    Notes: Titanium boron nitride (Ti,B)N films have been prepared by depositing Ti and B atomvapor on a (100) Si single crystal and an amorphous glass substrates under simultaneous irradiationof N ions, that is ion mixing and vapor deposition (IVD) technique. The transport ratio of metalsand ion, (Bva+Tiva)/Nion, was fixed at 4, and the film thickness was supposed to be 1 [removed info]m. With anincrease in the evaporation ratio of B and Ti, Bva/Tiva, from 0 to 3, microstructure of the film waschanged from fine triangular to granular morphology. It was confirmed by the XPS analysis that Nions were preferentially coordinated with Ti atoms when the Bva/Tiva was relatively low otherwisenitriding of B probably occurred. This result was also supported by the XRD spectra of the films.Knoop hardness of the (Ti,B)N films was strongly dependent on the Bva/Tiva. The highest hardnessof approximately 5000 was observed at the Bva/Tiva=0.3; this implies that a small amount of alloyingB considerably increases the film hardness

    Notes: Abstract  Mos plays a crucial role in meiotic cell division in vertebrates. In Xenopus, Mos is involved in the initiation of oocyte maturation as an initiator and in the arrest at the metaphase II stage (MII) as a component of the cytostatic factor (CSF). The function of Mos is mediated by MAP kinase (MAPK). We investigated the function of the Mos/MAPK pathway during goldfish oocyte maturation induced by 17α,20β-dihydroxy-4-pregnen-3-one (17α,20β-DP), a natural maturation-inducing hormone in fishes. Mos was absent in immature goldfish oocytes. It appeared before the onset of germinal vesicle breakdown (GVBD), increased to a maximum in mature oocytes arrested at MII and disappeared after fertilization. MAPK was activated after Mos synthesis but before maturation-promoting factor (MPF) activation, and its activity reached maximum at MII. Injection of either Xenopus or goldfish c-mos mRNA into one blastomere of 2-cell-stage Xenopus and goldfish embryos induced metaphase arrest, suggesting that goldfish Mos has a CSF activity. Injection of constitutively active Xenopus c-mos mRNA into immature goldfish oocytes induced MAPK activation, but neither MPF activation nor GVBD occurred. Conversely, the injection of goldfish c-mos antisense RNA inhibited both Mos synthesis and MAPK activation in the 17α,20β-DP-treated oocytes, but these oocytes underwent GVBD. These results indicate that the Mos/MAPK pathway is not essential for initiating goldfish oocyte maturation despite its general function as a CSF. We discuss the general role of Mos/MAPK during oocyte maturation, with reference to the difference in contents of inactive MPF (pre-MPF) stored in immature oocytes.

    Notes: Summary The method of the alternate culture and animal passage was introduced in the study of human glioma. For animal passage the hereditary asplenic-athymic (lasat) mice were used as a carrier. Because the lasat mice have practically no cellular and only little humoral immunity, the rate of tumor take was expected to be raised, and successful results were obtained. Ultrastructural and immunohistochemical (GFA protein) studies were also done. The overgrowth of stromal elements in reculture of tumors in lasat mice was less vigorous than in athymic nude mice. After four passages through lasat mice, an established human glioma cell line, KNS-42, showed numerous well differentiated cell nests, and this alternate culture and animal passage suggested to enhance the differentiation and growth capacity. After three passages through lasat mice, the tumor line, KNS-42-L, produced tumors also in athymic nude mice and their histological features were essentially the same as those in lasat mice, and hence the lasat mice could be saved.

    Notes: Abstract Objective: To evaluate the role of surfactant in the mechanism and treatment of acute lung injury caused by inhalation of fabric protector. Design: Prospective, randomized study. Setting: University laboratory. Interventions: In vitro experiment: a porcine surfactant suspension (10 mg · ml−1) was exposed to a fabric protector aerosolized with an ultrasonic nebulizer for 1 min. Minimum surface tension (γ min) was sequentially measured using pulsating bubble equipment. Animal experiment: 14 adult rats were anesthetized with pentobarbital and mechanically ventilated with pure oxygen. Then, all rats inhaled fabric protector aerosolized with the nebulizer for five breaths. Three hours after inhalation, the rats were randomly assigned to two groups: a surfactant group (n = 7), in which surfactant (100 mg · kg−1) was replaced, and a control group (n = 7), in which no substance was given. Measurements and results: In vitro experiment: exposure to fabric protector aerosol increased the mean γ min of the surfactant from 1.7 to 19.2 mN · m−1 (n = 5, p 〈 0.05). Animal experiment: the mean partial pressure of oxygen in arterial blood (PaO2) in all rats decreased from 62.8 to 17.1 kPa at 3 h after inhalation. The PaO2 in the surfactant group increased to 49.8 ± 11.1 (SD) kPa at 30 min after surfactant replacement (p 〈 0.05), while the PaO2 in the control group remained below 20 kPa. Conclusions: Impairment of surfactant is a factor involved in the development of acute lung injury caused by inhalation of fabric protector. Surfactant replacement may be therapeutic for such injuries.

    Notes: Abstract  Genes for dimeric and tetrameric human metallothionein (hMT) were designed and successfully overexpressed in Escherichia coli to generate functional oligomeric hMTs. An hMT synthesized with prokaryotic codons, a linker encoding a gly-gly-gly tripeptide, and Met-deficient hMT-II was ligated to create a dimeric hMT, from which a tetrameric hMT was then constructed. The increased molecular size of the constructs resulted in improved stability and productivity in E. coli. The oligomeric proteins formed inclusion bodies which were dissolved with dithiothreitol, and the purified apo-metallothioneins were reconstituted with Cd or Zn ions in a reducing condition. The oligomeric hMT proteins incubated with Cd ions showed a typical Cd-thiolate absorbance peak at 245–255 nm. The dimeric and tetrameric hMT proteins exhibited both Cd and Zn binding activities that were respectively two and four times higher than those of the hMT-II monomer protein. These novel oligomeric hMTs may be useful in bioremediation for heavy metals.

    Notes: Summary Extracellular and intracellular recordings were made from spinocerebellar tract neurones of the central cervical nucleus (CCN) in C1–C3 segments of the anaesthetized cat. These neurones were identified by antidromic activation from the cerebellar peduncle. Stimulation of the ipsilateral dorsal root elicited extracellular spikes or EPSPs with a monosynaptic latency in almost all CCN neurones in the same segment (segmental input). Late excitatory effects were also observed in about one third of CCN neurones. The monosynaptic EPSP was occasionally followed by an IPSP. The excitatory input from the dorsal root to CCN neurones was extended over several segments for some CCN neurons (extrasegmental input). Monosynaptic excitation was evoked in CCN neurones after stimulation of dorsal neck muscle nerves as well; i.e. splenius (SPL), biventer cervicis and complexus (BCC), rectus capitus dorsalis, and obliquus capitus caudalis. Thresholds for this excitation were near the threshold of the nerve, suggesting that it originated from group I fibres. The component of excitation added after strong stimulation of neck muscle nerves would be attributed to group II fibres. When a CCN neurone received excitatory input from the nerve of one muscle, it was generally not affected by stimulation of other nerves in the same segment. Such muscle specificity of segmental input was the principal pattern of connexion of neck muscle afferents with CCN neurones. In some cases, however, excitatory convergence from SPL and BCC nerves onto single CCN neurones or excitation from the SPL nerve and inhibition from the BCC nerve were also observed. Nearly half of the CCN neurones received input from one muscle nerve of the same segment and not from the afferent of the same muscle of different segments, indicating a segment specificity of input. In the remaining CCN neurones, weaker excitatory effects were induced from afferents of different segments as well. In such extrasegmental effects, inputs to CCN neurones from caudal segments predominated in frequency over those from rostral segments. The origin of extrasegmental input was generally confined to the same muscle. Low threshold muscle afferents from the SPL and BCC were intraaxonally stained with HRP. The collaterals of the stained fibre distributed branchlets and terminals to the CCN, laminae VII, VIII, and motor nuclei. Two fibres responding to local muscle prodding or stretch showed a similar morphology. The findings indicated that muscle spindle afferents from primary endings projected to the CCN.

    Notes: Summary Excitatory postsynaptic potentials (EPSPs) were recorded intracellularly from hindlimb motoneurones of the anaesthetized fresh water turtle. The EPSPs were evoked from low threshold muscle afferents and the amplitudes saturated for stimuli less than two times the nerve threshold. The segmental latencies of these EPSPs, measured from the initial positive peak of the triphasic cord dorsum potential to the onset of the EPSP, ranged from 1.5 to 3.1 ms. The intraspinal conduction time of afferents was estimated by recording afferent volleys in the grey matter along the vertical course of intraspinal afferent fibres. The synaptic delay was estimated by subtracting the latency of the afferent volley at the deepest region of the dorsal horn from the segmental latency of the EPSP (in the range from 1.6 to 2.1 ms) recorded in the same microelectrode track. The average value was 0.99 ms (range: 0.9–1.1 ms), which was close to the known synaptic delay of cold-blooded animals. Therefore, the EPSPs in this range of segmental latencies were regarded as monosynaptic. Taking account of the intraspinal afferent conduction time (0.8 ms on average) and another synaptic delay, the latency for disynaptic transmission would be 2.8 ms or more. Thus, EPSPs having segmental latencies of 1.5–3.1 ms were suggested to be almost all monosynaptic in nature, at least under the present conditions of deep anaesthesia. On the basis of the above criteria for the monosynaptic nature of EPSPs, the pattern of convergence of monosynaptic excitatory inputs from various muscle afferents was investigated. Monosynaptic EPSPs were induced from the homonymous muscle nerve and the nerve innervating the synergist at the same joint. The heteronymous EPSPs were also found between muscles within each group of the anterodorsal musculature and the posteroventral musculature. No monosynaptic connexions were found between anterodorsal and posteroventral muscles except between the muscles innervated by the peroneal and the tibial nerve.

    Notes: Summary The pharmacological action of a leukotriene antagonist FPL 55712 was studied on canine cerebral arterial segments. FPL 55712 at concentrations of 10−6M and 10−5M showed dosedependent inhibition of the constrictor responses of the artery to prostaglandin F2α. The constrictor responses of the artery to either serotonin or haemoglobin were suppressed by 10−5M FPL 55712, while 10−6M FPL 55712 failed to alter the responses. The KCL-induced constriction was not affected by treatment with either 10−6M or 10−5M FPL 55712. The present results suggest that FPL 55712 has vasodilator properties other than those related to the inhibition of leukotrienes.

    Notes: Summary Plasma and cerebrospinal fluid CEA detemination was done in 97 patients with neurosurgical disorders. Elevated titres were found in 14 of 64 patients with brain tumours. CEA levels were elevated significantly in patients with metastatic brain tumours. Following treatment, the values fell in three patients with ependymoma, medulloblastoma, and unverified brain tumour. This study suggests that CEA levels may be of value in the differential diagnosis of primary and metastatic brain tumours, and useful in the evaluation of patients with brain tumours after treatment. CEA in the cerebrospinal fluid was absent in eight patients with brain tumours.
