    Notes: An ion accelerator, purpose built to produce beams at energies down to 10 eV with current densities in the 10–100 μA cm−2 range, is described. Fitted with dual ion source assemblies, the machine enables ultralow energy ion implantation and the growth of films and multilayers to be carried out under highly controlled conditions. The accelerator delivers ion beams into an ultrahigh vacuum chamber, containing a temperature controlled target stage (range −120 to +1350 °C), where they are used to study the fundamental physics relating to the interaction of ultralow energy ions with surfaces. This knowledge underlies a wide range of ion-beam and plasma-based technologies and, to illustrate its importance, results are presented from investigations designed to determine the optimum conditions for the growth of diamond-like and aluminum films by ion-beam deposition and the formation of ultrashallow junctions in semiconductors by 2.5 keV As+ implantation. The later investigation shows how transient arsenic diffusion, which occurs during post-implant thermal processing, can be controlled by manipulating the substrate temperature during implantation. © 2000 American Institute of Physics.

    Notes: Sm–Nd garnet-whole rock geochronology, phase equilibria, and thermobarometry results from Garnet Ledge, south-eastern Alaska, provide the first precisely constrained P–T–t path for garnet zone contact metamorphism. Garnet cores from two crystals and associated whole rocks yield a four point isochron age for initial garnet growth of 89.9 ± 3.6 Ma. Garnet rims and matrix minerals from the same samples yield a five point isochron age for final garnet growth of 89 ± 1 Ma. Six size fractions of zircon from the adjacent pluton yield a concordant U–Pb age of 91.6 ± 0.5 Ma. The garnet core and rim, and zircon ages are compatible with single-stage garnet growth during and/or after pluton emplacement. All garnet core–whole rock and garnet rim-matrix data from the two samples constrain garnet growth duration to ≤5.5 my. A garnet mid-point and the associated matrix from one of the two garnet crystals yield an age of 90.0 ± 1.0 Ma. This mid-point result is logically younger than the 90.7 ± 5.6 Ma core–whole rock age and older than the 88.4 ± 2.5 Ma rim-matrix age for this sample. A MnNaCaKFMASH phase diagram (P–T pseudosection) and the garnet core composition are used to predict that cores of garnet crystals grew at 610 ± 20 °C and 5 ± 1 kbar. This exceeds the temperature of the garnet-in reaction by c. 50 °C and is compatible with overstepping of the garnet growth reaction during contact metamorphism. Intersection of three reactions involving garnet-biotite-sillimanite-plagioclase-quartz calculated by THERMOCALC in average P–T mode, and exchange thermobarometry were used to estimate peak metamorphic conditions of 678 ± 58 °C at 6.1 ± 0.9 kbar and 685 ± 50 °C at 6.3 ± 1 kbar, respectively. Integration of pressure, temperature, and age estimates yields a pressure-temperature-time path compatible with near isobaric garnet growth over an interval of c. 70 °C and c. 2.3 my.

    Notes: We report a rare case of cellular angiolipoma of the breast in a 73-year-old woman with abnormal mammographic findings. Histopathologic examination of the tumor revealed cellular spindle cell areas associated with collapsed and patent blood vessels in a background of mature adipose tissue. A salient feature of this tumor, which was an important clue to the diagnosis, was the presence of intravascular fibrin thrombi. Immunohistochemical studies showed reactivities for endothelial and muscle markers, highlighting the obscured angiomatous component in the cellular areas of the tumor. Failure to recognize this entity may potentially lead to diagnostic pitfalls that include angiosarcoma and Kaposi's sarcoma, two of the better recognized entities in this region.

    Notes: Maturation of Cultured Neonatal Rat Ventricular Myocytes. Introduction: Cultured neonatal rat ventricular myocytes (NRVM) reestablish gap junctions as they form synchronously and spontaneously beating monolayers, thus providing a useful model for studying activation and repolarization. Methods and Results: We used the multielectrode array data acquisition system with 60 unipolar electrodes to investigate the functional organization of cultured NRVM, by determining propagation and repolarization patterns. Activation maps were constructed from the local activation times at each electrode. During days 3 to 8 in culture, QRS amplitude and dV/dtmax increased with age. Concomitantly, with the culture maturation, QT interval (representing action potential duration) decreased, and T wave amplitude and slopes of the T wave ascending and descending limbs progressively increased. The changes in conduction velocity were different than those of the electrogram properties, slightly increasing during the first 3 to 5 days and gradually declining toward day 8 in culture. Conclusion: Establishment of uniform activation patterns in spontaneously firing or driven myocytes in monolayer cultures is accompanied by organization of activation and repolarization whose evolution appears in concert with that of a mature connexin43 staining pattern. The experimental techniques developed in this study provide useful tools to investigate the complex relations among gap junctions, conduction velocity, and propagation patterns, as well as a means to learn how gap junctional remodeling under pathophysiologic conditions predisposes the myocardium to arrhythmias.

    Notes: Previous studies have demonstrated opposing roles for adenosine A1 and A2A receptors in the modulation of extracellular levels of glutamate and dopamine in the striatum. In the present study, acute systemic administration of motor-activating doses of the A2A receptor antagonist MSX-3 significantly decreased extracellular levels of dopamine and glutamate in the shell of the rat nucleus accumbens (NAc) and counteracted both dopamine and glutamate release induced by systemic administration of motor-activating doses of either the A1 receptor antagonist CPT or caffeine. Furthermore, exposure to caffeine in the drinking water (1 mg/mL, 14 days) resulted in tolerance to the effects of systemic injection of CPT or caffeine, but not MSX-3, on extracellular levels of dopamine and glutamate in the NAc shell. The present results show: first, the existence of opposite tonic effects of adenosine on extracellular levels of dopamine and glutamate in the shell of the NAc mediated by A1 and A2A receptors; second, that complete tolerance to caffeine's dopamine- and glutamate-releasing effects which develops after chronic caffeine exposure is attributable to an A1 receptor-mediated mechanism. Development of tolerance to the dopamine-releasing effects of caffeine in the shell of the NAc may explain its weak addictive properties and atypical psychostimulant profile.

    Notes: Adenosine, by acting on adenosine A1 and A2A receptors, exerts opposite modulatory roles on striatal extracellular levels of glutamate and dopamine, with activation of A1 inhibiting and activation of A2A receptors stimulating glutamate and dopamine release. Adenosine-mediated modulation of striatal dopaminergic neurotransmission could be secondary to changes in glutamate neurotransmission, in view of evidence for a preferential colocalization of A1 and A2A receptors in glutamatergic nerve terminals. By using in vivo microdialysis techniques, local perfusion of NMDA (3, 10 µm), the selective A2A receptor agonist 2-p-(2-carboxyethyl)phenethylamino-5′-N-ethylcarboxamidoadenosine (CGS 21680; 3, 10 µm), the selective A1 receptor antagonist 8-cyclopentyl-1,3-dimethylxanthine (CPT; 300, 1000 µm), or the non-selective A1-A2A receptor antagonist in vitro caffeine (300, 1000 µm) elicited significant increases in extracellular levels of dopamine in the shell of the nucleus accumbens (NAc). Significant glutamate release was also observed with local perfusion of CGS 21680, CPT and caffeine, but not NMDA. Co-perfusion with the competitive NMDA receptor antagonist dl-2-amino-5-phosphonovaleric acid (APV; 100 µm) counteracted dopamine release induced by NMDA, CGS 21680, CPT and caffeine. Co-perfusion with the selective A2A receptor antagonist MSX-3 (1 µm) counteracted dopamine and glutamate release induced by CGS 21680, CPT and caffeine and did not modify dopamine release induced by NMDA. These results indicate that modulation of dopamine release in the shell of the NAc by A1 and A2A receptors is mostly secondary to their opposite modulatory role on glutamatergic neurotransmission and depends on stimulation of NMDA receptors. Furthermore, these results underscore the role of A1 vs. A2A receptor antagonism in the central effects of caffeine.

    Notes: The endoplasmic reticulum (ER) and the Golgi comprise the first two steps in protein secretion. Vesicular carriers mediate a continuous flux of proteins and lipids between these compartments, reflecting the transport of newly synthesized proteins out of the ER and the retrieval of escaped ER residents and vesicle machinery. Anterograde and retrograde transport is mediated by distinct sets of cytosolic coat proteins, the COPII and COPI coats, respectively, which act on the membrane to capture cargo proteins into nascent vesicles. We review the mechanisms that govern coat recruitment to the membrane, cargo capture into a transport vesicle, and accurate delivery to the target organelle.

    Notes: We present a high-spatial-resolution subsurface microscopy technique that significantly increases the numerical aperture of a microscope without introducing an additional spherical aberration. Consequently, the diffraction-limited spatial resolution is improved beyond the limit of standard subsurface microscopy. By realizing a numerical aperture of 3.4, we experimentally demonstrate a lateral spatial resolution of better than 0.23 μm in subsurface inspection of Si integrated circuits at near infrared wavelengths. © 2001 American Institute of Physics.

    Notes: We compare the degradation of InAs/GaAs quantum well (QW) and quantum dot (QD) laser diodes following irradiation by high energy (8.56 MeV) phosphorous ions. Over a fluence range of 108–1011 ions/cm2, the degradation of the low temperature QD photoluminescence and electroluminescence emission is greatly suppressed relative to that of QW based devices (×100 and ×1000, respectively at the highest dose studied). Irradiated QD laser diodes demonstrated lasing action over the entire range of fluences, and 2 orders of magnitude beyond the maximum dose sustainable by QW devices. The improved damage response of QD based structures results from efficient collection and localization of electrons and holes by QDs in the active region, which limit carrier transfer to nonradiative centers. This work suggests the suitability of QD device architectures for use in radiation environments, and in high power applications, wherever nonradiative processes promote the degradation or failure of traditional QW devices. © 2000 American Institute of Physics.
