    Notes: Efficient and well-regulated intercellular communication is essential to organize the activities of the different cell populations involved during growth and concomitant structural maturation of the skeleton. For direct cell-cell communication gap junctions composed of connexins (Cx) are present. So far 21 Cx types have been identified in mammals, which differ in molecular permeability as well as tissue expression pattern. To characterize the Cx distribution in the developing canine skeleton, samples from humerus, scapula and lumbar vertebrae of Beagle dogs aged 1.5, 3, 4.5 months and 1.5 years were fixed in Bouin's solution, decalcified with 25% EDTA and embedded in paraffin wax. Using immunohistochemistry and in situ hybridization, the expression of Cx 26, 30, 31, 32, 43, and 45 was studied in bone tissue plus associated epiphyseal and growth plate cartilage. Of all bone cells, osteoblasts showed the largest Cx expression repertoire: they were positive for Cx 26, 30, 32 and 43. After transformation into bone lining cells Cx43 expression was lost while in osteocytes only Cx 26 and 32 was detected. A wide variety of Cx reactions was also found in osteoclasts: Cx 26, 30, 32, and 43. In contrast, chondrocytes showed a much more restricted Cx expression pattern. Positive reactions were only seen in hypertrophic chondrocytes of epiphyseal cartilage (Cx 26, 31, 32) and in the proliferative (Cx32) and hypertrophic zone (Cx 26 and 32) of growth plate cartilage. Additionally, Cx31 was strongly expressed in the cellular layer of the periosteum covering areas of intensive bone modelling in the puppies. Age- and microlocation-dependent variations in expression and/or staining intensity were also observed in the other Cx types. These results demonstrated that canine bone cells are capable of synthesizing a wide variety of Cx types during skeletal development, thus propagating different cellular signals via gap junctions in a differentially regulated and requirement orientated manner.

    Notes: The aim of this study was to describe morphological differences between the lateral collateral ligament (LCL) and the medial collateral ligament (MCL) of the canine elbow joint. Forty forelimbs of 22 dogs (adult German shepherd dogs and shepherd mongrels) were dissected. The length and width of defined segments of the ligaments (i. e. total length, areas of origin and insertion, ‘free part’, ‘humeral part’, ‘antebrachial part’) were measured in extension (160°) and flexion (30°). Statistical analyses of the data were performed using the data analysis software SAS 8.0. In addition the collateral ligaments of ten forelimbs of five shepherd mongrels were studied histologically. The LCL differs from the MCL in the following morphological items: a) the areas of origin and insertion are larger and in consequence the ‘free part’ of the lateral ligament is shorter; b) the caudal crus of the LCL is fan-shaped with a broad insertion area at the ulna whereas the caudal crus of the MCL remains slim; c) in the LCL the ‘humeral part’ varies in length depending on extended or flexed position of the joint, which is due to the spiral shape of the capitulum humeri; d) the microscopical structure of the LCL is organized less tightly; the collagen fibre bundles cross in varying angles. Referring to these details the LCL seems to allow and to limit a slight rotation of the forearm when the elbow joint is extremely flexed.

    Notes: In the eccrine glands of the digital pads of the North American raccoon (Procyon lotor), ultrastructural localization of glycoproteins with various saccharide residues was studied using preferably lectin cytochemical methods. Secretory granules observed in the dark cells exhibited glycoproteins with different terminal sugars (α-d-mannose, β-d-galactose, β-N-acetyl-d-glucosamine and sialic acid). The cytoplasm of the clear cells contained numerous glycogen particles. Several sugars were also detectable in the surface coat of the plasma membrane of the secretory cells. The results obtained could be helpful in understanding secretion production and cell related secretion functions of the eccrine glands of the raccoon digital foot pads.

    Notes: Summary The buccal absorption of procainamide was studied in 4 normal, male volunteers over a pH range of 5–11. In another investigation the same 4 volunteers collected their urine for 24 h after taking 250 mg of procainamide hydrochloride orally on 3 separate occasions, during which urine pH was uncontrolled, acidified by ammonium chloride ingestion or alkalinised by sodium bicarbonate ingestion. The procainamide remaining after buccal absorption and present in the urine samples was determined spectrophotometrically. It was found that neither the buccal absorption nor the renal excretion of procainamide was substantially altered by changes in pH.

    Description / Table of Contents: Zusammenfassung Der Vorteil der vorliegenden Perfusionsstudie des Pankreas in vitro ist der, daß er eine Vorhersage der funktionellen Lebensfähigkeit des perfundierten Pankreas in direkter Beziehung zur spezifischen Organfunktion erlaubt. Das Pankreas mit Duodenum wurde Hunden entnommen und für 3 Std mit einer Blut-Ringer-Lactatlösung mit einem Membranoxygenator, einer Pulspumpe und einem Wärmeaustauscher bei 37°C perfundiert. Aufgrund der vorliegenden Untersuchungen erweisen sich der Insulinausstoß nach Glucosegabe und die Pankreassekretion nach Sekretin-Reiz als besonders zuverlässige Kriterien in der Beurteilung der Lebensfähigkeit des Organs. Der durchschnittliche Sauerstoffverbrauch des Pankreas liegt bei 0,77 ml 02/100 g. Ein durchschnittlicher Flow von 80 ml/min bei einem Druck von 75 mm Hg erweist sich in der vorliegenden Versuchsanordnung als nützlich. Makroskopische und histologische Untersuchungen bestätigen die Ergebnisse.
    Notes: Summary An advantage of the current perfusion-test in vitro is that it allows prediction of the functional viability of the perfused pancreas in direct relation to the specific organ function. The canine pancreas including the duodenum has been perfused for 3 h with a membrane oxygenator, a pulsatile pump and a heat exchanger at a temperature of 37°C. Among the criteria used in this study the insulin-output into the venous blood in response to glucose administration as well as pancreassecretion after secretin stimulation were found to be the most reliable indications as to the viability of the pancreas. This is further supported by an average oxygen consumption of 0.77 ml 02/100 g at a constant flow of 80 ml/min and a pressure of 75 mm Hg, and the macroscopical and histological findings.

    Description / Table of Contents: Zusammenfassung Vier Gruppen mit insgesamt 124 Wistar-Ratten (Körpergewicht 100–120 g) erhielten im Bereich der Schwanzwurzel eine intracutane Injektion von 0,08 — 0,1 ml kompletten Freundschen Adjuvans. Eine zweite gleichartige Injektion wurde bei je einer Gruppe 25, 39, 52 bzw. 101 Tage nach der ersten Applikation vorgenommen. Alle Tiere wurden vom Tage der Erstinjektion bis mindestens 50 Tage post reinjectionem regelmäßig mikroskopisch auf extraoculare und spaltlampenmikroskopisch auf Augenveränderungen untersucht. Folgende Ergebnisse wurden erzielt: 1. Nach Reinjektion war die Latenzzeit arthritischer Veränderungen verkürzt. 2. Frühe Reinjektion führte neben einem unterhaltenden Effekt auf die Arthritis zu einer Vermehrung der Uveitis-Fälle und größerer Rezidiv-Häufigkeit. Der Anteil der Primär-Uveitiden war höher als der Anteil der Zweiterkrankungen. 3. Späte Reinjektion (52. bzw. 101. Tag p.i.) führte zu einem milderen Arthritis-Verlauf, wobei die Anzahl der erneut betroffenen Gelenke die Zahl erstmalig reagierender Gelenke erreichte oder deutlich übertraf. Uveitiden waren weniger häufig und weniger stark remittierend aber nicht milder als nach Erstinjektion. Die relative Häufigkeit von Zweiterkrankungen war indessen wesentlich höher als die der Primär-Uveitiden. Der Gedanke an einen lokal disponierenden Einfluß des Primäreffekts liegt nahe.
    Notes: Summary Four groups of 124 Wistar rats altogether (average body weight 100–120g) received intracutaneous injections of 0.08–0.1 ml Freund's complete adjuvant in the region of the tail head. A second injection of the same kind was given 25, 39, 52 or 101 days after the first one. From administration of the first injection until at least 50 days post reinjectionem all animals were examined macroscopically for extra-ocular lesions and biomicroscopically for eye lesions at frequent regular intervals. Finally serial sections of 44 eyes were examined by light microscopy. The following results were obtained: 1. After reinjection the latent period of arthritic changes was reduced. 2. Early reinjection, besides maintaining arthritis caused a higher incidence of uveitis and relapse. There was a higher proportion of primary uveitis than of recurrence. 3. Late reinjection (days 52 and 101 post injectionem) was followed by a milder course of arthritis, while the number of joints reacting for the second time equalled or distinctly exceeded that of joints inflamed for the first time. Cases of uveitis were less frequent and less prone to temporary remission but were no milder than those following the first injection. The frequency of recurrence of lesions was, however, significantly higher than that of primary uveitis. It may be that the primary effect predisposes the area to recurrence.

    Description / Table of Contents: Zusammenfassung Groß-Chinchilla-Kaninchen (1,5–2,5 kg KG) erhielten interscapulär eine einzige Injektion von respektive: a) Einer Emulsion von 50 mg (Naßgewicht) frischer homologer Retina in 0,4 ml kompletten Freundschen Adjuvans (110 Tiere); b) einer Suspension von 50 mg homogenisierter, frischer homologer Retina in 0,4 ml physiologischer Kochsalzlösung (10 Tiere); c) einer Emulsion von 50 mg frischer homologer Retina in 0,4 ml inkompletten Freundschen Adjuvans (10 Tiere); d) 0,4 ml kompletten Freundschen Adjuvans ohne Retina (20 Tiere). Vor Beginn des Versuchs wurden alle Kaninchen und post injectionem 90 Versuchs-sowie sämtliche Kontrolltiere über 40 Tage täglich auf Augenveränderungen (Ophthalmoskop, Spaltleuchte) und neurologische Ausfälle untersucht. 63 von 90 über die volle Dauer des Experiments beobachteten Tiere reagierten nach einer Latenzzeit von 12–39 Tagen mit einer Chorioretinitis. Die schwersten Entzündungen traten in der Regel während der 3. Woche post injectionem auf und erreichten binnen 60 Std ihr Maximum. Die klinischen Bilder der Erkrankung werden beschrieben und in drei Erscheinungsformen gruppiert. Klinische Zeichen einer Beteiligung der vorderen Augenabschnitte oder des ZNS wurden nicht gefunden. Die beschriebenen Veränderungen waren ausschließlich nach Injektion homologer Retina in komplettem Freundschen Adjuvans, jedoch nicht nach Injektion der Kontrollpräparate b, c, und d zu beobachten.
    Notes: Summary Great Chinchilla rabbits (body weight 1.5–2.5 kg) were given interscapularly a single immunizing dose of, respectively: a) 50 mg (wet weight) of freshly dissected homologous retina emulsified in 0.4 ml of Freund's complete adjuvant (110 animals); b) 50 mg homologous retina as above incorporated in 0.4 ml saline (10 animals); c) 50 mg homologous retina as above incorporated in 0.4 ml of Freund's incomplete adjuvant (10 animals); d) 0.4 ml Freund's adjuvant (20 animals). Before starting the experiment, and at daily intervals thereafter, all animals were examined for eye lesions (ophthalmoscope, slit lamp) and for neurological deficits. In 90 experimental and in all control animals these examinations were carried out over a period of 40 days. 63 of 93 experimental animals developed ocular lesions characterized by chorioretinic patches of varying size in all parts of the fundus. The time of disease onset varied from 12 to 39 days after immunization. The most violent inflammations appeared as a rule in the third week after inoculation and reached maximum severity within 60 hours. Three types of clinical pictures are described. No involvement of the anterior segment or of the central nervous system was noted clinically in any of the animals. In contrast to the control preparations tested (b, c, d), only homologous retina in Freund's complete adjuvant was effective.
