Notes: Summary The possibility that liver sinusoidal cells are derived from the bone-marrow was investigated in chimeric mice. H2k-positive bone-marrow cells from F1 (B10.BR × B10.D2) hybrid mice were transplanted into irradiated H2k-negative parental mice (B10.D2), and the liver examined immunohistochemically for the presence of H2k-positive cells, with the help of an anti-H2k monoclonal antibody. With the passage of time (from the fifth week onwards), increasing numbers of transplanted bone-marrow cells enter the liver sinusoids, undergo alteration in their shape, and remain there, probably replacing sinusoidal lining cells. DNA-synthesising cells in the sinusoids were observed, suggesting, in addition, local cell proliferation. The replacement of sinusoidal cells from bone-marrow was greatly accelerated after liver damage had been induced by sublethal doses of endotoxin (LPS), and proliferation was also enhanced after treatment with LPS. These results strongly suggest that the bone-marrow participates in the replacement of liver sinusoidal cells.

Notes: Summary The mechanism of cell replacement of non-parenchymal liver cells (NPLC) was investigated in an attempt to answer the question: to what extend are NPLC replaced by proliferation of local resident cells and to what extend by cells originating from the bone-marrow. The bone-marrow of mice was used, and H2k positive cells from F1 (B10.BR X B10.D2) hybrid mice were transplanted into irradiated H2k negative parent animals. Their NPLC were isolated and tested immunocytochemically with a monoclonal anti-H2k antibody for the presence of H2k positive cells. During the whole of the experiment (from the 5th to the 20th week following transplantation) H2k positive cells (macrophages and non-macrophages) were present, and made up an average of nearly one third of the NPLC. The H2k positive (immigrant) non-macrophages showed essentially more active DNA synthesis than these H2k negative cells. To increase cell turnover, we injected one group of animals with endotoxin. At the time of maximum replacement, more than 50% of the NPLC (macrophages and non-macrophages) were recruited from cells of bone-marrow origin. Noteworthy here was the proportionate level of the H2k positive macrophages (more than 70% of all liver macrophages). The DNA synthesis of both the H2k positive macrophages and non-macrophages was more than twice that of the H2k negative NPLC during the regeneration process following administration of endotoxin. Our observations suggest that the bone-marrow contributes significantly to the replacement of macrophages and non-macrophages of NPLC in both health and disease.

Notes: Abstract To achieve its health goals, the United States must reduce the disproportionate burden of illness and poor health borne by urban populations. In the 20th century, patterns of immigration and migration, changes in the global economy, increases in income inequality, and more federal support for suburbanization have made it increasingly difficult for cities to protect the health of all residents. In the last 25 years, epidemics of human immunodeficiency virus infections and substance abuse and increases in homelessness, lack of health insurance, rates of violence, and concentrations of certain pollutants have also damaged the health of urban residents. Several common strategies for health promotion are described, and their relevance to the unique characteristics of urban populations is assessed. To identify ways to strengthen health promotion practices in U.S. cities, lessons have been taken from five related fields of endeavor: human rights, church- and faith-based social action, community economic development, youth development, and the new social movements. By integrating lessons from these areas into their practice, public health professionals can help to revitalize the historic mission of public health, contribute to creating healthier cities, and better achieve national health objectives.

Notes: Abstract The nicotine dehydrogenase from Arthrobacter oxidans was purified 40-fold to homogeneity with 26% recovery. SDS-polyacrylamide gel electrophoresis of the enzyme revealed three protein bands corresponding to Mr of 82 000, 30 000 and 15 000. The Mr of the native enzyme was calculated to be 12 0000 by gel chromatography. The enzyme contained about 1 FAD, 1 molybdenum, 4 iron and 2 labile sulfur.

Notes: Summary We report on incidental findings during family screening of two kindreds with multiple endocrine neoplasia type IIa. Pheochromocytoma and medullary thyroid carcinoma of considerable size were detected. The results underline the importance of early diagnosis of the syndrome, since the afflicted may be almost or wholly asymptomatic. High resolution chromosome banding studies were carried out in both families, but no abnormality was found. Linkage analysis using DNA markers closely related to the chromosomal locus at chromosome 10 was carried out and was positive in two asymptomatic offspring of one family, whereas the markers were not informative in a second family. We recommend early linkage analysis for establishing the genetic status in offspring of multiple endocrine neoplasia type IIa families to identify for further screening those who are predicted to be gene carrier.

Notes: Abstract It is shown that to each locally normal state of a boson system one can associate a point process that can be interpreted as the position distribution of the state. The point process contains all information one can get by position measurements and is determined by the latter. On the other hand, to each so-called Σc-point processQ we relate a locally normal state with position distributionQ.

Notes: Abstract Antibodies raised against the glycine decarboxylase proteins P1, P2, P3, and the selenoprotein PA, a component of the glycine reductase complex, were used for immunocytochemical localization experiments. Cells of Eubacterium acidaminophilum from logarithmic growth phase were fixed in the growth media with paraformaldehyde and glutaraldehyde. Fixed cells were embedded by the low-temperature procedure using Lowicryl K4M resin, and the protein A-gold technique was applied for the localization experiments. The vicinity of the cytoplasmic membrane contained about 27% of all gold particles when proteins P1 and P2 were to be localized, 50% for protein PA, and 61% for protein P3. Double immunocytochemical labeling experiments gave evidence for the existence of a protein P1/P2 complex located predominantly in the cytoplasm, and a P3/PA complex located at the cytoplasmic membrane. Only in very few instances the labels for proteins P3 and P1 were seen very close together in respective doublelabeling experiments. These results indicate that glycine decarboxylase does not occur in this organism as a complex consisting of all four proteins, but that protein P3, the atypical lipoamide dehydrogenase, takes part in both the glycine decarboxylase as well as in the glycine reductase reaction.
