    Notes: Amorphous LaNi5 films were prepared using a sputtering method. Mechanical properties and hydrogenation characteristics of the films were investigated and the results were compared with those of the deposited films. The density of the sputtered LaNi5 film was about 6 g/cm3, which was larger than that of the evaporation deposited film but smaller than that of crystalline LaNi5. Thermal conductivities of amorphous films were larger than those of bulk samples. Mechanical strength of the sputtered films deposited on various substrates was measured after 100 cycles of hyrogen absorption–desorption process. The result was in the following order for the substrates used: Ni foil〉Ni〉Al foil〉Cu foil〉Cu〉Al〉glass. The hydrogen content in a sputtered film with a thickness of 1.3 μm was found to be 2.4 H/LaNi5. The hydrogen concentration in the film increased monotonically with increasing the hydrogen pressure in both cases between 333 and 363 K.

    Notes: A male infant with partial DiGeorge syndrome responded to weekly administration of levamisole (2.5 mg/kg of body weight) with an increase of circulating E-rosette-forming T cells. Thymic hormone activity in plasma appeared to be elevated to a near-normal level of 11.6 ng thymopoietin equivalent/ml after levamisole administration. The in vitro incubation studies indicated that levamisole by itself had no E-rosette-promoting ability, but a dialysable and relatively heat-stable plasma factor induced by levamisole both in the patient and in heallhy individuals had E-rosette-promoting activity for the patient's lymphocytes. Such a plasma factor, however, could not be induced in all four thymectomized myasthenic subjects examined, suggesting a thymus-dependent nature of the plasma factor. These results suggest that levamisole might mediate an increased secretion of humoral factor(s) with E-rosette-promoting activity, even from such a rudimentary thymus as in the partial DiGeorge syndrome.

    Notes: A single oral dose of 150 mg levamisole was administered to five healthy adults. Circulating Fc(IgG) receptor-bearing T cells (Tγ cells) increased for 5 days after levamisole intake, but total E rosette-forming cells showed no significant alterations. The generation of immunoglobulin-producing cells in the peripheral blood lymphocytes (PBL), which was induced in the in vitro pokeweed mitogen (PWM)-stimulated cultures, was significantly suppressed for 5 days after levamisole administration. Suppressor T-cell activity on B-cell differentiation, which was induced by levamisole intake, was evaluated by co-culturing with allogeneic untreated adult PBL in the PWM system in six other volunteers. A seemingly dose-dependent suppression on B-cell differentiation was exerted by T cells isolated on day 3 of levamisole treatment, but not by T cells which were isolated before or on day 14 of the experiment. When T cells were fractionated into two subsets with regard to the presence or absence of Fc(IgG) receptors, suppressor T-cell activity appeared to be generated by levamisole largely in T cells lacking Fc(IgG) receptors, but not in Tγ cells.

    Notes: In tuberculin-sensitive individuals, IgG Fc receptor (FcR)-hearing lymphocytes in the peripheral blood increased transiently following PPD-tuberculin skin test. This rise in circulating FcR-bearing cells appeared to peak about 36–48 h after the intradermal inoculation of PPD and seemed to occur largely in the T cell population. Skin test-negative individuals showed no significant changes in their circulating FcR-bearing cells following PPD inoculation. Peripheral blood lymphocytes from PPD-sensitive individuals were fractionated into non-T cell and T cell-enriched populations by E rosette sedimentation technique. FcR-bearing cells in the T cell-enriched population were eliminated by EA rosette sedimentation: i. e. FcR-negative T cells. Then, equal numbers (1 × 105 cells each) of non-T cells and unfractionated or FcR-negative T cells were recombined in culture. Prior to PPD inoculation, there was no significant difference between these two cell mixtures in the in vitro cellular response to PPD or mitogens. When these cell populations were obtained. 16–48 h after PPD inoculation, however, the combination of non-T tells and FcR-negative T cells responded to PPD much better than the combination of non-T cells and unfractionated T cells, whereas the mitogen-induced cellular proliferation of these two cell mixtures did not differ from each other.

    Notes: Four clonal lines of ayu, Plecoglossus altivelis Temminck & Schlegel, were produced through gynogenesis, mixed before hatching and reared communally. After 10 months, a randomly taken sample was subjected to a standardized shallow-water stressor. Haematocrit, haemoglobin, red blood cells count (RBC) and mean corpuscular volume (MCV) were obtained from stressed and non-stressed fish. DNA fingerprinting was used to confirm the clonal nature of the organisms and to identify the clonal line to which each fish belonged.We observed significant differences between clonal lines mostly in the haematocrit and MCV measured under no-stress conditions. Such differences are suggested to represent mainly genetic variance, on account of the common environment provided to all the experimental groups.The stress response ratio was lower than expected, mainly due to some unexpectedly high non-stress values.Heritability values (h2) were medium to high for the no-stress measurements (mean 0.238) and very low or zero for the stressed groups’traits (excepting one high value of 0.484).We conclude that the use of communally reared clonal lines represents a good tool for the characterization of the physiological traits, thus allowing for their utilization as genetic selection criteria.

    Notes: Abstract. Triploidy was induced in three full-sib families of sea bream, Pagrus major (Temminck & Schlegel), by subjecting the fertilized eggs to cold-shock treatment at 0°C for a 12 min duration, starting 3 min after insemination. Results showed that this treatment gave 100% triploid fish with reasonable survival rates of more than 61% up to first feeding, Triploidy induction was proved by the use of allozyme markers at three (ADH*, EST* and 6PGD*) loci and red blood cell size analysis.At 2 months of age, the survival of the triploid group was lower than that of the diploids but it was comparable at 10 months. No differences in growth rate between diploid and triploid full-sibs were observed up to the age of 10 months. However, differences in body weight between families were noticed both in diploid and triploid groups when cultured in the same net cage. This possibly suggests a high heritability for the trait examined in the broodstock. The gonads of the triploid fish were significantly smaller than those of the diploids, Histological analysis of the gonads revealed sterility in triploid fish.

    Notes: Acetylglucosoaminyltransferase V (GNTV), an enzyme responsible for branching of asparagine-linked oligosaccharides is associated with tumor invasion and metastasis. Beta1–6 branched oligosaccharides, detected with the lectin, LPHA, are associated with tumor progression in human breast and colon cancer. They were found to be common to 22 types of human cancers, and were particularly expressed in metastases. Matriptase is a proteolytic inducer of both the HGF/cmet and plasminogen activator pathways, highly associated with metastasis. A potential prometastatic effect of GNTV was found to be due to modification of active matriptase by adding beta1–6 GlcNAc branching. We investigated expression of LPHA, matriptase and GNTV in pathology specimens of primary invasive and metastatic malignant melanomas. Sequential sections were stained with anti-GnT-V, anti-matriptase, or biotinylated LPHA. Our results showed variable staining patterns, from involving only small regions, to encompassing the entire tumor. The expression of all three markers was closely associated, suggesting that metastatic pathways might have been activated in the staining areas of these tumors. To our knowledge, this is the first report of matriptase expression in malignant melanoma, as well as the first demonstration of an association between matriptase, GNTV and beta 1,6-branched oligosaccharides in vivo.
