    Notes: Summary Distilled water rinsing after stimulation of the newt pit organ with divalent chloride salts produced neural off-responses. The off-response was selectively depressed by HgCl2 treatment and was not suppressed by a water rinse containing NaCl. The results indicate that the off-response might be due to the removal of the divalent cations bound to the receptor membrane.

    Notes: Summary Arylsulfatase activities in biological materials are too low to be detected by the methods available hitherto. A sensitive and specific assay method for arylsulfatase A (AS-A) has been developed in the present study. Ascorbate-2-sulfate is known to be a specific natural substrate of AS-A; the ascorbic acid liberated by the action of AS-A was quantitatively determined using HPLC equipped with an amperometric detector. The method was used to analyze the activity of AS-A in biological materials.

    Notes: Abstract The administration of 200 nM K252a to HuH7 suppressed the proliferation of the cells almost completely. The uptake of [3H]thymidine was inhibited, and flow cytometry revealed only one peak at 2C on day 3 after treatment with 100 nM K252a. The expression of proto-oncogene c-myc was not reduced. Despite the blockage at G1, both the size of the cells and the amount of cell protein had increased by 4 times by day 3 after treatment with K252a, while the cells secreted albumin and α-fetoprotein into the medium as usual. These results show that K252a can increase the cell size of HuH7 without losing its function by blocking the cell cycle at G1 phase.

    Notes: Summary Conventional chemical fixation and paraffin-embedding procedures give good preservation of morphology, although the antigenicity of many proteins in the tissue sample is destroyed. On the other hand, fresh frozen sections can preserve the antigenicity, but provide poor morphological preservation. To overcome this dilemma, cryofixation and freeze drying were used on human skin tissue, applying methodology which has only been used to study lymphoid tissue. First, fresh human skin was cryofixed in liquid isopentane (−160° C) cooled by liquid nitrogen. The skin was then freeze-dried at −40° C and 10−2 atmospheric pressure for 72 h, followed by embedding in paraffin. Sections 4 μm thick taken from this cryofixed, freeze-dried, and paraffin-embedded skin were stained with hematoxylin-eosin or used for immunolabeling with antibodies against basement membrane antigen, including type IV and type VII collagen, bullous pemphigoid antigen, epidermolysis bullosa acquisita antigen, and GB3 antigen. The morphological preservation of these sections was as good as that of routine formalin-fixed and paraffin-embedded skin sections. The basement membrane was clearly immunostained with all antibodies used, and the intensity of the reaction was as strong as that seen in frozen sections. Evaluation of antigen distribution in conjunction with the detailed skin structure was therefore possible in the same sections.

    Notes: Abstract Objective To investigate the effect of PaCO2 on cerebral blood flow (CBF) in chronic obstructive pulmonary disease (COPD). Design Before-after trial. Setting General ICU in a regional hospital. Patients 7 patients undergoing mechanical ventilation because of an exacerbation of COPD. Intervention CBF and cerebral metabolic rate of oxygen (CMRO2) of COPD were measured before and after hyperventilation and were compaired by those of normal patients. CBF was measured by the Kety-Schmidt technique using 15% N2O. Measurements/results Hyperventilation produced a significant reduction in CBF in COPD with no concomitant change in CMRO2. CMRO2 in COPD was significantly lower than those in normal patients. The regression equation was shifted significantly more to the right in COPD. Conclusion The sensitivity of CBF in CO2 remained but CMRO2 was reduced markedly in COPD patients.

    Notes: Abstract Results of the measurement of flow properties in a two-dimensional turbulent wall jet which is injected into the turbulent boundary layer in the direction opposite to that of the boundary layer flow are presented by varying the ratio of the jet issuing velocity to the mainstream velocity λ. This flow includes the flow separation and the recirculating flow, and so it requires the magnitude and direction of instantaneous velocity be measured. In the present work, a tandem hot-wire probe is manufactured and its characteristics are examined experimentally. With the use of this probe the change in the penetration length of the jet with the velocity ratio is investigated. It is clarified that two regimes of flow patterns exist: in the low velocity ratio the penetration length of the jet changes little with λ, and in the high velocity ratio it goes far from the nozzle with increasing λ. Streamlines, turbulence intensity contours and static pressure contours are presented in the two typical velocity ratios corresponding to each of two flow patterns, and they are compared.

    Notes: Summary A horseradish peroxidase (HRP) labeled antibody method was developed for use with a monoclonal antibody to detect estrogen receptor (ER) in mouse tissue. Combined use of HRP labeled F(ab′)2 fragment absorbed with mouse liver protein to minimize background staining and imidazol-DAB reaction gave the most reliable and sensitive immunostaining. The method was applied to uterine, vaginal, pituitary and liver tissues in ovariectomized adult mice. In uterus and vagina, ER was recognized in nuclei of epithelial cells, stromal cells and smooth muscle cells of the muscle layer and blood vessels. Liver tissue showed positive nuclear immunostaining in parenchymal cells; however, no reaction was present in endothelial cells, Kupffer cells, bile ductal cells, and smooth muscle cells of blood vessels. ER was localized in the nuclei of anterior pituitary cells while weak reaction was also recognized in cells of the intermediate lobe. No staining was detected in the posterior pituitary. Results demonstrate that both occupied and unoccupied ER are localized in the cell nucleus from several target tissues. Weak immunostaining in samples could not be enhanced by multiple procedures. It is suggested that nuclear ER is partially hidden by nuclear components such as nuclei acid and chromatin proteins.

    Notes: Summary C-terminal amino acid sequence analysis seemed to be established procedure, as the counterpart of Edman's N-terminal sequencing method. However, poor recovery of the C-terminal amino acids in the reaction in homogeneous solution suggested further improvement of the method. In the present study, N-terminal amino acid was fixed covalently to the controlled pore glass (CPG) beads and the C-terminal amino acid was activated (by treating with acetic anhydride), coupled with thiocyanate to form thiohydantoin (TH) ring at the C-terminus. Then, the C-terminal amino acid was split off as the corresponding TH derivative, and analyzed by HPLC. Hydrolysis of the TH derivative was achieved at 60°C in the presence of 2 M HC1 for 2 h. Solid phase fixed peptide was washed simply with acetone, and dried for the next cycle of the reaction. So far obtained results in the heterogeneous mixture are not satisfactory in terms of the recovery of the C-terminal TH, and improvement of the recovery and further steps are under progress.

    Notes: Summary Both Taylor vortex flows in a symmetric or asymmetric system exhibit various patterns (cell modes). They can be classified by the process of cell formation, the number of cells and the direction of flow for the cell, into primary modes or secondary modes, and normal modes or anomalous modes. Following the previous report in which flows in a symmetric end condition were classified, in the present work, for flows in an asymmetric end condition, the Reynolds number at which a secondary mode bifurcates into another mode is experimentally investigated, and the bifurcation of the Taylor vortex flows in an asymmetric system when the Reynolds number is gradually decreased is presented in a bifurcation diagram.

    Notes: Summary The laminar boundary layer which develops on a rotating thin cylinder fitted with an aft-section rotating with an angular velocity different from that of a fore-section is examined numerically. This problem concerns a relaxation process of the boundary layer subjected to a sudden circumferential rate of strain. Two methods are adopted: one is to regard the flow on the aft-cylinder as perturbations of that on the fore-section, and the other is to approximate the discontinuous change in the angular velocity by the cumulative normal distribution function. It is shown that the flow fields are largely influenced by the degree of favorable pressure gradient produced in the boundary layer. Especially, in case of flow passing onto the aft-section with a smaller angular velocity, adverse pressure gradient is induced immediately after the junction between two cylinders, and when the degree of the discontinuity is increased, flow separation can be provoken.
