    Notizen: Summary The trace element content of kidneys has been documented in very few international centres. Significant differences can be demonstrated between the cortical and medullary levels of calcium, zinc and cadmium. For the West of Scotland the values for the latter are similar to those found in Scandinavia. There are significantly higher cadmium contents in subjects who smoke and who have evidence of ischaemic heart disease.

    Notizen: Summary After initial evaluation of a manual fluorescence microscopy system on a variety of urines the method was automated and subsequently tested in a population survey of urinary tract infection in schoolgirls. This automated Bactoscan system allowed a rapid analysis of urine samples and with the introduction of modifications to the staining protocol it correctly eliminated 91% of samples as being not significantly infected.

    Notizen: Summary The prevalence of urinary tract infection in a modern Scottish town has been ascertained in female school children aged 5–12. In 2,234 girls the initial prevalence of urinary tract infection is 3.3%. In addition to the information on urinary tract infection the social and personal history was available including data on incontinence, enuresis and current antibiotic therapy. The necessity to have a system available which allows a cheap and rapid assessment of population surveys is made.

    Notizen: Summary Cadmium induces a variety of effects on kidney tubules including hypercalciuria. This is accompanied by hyperzincuria. The present work shows that both of these biochemical abnormalities can be corrected but where a state of cadmium-induced-anaemia exists urine zinc and calcium excretion do not follow the expected parallel course in response to iron therapy as they do with bendrofluazide.

    Notizen: Summary Cadmium injection in rats affects the genito-urinary system. The renal effects are generally considered to be mainly tubular. Light microscopy of 29 specimens confirms cellular damage in the glomeruli with changes in glomerular capillaries. Electron microscopic changes in the glomeruli confirm changes in the podocytes, thickening of the basement lamina, the occurrence of electron dense deposits in association with the capillary endothelium with platelet and polymorphonuclear cell accumulations which suggests opening of the endothelial junction of the glomerular vessels.

    Notizen: Previous studies by Stephens and McNulty and Strecker and Stephens have demonstrated that foil barriers placed between the mesonephros and lateral plate at stages 12 to 15 inhibited limb development, but foil barriers placed between the neural tube and somites at stages 11 to 12 resulted in limbs with normal skeletal patterns. It was concluded that some influence present in the paraxial region of the embryo at stages 11 to 15 is necessary for normal limb development. The present study was undertaken to localize that influence more precisely. Foil barriers were placed in the lateral edge of the somites or segmental plate of stage 10 to 15 chick embryos. Barriers placed into stage 13 to 15 embryos resulted in chicks with normal limbs, but barriers placed into stage 10 to 11 embryos resulted in chicks with defective limbs. Barriers inserted just lateral to Hensen's node at stages 6 to 8 resulted in embryos with defective or absent wings. We also grafted stage 4 to 9 presumptive limb territories with and without Hensen's node. Explants without Hensen's node formed limb-like structures in 1% of the cases. Explants with Hensen's node formed limb-like structures in 27% of the cases. When barriers were implanted and a node was placed on the lateral side of the barrier, limbs formed in 40% of the cases. These data suggest a medial to lateral progression of some as yet unknown morphogenetic influence necessary for normal limb development and we hypothesized that the influence may initially emanate from Hensen's node.

    Notizen: The structure of human interleukin 4 (IL-4) was predicted utilizing a series of experimental and theoretical techniques. Circular Dichroism (CD) spectroscopy indicated that IL-4 belonged to the all α-helix class of protein structures. Secondary structure prediction, site-directed mutagenesis, and CD spectroscopy suggested a predominantly α-helical structure, consistent with a four-helix bundle structural motif. A human/mouse IL-4 chimera was constructed to qualitatively evaluate alternative secondary structure predictions. The four predicted helices were assembled into tertiary structures using established algorithms. The mapping of three disulfide bridges in IL-4 provided additional constraints on possible tertiary structures. Using accessible surface contact area as a criterion, the most suitable structures were right handed all antiparallel four-helix bundles with two overhand loop connections. Successful loop closure and incorporation of the three disulfide constraints were possible while maintaining the expected shape, solvent accessibility, and steric interactions between loops and helices. Lastly, energy minimization was used to regularize the chain.

    Notizen: Advent of the multichannel plate and position sensitive detector has made possible true single photon counting imaging tubes. We have investigated the application of these detectors in studies of the ultraweak light emission of biological materials. Initially, we focussed our efforts on two objectives: (1) obtaining single photon counting images of living tissues using only the light (chemiluminescence) emitted by the specimen and (2) developing means of obtaining well-resolved spectra of weakly emitting sources. We have obtained a variety of images. One striking result of this work is the first observation of tissue specific localization of photon emission in situ. Using this detector we have also obtained the first well-resolved spectra of some important ultraweak emission processes. These results illustrate the potential use of single photon imaging in bioluminescence and chemiluminescence research.

    Notizen: Scanning electron microscopy (SEM), especially low-voltage (1 KeV) high-resolution SEM, can be used in conjunction with stereo pair high-voltage (1 MeV) transmission electron microscopy (HVEM) of whole spread cells or thick sections effectively to correlate surface structure with internal structure. Surface features such as microvilli, pits, pseudopodia, ruffles, attached virus, and other surface-related morphologic characteristics can be identified using SEM, while underlying cytoskeletal structure and organelle organization can be viewed by HVEM of the same preparation. However, the need to “prepare” cells for electron microscopy precludes observation in the living state. The use of several types of video-enhanced light microscopy (VLM) permits observation of living cells such that certain surface and internal features can be observed at a relatively high level of resolution or detection. Thus, changes in living cells can be followed, and at appropriate times the cells may be chemically fixed or rapidly frozen and prepared for ultrastructural examination by electron microscopy.We have utilized VLM in conjunction with SEM and HVEM to correlate changes in shape and surface structure with changes in the internal structure of platelets. In addition, we have found it advantageous to use colloidal gold-labeling procedures, because these markers are detectable by all three forms of microscopy. Using this approach we have labeled platelet membrane GPIIb/IIIa, a receptor for RGD-containing adhesive proteins, with gold-fibrinogen or gold-anti-IIb/IIIa. The initial binding and subsequent movement of gold-fibrinogen-IIb/IIIa complexes in living platelets was followed by VLM. The movement of individual labels could be mapped. Subsequent observation by low-voltage (1 KeV), high-resolution SEM and HVEM permits visualization of the same individual receptors tracked by LM. The final position on the membrane or the position-in-transit when fixative was added was determined relative to surface ultrastructure (SEM) and internal, particularly cytoskeletal, ultrastructure (HVEM).
