    Notes: Tissues peripheral to well-developed papules of colloid milium were chosen for biopsy and electron microscopy examination. Evidence was presented that colloid is derived from elastic fibres through sequential degenerative changes. In the upper to middle dermis of the peripheral tissue, a number of abnormal masses in the vicinity of small colloid depositions were observed. Ultrastructurally, those abnormal masses were degenerating elastic fibres in which “electron-dense layers” of normal elastic fibre increased in amount, although their electron density diminished. Electron-light layers were gradually diminished in quantity. Degenerated elastic fibres finally became granulo-fibrillar and were indistinguishable from colloid. Many fibrils which closely resembled 10 nm tubular microfibrils of normal elastic fibre were observed in the granulo-fibrillar substance. Since one stage of the degeneration of elastic fibre which eventually leads to the formation of colloid milium was very similar to that of actinic elastosis, a direct role of sunlight in the formation of colloid milium is suggested.

    Notes: Regulation of acute-phase serum amyloid A (A-SAA) synthesis by proinflammatory cytokines and steroid hormones in human aortic smooth muscle cells (HASMCs) is distinct from that in HepG2 cells. To study the cis- and trans-activating promoter element involved in the SAA1 gene expression by HASMCs and HepG2 cells, we constructed plasmid vectors for luciferase reporter gene assay with varying lengths of SAA1 upstream regulatory region (up to 1431 bp), and examined their response to proinflammatory cytokines and/or steroid hormones. The corresponding vectors with the SAA4 upstream regulatory region served as controls. The presence of proposed transcriptional regulatory factors binding to these regions was confirmed immunohistochemically.The sequences of 1478 and 1836 bp of the SAA1 and SAA4 5′-flanking regions were determined, respectively. SAA1 promoter transcription in cultured HASMCs was upregulated not by proinflammatory cytokines, but rather by glucocorticoids. This differed from HepG2 cells, in which SAA1 promoter transcription was upregulated synergistically by proinflammatory cytokines and glucocorticoids. The promoter activity of a series of truncated SAA1 promoter constructs measured using the reporter gene assay showed that the 5′-region from −252 to −175, containing a consensus site for CCAAT/enhancer binding proteins α,β (C/EBPα,β), was essential for SAA1 induction in HASMCs. In HepG2 cells, the 5′-region from −119 to −79, containing a nuclear factor kappa-B (NFκB) consensus sequence, was essential for the induction. The functional significance of the C/EBP site as indicated by the immunohistochemical result was that in HASMCs anti-C/EBPβ reactivity was shifted from the cytoplasm to the nuclei.We have, therefore, demonstrated that the region containing the C/EBPα,β consensus binding site between the bases −252 and −175 is important for the glucocorticoid-induced SAA1 gene expression in HASMCs but not in HepG2 cells.

Pancreatic secretion has been monitored in the isolated, blood-perfused canine pancreas, and the effects of depletion of serum calcium by infusion of EGTA on the increases in secretion produced by intra-arterial injections of dopamine and secretin have been investigated.2. Under resting conditions in preparations in dogs fasted for 24 h, the mean rate of pancreatic secretion was 16.4 μ1/min (s. e. m. = 2, n= 12). The mean concentrations of protein, bicarbonate and chloride in the pancreatic juice were 53.8 mg/ml (s. e. m. = 4.5), 18.0 mmol/1 (s. e. m. = 1.1) and 122.5 mmol/1 (s. e. m. = 7.5), respectively. Infusion of EGTA had no effect on resting secretion.3. Secretion elicited by dopamine or secretin was diminished about 50% during the intra-arterial infusion of EGTA (10−2 mM/ml) in the perfusing blood. The protein concentration in the secretion was diminished to a similar extent. The concentrations of bicarbonate and chloride in the pancreatic juice was not modified by EGTA infusion.4. Concomitant infusion of an equimolar concentration of CaCl2 solution abolished the inhibitory effects of EGTA infusion on the secretory responses to dopamine or secretin.5. The results suggest that dopamine and secretin influence the exocrine secretions of the pancreas by actions on both acinar and ductular cells. Acinar cell secretion is more susceptible to depletion of serum calcium levels than is secretion from ductular cells.

    Notes: Summary Exercise-induced anaphylaxis (EIA) is a form of physical urticaria that is induced by exercise. A 16-year-old Japanese boy had a 4-year history of recurrent wealing and dyspnoea after physical exercise such as jogging, playing handball or riding a bicycle in winter. The episodes were not associated with ingestion of foods including wheat or soya bean. A provocation test, with 15 min of exercise and 2 min of cold stimulation immediately before or immediately after the exercise, elicited a weal that was localized to the test area. A challenge test with ingestion of boiled soya beans and exercise did not elicit a weal. Therefore, in this case, cold exposure, but not food ingestion, was essential for inducing EIA. Cold-dependent EIA is different from cold urticaria, food-dependent EIA, cholinergic urticaria and cold-induced cholinergic urticaria, and may be a distinct entity.

    Notes: The molecular sizes of secreted and cell-associated plasminogen activators from four cultured cell types were determined using an SDS-PAGE technique in which plasminogen and casein were included during polymerization of the polyacrylamide gel. The major bands of plasminogen activators secreted by human neonatal epidermal cells, human adult epidermal cells and transformed human squamous cells migrated the same distance as the high molecular weight band of authentic urokinase, indicating that the apparent molecular weight of these plasminogen activators was approximately 55,000 daltons. Plasminogen activator extracted from normal adult human epidertnis also migrated with this major band of plasminogen activator, and a minor higher molecular weight band was also detected. In contrast, plasminogen activators secreted by transformed mouse squamous cells migrated between the high molecular weight band (∼ 55K) and the low molecular weight band of urokinase (∼ 32K), indicating that plasminogen activators of mouse epidermal cells differ from those of human epidermal cells. The mobility of the major bands of plasminogen activators detected in cell lysates of the four cell types was identical to that of secreted plasminogen activators.

    Notes: Abstract Amorphous ternary palladium-based alloys containing platinum group metals as an additional element were prepared by rapid quenching from the molten state and their anodic characteristics were investigated in a 4 mol dm−3 NaCl solution of pH 4 and 80° C. The amorphous alloys containing sufficient quantities of rhodium, platinum or iridium were passivated by anodic polarization and their corrosion rates at high current densities in the chlorine evolution region were extremely low. This fact was attributed to the formation of a highly protective passive film due to both the transformation to the amorphous structure and the addition of rhodium, platinum or indium. The electrocatalytic activities for chlorine evolution of amorphous alloys were higher than those of pure platinum group metals except palladium. In particular, the amorphous Pd41Ir40P19 alloy had the desired stable, high electrocatalytic activity for chlorine evolution and the high overvoltage for oxygen evolution.
