Chemical Synthesis of Deoxyoligonucleotides with 5′-Phosphate Group on a Polymer SupportTwo new methods are described for attaching nucleotides via the 5′-phosphate group to a polymer support: The first procedure uses a ribocytidine as connecting link; cleavage is carried out by hydrolysis with trifluoroacetic acid, periodate oxidation and β-elimination. The principle of the second method is the reaction of chloromethyl polystyrene with a nucleoside-5′-phosphorothioate leading to S-alkyl-5′-phosphorothioate; cleavage occurs under mild conditions with iodine/water.Both concepts were successfully used for synthesizing of the 5′-phosphate containing oligonucleotides (pdT)5, pdTpdApdT, pdTpdCpdT, and pdTpdGpdT.

Zwei neue Methoden zur Verknüpfung von Nucleotiden über die 5′-Phosphatgruppe mit einem polymeren Träger werden beschrieben: Im einen Falle dient ein Ribocytidin als Zwischenglied und erfolgt die Wiederabspaltung durch Hydrolyse mit Trifluoressigsäure, Perjodat-Oxidation und β-Eliminierung, im zweiten Falle wird ein Nucleosid-5′-phosphorothioat mit einem Chlormethylpolystyrol unter S-Alkylierung umgesetzt, und die Abspaltung gelingt unter milden Bedingungen mit Jod/Wasser.Die Brauchbarkeit beider Konzepte für die Darstellung von Oligonucleotiden mit 5′-Phosphatgruppe wurde durch die Synthese der Oligonucleotide (pdT)5, pdTpdApdT, pdTpdCpdT und pdTpdGpdT demonstriert.
