    Notes: Cyclotron resonance in GaAs/AlGaAs heterostructures, containing a high-mobility two-dimensional electron gas (2DEG), has been investigated with the help of a resonator technique over the frequency range 26–34 GHz. Heterostructures with mobilities from 2×105 to 1.6×106 cm2/V s have been measured. The microwave conductivity of the 2DEG is well described by the Drude model and momentum relaxation time values are found to be close to dc values. The cyclotron mass was found to be 0.068±0.002m0.

    Notes: A noninvasive voltage probe, consisting of a one-dimensional channel, has been fabricated within a modulation doped GaAs/AlGaAs heterostructure. With precision piezoelectric scanning equipment this probe has been brought to within 14 nm of a semiconductor device which includes surface gates and a 300 nm deep two-dimensional electron gas. Measurements of voltages applied to these conducting layers have been made at room temperature and at 4.2 K using the sensitive conductance of the one-dimensional channel. A voltage resolution of 0.48 mV has been observed at 4.2 K. Probe conductance measurements have also been made as a function of probe–sample separation. The conductance–separation data were fitted using a simple parallel plate capacitor model and a height resolution of 4 nm was calculated. Images of the sample layers have been obtained and a lateral resolution of 760 nm observed. © 2001 American Institute of Physics.

    Notes: We describe a detailed device fabrication technique for the formation of a lateral quantum dot using a multilayered gated design. In our versatile system, a quantum dot is electrostatically defined by a split gate and two overlaying finger gates which introduce entrance and exit barriers to the dot. Periodic and continuous conductance oscillations arising from Coulomb charging effects are clearly observed in the transport properties at low temperatures. © 2000 American Institute of Physics.

    Notes: We report on the fabrication and electron transport in a novel heterostructure in which layers of AlGaAs and GaAs have been grown after a submicron free-standing GaAs wire has been formed. Electronic conduction at low temperatures in this material is shown to be consistent with three-dimensional hopping conduction with a transition to one-dimensional hopping at temperatures below 1 K.

    Notes: We have studied the quantized conductance of a one-dimensional ballistic channel in the two-dimensional electron gas of a back-gated GaAs/AlGaAs heterostructure. A standard Schottky split-gate fabricated with electron-beam lithography techniques is used to define the one-dimensional channel, but we incorporate an epitaxially grown in situ back-gate, situated ∼1 μm below the electron gas, to provide additional control of the carrier density. Quantized conductance steps can be induced by changing the bias on either gate, highlighting the self-consistent nature of the electrostatics involved. We show that we can, in principle, achieve independent control of the one-dimensional carrier density and channel width.

    Notes: Abstract Coupling of a soft X-ray laser beam with a relaying concave mirror in a sequentially pumped amplifier geometry using the Ne-like Ge system has been studied experimentally. Preliminary observations indicate an increase in the spatial coherence of the amplified relayed beam. In addition, near-field imaging of one of the amplifier plasmas shows a double-lobed intensity pattern of the emergent beam indicating refractive guiding of the amplified beam with components both normal and tangential to the target surface.

    Notes: Abstract A perfusion system for production of monoclonal antibodies was developed using an externally-mounted, hollow-fibre cartridge. The experimental apparatus was operated for 420 h and demonstrated increased steady-state viable cell concentration with increase in perfusion rate. Antibody titres were up to three times those measured for batch cultures and specific antibody productivity was doubled. The procedure was successfully scaled to a 10 dm3 system which produced antibody under conditions of Good Manufacturing Practice (GMP). A calculation of productivity between the scaled perfusion system and 260 dm3 batch cultures resulted in comparable antibody production, whereas the perfusion allowed a halving in medium utilisation. Reactivity assays conducted on the purified antibody from both batch and perfusion cultures showed no evidence of proteolysis or altered antibody activity in the final perfusion product. This study provides additional support for the use of homogeneous perfusion cultures in production of monoclonal antibodies under GMP conditions.

    Notes: A comprehensive 13C-NMR method for the analysis of composition in the most common commercial polyethylene copolymers has been established. The method covers ethene copolymers with propene, butene-1, hexene-1, octene-1, and 4-methyl pentene-1 in the composition range of 1-10 mol %. The chemical shift assignments and T1 values of the resonances of the copolymers are presented. Results of precision studies and interlaboratory analyses showed that the molar composition could be determined with a relative precision at 2δ of about 6%. This method is being proposed to ASTM as Method X70-8605-2.

    Notes: The use of solvent-free systems in the oil and fats industry is commonplace. Initial studies on interesterification were carried out in solvent systems because the lipase was immobilized solely by adsorption onto particles of diatomaceous earth. In this study, the mass transfer characteristics associated with the continuous interesterification of olive oil in a solvent-free system have been examined, for lipase immobilized on the three ion-exchange materials: Duolite ES562, Duolite ES568, and Spheroil DEA. The process of immobilization is influenced by the internal structure of the material and this in turn influences the interesterification activity of the catalyst. Individually prepared catalysts for the three support materials have shown that external mass transfer limitations are unlikely even at low flowrates.In the case of Spherosil DEA, with a mean pore diameter of 1480 Å, the wide pores would be expected to reduce internal mass transfer limitations; however, it is more likely that the reduction in activity with increased catalyst loading is due to the lipase molecules being immobilized in a tightly packed monolayer. In such a situation, some active sites of the lipase molecules would become inaccessible to substrate molecules leading to an observed reduction in activity. For Duolite ES568, the observed results are very similar to those seen for Spherosil DEA, however, the pore structure of this support material indicate that some internal mass transfer limitations may also be occurring. Yet the contribution of the individual effects cannot be determined. The results observed for the support Duolite ES562 are different than those observed for the other materials and reflect the heterogeneity of Duolite ES562. The large proportion of narrow pores in the support mean that, for the catalysts examined, immobilization is most likely to have occurred in the external pores of the particles, and as such no internal mass transfer limitation is observed.It is clear that for interesterification the material chosen for enzyme immobilization will have an important role in determining the catalyst efficiency. External mass transfer limitations are very minor and observed internal mass transfer limitations may be caused by both internal mass transfer and the manner in which the immobilization process occurs. © 1994 John Wiley & Sons, Inc.
