    Notes: Abstract The present study in 10 breast cancer patients supports the concept that newly synthetized, nascent histamine is involved in tumour growth. Histidine decarboxylase (HDC) activity is increased in mammary tumour tissue compared to healthy mammary gland-, skin- and muscle tissue in all but one patient studied. The newly formed histamine is probably not stored in the tumor tissue. Significantly decreased histamine concentrations were measured in parallel samples in the tumour tissue. Moreover, the preliminary results from urinary analysis of histamine and Nτ-methylhistamine in 3 of the 10 patients studied showed a significant decline after tumour extirpation compared to preoperative values.

    Notes: Abstract Following numerous reports of high histidine decarboxylase (HDC) activity in tumour-bearing animals, the present work was designed to determine the activity of this enzyme in normal and tumour tissues in a series of ten surgical patients with colorectal carcinoma. Significantly increased HDC activity, almost double that of normal tissues, was found in specimens from extirpated human tumours. These results, obtained under reliable sampling and measurement conditions indicated that changes in the enzymic activity of HDC may have a significant role in the development of colorectal tumour cells. Inhibition of the enzyme activity in human cancer subjects may retard or impede tumour growth and perhaps limit the spread of metastases.

    Notes: Abstract Details of a new technique in pulsed wire anemometry, developed to allow measurement of two-point spatial velocity correlations in highly turbulent flows, are described. An outline of the interface devices necessary for linking two anemometers to a microcomputer is given and examples of the use of the technique are presented. Firstly, measurements of spatial correlations with and without time delay in the near wake of a nominally two-dimensional cylinder normal to a uniform stream confirmed the viability of the technique. Secondly, measurements in the highly turbulent, separated region behind a normal flat plate fitted with a downstream splitter plate are presented as a demonstration of the effectiveness of the technique in such ‘difficult’ flows. We believe that these are the first direct measurement of spatial correlation functions within a separated flow.

    Notes: Summary A simultaneous distillation-extraction apparatus proposed by Godefroot has been used for the GC study of volatile components of cheese. 5–10 g of cheese provide a fraction that can be directly injected into a GC or a GC/MS. Two capillary columns (SE-30 and SP-1000) were evaluated, the second one being adequate for quantitative determinations. Camphor was used as internal standard. The coefficients of variation ranged from 1.4 to 11.8. The technique can be used as a fast method for the characterization of cheeses from their volatile component concentrations.

    Notes: Summary Tetramethylammonium hydroxide in methanol has been proposed as a catalyst for preparing methyl esters of both free fatty acids and glycerides of fats. Nevertheless, procedures normally recognized as suitable can be inadequate when the fat under study contains noticeable amounts of low relative molecular mass esters. The method using tetramethylammonium hydroxide has been examined and modified in order to analyze milk fats by capillary GC. The pyrolysis methylation of free fatty acids in capillary injectors has been also studied. The modified method was satisfactory for both neutral and acid milk fats.

Pentoses were separated on SE-54 and hexoses on OV-225. The four components of the tautomer equilibrium mixture have been identified using GC/MS and NMR data.

    Notes: A perifusion system was designed in order to study glucose and glutamine metabolism by freshly harvested Ehrlich ascites tumour cells in steady state conditions. Cells were perifused in the presence of 5 mM glucose, 0·5 mM glutamine or 5mM glucose and 0·5 mM glutamine. The results in steady state reveal that both substrates glucose and glutamine are continuously wasted by tumour cells, excreting two moles of lactate per mol of glucose and one mol of glutamate and ammonia per mol of glutamine consumed into the medium. Glutamine consumption in the presence of glucose was higher than with glutamine alone.

    Notes: This paper considers the use of various finite differencing schemes for the computation of flows involving regions of recirculation. Standard first-order hybrid schemes, vector (or skew) schemes and second-order schemes are used to predict laminar flows in a channel containing a constriction and over a normal flat plate with a downstream splitter plate. In the former case the results are compared with those of other workers and with the implications of analytic theories for the viscous dominated flow around the sharp corner.Attention is concentrated on the effects of errors arising from the use of non-uniform grids and it is shown that higher-order differencing schemes are generally much less susceptible to these than the simpler schemes. The major conclusion is that for flows containing regions where pressure gradients largely balance the convective terms in the momentum equations, in addition to other regions where convection and diffusion balance, higher order differencing schemes are likely to be essential if accurate predictions are required on grids without excessive numbers of nodes. It is argued that similar conclusions must hold for high Reynolds number turbulent flows.
