    Notes: We developed a two-dimensional spatial resolved high-speed UV sampling camera (HISAC) to study energy transport in laser-produced plasmas. The HISAC is composed of an optical bundle of fibers coupled to a streak camera to obtain a two-dimensional spatial resolution with a temporal resolution of less than a few 10 ps. This HISAC system was demonstrated in the experiments on inferred uniformity measurements of laser-ablation pressures as well as on energy transport in ultraintense laser-plasma interactions. Two-dimensional shock heating was temporally resolved in a hemispherical target, resulting in the distribution of the ablation pressure as a function of laser incidence angle. © 1999 American Institute of Physics.

    Notes: A toroidal electrostatic analyzer with a two-dimensional position-sensitive electron detector has been developed for the measurement of differential excitation cross sections in electron-ion collisions. The electron position is resolved simultaneously in the energy and angular dispersive directions of the analyzer. The present electron analyzer is specially designed with a narrow curved entrance slit and a wide exit slit coupled with an exit lens system after the toroidal deflector, which can give a wide energy dispersed range of 20% of the mean pass energy and a large angular range extending from 30° to 120°. The performance of the spectrometer has been studied by measuring the scattered electrons from Ar atoms and from a highly oriented pyrolytic graphite surface, indicating the usefulness of the present analyzer in electron spectroscopy. A brief description of the method for a spectrum transformation from Cartesian coordinates to polar coordinates is also presented. © 1999 American Institute of Physics.

    Notes: We developed a 19.6 nm laser x-ray laser grid-image refractometer (XRL-GIR) to diagnose laser-hole boring into overdense plasmas. The XRL-GIR was optimized to measure two-dimensional electron density perturbation on a scale of a few tens of μm in underdense plasmas. Electron density profiles of laser-produced plasmas were obtained for 1020–1022 cm−3 with the XRL-GIR and for 1019–1020 cm−3 from an ultraviolet interferometer, the profiles of which were compared with those from hydrodynamic simulation. By using this XRL-GIR, we directly observed laser channeling into overdense plasmas accompanied by a bow shock wave showing a Mach cone ascribed to supersonic propagation of the channel front. © 1999 American Institute of Physics.

    Notes: Production of tissue factor (TF) in response to lipopolysaccharide (LPS) was examined in human umbilical vein endothelial cells (HUVECs) transfected with human CD14 DNA. The expression of CD14 on HUVECs dramatically enhanced the production of TF at a low concentration of LPS in the absence of fetal calf serum (FCS). On the other hand, mock-transfected HUVECs did not respond to even a high concentration of LPS. TF production in CD14-expressing HUVECs was significantly inhibited by anti-CD14 monoclonal antibody. Addition of FCS to the culture of CD14-expressing HUVECs markedly augmented the LPS-induced TF production, whereas only a marginal effect was observed in mock-transfected HUVECs. The findings suggested that the integration of membrane CD14 rendered HUVECs highly sensitive to LPS in the production of TF irrespective of the presence of FCS.

    Notes: Abstract. The time courses of fecal excretion and tissue distribution of metabolites derived from 2,2′,4,5,5′-pentachlorobiphenyl (CB101) and 2,2′,3,4′,5′,6-hexachlorobiphenyl (CB149) were investigated in male Wistar rats. The metabolism of both congeners involved primarily hydroxylation at the 3-position, and methylthiolation at the 4-position. Metabolites distributed in tissue were dominated by different ratios of 3- and 4-methylsulfonyl (MeSO2) metabolites. The 3-/4-MeSO2 metabolite ratios in liver and adipose tissue for both congeners were 0.41–0.61 at day 4, and then increased to 0.85–1.00 for up to day 42. In contrast, the ratios in lung were 0.03–0.04, and then decreased to 0.01. Compared to the unchanged PCBs at day 42, the distribution ratios of 3-MeSO2 metabolites were greater in the order of liver (0.46 for CB101 and 0.21 for CB149) 〉 kidney 〉 blood 〉 lung 〉 adipose tissue, whereas those of 4-MeSO2 metabolites were in the order of lung (9.50 for CB101 and 4.00 for CB149) 〉 kidney 〉 blood 〉 liver 〉 adipose tissue, indicating the different binding affinity of 3-MeSO2 metabolites in liver from that of 4-MeSO2 metabolites in lungs of rats. Furthermore, the structure-tissue affinity relationship for 3-MeSO2 metabolites was investigated, following the administration of 11 3-MeSO2-PCB congeners to rats. The results indicated that the retention potential of 3-MeSO2 metabolites in the liver largely depends on the ortho-chlorine substitution in the biphenyl ring rather than the degree of chlorination.

    Notes: Abstract An exceedingly rare case of prenatally diagnosed cystic neuroblastoma (NB) is described. Prenatal ultrasonography at 28 weeks' gestation revealed a cystic right suprarenal mass that measured 15 × 17 mm in diameter. The mass increased parallel to fetal size until it was 41 × 44 mm in diameter at 39 weeks' gestation. Magnetic resonance imaging performed prenatally and after delivery showed two different intracystic intensities with fluid levels, suggesting intracystic hemorrhage. The patient underwent a right adrenalectomy at 20 days of age, and the resected tumor was diagnosed as a poorly-differentiated ganglioneuroblastoma. Eight months after surgery, the patient is generally healthy with no evidence of recurrence. We reviewed 25 additional cases of prenatally diagnosed adrenal cystic NB and examined the clinical features of this rare entity.

    Notes: Abstract  Five hundred and thirteen cases of gastric carcinoma were investigated for the presence of viral RNA, and the clinico-pathological data, geno-type, BamHIF restriction fragment polymorphism (RFLP) and specific LMP-1 30 bp gene deletion were also examined. EBVs detected in lymphocytes in 20 normal gastric mucosa, 7 lymphoma cell lines (LCLs) maintained in severe combined immunodeficiency (SCID) mice and 18 non-Hodgkin’s lymphomas were compared with those in the gastric carcinoma cases. Thirty-three cases (6.4%) were demonstrated to be positive for EBV by means of EBER-1 RNA in situ hybridization. Clinico-pathological data showed no statistically significant difference in histological grading, location of cancer and status of vessel and lymphatic invasion between the EBV-positive and -negative groups, although the former significantly predominated in the submucosal invasion group (submucosal vs mucosal P=0.021; submucosal vs advanced cancer P=0.033). Some of these data were different from corresponding data in earlier reports. In cases that were evaluated by molecular biology, type A, wild-type F and LMP-1 gene deletion predominated except one in 21 informative cases, one in 24 and two in 16, respectively. EBVs detected in lymphocytes in normal gastric mucosa, LCLs in SCID mice and non-Hodgkin’s lymphoma were also predominantly affected by type A, wild-type F and LMP-1 gene deletion with few exceptions. The results indicate a lack of genetic differences among EBVs in gastric carcinoma, normal population, LCLs of SCID mice and non-Hodgkin lymphomas. Some EBV infections in gastric carcinomas may be transient, especially in the submucosal invasion group.

    Notes: Abstract The effects of exogenous basic fibroblast growth factor (bFGF) on the repair of full-thickness cartilage defects were examined. Four-millimeter diameter, cylindrical defects were made in rabbit articular cartilage and were filled with human recombinant bFGF. The addition of bFGF to the defect induced the formation of a thick cartilage layer composed of chondrocytes and a metachromatic-stained matrix after 6 weeks. The score of the bFGF-treated tissue, as evaluated by a semiquantitative histological scale, was significantly higher than that of the untreated tissue. At 24 weeks, the cartilage-like matrix that contained the proteoglycans and type II collagen was thicker in the bFGF-treated tissue than in the untreated tissue. Immunohistochemical analysis of the tissues at 6–12 weeks with an anti-bFGF monoclonal antibody suggested that a single application of bFGF increased the number of differentiating chondrocytes that synthesized bFGF at a high level. In contrast, immunostaining of the tissues at 6–12 weeks with a monoclonal antibody against proliferating cell nuclear antigen showed that the number of proliferating cells in the bFGF-treated tissue was fewer than in the untreated tissue. These findings suggest that administration of bFGF into cartilagenous defects promotes the differentiation of chondrocytes and their matrix synthesis, and that this growth factor is useful for improving cartilage repair.

    Notes: Abstract Immunocytochemical examination for mitochondrial protein and cytochrome oxidase was performed to demonstrate oncocytes in normal adenohypophysis obtained from 28 patients of various age. A small number of solitary large epithelial cells showed intense cytoplasmic granular immunoreactivities for mitochondrial protein and cytochrome oxidase. The proportions of the cells positive for the former and the latter ranged from 0% to 5.9% (mean ± SD; 1.5 ± 1.7%) and from 0% to 4.9% (1.4 ± 1.6%), respectively. These cells were either absent or extremely rare in young patients (under 10 years) but tended to increase in number with age (P 〈 0.0001). On the other hand, the mirror section technique showed that most of these cells were negative for adenohypophysial hormones, but a few of them were faintly positive for: α-subunit (8.0%), β-subunits of follicle-stimulating hormone (4.8%), luteinizing hormone (2.5%), thyroid-stimulating hormone (1.0%), and growth hormone (0.5%), and were negative for prolactin and adrenocorticotropic hormone. We considered that these cells represent oncocytes that exist in varying numbers in normal adenohypophysis. It was suggested that oncocytes in normal adenohypophysis share various common features with tumorous oncocytes of pituitary oncocytomas.
