    Notes: Abstract Transport of nickel ions was studied in Alcaligenes eutrophus. Two transport systems for nickel ions exist to satisfy the nickel demand for the lithotrophic hydrogen metabolism. A major nickel transport activity exhibited an apparent affinity constant (K m) of 17 μM nickel chloride. This activity was competitively inhibited by Mg2+, Mn2+, Zn2+, and Co2+. A minor nickel transport activity was determined in the presence of high (0.8 mM) magnesium. This activity was not inhibited by Zn2+ or Mn2+; its K′ m was determined to be 0.34 μM nickel chloride. These kinetics suggested a second transport system in A. eutrophus. The membrane potential of A. eutrophus was decreased upon the addition of ammonium ions leading to a decreased nickel transport. This inhibition could be reversed by fructose or by hydrogen indicating an energy dependent nickel transport. Protonophores inhibited the nickel transport. However, inhibitors of ATP synthase like dicyclohexylcabodimide or venturicidin had little or no effect on nickel transport. These data indicated that the transport was coupled to the proton motive force.

    Notes: Abstract Chromosomal mutants of Alcaligenes eutrophus unable to grow with molecular hydrogen as the energy source also failed to grow with nitrate as the terminal electron acceptor or as a nitrogen source. The mutants (Hno−) (i) formed neither soluble nor particulate hydrogenase antigens, (ii) expressed only about 50% the wild type level of ribulosebisphosphate carboxylase activity, and (iii) transported nickel, an essential constituent of active hydrogenase, at a significantly lower rate than wild type cells. Moreover, the mutants grew very slowly with urea as nitrogen source and did not express urease. Growth on formamide was also affected and formamidase activity was induced to only a very low level. Growth of the Hno− mutants on succinate, glutamate, fumarate, and malate was significantly slower than wild type, and a reduced rate of succinate incorporation into the mutant cells was demonstrated. The highly pleiotropic phenotype of Hno− mutants is indicative of a chromosomal gene with a considerable physiological importance. It affected the expression of both chromosomal and megaplasmid encoded systems of energy, carbon, and nitrogen metabolism. Thus, the hno mutation restricts the metabolic versatility but does not affect the basic metabolic functions of the organism.

    Notes: Abstract Total protein of Alcaligenes eutrophus was analyzed by two-dimensional protein map. Cells grown at 30° C expressed hydrogen-oxidizing (Hox) ability mediated by a soluble (Hos) and a particulate hydrogenase (Hop). Hox ability was not expressed at 37° C (HoxTs). The six subunits of the two hydrogenases were identified. Besides these six subunits eight peptides were not or hardly detected at 37° C. The mutant HF117 which expressed Hox ability at 37° C (HoxTr), formed the hydrogenase peptides and five of the eight peptides. These peptides designated B, C, E, F, and H were characterized by their isoelectric point and molecular mass (M r); their M r were 18 800, 45 400, 41 900, 39 400, and 40 600, respectively. The five peptides were not formed in regulatory Hox− mutants, and not formed in mutants cured of plasmid pHG1, carrying the genetic information for hydrogenase formation. Strain HF160, carrying a Tn5 insertion in a gene essential for Hos expression specifically did not form the B-peptide. All peptides were found in the soluble fraction of cell extracts, the F-peptide was also detected in the particulate fraction. The function of the new Hox-peptides is presently unknown.

    Notes: Abstract In developing a rheological constitutive equation for crosslinking polymers the determination and explanation of time constants are of great importance. For stoichiometrically balanced and imbalanced reactions of polydimethylsiloxane and polyurethane model polymers with appropriate “cross-linkers”, it is shown that the experimental data for the storage and loss moduli are well fitted within a large frequency range by a linear rheological constitutive equation with a truncated power law relaxation function. Special attention is given to the problem of estimating the different time constants. A method is proposed for the determination of the terminal relaxation time (in the case below the gel point) and a characteristic retardation time (in the case above the gel point) from the intersection points of the high and low frequency asymptotes of the storage modulus.

    Notes: Summary The cytoplasmic hydrogenase from Alcaligenes eutrophus (E.C.1.12.1.2.) was used to achieve continuous NADH regeneration. Cells were grown under heterotrophic conditions in a medium containing fructose and glycerol as carbon sources. A cell suspension with about 1 unit/ml activity was obtained. The enzyme was purified 20 fold with a 72 % yield and a final specific activity of 21 units/mg. The hydrogenase was covalently grafted onto corn stover particles for reactor application. The immobilized derivative shows a higher stability than the free enzyme. This hydrogenase preparation was used in combination with L-lactate dehydrogenase, to achieve the continuous reduction of pyruvate into lactate. For this purpose, both enzymes were simultaneously immobilized onto corn stover. Continuous NADH regeneration could thus be obtained over periods of time lasting over 45 hours, corresponding to 111 NADH regeneration cycles.

    Description / Table of Contents: Résumé Cet article a porté sur 10079 participants provenant de 52 centres répartis dans 32 pays. Ces résultats, récemment publiés, montrent qu'une association linéaire simple entre sel alimentaire et tension artérielle n'est plus un modèle pertinent. De plus, la surcharge pondérale et la consommation d'alcool agissent indépendamment sur la pression artérielle. Les résultats de l'étude doivent être interprétés prudemment dans le cadre d'une analyse épidémiologique soigneuse.
    Abstract: Summary The recently published Intersalt Study comprising 10079 participants from 52 study centers in 32 countries demonstrated that the simple linear relationship so far postulated for the association between electrolytes and hypertension is history. In addition overweight and alcohol consumption make an independent contribution to the level of individual blood pressures. The study results have to be interpreted carefully in the framework of a differentiated epidemiologic analysis.
    Notes: Zusammenfassung Die bei 10079 Teilnehmern der an 52 Zentren in 32 Ländern durchgeführten Intersalt Studie erhobenen Daten zeigen, dass die bisher postulierten linearen Beziehungen zwischen Elektrolytausscheidung und Hypertonie der Vergangenheit angehören. Darüber hinaus leisten Übergewicht und Alkoholkonsum einen unabhängigen Beitrag zur Höhe des individuellen Blutdruckes. Die Studienergebnisse müssen im Rahmen einer differenzierten epidemiologischen Bewertungsweise interpretiert werden.
