    Notes: Abstract This study describes clinical signs and symptoms in 16 patients with the DiGeorge syndrome (DGS). Diagnosed on the basis of typical facial stigmata, a broad spectrum of severity is seen with respect to congenital heart disease, hypoparathyroidism and immunologic parameters. A simple index of severity is introduced that clearly differentiates complete forms of the syndrome (cDGS) with poor prognosis from partial forms of the syndrome (pDGS). Of 13 pDGS patients, 12 are still living; 8 underwent corrective heart surgery without infectious complications. Moderate to severe mental retardation is seen in all pDGS patients. Due to the lack of thymus function, immunodeficiency is a result of cDGS, whereas immunoregulatory disturbances (hypergammaglobulinaemia, high titres of specific antibody production) prevail in pDGS patients.

    Notes: Abstract We present immunological in vitro data on a group of 16 patients with the DiGeorge sequence (DGS). Detailed tests gave evidence that the decrease in number and function of mature T-cells is closely correlated with phenotypic expression and represents a wide immunological spectrum ranging from normal function to severe combined immunodeficiency with B-cells. Careful immunological testing in DGS may help to plan therapeutic procedures (i. e. thymic factor therapy or bone marrow transplantation) may also have prognostic significance and help to gain insight into mechanisms operative in disturbances of T-cell regulation.

    Description / Table of Contents: Zusammenfassung Ein atypischer Fall von myatrophischer Lateralsklerose wurde beschrieben, dessen Besonderheit auf einer Frühmanifestation, auf einer länger dauernden Asymmetrie der Initialsymptome, auf dem Fehlen einer bulbären Symptomatik bei sonst sehr fortgeschrittener Symptomatik, auf einem ungewöhnlich langen Verlauf und auf konstanten Zeichen einer entzündlichen Reaktion im Liquor beruht. Die konstante Liquor-Pleocytose war Anlaß, ausgedehnte virologische Untersuchungen einzuleiten, die im Versuch einer Virusisolierung gipfelten. Es wurde schließlich aus dem Liquor ein Virus isoliert, welches dem TbE-Komplex der Arbovirus-Gruppe B (tick-borne Flaviviren) zugehört. Etwa 70% einer Gruppe von 19 ALS-Patienten aus Hamburg hatten mit diesem Virus offenbar Kontakt. Das Antikörpermuster des Falles ermöglichte eine Erklärung seiner Besonderheiten.
    Notes: Summary An atypical case of amyotrophic lateral sclerosis (ALS) is described, characterized by early manifestation, a long lasting course with asymmetry of the lesions, absence of bulbar symptoms in the presence of an otherwise very advanced symptomatology, and constant signs of an inflammatory reaction in the CSF which was the reason to initiate extensive virological studies, including procedures for virus isolation. A virus, belonging to the TbE complex of arbovirus group B (tick-borne flaviviruses), was finally isolated from the CSF. About 70% of the ALS cases in Hamburg/W. Germany, examined for antibodies, appearently had contact with this virus. The antibody pattern found made it possible to explain this exceptional case.

    Description / Table of Contents: Zusammenfassung Aus dem Liquor eines Patienten mit mehrjähriger amyotropher Lateralsklerose, aber mit langdauernder Pleozytose, wurde ein Virus isoliert und durch Serienblindpassagen an Mäusebabys adaptiert. Es gehört zum sog. TbE-Komplex der Togavirus-Gruppe B. Gehirnhomogenat von Babymäusen der 13. Passage wurde auf Mäuse, Hamster und Cynomolgus-Affen verimpft. Bei den Hamstern entwickelten sich nach subkutaner Impfung Abmagerung und Parese der Hinterbeine. Histologisch fanden sich bei mehreren Tieren petechiale Blutungen und Zelluntergänge mit entzündlicher Reaktion in der grauen Substanz. Versuche mit wiederholten Impfungen von Hamstern mit verschiedenen Flavivirusstämmen in hoher Verdünnung sprechen dafür, daß sich bei dieser Versuchsanordnung Krankheitsbilder nach Art der Slow-Virus-Erkrankungen entwickeln mit fehlender enzephalitischer Reaktion und vorwiegendem Zelluntergang in Vorderhornzellen mit nur geringer entzündlicher Komponente. Intracerebrale Verimpfung von Schu-Virus erzeugte bei Cynomolgus-Affen eine stürmische Meningoencephalitis mit epileptischen Erscheinungen, Ataxie, spastischen oder schlaffen Lähmungen. Nach subkutaner Applikation des Virus kam es zur langsamen Ausbreitung der Infektion entlang des peripheren Nerven über die vorderen Wurzeln ins Rückenmark mit Untergang der motorischen Vorderhornzellen. Schließlich wurde auch das Gehirn erreicht. Die in den Versuchstieren gefundenen histologischen Läsionen lassen den Schluß zu, daß der Patient, aus dessen Liquor das Virus isoliert wurde, an den Folgen der Infektion mit diesem Virus erkrankt war. Daraus ergibt sich die Schlußfolgerung, daß zumindest ein Teil der als amyotrophe Lateralsklerose diagnostizierten Fälle ihre Erkrankung einer Arbovirusinfektion verdanken.
    Notes: Summary A virus isolated from the CSF of a patient who had amyotrophic lateral sclerosis for 7 years, and prolonged pleocytosis in the CSF, was adapted to suckling mouse brain by subsequent serial blind passages. This Schu virus belongs to the tick-borne encephalitis complex of the genus Flavivirus (Togaviridae). Suckling mouse brain homogenate of the 13th passage was used for transmission experiments in various species of laboratory animals. Golden hamsters infected subcutaneously fell ill after a number of months, lost weight, and had paresis of the legs. Histologically they had petechial hemorrhages in different parts of the CNS and inflammatory changes in the gray substance of the spinal cord. Pilot studies with repeated inoculations of small doses of different flavivirus strains suggest a course of the disease in experimental animals which resembles slow-virus infections insofar as no encephalitis is produced and degenerative changes of the anterior horn cells prevail over inflammatory signs in the spinal cord. After intracerebral application of Schu virus, cynomolgus monkeys developed the typical lesions of togavirus panencephalitis with epileptic seizures, ataxia, and paresis. After subcutaneous application, the virus seems to spread along peripheral nerves to anterior spinal roots and spinal cord, where mainly motor neurons of the anterior horn are damaged, and from there to the brain. The histological findings are such that one may assume the disease of the patient was due to the infection with the virus isolated from his CSF. Therefore, the hypothesis may be advanced that at least some of the cases diagnosed as amyotrophic lateral sclerosis are due to a togavirus infection.

    Notes: The effects on viscose ripening of a variety of materials are reported. (1) Normal constituents (Na2CO3, Na2CS3, NaCl) appeared not to affect ripening; however, Na2S retarded ripening. By-product removal appeared to retard ripening. (2) Oxidizing agents (H2O2, Na2Sx) accelerated ripening; reducing agent (Na2SO3) retarded ripening; this is suggestive of possible addition product formation. (3) Na2SO4, (CH3)2SO, KClO3, Na2BH4 showed no effect on ripening. (4) Salts which precipitated as hydroxides or carbonates did not appear to affect ripening. (5) Salts which formed insoluble sulfides resulted in an initial increase in ripening rates. In later stages, ripening proceeded normally, probably after the salts were removed from solution as insoluble sulfides. (6) FeCl3, BiCl3 and Na3AsO3 appeared to retard ripening. (7) A comparison of the effect on ripening of the trivalent state of elements in group V of the periodic table showed that the rate of ripening decreased with the oxidation potential of the element. (8) The addition of sodium zincate retarded ripening and suggested possible cross-linking of xanthate and by-product sulfur, or of two xanthate groups. (9) Addition of tin and chromium salts at the 0.5% level resulted in gelation of the viscose within 30 min. after the addition.

    Notes: Mass spectrometry (MS) with electrospray ionization (ESI) has shown utility for studying noncovalent protein complexes, as it offers advantages in sensitivity, speed, and mass accuracy. The stoichiometry of the binding partners can be easily deduced from the molecular weight measurement. In many examples of protein complexes, the gas phase-based measurement is consistent with the expected solution phase binding characteristics. This quality suggests the utility of ESI-MS for investigating solution phase molecular interactions. Complexes composed of proteins from the human immunodeficiency virus (HIV) have been studied using ESI-MS. Multiply charged protein dimers from HIV integrase catalytic core (F185K) and HIV protease have been observed. Furthermore, the ternary complex between HIV protease dimer and inhibitor pepstatin A was studied as a function of solution pH. Zinc binding to zinc finger-containing nucleocapsid protein (NCp7) and the NCp7-psi RNA 1:1 stoichiometry complex was also studied by ESI-MS. No protein-RNA complex was observed in the absence of zinc, consistent with the role of the zinc finger motifs for RNA binding. Proteins Suppl. 2:28-37, 1998. © 1998 Wiley-Liss, Inc.

    Notes: Polarographic microcoaxial needle electrodes were used to measure internal profiles of dissolved oxygen tension (Po2) within single Ca-alginate beads of different diameter containing entrapped cells of Saccharomyces cerevisiae. For the investigations, single beads coming from variable growing conditions and distinct cultivation stages were fixed in a special holding device. In dependence on microbial growth steep oxygen gradients were observed. The Oxygen penetration depth at steady state lay between 50 and 100 μm. After 8 h of cultivation time, the anaerobic space within the beads (φ 2 mm; cultivation in a packed bed reactor) is beginning at ∼ 130 μm, whereas the anaerobic space within the beads (φ 2 mm) coming from the shaker flask culture is located ∼440 μm below the bead surface. Surprisingly, steep gradients were also observed, when recording profiles from cell-free Ca-alginate beads of different diameter and alginate concentrations. The steep oxygen gradients apparently had to be interpreted as pseudo-Po2-gradients. These results were borne by several effects, such as formation of artifacts and diffusion barriers in front of the electrode tip or oxygen “availability” at the tip and consumption of oxygen by the electrode itself. These phenomena could be documented by microscopic observation and photography. Thus, to obtain real Po2-profiles it is important to be exactly informed about the physical, chemical, and biological properties of the material to be investigated. Furthermore, it is necessary to apply a special stepwise puncture technique with distinct step-in/step-out movements of the electrode: e.g., unidirectional or contradirectional puncture techniques. © 1994 John Wiley & Sons, Inc.
