    Notes: The intensity dependence of the transient photovoltaic response in the nanosecond region and the spectral dependence of the continuous wave (cw) photovoltaic response in Al/α-SnPc/ITO sandwich cell have been reported. The cw photovoltaic action spectrum is similar to the absorption spectrum. The magnitude, polarity, and response time of the transient photovoltage are found to be dependent on the intensity and wavelength of the incident light, which offers a potential application in the area of light-controlled nonlinear optoelectronic detectors. © 1996 American Institute of Physics.

    Notes: We have demonstrated that the supercurrent distribution in current-biased YBa2Cu3O7−δ thin films can be obtained by measuring the radiation power of THz electromagnetic pulses excited with femtosecond laser pulses. As the radiation power is proportional to the square of the bias current density at the laser spot position, the two-dimensional current distribution can be obtained from the intensity distribution of THz radiation by scanning the laser spot. The characteristic supercurrent distribution is analyzed by using the critical-state model. © 1999 American Institute of Physics.

    Notes: Tip surfaces of n-type Si field emitter arrays (FEAs) have been anodized to obtain n-type porous layers on the top surfaces of the Si emitters. The gate voltage required for emission could be lowered by tip anodization and the emission current was enhanced by a factor of up to 10. Fowler–Nordheim plots for the FEAs before and after tip anodization revealed that the work function of the tip could be decreased and the field conversion factor could be increased by the process. © 1995 American Institute of Physics.

    Notes: We demonstrated the vector mapping of the supercurrent flow in YBa2Cu3O7−δ thin films using a terahertz (THz) radiation imaging system. A wire-grid polarizer was employed to detect the vector components of the THz radiation. The two THz radiation images obtained through the polarizer tilted by +45° and −45° with respect to the detector's orientation were successfully transferred into the vector image of the supercurrent distribution. The observed distributions in the current-biased and the vortex-penetrated films were qualitatively explained using Bean's critical-state model [E. H. Brandt and M. Indenbom, Phys. Rev. B 48, 12893 (1993)]. © 1999 American Institute of Physics.

    Notes: FK506 (tacrolimus) is a natural product now approved in the US and Japan for organ transplantation. FK506, in complex with its 12 kDa cytosolic receptor (FKBP12), is a potent agonist of immunosuppression through the inhibition of the phosphatase activity of calcineurin. Rapamycin (sirolimus), which is itself an immunosuppressant by a different mechanism, completes with FK506 for binding to FKBP12 and thereby acts as an antagonist of calcineurin inhibition. We have solved the X-ray structure of unliganded FKBP12 and of FKBP12 in complex with FK506 and with rapamycin; these structures show localized differences in conformation and mobility in those regions of the protein that are known, by site-directed mutagenesis, to be involved in calcineurin inhibition. A comparison of 16 additional X-ray structures of FKBP12 in complex with FKBP12-binding ligands, where those structures were determined from different crystal forms with distinct packing arrangements, lends significance to the observed structural variability and suggests that it represents an intrinsic functional characteristic of the protein. Similar differences have been observed for FKBP12 before, but were considered artifacts of crystal-packing interactions. We suggest that immunosuppressive ligands express their differential effects in part by modulating the conformation of FKBP12, in agreement with mutagenesis experiments on the protein, and not simply through differences in the ligand structures themselves.

    Notes: We have synthesized a series of non-macrocyclic ligands to FKBP12 that are comparable in binding potency and peptidyl prolyl isomerase (PPIase) inhibition to FK506 itself. We have also solved the structure of one of these ligands in complex with FKBP12, and have compared that structure to the FK506–FKBP12 complex. Consistent with the observed inhibitory equipotency of these compounds, we observe a strong similarity in the conformation of the two ligands in the region of the protein that mediates PPIase activity. Our compounds, however, are not immunosuppressive. In the FKBP12–FK506 complex, a significant portion of the FK506 ligand, its `effector domain', projects beyond the envelope of the binding protein in a manner that is suggestive of a potential interaction with a second protein, the calcium-dependent phosphatase, calcineurin, whose inhibition by the FKBP 12–FK506 complex interrupts the T-cell activation events leading to immunosuppression. In contrast, our compounds bind within the surface envelope of FKBP12, and induce significant changes in the structure of the FKBP12 protein which may also affect calcineurin binding indirectly.

    Notes: Titanium boron nitride (Ti,B)N films have been prepared by depositing Ti and B atomvapor on a (100) Si single crystal and an amorphous glass substrates under simultaneous irradiationof N ions, that is ion mixing and vapor deposition (IVD) technique. The transport ratio of metalsand ion, (Bva+Tiva)/Nion, was fixed at 4, and the film thickness was supposed to be 1 [removed info]m. With anincrease in the evaporation ratio of B and Ti, Bva/Tiva, from 0 to 3, microstructure of the film waschanged from fine triangular to granular morphology. It was confirmed by the XPS analysis that Nions were preferentially coordinated with Ti atoms when the Bva/Tiva was relatively low otherwisenitriding of B probably occurred. This result was also supported by the XRD spectra of the films.Knoop hardness of the (Ti,B)N films was strongly dependent on the Bva/Tiva. The highest hardnessof approximately 5000 was observed at the Bva/Tiva=0.3; this implies that a small amount of alloyingB considerably increases the film hardness

    Notes: Abstract A clone that can complement both Escherichia coli hemB and hemL mutations was found among plasmids containing the Propionibacterium freudenreichii hemB gene, which encodes δ-aminolevulinic acid dehydratase. The regions upstream and downstream of the hemB gene were sequenced. Two open-reading frames (ORF1 and -2), which were similar to the hemY gene encoding protoporphyrinogen oxidase and the hemH gene encoding ferrochelatase from Bacillus subtilis, were found upstream of the hemB gene. ORF1 and -2 complemented the E. coli hemG mutation, defective in protoporphyrinogen oxidase, and the hemH gene respectively. Since ORF1 had no homology to hemG, the gene was designated hemY. The hemYHB genes appeared to be within the same transcription unit. Downstream from the hemB gene, three open-reading frames were found. One of these, transcribed in the same direction as the hemB gene, was identical to be the hemL gene, which encodes glutamate-1-semialdehyde 2,1-aminomutase. The other two open-reading frames, located between the hemYHB and hemL genes, were transcribed divergently, and their deduced amino acid sequences showed similarities to a membrane-bound transport protein and a transcriptional regulatory protein respectively. The two genes may thus be involved in hem transport and the regulation of hem gene expression respectively, and were tentatively named hemX and hemR. Although hemX and hemL are unlikely to be part of the same operon, hemYHBXRL␣are clustered on the P. freudenreichii chromosome.

    Notes: Abstract We succeeded expressing the streptomyces cholesterol oxidase gene (choA), a potent insecticidal protein active against boll weevil larvae, in tobacco cells. A plasmid, pBC4, containing the choA gene under the control of the 35S promoter of cauliflower mosaic virus, was introduced into tobacco cells (BY-2) using a pneumatic particle gun. Stable integration of the choA gene in the genome of the transformed callus line was confirmed by Southern hybridization analysis. Northern blot analysis revealed material corresponding in size to the choA transcript in the transformed cells. Cholesterol oxidase activity in transformed callus was about eightfold higher than that in control cell line.
