Summary In the serum and plasma of adults patients suffering from osteopetrosis we found a 6–8-fold increased activity of acid phosphatase and aldolase. The increase of lactase dehydrogenase was minor. The phosphatases had three maxima of activity in the acid pH range using 0,2M acetate buffer. Starch gel electrophoresis showed separation of three fractions. The substrates phenylphosphate,β-glycerophosphate andα-glycerophosphate are hydrolysed in the order listed with decreasing velocity. l-tartrate inactivated the phosphatases to 40%, but cystein to about 70%. In all patients an increase of citric acid, pyruvic acid and lactic acid was observed.

Zusammenfassung 1. In Serum und Plasma von Patienten, die an Marmorknochenkrankheit litten, wurden Erhöhungen der Aktivität der „sauren“ Phosphatasen bis auf das Sechsfache der Norm, beträchtliche Aktivitätserhöhung der Aldolase und geringfügige der Lactatdehydrogenase festgestellt. 2. Die saure Plasmaphosphatase besitzt drei Aktivitätsmaxima bei pH 5,1, 4,7 und 4,0. Saure Serumphosphatase wird durch Stärkegelelektrophorese in drei Fraktionen aufgespalten und greift Phenylphosphat,β-Glycerophosphat undα-Glycerophosphat in der Reihenfolge ihrer Nennung mit abnehmender Geschwindigkeit an. Diese Plasmaenzyme wurden durch l-Tartrat zu 40% und durch Cystein zu 70% gehemmt, was sie von Phosphatasen anderer Herkunft unterscheidet. 3. Bei den untersuchten Patienten waren die Konzentrationen von Citronensäure und Brenztraubensäure im Serum stark, die der Milchsäure leicht erhöht.
