    Description / Table of Contents: Summary A vascular etiology of Parkinson’s disease (PD) has been long debated. In order to search for an ischemic basis of PD we assessed the clinical symptomatology of a consecutive group of 60 PD patients and compaired their frequency of cerebrovascular risk factors, carotid atherosclerosis and ischemic brain lesions with age-matched groups of stroke patients and normals. There were 16 (27%) subjects with PD who also had symptoms of cerebrovascular disease. The frequencies of carotid stenoses, ischemic brain lesions and most of cerebrovascular risk factors seen in the latter group was comparable with those of stroke patients and significantly higher than in the investigational subsets of patients with ”pure” PD and normals. Only one (1.6%) individual with PD presented signs suggestive of an ischemic etiology of parkinsonism. These findings suggest that cerebrovascular disease occurs in approximately one fourth of patients with PD, but seldomly is causally related.
    Notes: Zusammenfassung Eine mögliche vaskuläre Genese des Morbus Parkinson steht immer wieder zur Diskussion. Ziel der vorliegenden Arbeit war die Erhebung einer möglichen vaskulären Ätiologie der Parkinson-Symptomatik bei einer konsekutiven Gruppe von 60 Parkinson-Patienten unter Einbeziehung der klinischen Symptomatik, Verteilung zerebrovaskulärer Risikofaktoren, Präsenz computertomographisch erfaßbarer morphologischer Hirnveränderungen und sonographisch darstellbarer Hirnarterienveränderungen und Vergleich der Ergebnisse mit denen einer altersvergleichbaren Insult- und Normalgruppe. 16 (27%) Parkinson-Patienten wiesen anamnestisch oder klinisch zusätzlich Zeichen einer zerebrovaskulären Insuffizienz auf. Die Frequenz der meisten zerebrovaskulären Risikofaktoren, zerebraler Infarkte und Karotisstenosen in dieser Patientengruppe war vergleichbar mit jener der Insultpatienten und signifikant höher als bei Parkinson-Patienten ohne Zeichen einer zerebrovaskulären Insuffizienz und Normalpersonen. Eine mögliche vaskuläre Ätiologie der Parkinson-Symptomatik konnte nur in einem Fall (1,6%) diskutiert werden. Zusammenfassend fand sich im Rahmen der vorliegenden Studie bei ungefähr einem Viertel der Parkinson-Patienten eine additive zerebrovaskuläre Erkrankung, ein kausaler Zusammenhang mit der Parkinsonsymptomatik kann jedoch als selten angesehen werden.

    Notes: Abstract  Four paroxysmal nocturnal hemoglobinuria (PNH) patients with severe thrombocytopenia, hemolytic anemia and neutropenia were treated using a combination of filgrastim (G-CSF) and cyclosporin. In all patients a trilineage response of hematopoiesis was achieved. In addition, the proportion of glycosyl-phosphatidylinositol (GPI)-deficient granulocytes decreased. All patients mobilized CD34+ hematopoietic progenitors into peripheral blood after starting treatment with G-CSF. The majority of early progenitors (CD34+CD38–) after mobilization into peripheral blood was found to be unaffected by the GPI-anchoring defect. No patient developed leukemia while under therapy. We conclude from these data that the combination of G-CSF and cyclosporin represents an efficient option for the treatment of hypoplastic PNH.

    Notes: Abstract  In this paper we report on the optimization of homoepitaxial InAs and InP growth in MOMBE (metalorganic molecular beam epitaxy). A correlation is made between good optical quality material and the observation of RHEED (reflection high energy electron diffraction) intensity oscillations. It will be shown, that in situ RHEED oscillations can be used to determine a growth parameter window in MOMBE.

    Notes: Abstract A drug tracer is most commonly applied to get information about the pharmacokinetics (PK) of a drug that is not confounded by an endogenously produced drug or an unknown drug input. An equally important use of tracers that has not been fully recognized is their use in the study of nonlinear PK behavior. In the present study a system analysis is applied to examine the interaction between drug molecules characteristic and intrinsic to any nonlinear process which enables the nonlinearity to be identified and modeled using a drug tracer. The proposed Tracer Interaction Methodology (TIM) forms a general developmental framework for novel methods for examining nonlinear phenomena. Such methods are potentially much more sensitive and accurate than previous methods not exploiting the tracer principle. The methodology proposed is demonstrated in a simulation study and with real data in a specific implementation aimed at determining the Michaelis-Menten (MM) parameters of nonlinear drug elimination while accounting for drug distribution effects. The simulation study establishes that the TIM-based, MM parameter evaluation produces substantially more accurate parameter estimates than a nontracer (NT) conventional method. In test simulations the accuracy of the TIM was in many cases an order of magnitude better than the NT method without evidence of bias. The TIM-based, MM parameter estimation methodology proposed is ideally suitable for dynamic, non-steady-state conditions. Thus, it offers greater applicability and avoids the many problems specific to steady state evaluations previously proposed. TIM is demonstrated in an evaluation of the nonlinear elimination behavior of erythropoietin, a process that likely takes place via receptor-based endocytosis. Due to its high sensitivity, accuracy, and intrinsic nonlinearity the TIM may be suitable for in-vivo studies of receptor binding of the many biotechnology produced peptide drugs and endogenous compounds displaying receptor-mediated elimination.

    Notes: Abstract Cluster is an ESA/NASA four-spacecraft mission designed to study plasma processes in three dimensions using the combined data from eleven instruments on each spacecraft. This mission requires the combination of many measured parameters, and the Cluster community have taken the unprecedented step of establishing a set of high quality data products from all instruments at spin (~ 4 s) resolution which will be produced and distributed throughout the mission lifetime. The Cluster Science Data System (CSDS) is based on a set of eight data centres which are implemented and funded through national programmes. As part of CSDS, a Joint Science Operations Centre (JSOC) has been established to facilitate the commanding of the 44 instruments. It is co-located with the UK data centre at the Rutherford Appleton Laboratory (RAL), Didcot, United Kingdom. ESA's contribution to CSDS includes the provision of the CSDS User Interface, a dedicated network (CSDSnet) to interconnect the data centres, and the co-ordination of all activities at CSDS level. A wide scientific community wishing to use Cluster data will have differing data rights, experience and means of access. Users will also include those working with data sets from other missions, e.g., Soho, Geotail, Wind, Polar, Interball, and Equator-S. The Cluster Science Data System is primarily designed to support multi-instrument and multi-spacecraft data analysis and it is distributed across six national data centres in Europe, one in the USA, and one in China. CSDSnet will be used to interconnect the European data centres, the Joint Science Operations Centre at Didcot and the spacecraft Operations Control Centre at ESOC in Darmstadt.

    Notes: Abstract The European Space Agency's Cluster programme is designed to study the small-scale spatial and temporal characteristics of the magnetospheric and near-Earth solar wind plasma. The programme is composed of four identical spacecraft which will be able to make physical measurements in three dimensions. The relative distance between the four spacecraft will be varied between 200 and 18000 km during the course of the mission. This paper provides a general overview of the scientific objectives, the configuration and the orbit of the four spacecraft and the relation of Cluster to other missions.

    Notes: Abstract The electric-field and wave experiment (EFW) on Cluster is designed to measure the electric-field and density fluctuations with sampling rates up to 36000 samples s-1. Langmuir probe sweeps can also be made to determine the electron density and temperature. The instrument has several important capabilities. These include (1) measurements of quasi-static electric fields of amplitudes up to 700 mV m-1 with high amplitude and time resolution, (2) measurements over short periods of time of up to five simualtaneous waveforms (two electric signals and three magnetic signals from the seach coil magnetometer sensors) of a bandwidth of 4 kHz with high time resolution, (3) measurements of density fluctuations in four points with high time resolution. Among the more interesting scientific objectives of the experiment are studies of nonlinear wave phenomena that result in acceleration of plasma as well as large- and small-scale interferometric measurements. By using four spacecraft for large-scale differential measurements and several Langmuir probes on one spacecraft for small-scale interferometry, it will be possible to study motion and shape of plasma structures on a wide range of spatial and temporal scales. This paper describes the primary scientific objectives of the EFW experiment and the technical capabilities of the instrument.

    Notes: Abstract Charging of the outer surface or of the entire structure of a spacecraft in orbit can have a severe impact on the scientific output of the instruments. Typical floating potentials for magnetospheric satellites (from +1 to several tens of volts in sunlight) make it practically impossible to measure the cold (several eV) component of the ambient plasma. Effects of spacecraft charging are reduced by an entirely conductive surface of the spacecraft and by active charge neutralisation, which in the case of Cluster only deals with a positive potential. The Cluster spacecraft are instrumented with ion emitters of the liquid-metal ion-source type, which will produce indium ions at 5 to 8 keV energy. The operating principle is field evaporation of indium in the apex field of a needle. The advantages are low power consumption, compactness and high mass efficiency. The ion current will be adjusted in a feedback loop with instruments measuring the spacecraft potential (EFW and PEACE). A stand-alone mode is also foreseen as a back-up. The design and principles of the operation of the active spacecraft potential control instrument (ASPOC) are presented in detail. Flight experience with a similar instrument on the Geotail spacecraft is outlined.
