a homogenous appearance. The possible significance of the thickening of the basal lamina is discussed in relation to the general high renal efficiency of the camel.

    Notizen: The fine structure of abembryonic and mural trophoblast cells of mouse embryos was analyzed during the initial stages of invasion of the endometrial stroma by the embryo (days 6-8 of pregnancy). On day 6 of pregnancy, most trophoblastic cells are flat and have spindle-shaped nuclei. A few large, round trophoblastic cells (giant cells) are present at the abembryonic pole. As pregnancy proceeds through days 7 and 8, the area occupied by the trophoblast becomes larger because of an increase in the trophoblastic cell population, growth of giant cells, and rearrangement of the latter cells into a network containing maternal blood. As flat cells transform into giant cells, their content of ribosomes, granular endoplasmic reticulum, Golgi complexes, lysosomelike bodies, and heterophagosomes increases. Reichert's membrane is always lined by cell bodies or by laminar processes of trophoblastic cells that are provided with small pores. Transformation of flat cells into giant cells is associated with an activation of the giant cells and their acquisition of invasive behavior.

    Notizen: The ultrastructure of the Malpighian tubules of the adult desert locust, Schistocerca gregaria, is described. Male and female adults possess about 233 tubules, which empty proximally into the midgut-ileal region of the alimentary canal by way of 12 ampullae. The tubules vary from 10 mm to 23 mm in length. About one third of them are directed anteriorly, attaching distally at the caeca, while the remainder are directed posteriorly, attaching to other tubules, the rectum or large tracheal trunks adjacent to the hindgut. The Malpighian tubules from all locations examined consist of three ultrastructurally distinct regions: proximal, middle, and distal, referring to their position relative to the midgut. All cell types possess ultrastructural features characteristic of ion transporting tissue, i.e., elaboration of the basal and apical membranes and a close association of these membranes with mitochondria. The distal and proximal segments are short (1.5-1.7 mm) and heavily tracheated, and each is composed of a single, distinct cell type. The middle region is the longest segment of the Malpighian tubule and is composed of two distinct cell types, primary and secondary. Both cell types are binucleate. The more numerous primary cells have large nuclei, contain laminate concretions in membrane-bound vacuoles, and possess large microvilli that contain mitochondria. The secondary cells are smaller and possess smaller nuclei. The microvilli are reduced and lack mitochondria. Secondary cells do not contain laminate concretions. The possible compartmentalization of ion and fluid transport function based on segmentation in the Malpighian tubules is discussed.

    Notizen: The morphology of the middle ear region including the basicranium and quadrate of tinamous is compared among ratites and flying birds belonging to the Procellariiformes, Sphenisciformes, Pelecaniformes, and Ciconiiforms. The middle ears of tinamous and ratites share a number of important characters including absence of a separate foramen for the glossopharyngeal nerve; eustachian tube, carotid artery, and stapedial artery encased in bone; and a metotic process with vascular canals or notches. Outgroup analysis confirms these characters as synapomorphies. These data support the position that the Tinami and Ratiti form a monophyletic assemblage.

    Notizen: The colonial marine bryozoan Membranipora membranacea produces a planktotrophic cyphonautes larva that is encased by a triangular bivalved shell. Following a relatively long free-swimming phase, the cyphonautes larva settles and undergoes a dramatic metamorphosis to become the sessile progenitor of the colony, referred to as the ancestrula. This paper examines the initial morphogenetic movements of metamorphosis that transform the cyphonautes larva into an incipient ancestrula. At the onset of metamorphosis, contractions of the striated median muscles situated along the anterior and posterior margins of the larva cause a retraction of the larval apical organ and a centripetal movement of the anterior and posterior ends of the larva. Concurrently, the ciliated corona at the base of the larva is pulled within the shell by contractions of the striated lateral muscles. As the larva assumes a more spherical shape, the posterior margins of the shell are spread apart, and the internal sac is everted. Eversion of the sac is apparently achieved by contractions of the lateral muscles that cause a buckling of the shell in the apical-basal direction. The neck region of the everted sac secretes adhesive granules that attach the larva to the substratum. Subsequently, contractions of the nonstriated sac muscles fold the valves of the shell over each other and draw the larva toward the substratum. The initial events of metamorphosis that culminate in the attachment and flattening of the larva are completed in 10-15 seconds. In the subsequent few minutes, the lateral edges of the everted sac fuse with the neighboring margins of the aboral epithelium underlying the shell and thus form the fully sealed body wall of the incipient ancestrula.

    Notizen: Available evidence provides little support for a recent proposal that the term “trophoblast” be applied solely to eutherian mammals. Arguments for such a restricted usage are based on a dichotomous interpretation of therian reproduction that underestimates the developmental, structural, and functional diversity of trophoblastic tissues occurring within the infraclass Eutheria. The occurrence of developmental patterns that are phenotypically intermediate between those of commonly studied eutherians and metatherians suggests that blastocyst development is not fundamentally different in marsupials and eutherians.The trophoblast of marsupials accomplishes most or all of the major functions of the eutherian trophoblast, including maternal-fetal physiological exchange, implantation, contribution to placental membranes, steroid metabolism, and possibly, immunological protection of the conceptus. Furthermore, application of the term “trophoblast” to marsupials is consistent with present and past usage, as well as with the original definition and etymological derivation of the term. Therefore, we recommend that the term “trophoblast” continue to be applied in a functional-morphological sense to the appropriate extraembryonic tissues of marsupials. Such use of functional (rather than taxonomic) criteria for application of this term avoids biasing interpretations of mammalian reproductive evolution.

    Notizen: Intercellular bridges joining cells contained in cysts of Chortophaga viridifasciata testes were studied with light and electron microscopy. Preparations consisted of expressed whole cells (living, or fixed and stained) as well as sections. The secondary spermatogonia of each cyst are joined centrally by persisting fused interzonal bodies (fusomes) of incompletely cleaved cells. Shifts in cell orientation during anaphase are apparently responsible for central as opposed to chain linkage of cells. In the primary spermatocytes, the central fusome is replaced by a chain linkage, apparently resulting from the breakdown of the fusome into its original interzonal body components. Intercellular bridges are also present in spermatids, but there is no evidence to indicate the time of their formation (in the immediately preceding meiotic divisions or in the secondary spermatogonial divisions). The function of the compact centrally situated fusome in the secondary spermatogonial cyst is discussed as it relates to synchrony, number of cell divisions, spermatodesm formation, and fertility.

    Notizen: Using laboratory-grown colonies of Plumatella emarginata, the formation of the floatoblast and the sessoblast was studied. Both types of statoblast develop in the funiculus. Toward the termination of development, the floatoblast secretes a gas and the float chambers are filled with the gas in about 20-30 minutes. The floatoblast thus complete is separated from the funiculus. Until early epidermal-disc-stage, distinction between a floatoblast and a sessoblast is impossible at least morphologically. Toward the late epidermal-disc stage, a future sessoblast becomes larger than a future floatoblast and attaches by its cystigenic side to the cystid. Very often it initially attaches to a lateral wall, then migrates to the basal wall of the cystid. Both the attachment to and the migration along the cystid wall are attained by peritoneal cells covering the sessoblast, specifically by those in the marginal zone of the cystigenic side. The sessoblast is separated from the funiculus precociously, shortly after attachment to the cystid. Then, it produces the capsule, followed by the formation of both the lamella, a homologue of the float in the floatoblast, and the attaching apparatus.Almost all polypides produce floatoblasts (up to 17 in number), but only a small portion of them produce both floatoblasts and sessoblasts. The number of sessoblasts produced by a single polypide is usually not more than 3 but occasionally reaches up to 6. When multiple sessoblasts are formed by a polypide, they are as a rule derived from primordia located adjacently on the funiculus, accordingly successively in a short period, but their arrangement on the basal wall of the cystid does not always correspond to that on the funiculus. Sessoblast formation is never associated with the death of the mother polypide. Ancestrulae derived from statoblasts never produce mature statoblasts, though may undergo gametogenesis. Several mosaic statoblasts consisting of floatoblast- and sessoblast-portions were found in some species of the Plumatellidae. A primordium of the statoblast seems to have a dual ability of differentiating into either a floatoblast or a sessoblast; but little has been known about the mechanism or factors controlling the formation of these two types of statoblasts.

    Notizen: The giant anterior salivary gland cells from the large mammalian blood-sucking, glossiphoniid leech, Haementeria ghilianii, can be subdivided into three morphologically and functionally distinct regions: (1) a soma, responsible for the synthesis and storage of secretory products; (2) a long cell process, responsible for the storage and intracellular transport of the secretory vesicles; and (3) the site of exocytosis at the process terminal. The giant somata are densely packed with secretory vesicles. Deep plasmalemmal invaginations invade the soma and form an extensive system of extracellular lacunae. The rough endoplasmic reticulum (ER) and the Golgi apparatus are organized in the cell periphery, near the highly branched nucleus, and along the lacunae. The somata taper into long processes extending over several centimeters to the proboscis tip. These contain secretory vesicles through their whole length. In the process periphery, the vesicles are completely ensheathed by a concentric subplasmalemmal smooth ER cisterna. This originates deeply within the soma and extends through the whole cell process to its terminal. The ER provides support for up to several hundred longitudinally oriented microtubules. Secretion occurs at the very tip of the cell processes, each of which terminates at the proboscis tip at the base of a cuticular pore.We found synapses close to the sites of exocytosis, providing morphological evidence for neuronal control of secretion.

    Notizen: Two fast-twitch fiber types are histochemically identified in the primary flight muscles of Artibeus jamaicensis. These are classified as type IIa and IIb according to an acid-preincubation staining protocol for myosin ATPase. All fibers in the bat flight muscles exhibit relatively intense staining properties for NADH-TR, suggesting a high oxidative capacity. The glycolytic potential of all fibers is rather low, as assessed by stains for alpha-GPD. This two-type histochemical profile appears to parallel biphasic electromyographic patterns observed in these muscles and leads us to propose that flight muscle histochemistry and activation are mediated by a “two-gear” neuromuscular control system. In contrast, earlier studies on Tadarida brasiliensis demonstrate the existence of a “one-gear” neuromuscular control system, exemplified by the presence of one fiber type. These observations are discussed with respect to the natural history and flight styles of several species.
