    Notes: Abstract From the ability to successfully manipulate the mouse genome has come important transgenic and gene-targeted knockout models that impact many areas of biomedical research. Genetically engineered mouse models geared toward the study of cardiovascular regulation have recently been described and provide powerful tools to study normal and compromised cardiac physiology. The genetic manipulation of the adrenergic receptor (AR) signaling system in the heart, including its regulation by desensitizing kinases, has shed light on the role of this signaling pathway in the regulation of cardiac contractility. One major finding, supported by several mouse models, is that in vivo contractility can be enhanced via alteration of myocardial AR signaling. Thus genetic manipulation of this critical receptor system in the heart represents a novel therapeutic approach for improving function of the failing heart.

    Notes: Bei Flughafer wurde der Emfluß verschiedener Applikationsorte zwischen Spitze und Basis des Sprosses auf die Penetration und Translokation von 14C-Diclofop-methyl untersucht. Die Penetration liegt an der Basis der Blattspreitc am 1. Blatt um 64% und am 2. Blatt um 95% höher als an der Spitze des jeweiligen Blattes. Die Translokation von 14C-Diclofop-mcthyl in basipeitaler Richtung ist begrenzt. Daher ist die Menge an 14C-Diclofop-methyl an der Sproßbasis, dem Hauptwirkort, um so höher, je näher das Herbizid zur Sproßhasis appliziert wird. Hinsichtlich einer möglichst optimalen Verteilung von 14C-Didofop-methyl in der Pflanze erwies sich die Applikation an die Basis del Blattsprciten als am günstigsten. Dies beruht u.a. darauf daß es bei diesem Applikationsort zu einem Einlaufen der Herbizidlösung in die Blattscheidc und infolgedessen zu einem mechanischen basipetalen Vordringen innerhalb dieser in den Bereich der mcristematischen Gewehe der Sproßbasis kommt. Als Konsequenz daraus ergibt sich die Forderung nach einer möglichst gezielten Deponierung von Diclofop-methyl in den Bereich der Sproßbasis.

    Notes: Background There is an increasing need for allergen inhalation systems to perform basic clinical research and test anti-allergic drugs under well-controlled conditions. This requires stability of environmental conditions like temperature and humidity, as well as allergen concentration and reproducible induction of allergic symptoms.Objective The aim of this study was to validate an environmental exposure unit for controlled human pollen inhalation studies in participants with seasonal allergic rhinitis.Methods Temperature, relative humidity, and air flow rate were kept constant with an air conditioning system. Pollen atmosphere was generated using a specially designed feeding system and monitored online by laser counter and offline using rotating rod samplers. Efficacy (total nasal symptom score, nasal air flow rate, nasal secretion) and safety (lung function) parameters were evaluated at different pollen concentrations and repeated allergen challenges.Results Temperature, humidity, and air flow rate in the environmental exposure unit remained constant within a range of 〈2%. The spatial distribution and the temporal stability of the pollen concentration varied only slightly over 4 h (±10% and 〈4%, respectively). Dose-dependent induction of allergic rhinitis symptoms, reduction in nasal air flow rate, and increase in nasal secretion were observed over time. These effects were reproducible from day to day. Lung function remained clinically normal at all concentrations and from day to day.Conclusions Thus, pollen exposure in the environmental exposure unit is an effective, reproducible, safe, and suitable method for single-centre clinical studies on the efficacy of anti-allergic treatment or basic clinical research.

    Notes: Abstract: Dendritic cells seem to be of major importance for the pathogenesis of psoriasis. They are increased in number in lesional psoriatic skin which is thought to be due to an increased influx from the peripheral blood regulated by chemotaxins. Using a biological/biochemical approach we have addressed the question whether psoriasis scale extracts contain proteinaceous chemotaxins for dendritic cells. Human monocytes differentiated into dendritic cells by culture with GM-CSF and IL-4 (MoDC) served as responder cells. Chemotactic activity for MoDC was purified by several HPLC-steps. The results of our study show that C5a/C5adesarg is the major chemotactic peptide for MoDC in psoriasis scale extracts. In comparison to other stimuli such as fMLP or monocyte chemotactic peptide 1 (MCP-1) C5a proved to be a most potent and efficient chemotaxin for MoDC. C5a co-eluted with MRP14/calgranulin B which is present in large amounts in psoriasis scale extracts as identified by amino acid sequencing. However, MRP14/calgranulin B did not possess any chemotactic activity for MoDC. Our results provide evidence that C5a/C5adesarg although not specific for dendritic cells seems to be the major chemoattractant for these cells in lesional psoriasis skin.

    Notes: Abstract A model representing a two- or a three-dimensional array of classical harmonic chains withnonlinear coupling between them is investigated. Physically real systems to which this model applies are discussed. The model exhibits soliton-like nonlinear modes. The influence of these nonlinear modes on the static and the dynamic correlation functions is calculated by generalizing techniques developed for strictly one-dimensional systems. In the static correlation functions these modes lead to minor quantitative changes only. In certain dynamic correlation functions, however, a central peak is found to occur due to the nonlinear modes. The total weight and the width of this peak are calculated for a real spin system.

    Notes: Summary The relationship between stroke volume (SV) measured simultaneously by impedance cardiography (SV1) and echocardiography (SVE) has been analyzed. Change in stroke volume in 6 healthy volunteers was induced by passive head-up tilting (80°) and by oral administration of the sympathomimetic drug amezinium. A correlation ofr=0.83 (p j 0.01) was found between SVI and SVE calculated by the equation: SVE=16.0+(0.528×SVI). Though stroke volume is overestimated by impedance cardiography, the method may be useful for the clinical pharmacologist interested in a simple non-invasive tool to estimate stroke volume and change in it.

    Description / Table of Contents: Zusammenfassung Nachdem psychophysische Experimente nahelegen, daß schon nach geringen Leuchtdichteänderungen die interokulären Latenzdifferenzen bei MS-Patienten beträchtlich zunehmen, untersuchten wir den Einfluß von Leuchtdichteänderungen auf die diagnostische Trefferrate der VEP bei der Multiplen Sklerose. Über einen Bereich von drei logarithmischen Einheiten brachten Leuchtdichteänderungen keine Zunahme der diagnostischen Ausbeute. Auch die Regressionskoeffizienten der Funktion VEP-Latenz gegen Leuchtdichte unterschieden sich bei MS-Kranken nicht von denen bei Gesunden. Da bei abnehmender Leuchtdichte die Trefferquote der VEP sogar deutlich abnimmt, muß diese Variable in der Klinik beachtet werden.
    Notes: Summary In view of the fact that with psychophysical methods monocular luminance changes may increase the detection rate of pathological interocularlatency differences in MS patients, we studied the influence of stimulus luminance on the detection rate of MS using checkerboard visual evoked potentials. Decrements of stimulus luminance covering a range of three log units were unable to increase the detection rate of VEP. Regression coefficients of the luminance-latency functions did not differ in MS patients and normals. Contrary to the hypothesis tested, the diagnostic significance of VEP decreases with decreasing stimulus intensity.
