    Notizen: The initial lymph vessels, the lymph capillaries and precollectors, are difficult to detect in normal skin using routine histological methods. A method is described in which specimens of the skin are fixed in an expanded condition. In 1.05 μm thick plastic-embedded sections the dilated lymph vessels of the skin can be demonstrated. The dermal lymph vessel network can be seen following the subepidermal injection of a coloured solution. The endothelial cells and subendothelial fibrous network can be examined using electron microscopy.

    Notizen: Two computer programs – one for calculation and graphical representation of the azimuthal angle ψ of all possible multiple reflections versus the wavelength λ (PSILAM) and one for the simulation of the Umweganregung-intensity variation during rotation of the crystal about the scattering vector of a `forbidden' or very weak reflection (PSIINT) – have been developed on the basis of the program UMWEG [Rossmanith (1985). Z. Kristallogr. 171, 253–254; (1986). Acta Cryst. A42, 344–348] with emphasis on suitability for inexpensive hardware. Examples of the excellent agreement between measurements and calculations and of the necessity of the combined application of both programs are given.

    Notizen: Summary We describe a method which makes it possible to study the expression of the trimeric glycoprotein (TGP, CD 103) on the surface of hairy cells (HCs) by electron microscopy (e.m.). The monoclonal antibody (mAb) Ber-ACT8 was used to identify the TGP. By testing mucosa of the small intestine, we found that a prefixation incubation with Ber-ACT8 was necessary. After isolation with a density gradient, Ber-ACT8 incubation, and fixation, the peroxidase-anti-peroxidase (PAP) method was used to make the TGP visible. The HCs showed a clearly discernible stain on their surface, a fairly constant distribution of the TGP, and a very good ultrastructural preservation. This method makes it possible to demonstrate the TGP or other unfixable antigens by e.m.

    Notizen: Abstract New dyes with characteristic fluorescence lifetimes have been developed for bioanalytical applications. Based upon the concept of “multiplex dyes,” we have designed rhodamine dyes with nearly identical absorption and emission spectral characteristics but different fluorescence lifetimes. Extending this principle to applications with laser diodes, new rhodamines with functional groups for covalent coupling of analytes have been developed. The new labels exhibit absortion and fluorescence beyond 600 nm and have a high quantum efficiency, even in aqueous buffer systems.

    Notizen: Summary Confocal laser scanning microscopy (CLSM) and fluorochromes were used to visualize the assimilate-conducting sieve cells of conifers in vivo. When still nucleate, the cytoplasm of these cells shows streaming and occupies the cell periphery including the pitlike, thin wall regions where sieve areas would develop. During differentiation the nuclear fluorescence and the central vacuoles disappear. At maturity and after ER-specific staining the sieve areas are the most conspicuous character of sieve cells. Those linking two sieve cells are covered on either side with prominent amounts of ER, while those leading to a Strasburger (=albuminous) cell show fluorescence on the sieve-cell side only. Within the sieve-area wall fluorescence appears also in the common median cavity which is part of the symplastic path between sieve cells. Electron microscopy (EM) depicts the ER as complexes of densely convoluted tubules of smooth ER, equally on either side of a sieve area, provided that the fixation of this sensitive tissue is appropriate. Purposeful wounding causes a swelling and vesiculation of the ER-tubules which is visible in both CLSM and EM. Electron micrographs of ER-complexes at sieve areas -in this paper demonstrated in vivo -have often been argued to be artefacts, since they should raise flow resistance considerably and are not consistent with the Münch hypothesis on phloem transport. The implications of this location for phloem transport are discussed.

    Beschreibung / Inhaltsverzeichnis: Zusammenfassung Mit den bisher üblichen Färbemethoden war es kaum möglich, sicher zwischen frühen ischämischen Herzmuskelschäden, autolytischen Veränderungen and auch intaktem Myocard ohne Kernanschnitt zu unterscheiden. 1986 wurde erstmals eine immunhistochemische Methode vorgestellt, mit der C5b-9-Komplement-Komplex als Nekrosemarker am Kryostatschnitt dargestellt wurde. In Fortentwicklung dieses Verfahrens wird eine Technik zur Anwendung am Routine-Paraffinschnitt beschrieben: Mit dem C5b-9-Komplement-Komplex gelingt es insbesondere, disseminierte Einzel- und Gruppenfasernekrosen des Myocards exakt and autolyseunempfindlich darzustellen. Die Anwendung dieser Methode am Paraffinschnitt eröffnet den Weg zur Routinediagnostik sehr früher ischämischer Herzmuskelschädigungen selbst in Fallen von “Sekundenherztod”.
    Notizen: Summary With conventional staining techniques it was nearly impossible to differentiate with certainty between early ischemic myocardial lesions, autolytic changes and intact myocardium without a nuclear section. In 1986 it was introduced an immunohistochemical method, which presented C5b-9 Complement Complex as a necrosis marker in cryostat sections. Our further development of this process has resulted in a technique for application on routine paraffin sections: C5b-9 Complement Complex is particularly well-suited for the exact and autolytically insensitive presentation of disseminated necroses of fiber bundels and single fibers of myocardium. The application of the technique on paraffin sections will lead to the use of the same in forensic medicine routine diagnostics of early ischemic myocardial lesions, even in cases of “sudden” cardiac death.
