    Notes: Abstract  It is generally accepted that early human prostate cancers reveal higher androgen dependency than do advanced ones. In the present study, we examined whether the animal model of prostate cancer has already lost androgen dependency at the early stages of carcinogenesis. At experimental week 46, androgen deprivation was induced in rats and the incidences of atypical hyperplasia and cancer were examined in the ventral, dorsolateral prostate, coagulating glands, and seminal vesicles. Androgen deprivation significantly lowered the incidence of atypical hyperplasia in all four organs. As for the incidence of cancer, no significant differences were observed in the coagulating glands and seminal vesicles. Regarding atypical hyperplasia, androgen deprivation significantly decreased the proliferative cell nuclear antigen labeling index in the coagulating gland and seminal vesicles. The presence of cancer was also decreased in the coagulating gland but not in the seminal vesicles. With control group specimens, more intense staining of androgen receptor was observed in atypical hyperplasias than in cancers. Compared with the atypical hyperplasias, the cancers revealed low androgen dependency at the early stages of carcinogenesis. The cancers in the seminal vesicles also revealed higher androgen independency than did those in the coagulating gland.

    Notes: We have found that a cyclic crystalline–amorphous phase transformation can occur in Co75Ti25 alloy powder when subjected to ball milling. The results have shown that a single amorphous phase of Co75Ti25 is obtained after 11 ks of mechanical alloying (MA) time. This amorphous phase transforms into a new metastable phase of bcc-Co3Ti upon milling for 86 ks. The bcc-Co3Ti is thermally stable and does not transform to any other phase(s) upon heating up to 1300 K. It however returns to the same amorphous phase of Co75Ti25 upon milling for 360 ks. Further milling leads to the formation of crystalline and/or amorphous phases depending on the MA time. © 1997 American Institute of Physics.

The purpose of the present study was to investigate the changes in superoxide dismutase (SOD) isoenzyme (Mn2+-SOD and Cu2+, Zn2+-SOD) activities, contents and mRNA expressions in rat skeletal muscle during endurance training and a single bout of exercise.2. Thirty-eight male Wistar rats were divided into untrained (U) and trained (T) groups. The T group rats were treadmill-trained for 9 weeks. The activity, content and mRNA expression of Mn2+-SOD and Cu2+, Zn2+-SOD were determined in the soleus muscle of each rat.3. Mn2+-SOD activity and content in the T group were significantly higher than in the U group, both at rest (22 and 21%, respectively) and after exercise (24 and 46%, respectively), while a single bout of exercise affected neither the activity nor content of Mn2+-SOD in either group.4. The content of Cu2+, Zn2+-SOD in both groups was not different at rest and after exercise, although its activity at rest was significantly higher in the T group than in the U group (by 29%).5. After exercise, the expression of Mn2+-SOD mRNA was markedly attenuated only in the U group (49%); the expression of Cu2+, Zn2+-SOD mRNA was not influenced by exercise.6. Our results suggest that adequate endurance training increases both the activity and content of Mn2+-SOD and that untrained rats are rather susceptible to oxidative stress during physical exercise. It thus appears that Mn2+-SOD provides a reliable index of physical training.7. The results obtained in the present study also suggest that muscle has the capacity of responding to training in such a manner as to reduce the potential harm arising from the accumulation of oxygen free radicals resulting from enhanced metabolic activity.

    Notes: Abstract By replacing CH4 with CD4, the isotope effect on the reaction of adsorbed hydrocarbon species with CO2 over Ni/Al2O3 catalyst was studied using pulse surface reaction rate analysis (PSRA). The first-order rate constant for this step was 1.45 times larger for CH4 than for CD4. The observed isotope effect suggests that the reaction of adsorbed hydrocarbon species with CO2 (or adsorbed oxygen) is rate-controlling for the reforming of CH4.

    Notes: Abstract Cell culture models of calcium phosphate renal stone formation were established using the MDCK cell line. Renal microliths were detected within pseudocysts in three-dimensional soft agar cultures, and were also observed in the basal region of cells lining the cell sheet, and immediately beneath domes or blisters in monolayers and collagen gel cultures. Light and scanning electron microscopy indicated that these microliths had a similar lamellated and spherical appearance to those in humans. These microliths were first detected microscopically after 21 days of culture, and were found to be composed of calcium phosphate by X-ray and microinfrared spectroscopic analyses. These culture models may provide a powerful new tool to study the pathogenesis of renal stone diseases and/or calcium phosphate stone formation in humans and animals.

    Notes: Abstract This study used reverse transcription polymerase chain reaction (RT-PCR) to examine heparan sulfate proteoglycan (HS-PG) mRNA expression levels during stone formation in the rat kidney. Total RNA in kidneys was extracted and converted to cDNA. PCR products were resolved by electrophoresis on 1.5% agarose gel and visualized with ethidium bromide. Fragment intensity and area were measured using an image analyzer. Control cyclophilin and HS-PG mRNAs were expressed in all samples examined as 235 by and 506 bp bands, respectively. Cyclophilin expression in the normal group was not significantly different from expression in the group that formed stones. However, the level of HS-PG mRNA expression apparently increased in calcium oxalate (CaOx) microlith. The findings suggest an association between CaOx nephrolithiasis and expression of HS-PG in the rat kidney.

    Notes: Abstract We have encountered a paternity case where exclusion of the putative father was only observed in the ABO blood group (mother, B; child, A1; putative father, O), among the many polymorphic markers tested, including DNA fingerprints and microsatellite markers. Cloning a part of the ABO gene, PCR-amplified from the trio’s genomes, followed by sequencing the cloned fragments, showed that one allele of the child had a hybrid nature, comprising exon 6 of the B allele and exon 7 of the O1 allele. Based on the evidence that exon 7 is crucial for the sugar-nucleotide specificity of A1 and B transferases and that the O1 allele is only specified by the 261G deletion in exon 6 of the consensus sequence of the A1 allele, we concluded that the hybrid allele encodes a transferase with A1 specificity, resulting, presumably, from de novo recombination between the B and O1 alleles of the mother during meiosis. Screening of random populations demonstrated the occurrence of four other hybrid alleles. Sequencing of intron VI from the five hybrid alleles showed that the junctions of the hybrid alleles were located within intron VI, the intron VI-exon 7 boundaries, or exon 7. Recombinational events seem to be partly involved in the genesis of sequence diversities of the ABO gene.

    Notes: Abstract. We compare the clinical efficacy of two approaches for balloon aortic valvuloplasty (BAV) in infants with critical aortic valve stenosis. The approaches were through the carotid artery and the femoral artery. Eight catheterizations for BAV were performed in seven consecutive patients with critical aortic stenosis: four BAVs were approached through the femoral artery and four through the right common carotid artery. We inserted a 5F sheath into the right common carotid artery by a cutdown procedure; after BAV the sheath was removed and the carotid arteriotomy sutured with 7-0 monofilament. Two cases in which the femoral artery approach was used resulted in failure to perform BAV; two cases had complications. All four cases with the carotid artery approach were successful, with no complications; aortography performed 3 months after one balloon valvuloplasty revealed a smooth, unobstructed right carotid artery. Use of the carotid artery approach may reduce serious complications with BAV and offers quicker, easier maneuvering in infants and neonates with critical aortic valve stenosis.
