    Notizen: Abstract The dissolution of fused calcium phosphate (FP) and transformation of P were monitored in a silt loam, sandy loam, sandy clay and clay soil (pH in water = 5.3 − 5.8) incubated with 0, 108 or 324 mg P, as FP kg−1 soil dry wt, for 10, 30 or 60 d. Soluble P was fractionated by sequential extraction using a chloride anion exchanger (P-resin) and solutions of NaHCO3 (PtCO3), NaOH (PtOH) and HCl (P-HCl). Inorganic P (PiCO3, PiOH) and organic P (PoCO3, PoOH) were also determined. The FP treatments significantly (P = 5%) increased both P-resin and P-HCl levels. Over time, the differences between P-resin or P-HCl levels and the controls declined, except in the silt loam soil in which P-resin levels nearly doubled. PtCO3 and PtOH levels did not markedly respond to FP applications. As incubation proceeded PtOH levels significantly (P = 5%) increased both as PiOH and PoOH. This PtOH buildup was accompanied by small declines in PtCO3 levels, with the declines being greater in the PoCO3 than in PiCO3 fraction. P-resin, PiOH and PoOH levels were negatively correlated with those of p-HCl (P = 5%, n = 36). P-resin levels were also negatively correlated with those of PoCO3. Of the small and large FP doses, 12–45% and 51–56%, respectively, had yet to react by the end of the experiment.

    Notizen: Abstract The time course of the microglial cell reaction in central nervous system primary sensory projection territories has been examined following peripheral nerve injury in the adult rat using qualitative and quantitative analysis of immunoreactivity with the monoclonal antibody OX-42, which recognises the complement receptor CR3. The regions examined included the gracile nucleus, the column of Clarke and the spinal cord dorsal horn (superficial and deep laminae separately) after unilateral sciatic nerve transection, and the spinal trigeminal nucleus following unilateral infraorbital nerve transection. In all territories examined a qualitative increase in OX-42 immunoreactivity was observed 24 h postlesion. Further, quantitative analysis revealed an exponential development of the OX-42 immunoreactivity, with a peak at one week postlesion, thereafter showing a slow exponential decline. Our results show that the signal (or signals) that induces the microglial cell response in primary sensory projection territories is rapid in comparison to previously described central degenerative changes following peripheral nerve lesions (transganglionic degeneration). These findings are compatible with the hypothesis that activated microglia play a pathogenetic role in the development of transganglionic degeneration.

    Notizen: Abstract Rat lumbar dorsal root ganglion neurones projecting to the nucleus gracilis in the brainstem were retrogradely labelled with Fluoro-Gold and analysed immunocytochemically for their expression of substance P-, calcitonin gene-related peptide-, galanin-, galanin message-associated peptide-, neuropeptide Y-, nitric oxide synthase- and carbonic anhydrase-like immunoreactivity as well as affinity to Griffonia (bandeiraea) simplicifolia lectin I — isolectin B4, RT97 and to choleragenoid. The analysis was made both in uninjured rats and in rats which had been subjected to unilateral sciatic nerve transection and partial resection 3 weeks earlier. The data showed that 6% of the L4 and L5 lumbar dorsal root ganglion cells that projected to the nucleus gracilis showed substance P-like immunoreactivity. Following nerve injury, none of the nucleus gracilis-projecting dorsal root ganglion cells showed substance P-like immunoreactivity. Nineteen per cent of the investigated cell population showed calcitonin gene-related peptide-like immunoreactivity in uninjured rats, but no nucleus gracilisprojecting calcitonin gene-related peptide-positive cells were found after nerve injury. Galanin- and galanin message-associated peptide-like immunoreactivity were found in 2% and 3%, respectively, of the Fluoro-Gold-labelled cell population normally and in 22% and 14%, respectively, after injury. No neuropeptide Y-positive cells were found in the Fluoro-Gold-labelled cell population normally, but after nerve injury, 96% of this population became neuropeptide Y-positive. Nitric oxide synthaselike immunoreactivity was found in 2% of the Fluoro-Gold-labelled cells normally and in 10% after injury. Two per cent of the Fluoro-Gold-labelled cells in the normal cases were stained by Griffonia (bandeiraea) simplicifolia lectin I — isolectin B4. After injury, however, no such double labelling was found. Thirty-four per cent of the Fluoro-Gold-labelled cell population was carbonic anhydrase positive normally, and 42% after injury. Seventy-five per cent of the Fluoro-Gold-labelled cells showed RT97 immunoreactivity normally and 12% after injury. Choleragenoid-like immunoreactivity was found in 99% of the Fluoro-Gold-labelled dorsal root ganglion cells normally and 81% after injury. Immunohistochemical visualisation of choleragenoid transganglionically transported from the injured sciatic nerve combined with neuropeptide Y immunocytochemistry showed that primary afferent fibres and terminals in the nucleus gracilis contain neuropeptide Y following peripheral nerve transection. Taken together, the results indicate that peripherally axotomised nucleus gracilis-projecting neurones undergo marked alterations in their cytochemical characteristics, which may be significant for the structural and functional plasticity of this system after injury.

    Notizen: Abstract In a study of hospital staff's perceived quality of care and worry, related to the use of medical equipment, a systems approach was used. This included training and user aspects on the equipment, environmental ergonomics, work organisation and psychosocial work conditions. In a survey study, 543 hospital staff members from seven departments with various degrees of equipment utilisation participated. By means of factor analysis, factors which were important to perceived quality of care and worry were determined. Adaptation of equipment and environment and training on the use of equipment were shown to be important. However, several organisational and psychosocial factors turned out to have a still greater impact. In the assessment of healthcare technologies which include medical equipment a holistic systems approach is therefore advocated.
