    Notes: Fowl meat processed in cans and flexible retortable pouches was evaluated for heat effects on sensory properties. A trained panel evaluated texture and off-flavor of canned boned chicken, uncooked fowl meat retorted in flexible pouches, cooked meat retorted in flexible pouches, and simmered fowl meat (control). Results indicated that the retort pouch process may offer a method for improving the texture of processed fowl meat from spent hens by adequately cooking to tenderize the meat but not overcooking it to the extent that meat chunks are reduced to fibrous, shredded, or stringy components. Off-flavor development was related to pre-cooking before retorting.

    Notes: Diskussion Publikationen über die Anwendung der epiduralen Morphinapplikation in der Geburstshilfe liegen bislang von englischen Arbeitsgruppen vor. So berichten Husemeier et al. über keine suffiziente Analgesie nach Injektion von 2 mg Morphin verdünnt in 8 ml Kochsalzlösung, während Perris et al. in 50% der Fälle eine sehr gute und in weiteren 43% eine mäßige Beeinflussung des Wehenschmerzes beobachten konnten. Das kardiozirkulatorische sowie respiratorische System blieben dabei ebenso unbeeinflußt wie die sympathische Aktivität der Schwangeren. Neben diesen durchaus positiven Untersuchungsergebnissen sind aber auch Mitteilungen publiziert worden, welche eine nach Naloxon reversible Depression der Respiration beschreiben, wobei der analgetische Effekt durch das applizierte Naloxon unbeeinflußt blieb. Unsere eigenen Erfahrungen mit der morphininduzierten Epiduralanalgesie ließen bislang keinerlei Nebenwirkungen am maternalen wie fötalen Organismus erkennen.

    Notes: Abstract The genetic variability of membrane proteins (structure-bound proteins) and cytosol proteins (water-soluble proteins) was investigated in two inbred strains of the mouse, C57BL/6J and DBA/2J. Membrane proteins and cytosol were isolated from the brain and liver of the mouse. The proteins were separated by two-dimensional electrophoresis. A high number of genetic variant proteins (brain, 30; liver, 72) was found in the cytosol. Most of these variants represented changes in the amount of proteins. Electrophoretic mobility changes occurred only in about 1% (brain, 6; liver, 9) of all protein spots of a two-dimensional pattern. In contrast to the cytosol proteins, no genetic variation was detected among the membrane proteins, not even for the quantitative characteristics of the protein spots. The results obtained for the two classes of proteins suggest that the degree of variability in the amount of proteins is related to the degree of variability in the structure of proteins.

    Notes: Abstract The effect of different cell culture parameters on two-dimensional polypeptide maps of human fibroblasts is considered. Improved culture methods are introduced to get better resolution and reproducibility. Based on these investigations, highly standardized techniques for the protein mapping of this tissue and also of hair root cell lysates are presented. These methods seem to be quite suitable for analysis of cellular synthesized proteins and study of variability in normal subjects and in patients with genetic disorders.

    Notes: Abstract A standardized technique for the two-dimensional polypeptide mapping of cultured human fibroblasts has been used for the study of cellular protein variations in healthy controls, patients with inborn errors of metabolism and some other genetic defects. The analysis of about 50 gels has established that this method is very reproducible and enables the examination of about 600 polypeptides in a single gel. The inter-individual variation has been rather low (about 3%). In the gels from patients with genetic defects only very minor qualitative variations have been observed.

Die Rate an kurativen Resektionen wird nicht beeinflußt.
    Notes: Summary Modern diagnostic procedures such as sonography and computer tomography combined with percutaneous transhepatic cholangiodrainage (PTCD) have changed the diagnostic and therapeutic management of patients with malignant obstructive jaundice. Use of invasive procedures such as angiography has been reduced, and an increasing number of patients with inoperable carcinoma of the pancreas no longer need explorative laparotomy to establish the diagnosis. The rate of curative resection does not change.

Aufgrund der hohen Anregungstemperatur in einem Plasma treten dabei die von der konventionellen Atomabsorptionsspektroskopie her bekannten chemischen Interferenzen nicht auf, so daß mit geringen Probenmengen (max. 1 ml Serum für den Nachweis dieser 6 Elemente) ein sehr empfindlicher und sehr gut reproduzierbarer Nachweis möglich ist.
    Notes: Summary The application of ICP-emission spectroscopy for the determination of Cu, Fe, Zn, Ca, Mg and Na in human and animal serum is described. Due to the very high excitation temperature in a plasma the well known chemical interferences in atomic absorption spectroscopy do not appear in this technique. It is possible to carry out very sensitive and very well reproducible measurements on very small amounts of the sample (the total sample consumption for the determination of the above mentioned six elements is less than 1 ml).

    Notes: Membrane proteins and cytosol proteins from organs (liver, brain) of inbred mice were separated by two-dimensional electrophoresis (2DE). The 2DE technique revealed high resolution of complex protein solutions by staining the proteins. The 2DE patterns of membrane proteins showed about 660 protein spots (polypeptides) for both liver and brain. The protein patterns of the cytosols revealed about 800 polypeptides for the liver and about 680 for the brain. The membrane proteins and cytosol proteins of the 2DE patterns represented two protein populations specific for these two cell components. They matched in 7 % (liver) or 23 % (brain) of the total number of membrane protein spots. The matched spots represented dissociable membrane proteins rather than contamination of the isolated membrane fractions by cytosol proteins. Protein patterns of the same cell fraction but from the two different organs investigated contained more than 50 % organ-unspecific polypeptides.Several conditions of our 2DE standard procedure were reinvestigated to test the usefulness for our technique of some methodical steps employed in 2DE by other authors. We found that many steps often used in sample preparations impaired rather than improved the 2DE pattern. In particular, addition of SDS to the protein sample resulted in a considerable loss of proteins during isoelectric foucing. The 2DE itself could be simplified by omitting several steps such as prefocusing, equilibration of focusing gels, forming gel gradients, autoradiography. This has some implications for the development of 2DE to a routine method.
