Notes: Summary Fusions of chicory leaf protoplasts were realized in order to obtain a higher number of tetraploid plants compared to the traditional doubling techniques with colchicine. Thanks to the fusion technique, using a mixture of PEG, DMSO and a solution of CaCl2 (pH 10.5), 25% tetraploid (2n=36) plants were obtained. Among the 167 regenerated plants, only one aneuploid of a near tetraploid level (2n=33) was identified.

Notes: Abstract The improvement of N2 fixation in legumes may lead to increased yields and reduced fertilizer requirement. Levels of N2 fixation were determined for three cultivars and nine progeny lines from two inbred backcross common bean (Phaseolus vulgaris L.) populations that were grown at Hancock, Wissconsin in 1984 and 1985 using 15N-depleted (NH4)2SO4. The high N2-fixing line ‘Puebla 152’ was the donor parent for both inbred backcross populations and the cultivars ‘Porrillo Sintetico’ and ‘Sanilac’ were the recurrent parents for populations 21 and 24, respectively. Total N yield, fixed N2 and % N derived from the atmosphere were determined for whole plants and plant parts at the R3 (50% bloom) and R9 (maturity) growth stages. Significant year-by-line interactions were found for N2 fixation traits among the population 21 lines and parents, but not for population 24 lines and their parents. Measures of N2 fixation at R3 were inadequate to predict N2 fixation at R9. Population 24 lines and parents differed for N2 fixation ability at R9, and fixed N2 was correlated with maturity. The recovery of an inbred backcross progeny line, 24-21, which matured earlier and fixed more N2 than the recurrent parent ‘Sanilac’ indicated that N2 fixation was heritable and that favorable alleles, independent of maturity, were recovered from a late-maturing, high N2-fixing donor parent by utilizing the inbred backcross breeding method. Since most fixed N2 and non-fixed N (〉80%) was found in the seeds at maturity, and most lines did not vary for the distribution of nitrogen throughout the plant, selection for improved remobilization of nitrogen to the seed to increase yield is impractical in this genetic material. The highest N2-fixing lines tended to have high and similar % Ndfa in all plant parts.

Notes: Abstract Somatic embryos and adult plants were regenerated from mesophyll protoplasts of a clone of chicory ‘474’ (Cichorium intybus L. x Cichorium endivia L.). Embryos were obtained in three different ways: - plating of 7-day-old protoplast cultures on PM-3 solid induction medium with 2.5 μM 2-isopentenyladenine (2-iP) and 0.5 μM naphthaleneacetic acid (NAA); - transfer of 1–2 mm microcolonies into liquid M-17 induction medium with 2.5 μM (2-iP) and 0.1 μM (NAA); - transfer of well-established protoplast-derived calluses onto M-17. Development of embryos was accomplished in agitated liquid Heller medium with 0.15 μM gibberellic acid and germination on solid Heller medium without growth regulators. The total time for plantlet (4-leaves stage) recovery, following protoplast isolation, was 13 to 15 weeks. After acclimatization all protoplast-derived plants of Cichorium ‘474’ were phenotypically normal and fertile.

Notes: Thermal Aqueous Liquid Chromatography (TALC) using pure water as a mobile phase should be a very attractive technique promoting the use of liquid chromatography in routine analysis, either in the laboratory or in future on-line process control. This simple form of chromatography, the selectivity of which depends on the fixed phase only, would allow the use of flame ionization as an almost universal, low-cost detector in liquid chromatography. The potential of the TALC technique is illustrated by selected examples.

Description / Table of Contents: Polymerization of acetoxy-2 styrene, parachloro acetoxy-2 styrene, paramethyl acetoxy-2 styrene and paranitro acetoxy-2 styrene was carried out by means of free radical initiators or by thermic reaction. The polymers therefrom showed molecular weights of about 200 000. Studies on the thermal behaviour reveal that these polymers do not undergo any change of phase, either vitreous or crystalline. Dielectric experiments show losses of about 10-3 radian; the losses are practically constant within a large scale of temperature and frequency. Over 320°K one notices a relaxation process of small magnitude and the presence of water brings about a marked change in the way the polymer reacts to dielectric treatment.

Notes: Water-borne enzymatic coating-based dry chemistries have been used for more than 20 years, especially by diabetics, for self-monitoring of blood glucose. Until now, it has been believed that enzymes work only in water and not in organic solvents. Synthesis of a hydroxylated acrylic polymer and the novel concept of dispersing enzymes have enabled us to design nonaqueous diagnostic coatings. These coating films gave excellent dose response and dynamic range. Additionally, these coatings can easily be ranged by a wide variety of antioxidants. In contrast, water-borne coatings are extremely difficult to range. Furthermore, nonaqueous coating films produce color signals that are independent of the reaction time with excellent thermal stability. Molecular forces and thermodynamical considerations have been used to explain the performance of the new dry chemistries. An organic reaction mechanism of ranging has also been proposed for the nonaqueous system. © 1993 John Wiley & Sons, Inc.

Notes: A lithographic process is described which increases the sensitivity of polymers to electron beam irradiation. The free radicals and peroxides produced in a film of polymer by irradiation are used to initiate the grafting of a monomer to form a graft copolymer, the solubility of which may be higher or lower than that of the initial polymer. Acrylic acid has been grafted into poly(methyl methacrylate) films to produce a copolymer which is insoluble in the solvents of PMMA. The sensitivity of this negative system depends on the conditions of grafting and on the conditions of development. A sensitivity of 2.8 10-8 C.cm-2 at 20 keV has been obtained.
