    Notes: Abstract This paper presents the results from a sedimentary survey undertaken in July 1992 in the Sado estuary, western coast of Portugal. Sediment samples were taken at 14 sites, corresponding to four organic enrichment gradients, three of which are located close to harbour facilities or to industrial or urban effluent outfalls. The fourth site is located away from direct sources of anthropogenic disturbance. An integrated approach was conducted in order to study the quality status of the superficial sediments, and included 1. the analysis of sediment descriptors and contaminants (grain-size, total organic matter, heavy metals, PCBs), 2. the evaluation of toxicity, using bioluminescent bacteria and the Microtox® solid-phase protocol and 3. the study of macrofaunal communities structure and composition. Most of the sediments showed low IC50 values (high toxicity), while benthic macrofauna did not indicate community disturbance in most of the sites, nor the contaminants analysed provided explanation for the potential toxicity observed. Most of the sediments showed high fines content. A statistical significant relation between fines and IC50 values was found, indicating the sensitivity of the toxicity assessment method for granulometry. This sensitivity might explain the lack of agreement between the IC 50 values and the sediment chemistry-infaunal communities structure. The potential grain-size sensitivity was further analysed using another test sediment from which artificial samples with different sand∶fines ratios were prepared. It is concluded that extreme caution should be used when applying the solid-phase Microtox® protocol to assess the toxicity of natural sediments, covering a range of granulometry and contamination situations.

    Notes: Groundstation antennas located in Western Australia have been used as elements in the Southern Hemisphere very long baseline interferometry (VLBI) experiment. These commercial antennas of 27.5 and 15 m diameter provide a very economical improvement to the imaging capability of this array and the practice of using such antennas can be extended to other sites in the Southern Hemisphere in preparation for the forthcoming space-VLBI experiments. © 1995 American Institute of Physics.

    Notes: Abstract Differential scanning calorimetry (DSC) was used to study the 7S and 11S globulin fractions extracted from lupin seed (Lupinus luteus) flour. In agreement with previous work on other lupin species, the isolate showed three denaturation peaks compared to the two observed with soy. By comparison with the isolated globulin fractions, the denaturation peaks at the two higher temperatures in the lupin isolate were assigned to the 11S and 7S globulins. The denaturation temperature of the lupin 7S globulin was about 10 K higher than that for the corresponding soy globulin, whereas the values for the 11S globulin were similar. All globulins displayed increasing thermal stability with decreasing moisture contents. Possible reasons for the differences in behaviour of soy and lupin protein isolates are discussed.

    Notes: Abstract A tetranucleotide tandem repeat locus on chromosome 19 (D19S253) was analysed. PCR products were detected by denaturing polyacrylamide gels with fluorescent-based technology. This study has confirmed a polymorphism with 9 alleles ranging from 209 to 241 by with a simple repeat structure arranged from 7 to 15 repeats. Family studies confirmed mendelian inheritance of alleles. The efficiency on DNA extracted from bloodstains and cigarette butts has been evaluated. The protocol has shown sensitivity and reproducibility.

    Notes: Aiming at encapsulation of a hydrosoluble drug, large unilamellar liposomes (LUV) of egg phosphatidylcholine (PC) were coated with a natural polysaccharide derivative, O-palmitoylpullulan (OPP), and its in vitro stability evaluated using fluorescent probes. This coating (in OPP/PC weight ratio of 3) improved significantly the in vitro stability of LUV by decreasing both the permeability and fluidity of the liposomal membrane.

    Notes: Abstract. External muscle and myenteric plexus from the small intestine of adult guinea-pigs were maintained in vitro for 3 or 6 days. Myenteric neurons and smooth muscle cells from such organotypic cultures were examined at the electron-microscopic level. An intact basal lamina was found around the myenteric ganglia and internodal strands. Neuronal membranes, nuclei and subcellular organelles appeared to be well preserved in cultured tissues and ribosomes were abundant. Dogiel type-II neurons were distinguishable by their elongated electron-dense mitochondria, numerous lysosomes and high densities of ribosomes. Vesiculated nerve profiles contained combinations of differently shaped vesicles. Synaptic membrane specializations were found between vesiculated nerve profiles and nerve processes and cell bodies. The majority of nerve fibres were well preserved in the myenteric ganglia, in internodal strands and in bundles running between circular muscle cells. No detectable changes were found in the ultrastructure of the somata and processes of glial cells. Longitudinal and circular muscle cells from cultured tissue had clearly defined membranes with some close associations with neighbouring muscle cells. Caveolae occurred in rows that ran parallel to the long axis of the muscle cells. These results indicate that the ultrastructural features of enteric neurons and smooth muscle of the guinea-pig small intestine are well preserved in organotypic culture.

    Notes: Abstract. The projections of myenteric neurons within the myenteric plexus of the guinea-pig small intestine were established using retrograde tracing in organotypic culture. Three days after applying the fluorescent dye DiI to a single internodal strand in the myenteric plexus, 500–1000 nerve cell bodies were labelled. Of these, 77% were located oral to the application site, 15% were located anally and 7% were located within 1 mm of this site. Three major morphological types of neurons could be distinguished. Dogiel type I neurons had lamellar dendrites and single axons, Dogiel type II neurons had large smooth cell bodies and several long processes, and filamentous neurons had smooth ovoid cell bodies, single axons and several filamentous dendrites. Dogiel type I, II and filamentous neurons accounted for 54.6%, 38% and 7.4% of all filled cells, respectively. Labelled nerve cell bodies were present up to 13 mm aboral to the DiI application site; all neurons more than 2 mm aboral had Dogiel type I features. On the oral side, Dogiel type I neurons were found up to 110 mm, Dogiel type II neurons up to 100 mm and filamentous neurons up to 80 mm. Neurons with 2 mm oral or aboral to the DiI application site were located up to 7 mm circumferentially and were mainly Dogiel type II cells. This work revealed remarkable polarity within the myenteric plexus, with a significant prevalence of myenteric neurons projecting anally for longer distances than those projecting orally. These long pathways are probably involved in the coordination of intestinal motility.
