Notes: Summary The medium composition was optimized on a shake-flash scale for production of cellulases by Gliocladium virens using the Graeco-Latin square technique. With the optimized medium 0.33 units of filter paper (FP)-cellulase, 1.52 units of β-glucosidase, and 30.45 units of xylanase were produced per millilitre of culture filtrate at 120 h of fermentation. In a laboratory fermentor the lag phase was much reduced and 0.25 units of FP-cellulase, 0.77 units of β-glucosidase, and 24.04 units of xylanase were obtained per millilitre of culture filtrate at 39 h. Characterization of the enzymes with respect to pH and temperature optima, and pH and heat stabilities indicated that cellulases and xylanase of G. virens have properties comparable to those of Trichoderma reesei and some other fungi.

Notes: Summary The feasibility of cellulase production by Trichoderma reesei using inexpensive lignocellulosic material was examined. Sulfite pulp used as standard substrate yielded 3.7 IU/ml filter paper units (FPU) and 2.15 IU/ml β-glucosidase. The yield was 185 FPU per gram total carbohydrate (CH) in the fermentation medium. Steam treated wheat straw (2%) gave 1.9 FPU/ml, 0.83 IU/ml β-glucosidase and 151 FPU/g CH, whereas the spent fibres remaining after enzymatic hydrolysis of steamed wheat straw gave 2.4 FPU/ml, 1.55 IU/ml β-glucosidase and 147 FPU/g CH. A good substrate (3%) was also the combustible fraction of municipal waste (BRAM) treated with NaOH, which gave 2.5 FPU/ml, 0.86 IU/ml β-glucosidase and 130 FPU/g CH. A further increase in the final enzyme titer is obtainable by increasing the substrate concentration. In shake cultures 5% steamed wheat straw gave 3.8 FPU/ml and 1.95 IU/ml β-glucosidase. Untreated wheat straw gave only low final enzyme titers and low yields of FPU/g CH. In the case of lignocellulosic substrates a constant pH-value of pH 6.0 during the fermentation gave optimal yields.

Notes: Summary The kinetic behavior of heterogenous microbial populations in biotechnological processes can be described with the aid of the Monod equation extended by a term kd for endogenous metabolism. The modified Monod equation is examined in detail to demonstrate the influence of the kd-term on the values of the Monod parameters μm and KS. The principles of kinetic analysis are also shortly considered in connection with fermenter design. The prerequisites for kinetic analysis together with parameter determination are demonstrated.

Notes: Summary The EMG enables diagnosis, follow-up and prognosis of vocal cord palsy as well as an indication for surgery. Using the zoom-endoscope by v. Stuckrad the EMG can be performed in local anesthesia. With a specially designed instrument the needle electrode can be transorally applied exactly in the posterior cricoarytenoid muscle as well as in other muscles of the larynx. As the patient can follow instructions both voluntary and spontaneous activity can be recorded under visual and acoustic control during inspiration, phonation and exspiration on a tape. Thus replay, control examination and electronic summation is possible.

Notes: Summary The importance and necessity of an endoscopic examination in case of suspected foreign body is demonstrated by endoscopic findings in the ear, nose and paranasal sinuses, the pharynx and esophagus as well as in the larynx, trachea and bronchial system. The principles, advantages and limitations of endoscopic foreign body removal are shown.

Notes: Summary Chronic and diffuse contact between inhalation noxae and the mucosa plays an important role in the pathogenesis of cancer of the larynx and lead us to expect a multilocular development of carcinoma and precancerous changes. This supposition was examined in some 100 larynx resection preparations. After standardized stratigraphic sub-divisioning of the larynx, it was lamellated and the individual zones studied histologically for epithelial dysplasias, varying degrees of differentation up to an including carcinoma, and for inflammatory changes. Certain relationships were found to exist between certain characteristics of growth of the carcinoma and the site, extent and histological classification of the tumour. The fact that, frequently, a macroscopically non-verifiable coexistance of precancer and carcinoma can be found, is taken account of in the modified conception for the taking of biopsies.

Notes: Summary Surgical speech rehabilitation after total laryngectomy by the three procedures of Prof. Staffieri is based on the principle of establishing a short fistula lined by pharyngeal mucosa between trachea and hypopharynx. It permits voluntary air pressure control for voice production while preventing aspiration during deglutition. In a single-stage procedure the “Neoglottis phonatoria” is established on the occasion of the laryngectomy at the top of the trachea. In cases of previous laryngectomy a shunt is established between the trachea dorsal wall and the Oesophagus, either cranially (direct internal shunt) or more caudally (retrograde internal shunt). The three methods are described, and the one-way function of the different fistulas are demonstrated. The percentage of successful voice restorations that remain free from swallowing difficulties in 700 patients, subjected to these techniques in Europe and Overseas is between 60 and 90%.

Notes: Summary The needle-biopsy taken in the pharynx and larynx is a simple and reliable procedure for the histological diagnosis of submucosal tumours. It can be performed either in local anaesthesia using the zoom-endoscope by v. Stuckrad or during microlaryngoscopy under general anaesthesie. This method however does neither replace the single biopsy of exophtic tumours nor the microlaryngoscopic excision of suspicious, precancerous lesions.

Notes: Summary Endoscopic methods as the zoom-epipharyngoscopy and the transnasal inspection of the epipharynx enable an exact examination and diagnosis in the organic or functional lesions of the pharyngeal tube. The physiological and pathological movements of the tubal torus, the pharyngeal tubal orifice and the soft palate are demonstrated.
