    Notes: The dynamics of polyimide ablation plume particles generated during ArF excimer laser (193 nm) ablation were investigated using time-resolved, laser light scattering and Raman spectroscopy. The evolution of plume particle size is determined over a range of 300–1150 μm above the polymer target surface. Raman-scattering measurements of the ablation plume allow in situ identification of the particle composition, which is equivalent to flame generated soot. The total mass of particulate carbon within the plume accounts for approximately 1% of the total carbon mass within the ablated polyimide. Scanning electron microscopy was used to examine the size and morphology of collected plume particles, which were found to correlate remarkably with the light-scattering results. The formation of particulate carbon within the ablation plume is apparently complete within 500 μm above the polymer surface, corresponding to a time scale of 30 μs, and then gives way to carbon oxidation at greater heights. It is concluded that particulate carbon is formed within the resulting ablation plume, with subsequent particle growth and oxidation processes being important in the evolution of the particles.

    Notes: Phase-matched optical second-harmonic (SH) generation was observed in GaN and AlN slab waveguides. Phase matching was achieved by waveguide modal dispersion. By tuning the output wavelength of an optical parametric amplifier, several phased-matched SH peaks were observed in the visible spectrum covering blue to red wavelengths. The peak positions are in agreement with the values calculated using the dispersive refractive indices of the film and substrate materials. © 1999 American Institute of Physics.

    Notes: The laser-induced breakdown spectroscopy technique has been extended to provide quantitative analysis of the mass and elemental composition of individual, submicrometer to micrometer-sized aerosol particles. A two-part approach was used for calibration of the overall mass concentration response, and of the characteristic plasma volume, equal to 2.5×10−4 cm3. Laboratory results are presented for submicrometer-sized particles containing a known concentration of magnetite. Additional data are presented for fine particulate matter measured in ambient air, including magnesium-containing particles with an average size of 313 nm. © 1998 American Institute of Physics.

    Notes: Self-assembled chromophoric multilayers having large second-order optical nonlinearities can be combined with linear guiding materials such as polymethylmethacrylate to produce two-component frequency doubling waveguides. This approach introduces considerable flexibility in optimizing the trade-off between the overlap of waveguide mode profiles and linear absorption. Extremely low waveguide propagation losses can be achieved because electric field poling and the accompanying poling-induced optical scattering are obviated. © 1997 American Institute of Physics.

    Notes: We present the results of large-area (20 cm2) diamond deposition from a scaled-up stagnation-flow reactor. The reactor uses a unique nozzle geometry that optimizes reagent gas usage. The premixed acetylene–oxygen–hydrogen flames were operated in a highly strained configuration, allowing uniform deposition of diamond with growth rates exceeding 25 μm/h. Substrate temperature control and flame stability of the chemical vapor deposition reactor are described. Diamond films were deposited on a molybdenum substrate with a surface temperature of approximately 1200 K and C/O ratio of 1.03. Diamond film growth results are presented, and film uniformity is assessed using micro-Raman spectroscopy. © 1996 American Institute of Physics.

    Notes: Thin films of potassium titanyl phosphate (KTiOPO4) for photonic applications have been fabricated on fused quartz substrates by pulsed excimer laser ablation. The textured films were optically uniaxial with the symmetry axis oriented normal to the film surface. The principle component of the second order electric susceptibility was oriented parallel to this symmetry axis and was determined to be about 16 times the magnitude of quartz for the fundamental wavelength of 1.064 μm. This configuration, with the largest nonlinear optical response for polarizations perpendicular to the film surface, is desirable in TM guided wave applications. Planar optical waveguide measurements were performed and the linear waveguide loss was measured. The frequency dependence of χ(2)(ω) was determined and found to be uniform for visible second harmonic wavelengths. © 1995 American Institute of Physics.

    Notes: Abstract In situ hybridization with rRNA-targeted, fluorescent (Cy3-labeled) oligonucleotide probes was used to analyze bacterial community structure in ethanol- or paraformaldehyde-fixed bulk soil after homogenization of soil samples in 0.1% pyrophosphate by mild ultrasonic treatment. In ethanol-fixed samples 37 ± 7%, and in paraformaldehyde 41 ± 8% of the 4′, 6-diamidino-2-phenylindole(DAPI)-stained cells were detected with the bacterial probe Eub338. The yield could not be increased by enzymatic and/or chemical pretreatments known to enhance the permeability of bacterial cells for probes. However, during storage in ethanol for 7 months, the detectability of bacteria increased in both ethanol- and paraformaldehyde-fixed samples to up to 47 ± 8% due to an increase in the detection yield of members of the α-subdivision of Proteobacteria from 2 ± 1% to 10 ± 3%. Approximately half of the bacteria detected by probe Eub338 could be affiliated to major phylogenetic groups such as the α-, β-, γ-, and δ-subdivisions of Proteobacteria, gram-positive bacteria with a high G+C DNA content, bacteria of the Cytophaga-Flavobacterium cluster of the CFB phylum, and the planctomycetes. The analysis revealed that bacteria of the α- and δ-subdivision of Proteobacteria and the planctomycetes were predominant. Here, members of the α-subdivision of Proteobacteria accounted for approximately 10 ± 3% of DAPI-stained cells, which corresponded to 44 ± 16 × 108 cells (g soil, dry wt.)–1, while members of the δ-subdivision of Proteobacteria made up 4 ± 2% of DAPI-stained cells [17 ± 9 × 108 cells (g soil, dry wt.)–1]. A large population of bacteria in bulk soil was represented by the planctomycetes, which accounted for 7 ± 3% of DAPI-stained cells [32 ± 12 × 108 cells (g soil, dry wt.)–1]. The detection of planctomycetes in soil confirms previous reports on the occurrence of planctomycetes in soil and indicates a yet unknown ecological significance of this group, which to date has never been isolated from terrestrial environments.

    Notes: Abstract We examined the abundance and spatial distribution of major phylogenetic groups of the domain Bacteria in hindguts of the Australian lower termite Mastotermes darwiniensis by using in situ hybridization with group-specific, fluorescently labeled, rRNA-targeted oligonucleotide probes. Between 32.0 ± 7.2% and 52.3 ± 8.2% of the DAPI-stained cells in different hindgut fractions were detected with probe EUB338, specific for members of the domain Bacteria. About 85% of the prokaryotic cells were associated with the flagellates of the thin-walled anterior region (P3a) and the thick wall of the posterior region (P3b/P4) of the hindgut, as shown by DAPI staining. At most, half of the EUB338-detected cells hybridized with one of the other probes that targeted a smaller assemblage within the bacterial domain. In most fractions, cells were found in varying numbers with probe ALF1b, which targeted members of the α-Proteobacteria, whereas substantial amounts of sulfate-reducing bacteria, gram-positive bacteria with a high DNA G+C content and members of the Cytophaga-Flavobacterium cluster of the Cytophaga-Flavobacterium-Bacteroides (CFB) phylum could be detected only in the wall fraction of P3b/P4. This clearly indicates that the hindgut microhabitats differ in the composition of their microbial community. In situ hybridization of cryosections through the hindgut showed only low numbers of bacteria attached to the P3a wall. In contrast, the wall of P3b was densely colonized by rod- and coccus-shaped bacteria, which could be assigned to the Cytophaga-Flavobacterium cluster of the CFB phylum and to the group of gram-positive bacteria with a high DNA G+C content, respectively. Oxygen concentration profiles determined with microelectrodes revealed steep oxygen gradients both in P3a and P3b. Oxygen was consumed within 100 μm below the gut surface, and anoxic conditions prevailed in the central portions of both gut regions, indicating that oxygen consumption in the hindgut does not depend on the presence of a biofilm on the hindgut wall.
