    Notizen: Abstract Cells of Bacillus subtilis 168+ were labeled with 32P-orthophosphate during the process of sporulation, germination and outgrowth. By two-dimensional gel electrophoresis, at least 30 protein species were found to be radioactively labeled; 30% of these were modified by phosphorylation. Significant changes in the protein phosphorylation pattern during growth and cellular differentiation could be demonstrated. Using γ-32P-ATP evidence for an ATP-dependent protein kinase was also obtained. Under these conditions 4 proteins with a molecular mass of 109 600; 103 100; 73 300 and 32 200 Da were found to be phosphorylated.

    Notizen: Summary Two monoclonal Beta-cell surface antibodies M10H6 und K14D10 were obtained by fusion of spleen cells of Balb/c mice with the myeloma cell line P30. The monoclonal antibody M10H6 was induced by immunization with rat insulinoma cells finally boostered with disintegrated rat islets, whereas the K14D10 was generated after immunization with porcine proinsulin. Both monoclonals belong to the IgG2A isotype and were screened with insulin-producing rat insulinoma cells by an indirect immunofluorescence test as well as by a cellular enzyme linked immunosorbent assay. In addition to the cell surface binding on living Beta cells the monoclonals react with islets on cryostat sections of rat pancreas. The anti-islet cytotoxic potential of these monoclonals was measured by 51Chromium-release in the presence of complement or Fc-receptor bearing leucocytes using 51Chromium-labelled rat islet cells as target. Both antibody secreting hybridomas were propagated in syngeneic mice resulting in high levels of islet cell surface antibodies in ascites and sera from the recipient. High anti-islet cytotoxicity was mediated by ascites fluid, but no mouse developed hyperglycaemia. Furthermore, the repeated injections of the monoclonals into rats did not exert a diabetogenic action and failed to reduce the pancreatic insulin content although the attraction of the K14D10 to the pancreatic islets in vivo could be demonstrated. We conclude that islet cell surface antibody-mediated Beta-cell lysis in vitro may not be relevant to Beta-cell destruction in vivo.

    Notizen: Summary To determine the local gastrointestinal absorption of a new synthetic somatostatin analogue (SMS 201-995 = Sandostatin), an intestinal tube was passed in eight healthy volunteers and on different days an aqueous solution was administered at four different locations: stomach, proximal duodenum, ligament of Treitz and jejunum. In a follow-up study, an oro-ileal tube was passed in six of the original volunteers and the drug solution was administered in to the terminal ileum. The aqueous solution of SMS was rapidly absorbed from the gastrointestinal tract after local application, and it was well tolerated. Absorption of the drug from the different sites was comparable, although there was a tendency to decreased peptide absorption after ileal administration. Absorption of the drug was quite variable between the subjects and the different locations. The dose-corrected systemic availability relative to subcutaneous administration in another study was 0.28%. However, significant plasma SMS concentrations were achieved, suggesting that oral delivery of the polypeptide may eventually be possible for long-term treatment of a variety of disorders.

    Beschreibung / Inhaltsverzeichnis: Summary Experiments on quantitative elemental analysis of glasses with muonic X-rays have been performed. The summed-up Lyman intensities were compared with the known abundances of the samples in order to deduce calibration numbers per element. In favourable cases a precision of the order of 1% of the respective abundance was obtained. The level of detection was about 100 ppm. The method is completely non-destructive and allows selection of certain parts (scanning), even well inside the sample. It may be combined with truly three-dimensional visualization.
    Notizen: Zusammenfassung Es wurden Experimente zur quantitativen Elementanalyse von Gläsern mit myonischer Röntgenstrahlung durchgeführt. Die Summe der Intensitäten in der myonischen Lymanserie wurde für die verschiedenen Elemente der Proben mit den bekannten atomaren Konzentrationen verglichen und damit für jedes Element eine Eichzahl erhalten. In günstigen Fällen konnte die Konzentration mit einer Genauigkeit von etwa 1% bestimmt werden. Die Nachweisgrenze lag bei 100 ppm. Die Methode ist völlig zerstörungsfrei. Es können auch ausgewählte Teile einer Probe analysiert werden (‘scanning’), selbst wenn sie weit innerhalb liegen. Die Methode kann mit einer dreidimensionalen Darstellung der Zusammensetzung verbunden werden.
