    Notes: AbstractPurpose: The ratio of Bcl-2 to Bax expression determines survival or death following an apoptotic stimulus. In order to establish a new predictor of the outcome of treatment for human cervical carcinoma, we investigated the relationship between the expressions of the Bax and Bcl-2 proteins and the response to radiotherapy after the administration of 10.8 Gy. Methods: A total of 44 patients with histologically proven carcinoma of the uterine cervix, including three with recurrent cervical stump carcinomas, were treated with definitive radiotherapy. The presence of mutations in exons 5–8 of the p53 gene was analyzed by a single-strand conformation polymorphism analysis and DNA sequencing. Results: Forty patients were found to have wild-type p53, and the remaining four had mutant p53. The Bax and Bcl-2 protein expressions prior to radiotherapy did not correlate with response and survival. However, the Bax and Bcl-2 protein expressions after radiotherapy correlated with both response and survival. Bax-positive tumors showed significantly better responses than the Bax-negative tumors after 10.8 Gy radiation (P = 0.0002). In contrast, the Bcl-2-positive tumors showed significantly poorer responses than the Bcl-2-negative tumors after radiation (P = 0.002). Increased Bax expression after the 10.8 Gy radiotherapy was found to be correlated with good survival (P = 0.04). In contrast, increased Bcl-2 expression after such radiotherapy was correlated with poor survival (P = 0.002). Conclusion: The levels of Bax and Bcl-2 expression after 10.8 Gy radiotherapy are useful prognostic markers in patients with human cervical carcinoma.

    Notes: Abstract The effects of chemoembolization with cisplatin on gynecological malignancy were investigated using rabbit uterine tumors. A group of 20 rabbits were subjected to inoculation of the uterus with 5×107 VX2 carcinoma cells and 4 weeks later were divided into four groups, each consisting of five rabbits: an untreated control group, a group given cisplatin intraarterially (IA), a group subjected to transcatheter arterial embolization (TAE) with Gelfoam particles and a group subjected to transcatheter chemoembolization (TACE) with Gelfoam particles plus 1 mg/kg cisplatin. All groups were examined histologically 2 days after treatment. The untreated control group was further investigated 4 weeks after inoculation. In the untreated control group, the tumor cell nuclei varied in size and were irregular in form, and multiple nuclei and nuclear division were also observed. No necrotic zones were found up to 4 weeks after inoculation. The IA group showed no necrosis, but a few apoptotic cells were scattered throughout the tumor. In the TAE group, necrosis was observed in the center of the tumors, but proliferating cells persisted at the periphery. In the TACE group, necrosis was observed in the central part with many apoptotic cells surrounding the necrotic region in layers. The proliferating cell nuclear antigen (PCNA) index was 95.88% in the untreated control group, 86.6% in the IA group, and 8.62% in the TACE group, indicating a significant reduction in cell proliferation in the TACE group. These findings suggest that TACE results in more effective cytotoxicity than the other two treatments in uterine cancer tumor transplants.

    Notes: Abstract Purpose: To determine the usefulness of a new platinum microcoil, the Berenstein Liquid Coil for vascular embolization. Methods: Nine patients underwent transcatheter arterial embolization with liquid coils. The occluded vessels included the splenic artery in four patients, gastroduodenal artery in two, arteriovenous malformation or fistula in two, and middle hepatic artery in one. All coils were delivered with a saline flush through a Tracker-18 catheter. Results: All arteries except one were successfully occluded in a one-stage procedure using liquid coils. No friction was observed between the liquid coils and the microcatheter regardless of the tortuosity of the artery. Platelet counts in all patients had increased after splenic embolization. In two patients with liver tumors, perfusion to the inappropriate area during intraarterial chemotherapy was prevented by selective embolization. Life-threatening bleeding was successfully treated by gastroduodenal artery occlusion. No complications were encountered. Conclusion: The Berenstein Liquid Coil provides easy, safe, and rapid vascular occlusion.

    Notes: Abstract We have sequenced a region (7 376-bp) of the mitochondrial (mt) DNA (54 kb) of the cellular slime mold, Dictyostelium discoideum. From the DNA and amino-acid sequence comparisons with known sequences, genes for ATPase subunit 9 (ATP), cytochrome b (CYTB), NADH dehydrogenase subunits 1, 3 and 6 (ND1, ND3 and ND6), small subunit rRNA (SSU rRNA) and seven tRNAs (Arg, Asn, Cys, Lys, f-Met, Met and Pro) have been identified. The sequenced region of the mtDNA has a high average A+T-content (70.8%). The A+T-content of protein-genes (73.6%) is considerably higher than that of RNA genes (61.3%). Even with the strong AT-bias, the genetic code employed is most probably the universal one. All seven tRNAs are able to form typical clover leaf structures. The molecular phylogenetic trees of CYTB and SSU rRNA suggest that D. discoideum is closer to green plants than to animals and fungi.

    Notes: Abstract During the course of mitochondrial DNA sequencing ofDictyostelium discoideum, a sequence with a tRNA-like structure and two genes for tRNAGin(UUG) and tRNATrp(CCA) were found downstream of the gene for large subunit rRNA. The existence of tRNATrp with CCA anticodon supports the finding that UGA codon is not a tryptophan codon inD. discoideum mitochondria. Interestingly, the tRNA gene-like sequence (t-element) has TTA at the anticodon position. Northern blot analysis showed that, in low molecular mass mitochondrial RNA fraction of growth-phase cells and developmental stage cells, a mature transcript of the ele ment could not be detected in the tRNA region on an ureadenatured 15% polyacrylamide gel, although there were several bands in the higher molecular mass region, indicating the actual transcription of the t-element. Southern blot analysis for total and mitochondrial DNA showed that the element exists as a single copy, only in the mitochondrial DNA but not in the nuclear DNA.

    Notes: Abstract A multicentre study on multicompartmental analysis of hepatic scintigraphy using technetium-99m labelled galactosyl serum albumin (GSA), which binds to the asialoglycoprotein receptor, was carried out at seven institutions in Japan. Seventy-four patients with liver disease received 3 mg (185 MBq) of99mTc-GSA by intravenous injection. Sequential scanning was performed 30 min after injection to obtain anterior images of the heart and liver, followed by single-photon emission tomography (SPET). The indices included in this analysis were hepatic blood flow (Q) and maximal receptor binding rate (R max), which showed a good correlation with semiquantitative ratio indices for99mTc-GSA, namely the retention rate in blood (HH15) and the hepatic uptake rate (LHL15).Q andR max also showed a significant correlation with other measures of hepatic function. When patients were grouped according to the severity of chronic liver damage (hepatocellular functional damage),Q was reduced in the moderate and severe groups, whileR max was reduced in proportion to the functional stage. Both parameters showed no inter-institution difference using analysis of co-variance with the functional stage as a co-variant. With regard to the hepatic uptake rate, anterior planar images and SPET images gave similar results forQ andR max. Acquisition times of 15 or 30 min provided the same results. The multicopartmental model analysis permitted comparable results to be obtained at institutions using different gamma cameras, and is therefore considered a universally applicable method. These results indicate thatQ andR max are useful general indices for evaluating the function reserve capacity of the liver.
