    Notes: Zusammenfassung 1. Bei kolloidchemischen und technologischen Untersuchungen an Färbebädern konnte wiederholt in den Farbstofflösungen Synäresis beobachtet werden. Die synäretischen Gebilde zeichneten sich, wie in anderen Fällen bereits von Liepatoff beobachtet, durch regelmäßige Konturierung und große Elastizität aus. Der angewandte Farbstoff war Benzopurpurin 4 B extra. 2. Eine nähere qualitative und quantitative Untersuchung ergab, daß die durch Synärese entstandenen Körper, entgegen der ursprünglichen Annahme, nicht aus einer homogen Gallerte bestanden, sondern sich aus einer Gelhaut und darin eingeschlossenem Sol zusammensetzten. Die äußere Flüssigkeitshülle, welche den Körper umgab, war nach längerer Zeit an Farbstoff zu einer ganz oder fast molekularen Lösung verarmt. Auch das im Inneren befindliche Sol besaß eine geringere Konzentration als die Ausgangslösung. 3. Es wurde versucht, eine Erklärung für das Zustandekommen solcher eigenartigen Bildungen von Synäresis zu geben, was auf mehrfachem Wege möglich schien. Anschließend wurde auch versucht, eine Erklärung für den Einfluß von Zusätzen, sog. Netzmitteln, zu liefern, da sich gezeigt hatte, daß ein solcher Zusatz entscheidend für die Bildungsgeschwindigkeit und Größe der Synäresekörper war.

    Notes: Summary The cytoplasmic hydrogenase from Alcaligenes eutrophus (E.C.1.12.1.2.) was used to achieve continuous NADH regeneration. Cells were grown under heterotrophic conditions in a medium containing fructose and glycerol as carbon sources. A cell suspension with about 1 unit/ml activity was obtained. The enzyme was purified 20 fold with a 72 % yield and a final specific activity of 21 units/mg. The hydrogenase was covalently grafted onto corn stover particles for reactor application. The immobilized derivative shows a higher stability than the free enzyme. This hydrogenase preparation was used in combination with L-lactate dehydrogenase, to achieve the continuous reduction of pyruvate into lactate. For this purpose, both enzymes were simultaneously immobilized onto corn stover. Continuous NADH regeneration could thus be obtained over periods of time lasting over 45 hours, corresponding to 111 NADH regeneration cycles.

    Notes: Alloxan is known to induce diabetic renal changes as well as causing nephrotoxic alterations. However, no ultrastructural study has been performed to differentiate diabetic verses toxic affects of alloxan to the tubule and/or glomerulus. Therefore the present study used the “protected” kidney model to prevent one kidney from being exposed to the alloxan while allowing the other to receive the drug immediately. In all experimental animals the right renal hilum was gently occluded for 5 minutes and then released. This was performed prior to the injection of alloxan. Subsequently, the left renal hilum was occluded at the time of, and for 5 minutes after, alloxan administration (40 mg/kg i.v.). The experimental rats were divided into three groups: untreated diabetics, diabetics treated with protamine-zinc-insulin, and alloxantreated rats that failed to become diabetic. Three groups of controls were included: one group received an equal volume of saline diluent as the experimental rats but no clamping of either renal hilum; another group received the saline and had the left renal hilum occluded for 5 minutes; and a third group had both the right and left renal hila occluded. All animals were followed and sacrificed after 9 weeks. Endogenous creatinine clearance did not change among groups. Alloxan-treated nondiabetic rats displayed marked interstitial nephritis in unprotected kidneys, while protected kidneys were normal. The diabetic state resulted in mesangial proliferation and focal glomerular basement membrane thickening as well as glomerular capilary endothelial abnormalities and visceral epithelial foot-process fusion. The endothelial changes consisted of focal areas showing a reduction in the size of endothelial fenestrae. All glomerular changes were ameliorated by insulin treatment. We conclude: (1) alloxan per se is distinctly nephrotoxic; and (2) the glomerular endothelium and epithelium are involved early in the course of experimental diabetes.

    Notes: Consideration was given to the effects of casting conditions upon the performance and structure of membranes prepared from CA-acetone-water-inorganic salt solutions. Treating casting solutions as polymer-solvent-nonsolvent ternary systems, the data on equilibrium phase separation conditions were plotted in a triangular diagram for solutions containing Al(NO3)3, KSCN, Mg(ClO4)2 or no additive. Measurements of casting solution viscosity, membrane thickness, freezing and nonfreezing water contents were used to supplement flux and retention data of membranes made by varying the inorganic salt and holding time in a systematic way. With the aid of scanning and transmission electron microscopy, it was concluded that membranes may consist of one, two, three, or four layers. The data and correlations obtained allow us to propose a mechanism of formation for each of these layers.
