    Notes: Summary Schwannomas are benign solitary tumours of the peripheral nerve sheaths. The occurrence of multiple schwannomas usually implies hereditary disease. The most frequent syndrome associated with multiple schwannomas is neurofibromatosis type 2 (NF2), which is defined by bilateral vestibular schwannomas. Schwannomatosis is a distinct disease characterized by multiple pathologically proven schwannomas in the absence of vestibular schwannomas. It is not currently known if the presence of multiple schwannomas confined to a limb may represent a mosaic form of NF2 or a distinct disease, because mutation analysis of these tumours is not routinely performed. We report a 31-year-old patient who presented with multiple slowly growing subcutaneous tumours on his left arm. His family history was negative for cutaneous tumours or central nervous system disease, and he did not have additional features of NF2. Magnetic resonance tomography and ophthalmological examination excluded vestibular schwannoma and eye stigmata of NF2. After resection of three tumours, histological analysis confirmed the diagnosis of benign schwannomas. Molecular genetic analysis by temperature gradient gel electrophoresis and microsatellite marker analysis demonstrated two distinct mutations of the NF2 gene (NF2) in two different schwannomas, with concomitant loss of heterozygosity in both tumours. In contrast, neither normal skin nor peripheral blood lymphocytes revealed mutations of NF2. The clinical and molecular genetic findings suggest that the diagnosis in our patient is schwannomatosis rather than segmental NF2 because the mutations found in different tumours were not identical. The possibility of a localized predisposition for the acquisition of NF2 mutations is discussed.

    Notes: A central role of iron in the pathogenesis of Parkinson's disease (PD), due to its increase in substantia nigra pars compacta dopaminergic neurons and reactive microglia and its capacity to enhance production of toxic reactive oxygen radicals, has been discussed for many years. Recent transcranial ultrasound findings and the observation of the ability of iron to induce aggregation and toxicity of α-synuclein have reinforced the critical role of iron in the pathogenesis of nigrostriatal injury. Presently the mechanisms involved in the disturbances of iron metabolism in PD remain obscure. In this review we summarize evidence from recent studies suggesting disturbances of iron metabolism in PD at possibly different levels including iron uptake, storage, intracellular metabolism, release and post-transcriptional control. Moreover we outline that the interaction of iron with other molecules, especially α-synuclein, may contribute to the process of neurodegeneration. Because many neurodegenerative diseases show increased accumulation of iron at the site of neurodegeneration, it is believed that maintenance of cellular iron homeostasis is crucial for the viability of neurons.

    Notes: Abstract Haemochromatosis is characterised by deposition of iron-containing pigment in various organs, but little is known about possible deposition in the brain and its clinical impact. We therefore investigated 14 patients with hereditary haemochromatosis with MRI, CT and transcranial ultrasound (TCS) and examined them neurologically. In six of the patients dense lesions were found within the lentiform nucleus on CT, all of whom displayed hyperechogenic lesions in the same area on TCS, as did one other patient. In these patients the relative signal intensities of the lentiform nucleus measured by MRI relaxometry were higher. No patient had clinical signs of basal ganglia disorders.

    Notes: Summary ¶ Purpose. In this prospective study the results of multimodal postoperative neuro-imaging were related to the survival of patients with high grade gliomas.  Methods. All 73 patients included underwent microsurgical tumour resection and had postoperative CT and transcranial sonography (TCS) examinations. In addition, 35 of the 73 patients received an early postoperative MRI. Patients were followed up for at least one year.  Findings. At the end of the 7 year study period 56 patients had died. The median survival time was 371 days. Survival rate was significantly higher in patients with anaplastic astrocytomas and inpatients displaying complete tumour resection on MRI (log-rank-test, p〈0.05) or a small postoperative residual tumour bulk on TCS (log-rank-test, p〈0.05). Cox proportional hazards model identified histological tumour grade, postoperative Karnofsky index, complete resection based on MRI and small postoperative residual tumour mass on TCS as independent predictors of survival.  Interpretation. This study demonstrates that early postoperative neuro-imaging has prognostic implications for the survival of patients with high grade gliomas. According to our results postoperative imaging with MRI and TCS is a valuable prognostic with regard to patient survival and should therefore be implemented in postoperative follow-up. It also helps to evaluate the efficacy of adjuvant therapy.

    Description / Table of Contents: Zusammenfassung Ein bis fünf Tage nach i. p. Applikation von 67-Gallium-Zitrat in mit Ehrlich Ascites Tumorzellen beimpften Mäusen wird die Konzentration von 67-Gallium in Asciteszellen und im Ascitesserum bestimmt. Die maximale Galliumanreicherung in Asciteszellen gegenüber der Galliumkonzentration im gleichen Volumen Ascitesserum wird am 2. bis 3. Tage erreicht und beträgt dann das 30fache. Ein Dialyseversuch des Asciteszellhomogenates ergab, daß innerhalb von 24 Std. bei pH 7.5 nur um 25% des Galliums dialysierbar sind. Dieser Wert ist ein Maximalwert und wird auch nach weiteren Stunden nicht mehr überschritten. Die restlichen 75% müssen an größere Moleküle oder Organellen der Zelle gebunden sein. Nach Differentialzentrifugation des Zellhomogenates ergibt sich folgende Verteilung der 67-Gallium-Gesamtaktivität in den Zellen: Cytoplasma 61%, Mikrosomen 18%, Kerne 9%, Mitochondrion 5%, Ribosomen 5% und Lysosomen 2%. Diese Werte entsprechen dem durch Zentrifugation auf dem diskontinuierlichen Dichtegradienten erhaltenen Ergebnis. Die Fraktionierung der Cytoplasmafraktion durch Gelfiltration auf Sephadex G-100 zeigt einen Aktivitätsgipfel, der synchron mit der Proteinfraktion läuft und 80% der Gesamtaktivität enthält. Bei den restlichen 20% der Gesamtaktivität handelt es sich wahrscheinlich um den dialysierbaren Galliumanteil. Bei der Zellfraktionierung nach Schmidt und Thannhauser (1945), modifiziert nach Schneider (1945), findet sich nahezu die gesamte Aktivität im Perchlorsäure-Überstand. Dies besagt, daß die Gallium-Bindung an die Makromoleküle innerhalb der proteinhaltigen Fraktionen säurelabil und wahrscheinlich nicht kovalent ist. Die Bindung von Gallium an Cytoplasmoproteine von Tumorzellen wird diskutiert.
    Notes: Summary Hyperdiploid Ehrlich ascites tumor cells (Bayer, Wuppertal-Elberfeld) were inoculated into the peritoneal cavity of NMRI-mice. When these cells were incubated in vivo together with 67-gallium-citrate the cell concentration of this isotope is greatest on the third day. On this day the cellular concentration of 67-Ga is about 30 times greater than that of the same volume of the surrounding ascites serum. 25% of the gallium within the cell homogenate is dialyzable, 25% probably is bound to larger molecules or cell organelles. After differential centrifugation of the ascites cell homogenate the following distribution of 67-Ga within ascites cells is found: cytoplasm 61%, microsomes 18%, nuclei 9%, mitochondria 5%, ribosomes 5% and lysosomes 2%. These results are confirmed by a centrifugation of the homogenate on a sucrose-gradient. After gel-filtration on Sephadex G-100 80% of the total activity of the cytoplasm-67-Ga is found within the protein peak. It is likely that the remaining 20% of cytoplasmic gallium is the dialyzable isotope. After fractionating the cells by the method described by Schmidt and Thannhauser (1945), most of the total activity is found in the perchloric acid supernatant. This result shows that the binding of gallium within the tumor cells is acid labile, loose and probably not a covalent one. The binding of gallium to cytoplasmic proteins in tumor is discussed.

Nach 4, 5, 6 und 10 Tagen wurden die Regenerate histologisch und histochemisch (Glykogen, Glucose-6-phosphat-Dehydrogenase und RNS) untersucht. In Abhängigkeit von der Actinomycin-D-Konzentration werden bestimmte Phasen der Regeneration gehemmt; dabei sind Proliferationsprozesse weniger empfindlich als Differenzierungsprozesse. Histochemisch werden bei Hemmung der wachstumsaktiven Areale die im Normalfall zu beobachtenden stofflichen Merkmale: Glykogeneinlagerung und nachfolgender Anstieg der G-6-P-DH-Aktivität und der RNS-Konzentration vermißt.
    Notes: Summary Fragments ofLineus sanguineus were incubated in seawater, which contained different concentrations of actinomycin-D (1.9 μg/ml, 0.95 μg/ml and 0.48 μg/ml). After 4, 5, 6 and 10 days the regenerates were studied by histological and histochemical methods (glycogen, glucose-6-phosphate-dehydrogenase and RNA). Corresponding to actinomycin-D concentration certain phases of regeneration are inhibited. The phases of proliferation are less sensitive to actinomycin-D than the phases of differentiation. If morphogenetic areas are inhibited, the normally demonstrable histochemical characteristics: accumulation of glycogen and later, the increase of G-6-P-DH activity and RNA concentration do not appear.

    Description / Table of Contents: Zusammenfassung Bei der Ermittlung der Bleilässigkeit von Bedarfsgegenständen erfolgt zunächst eine Säurebehandlung des Gegenstandes, dann wird im Säureauszug der Bleigehalt ermittelt. Es werden verschiedene Verfahren der Säurebehandlung aufgeführt, wobei der Kaltextraktion mit 4 % iger Essigsäure über 24 Std der Vorzug gegeben wird. Die Bleibestimmung im essigsauren Auszug wird mit Hilfe der AAS durchgeführt. Das Chromatverfahren nach DIN 51031 wird in einigen Punkten geändert. Nach dem abgewandelten Chromatverfahren werden die gleichen Bleigehalte wie mit Hilfe der AAS erhalten. Sodann wird auf die rechtliche Beurteilung der vom Gebrauchsgegenstand abgegebenen Bleimenge unter Berücksichtigung der internationalen Gesetzgebung hingewiesen. Die Rechtslage in der BRD ist unbefriedigend. Es wird vorgeschlagen, die im Essigauszug enthaltenen Bleimengen auf die den Speisen zugewandte Oberfläche des Gebrauchsgegenstandes zu beziehen und eine Höchstmenge von 2 mg Blei je dm2 Oberfläche zu tolerieren.
    Notes: Summary The determination of lead solubility from ceramics is realized by extraction with acetic acid (4 % w/w in water, 24 hours at room temperature) and by following determination of lead by atomic absorption. The supplementary analysis by iodometric determination as lead chromate (DIN 51031) is varied in some points, so that both ways give the same results. The legal valuation of lead solubility is discussed in an international view.
