    Notizen: Summary A codominantly inherited mutation of the lactate dehydrogenase (LDH) in the C3H mouse causes a severe hemolytic anemia in homozygous mutants, whereas viability and fertility are close to normal. Investigation of multipotent hemopoietic stem cells (CFU-S), myeloid (GM-CFC) and erythroid progenitors (BFU-E, CFU-E) in femur and spleen indicates a general shift from bone marrow to splenic hemopoiesis. In terms of total body hemopoiesis, however, the BFU-E pool is 1.4- and the CFU-E pool 19-fold enlarged, whereas CFU-S and GM-CFC show little or no deviation from normal. It is concluded that this mouse mutant is an appropriate model of long-term hemopoietic stress showing that compensation in this severe hemolytic anemia is achieved primarily by an increase of the number of the most mature erythroid progenitors.

    Notizen: Summary Murine interleukin 3 (IL-3)-dependent permanent mast cell lines derived from normal mouse bone marrow were established using pokeweed mitogenstimulated spleen cell conditioned medium (SCM) as a source of IL-3. When propagated continuously in media containing a high concentration of IL-3 (20% SCM or 20 U/ml murine recombinant IL-3 (rIL-3)), all the cell lines remained strictly factor-dependent in vitro and non-tumorogenic in vivo. However, we were able to reproducibly generate autonomous sublines from cultures supplemented with low amounts of IL-3 (1% SCM or 2 U/ml rIL-3). Abrogation of exogeneous growth factor dependency was always associated with neoplastic transformation. In newly generated autonomous sublines an autocrine mechanism of growth regulation was evident in vitro.

    Notizen: Summary The proliferation kinetics of erythropoiesis and of myelopoiesis have been studied in a case of congenital dyserythropoietic anemia type II (HEMPAS) by means of quantitative 14-C autoradiography, Feulgen cytophotometry and 59-Fe ferrokinetics. Increased total erythropoietic activity and ineffective erythopoiesis was demonstrated by ferrokinetics. Quantitative 14-C autoradiography showed a generally delayed proliferation rate of erythroid cells, most evident in the polychromatic compartment. A deficiency of cell production of 25% was detected among the polychromatic erythroblasts. Part of this fraction is represented by cells still capable of passing to the successive stages of maturation. We conclude that only part of the deficiency of cell production in the polychromatic compartment represents real cell destruction. Most of the measured ineffectiveness is confined to later stages of maturation, such as orthochromatic erthroblasts and marrow reticulocytes.
