    Notes: In vitro cervical dilatations were carried out on 33 uteri removed by hysterectomy for conditions not involving the cervix from parous women in their reproductive span of life. A specially designed force measuring dilator was used for all the tests. The analysis of the results involved the application of a thick-walled cylindrical stress determination technique. Using this approach it was possible to calculate quasi-elastic moduli for different degrees of dilatation and obtain theoretical distributions of radial and circumferential stress across the cervical wall. These results indicated a yielding of the cervical tissue at about 9 to 11 mm. dilatation in a significant number of cases.

    Notes: Forearm skin collagen, dermal thickness and collagen density were measured in a large number of normal subjects as a standard reference for future studies. Skin collagen decreased with age and was less in the females at all ages. There is a direct relationship between skin collagen and dermal thickness but variations in collagen density in disease limit the use of dermal thickness as a guide to changes in its collagen content.

The effects of pentaerythritol trinitrate (pentrinitrol) and glyceryl trinitrate on myocardial oxygen consumption and myocardial and systemic haemodynamics were studied in anaesthetized open-chest dogs. An in vivo oximeter in the coronary sinus permitted continuous determination of arteriovenous oxygen difference and myocardial oxygen consumption. All parameters were determined simultaneously at various intervals after drug administration.2. Myocardial oxygen consumption was diminished by both nitrates for more than 16 min. Changes in arteriovenous oxygen difference and coronary sinus oxygen content were variable between drugs. Following an initial transient increase, coronary blood flow was reduced by both nitrates. Aortic blood pressure, aortic blood flow, left ventricular end-diastolic pressure and left ventricular dP/dt were also reduced. Heart rate and contractile force were not appreciably altered by either nitrate.3. The decrease in myocardial oxygen consumption appears to be associated with the haemodynamic profile of these drugs. Both nitrates produced comparable reductions in preload (left ventricular end-diastolic pressure) and afterload (aortic pressure) as well as dP/dt.

The effect of intravenous pentaerythritol trinitrate and glyceryl trinitrate on left ventricular subepicardial (epi) and subendocardial (endo) Po2 and perfusion were compared in anaesthetized open-chest mongrel dogs. Tissue Po2 was determined simultaneously at a depth of 3 mm (epicardial) and 9 mm (endocardial) with small platinum electrodes by polarography. In a separate series of dogs tissue perfusion of those regions was measured by hydrogen (H2) clearance using similar electrodes.2. Both nitrates increased endocardial Po2 while epicardial Po2 was not altered. Perfusion was determined at the point of the maximal rise in endocardial Po2 (4–7 min after injection of either nitrate). At that period average coronary artery inflow and epicardial perfusion were decreased but endocardial perfusion was not significantly altered.3. Using the data on Po2, hydrogen clearance and intercapillary distance, the effect of the nitrates on transmural metabolism (oxygen consumption) was estimated by Krogh analysis. Basal endocardial metabolism was 20–30% higher than epicardial metabolism. The nitrates reduced metabolism in each region. The absolute decrease in oxygen consumption was greater in the endocardium.4. The results show that both pentaerythritol trinitrate and glyceryl trinitrate improve endocardial oxygenation by producing a more favourable balance between perfusion and oxygen requirements in that region.

    Notes: Summary The intrarenal distribution of citrate was evaluated in the dog during antidiuresis and osmotic diuresis, by using the specific citrate assay method of Moellering and Gruber. The measurements were made on tissue samples taken from four different regions throughout the kidney: cortex, outer and inner medulla, and papilla. During antidiuresis, a characteristic distribution of citrate was observed with highest levels in the papilla and lowest ones in the outer medulla. A medullary concentration gradient for citrate was found. Mannitol greatly decreased papillary citrate and sodium, but no changes in outer and inner medullary citrate occured. The results could not be explained by the citrate contained either in the trapped urine or blood in the tissue. It is suggested that citrate accumulation in the inner regions of renal medulla may be accounted for by countercurrent mechanisms or regional differences in renal citrate metabolism.

    Description / Table of Contents: Zusammenfassung Mit Hilfe eines Clot-Lysis-Tests war es möglich, Streptokinasekonzentrationen in Humanplasma quantitativ zu erfassen. Das Testsystem bestand aus Rinderfibrinogen, Rinderplasminogen, Human-Euglobulin, EDTA, Humanplasma (mit unbekanntem Streptokinaseanteil) und Thrombin. Plasminogen und Fibrinogen lagen stark konzentriert vor, so daß Plasminogen und Fibrinogen des zu testenden Humanplasmas keine Rolle spielten. Ein Zusatz von EDTA verhinderte, daß Gerinnungsfaktoren und Thrombocyten des auszutestenden Humanplasmas auf die Clot-Lysis-Zeit einen Einfluß nahmen. Die Fehlerbreite der Methode für 2 E Streptokinase lag beis=±0,19 E pro ml Plasma, für 5 E Streptokinase beis=±0,47 E pro ml Plasma und für 20 E Streptokinase beis=2,50 E pro ml Plasma. Plasmen von Patienten, die sich einer fibrinolytischen Therapie unterziehen mußten, wurden auf ihren Streptokinasegehalt untersucht. Es wurden Streptokinasekonzentrationen von 0,7 E bis 15 E pro ml Plasma gemessen. Die Halbwertszeit für die Elimination der Streptokinase nach Beendigung der Infusionstherapie lag bei 18 min. Der Verfall von 5 E Streptokinase pro ml Plasma wurde über einen unterschiedlichen Zeitraum (15 und 60 min, 24 und 48 Std) und Inkubation bei Zimmertemperatur und +37° C untersucht. Es zeigte sich, daß bei Zimmertemperatur eine schnelle Verarbeitung zur Vermeidung von Aktivitätsverlusten notwendig ist. Im eingefrorenen Zustand geht keine Streptokinase verloren.
    Notes: Summary A method of a quantitative determination of plasma streptokinase concentrations in patients undergoing streptokinase infusion is described. The principle of this method is based on the clot lysis time recorded by the thromboelastograph. The test clot constituents were bovine fibrin, bovine plasminogen, human euglobulin, EDTA, human plasma (of unknown streptokinase quantity) and thrombin. As rather high concentrations (fixed excess) of plasminogen (euglobulin) and fibrinogen were present in the test coagulum, no interference with changing plasminogen and fibrinogen levels of the patient's plasma was observed. Furthermore, due to high EDTA concentrations, no interaction with platelet functions and coagulation factors took place. The standard deviation in measuring 2 u streptokinase in 1 ml human plasma was determined ass=±0.19 u/ml, of 5 u streptokinase ats=±0.47 u/ml and of 20 u streptokinase ats=±2.5 u in 1 ml of human plasma. Plasma samples of patients undergoing fibrinolytic treatment were investigated with regard to their streptokinase content. Streptokinase concentration values varied between 0.7 u and 15 u/ml plasma. The average half life of streptokinase in the organism was 18 min. The decay of streptokinase in plasma at different temperatures and over various periods of time was also determined. A considerable loss of streptokinase in the plasma during storage at room temperature could be observed. Therefore, the determination procedures should be carried out without delay.

    Description / Table of Contents: In order to define the influence of an allenic type conjugation, the NMR spectra of vinylallene and of some of its methyl derivatives are analysed The signs of the coupling constants are also determined. These compounds are characterised by a planar s-trans structure of the two noncumulated double bonds. A discussion of the parameters, and especially of the methyl replacement effects, evidences the importance of the π type contributions. The theoretical calculations of the coupling constants by the methods of finite perturbation CNDO and INDO corroborate this interpretation.
    Notes: Afin de préciser l'influence d'une conjugaison de type allénique les spectres RMN du vinylallène et de plusieurs de ses dérivés méthylés ont été analysés et les signes des constantes de couplage ont été déterminés. Ces composés présentent une structure s-trans plane des deux doubles liaisons non cumulées. Une discussion des paramètres et no amment de l'influence des substitutions méthylées fait ressortir une forte prédominance des contributions de type π. Cette interprétation est corroborée par les calculs théoriques des constantes de couplages, effectués par la méthode de perturbations finies CNDO et INDO.

    Description / Table of Contents: A total analysis of the NMR spectra of 6-deoxy-L-hexopyranoses in the α-configuration and of the corresponding β-anomers was carried out. The parameters obtained are characteristic of a 1C (L) chair conformation, having the anomeric substituent in an axial orientation for the methyl α-fuco-, α-rhamno- and α-chinovopyranosides and for the α-fuco- and α-rhamnopyranosyl chlorides. The structure is also of a 1C (L) chair type for the methyl β-fuco- and β-chinovopyranosides; the geometry is the same for the β-fuco- and β-rhamnopyranosyl chlorides despite the anomeric effect of a chlorine atom. However, the NMR parameters of the β-chinovopyranosyl chloride are not explicable on the basis of a chair conformation with an equatorial chlorine or a boat structure.
    Notes: Une analyse complète des spectres RMN de désoxy-6 L-hexopyrannoses de configuration α et des anomères β correspondants est réalisée. L'ensemble des paramètres obtenus confirme l'existence d'une structure chaise 1C (L) présentant une disposition axiale du substituant anomère dans le cas des α-fuco-, α-rhamno- et α-chinovopyrannosides de méthyle et des chlorures de α-fuco- et α-rhamnopyrannosyle. La structure reste de type chaise 1C (L) dans le cas des β-fuco- et β-chinovopyrannosides de méthyle. Il en est de même en dépit de I'effect anomère dû à I'atome de chlore, pour les chlorures de β-fuco-et β-rhamnopyrannosyle. Par contre les paramètres RMN du chlorure de β-chinovopyrannosyle ne semblent interprétables ni sur la base d'une géométrie chaise à chlore équatorial ni par une structure bateau.

    Notes: The principles of the theoretical calculations of the carbon atom screening constant are reviewed. The Independent Electron, SCF—MO and Valence Bond formulae are briefly discussed, and the relative importance of the different terms involved in an approximate decomposition of σ13C are pointed out. The variations of the excitation energy ΔE, the mean distance between the nucleus and 2p electrons, and the electronic charges and bond orders with the structure are presented. The nature of the steric effect is also explained. In a second part the various empirical correlations which make an evaluation of δ13C from structural and physical properties possible are shown. The actual substituent parameters of the main classes of compounds are listed and worked examples given to make the use of the tables easier. The correlations between δ13C and the electronic charge, electronegativity, Hammett-Taft constants, electric field effects, geometrical parameters, spectroscopic data (electronic transitions, screening of other nuclei) and pH are also discussed with a view to appraising the carbon screening.
